Combined Effect of Aging and Irradiation on Physicochemical Quality of Pork Shoulder

  • The effect of combined electron-beam irradiation and aging temperature of pork on microbiological and physicochemical properties was investigated. The samples from pork shoulder were irradiated with 0 or 2 kGy, vacuum-packaged, and assigned randomly to an aging temperature ($2^{\circ}C$, $10^{\circ}C$, or $25^{\circ}C$) during 8 d. On 4 d of aging at $25^{\circ}C$, total aerobic bacteria of non-irradiated ones reached 7 Log CFU/g which is no salable levels. Shear force values of irradiated meat after aging for 2 and 4 d at $25^{\circ}C$ was lower than those aged at $2^{\circ}C$. Irradiated samples at $2^{\circ}C$ had lower cooking loss after 2 and 8 d of aging, compared with other aging temperatures. Irradiation did not accelerate 2-thiobarbituric acid reactive substance (TBARS) value when aged up to 4 d. Irradiated samples aged at $10^{\circ}C$ and $25^{\circ}C$ for 8 d scored significantly higher TBARS values. With an increased aging period, $a^*$ and $b^*$ in irradiated samples at $2^{\circ}C$ slightly increased, but irradiation caused negligible changes in meat color. The highest contents of a desirable nucleotide flavor compounds (inosine-5-phosphate) were observed in pork at $2^{\circ}C$ when aged for 4 and 8 d, while the lowest contents were observed at $25^{\circ}C$. Aging in irradiated pork for 8 d at $2^{\circ}C$ resulted in optimal condition with improved meat quality and minimal microbiologically negative defect.

Questionable Reliability of Malondialdehyde to Measure Oxidative Stress in Sjögren's Syndrome: Preliminary Study

  • Purpose: To investigate the expression of malondialdehyde (MDA), lipid peroxidation marker for oxidative stress (OS), in autoimmune Sjögren's syndrome (SjS) by utilizing the SjS-prone C57BL/6.NOD-Aec1Aec2 (B6DC) mouse and the SjS patient plasma samples. Methods: The MDA concentrations in the lysates of the submandibular gland, liver, and serum samples from the SjS-prone B6DC mouse model were compared with those from the C57BL/6J as a control. A thiobarbituric acid reactive substance (TBARS) assay kit was used to measure MDA. Plasma samples from five SjS patients and five control subjects were also evaluated. Results: The MDA concentrations in experimental animals and controls were not significantly different. There were no significant differences between the plasma of SjS patients and of controls. Conclusions: The expression of MDA was investigated in the organs from the SjS-prone B6DC mouse for the first time and in the plasma samples of SjS patients. No significant differences were observed between SjS and control samples when MDA was the target molecule with the TBARS assay. MDA may not be a reliable marker to measure OS contrary to the published studies involving OS of SjS.

울산지역 노인의 생활습관에 따른 항산화 영양소 섭취실태 및 혈액내 항산화 영양 상태 비교 (The Effects of Smoking, Drinking and Exercise on Autioxidant Vitamin Intakes and Plasma Antioxidant Status in Elderly People Living in Ulsan)

  • Body antioxidant status is an important factor in the prevention of many chronic diseases caused by oxidative stress, especially in the elderly and is affected by health-related habits, such as smoking, drinking and regular physical activity. The aim of this study was to investigate the relationship between these health-related habits and plasma antioxidant status in the elderly. Plasma antioxidant status was examined by determining plasma levels of antioxidant vitamins (vitamin C, A, E, $\beta$ -carotene), total antioxidant status (TAS) and thiobarbituric acid-reactive substance (TBARS) . The subjects included 225 elderly persons aged over 60 yews (63 males, 162 females) living in the Ulsan area. They were interviewed to collect data on their general characteristics and health behaviors such as smoking, exercise and alcohol consumption by means of questionnaires. Their dietary intakes were obtained by means of semi-quantitative food frequency questionnaires (FFQ) The study population was divided into two or three groups according to their smoking, drinking, and exercise status. The ratio of smoker, drinker and exerciser was 16.7%, 31.0% and 44.2% respectively. The dietary antioxidant vitamin intakes were not significantly different among groups of smoking and drinking status, but tended to be higher in non-smokers and non-drinkers than in smokers and drinkers. Plasma vitamin C and $\beta$ -carotene levels were significantly higher in non-smokers, but Plasma vitamin A and TBARS levels were significantly lower in non-smokers than in smokers. Plasma TAS was not significantly different among the smoking groups, but showed a tendency to decrease with an increase in the number of packyear. Plasma vitamin C and $\beta$ -carotene levels of the non-drinkers were higher than those of drinkers and past-drinkers, but plasma vitamin A, C, E, TAS and TBARS showed no difference among the groups of drinker. All vitamin intakes of the exercisers were slightly higher than those of the non-exercisers, but vitamin C intake was significantly higher in female exercisers than in non-exercisers. Plasma $\beta$ -carotene levels were significantly higher in male exercisers and plasma vitamin A, C, E, TAS and TBARS levels tended to be higher in exercisers than in non-exercisers. These results suggested that change to non-smoker, modulation of alcohol consumption and regular exercise could enhance antioxidant defences against reactive oxygen species and might increase the likelihood of a healthier life span.

버찌(Prunus yedoensis) 추출물의 항산화 활성 평가 및 돈육 패티에 이용 (Antioxidant Activities of Water or Methanol Extract from Cherry (Prunus yedoensis) and Its Utilization to the Pork Patties)

  • 본 연구는 버찌를 메탄올과 물로 추출한 후 항산화 활성을 측정하여, 버찌의 메탄올과 물 추출물을 돈육패티에 첨가하여 육제품의 천연항산화제의 가능성을 알아보기 위하여 본 연구를 수행하였다. 버찌의 메탄올 추출물과 물 추출물의 총 페놀성 화합물 함량은 버찌 메탄올 추출물이2.17 g/100 g, 물 추출물이 2.77 g/100 g값을 나타내었다. 1,1-Diphenyl-2-picrylhydrazyl(DPPH) radical scavenging activity에서 버찌 메탄올 추출물이 대조구인 ascorbic acid와 농도별로 차이를 보이지 않았고, 2% 농도에서 대조구와 처리구간에 유사한 DPPH radical scavenging activity를 보였다. 환원력은 L-ascorbic acid에 비하여 낮은 수치를 보였으며 추출용매에 따른 환원력의 차이는 미미하였다. 철이온 흡착력은 버찌 메탄올 추출물이 19.8-94.0%, 버찌 물 추출물은 17.8-93.5%를 나타내었다. 버찌 메탄올 추출물은 0.5%의 농도까지 증가하다가 최종 농도인 2%까지 유의적인 차이를 보이지 않았고, 물 추출물은 농도가 증가할수록 철이온 흡착력이 증가하는 경향을 보였다. 특히 낮은 농도에서는 차이를 보였지만 농도가 증가할수록 대조구와 유사한 수치를 보였다. 버찌의 메탄올 추출물을 각각 0.5%, 1.0% 첨가한 돈육 패티에서 pH와 색도(명도, 적색도, 황색도)가 감소하였고, 지방산패도를 측정하기 위해 수행한 thiobarbituric acid reactive substances(TBARS) 결과에서 malondialdehyde(MDA) 함량이 대조구와 butylated hydroxytoluene(BHT)를 첨가한 참조구에 비해 낮은 결과를 나타내었으나, 저장 중 총균수나 대장균군수에는 효과가 없는 것으로 나타났다. 이러한 결과를 종합하면 버찌 추출물이 항산화 효과를 보여 천연 항산화제로써의 기능성 육제품 개발에 이용 가능할 것으로 평가된다. 하지만 버찌 추출물을 첨가하면 색도를 저하시키므로 이에 대한 보완 연구가 필요할 것으로 판단된다.

사람 LDL의 지질과산화에 의한 geraniin의 항산화 효과 (Antioxidative Role of Geraniin in Lipid Peroxidation of Human LDL)

  • 본 연구는 동맥 경화의 원인으로 알려진 사람 oxidized low density lipoprotein (LDL)에 대한 geraniin의 산화 억제 효과에 대하여 실험하였다. 사람 LDL을 C $u^{2+}$유도 LDL로 산화 시킬 때 50와 100 $\mu\textrm{g}$/ml 농도의 geraniin를 첨가하여 TBARS을 측정한 결과 LDL에 대한 항산화가 높았으며 용량 의존형으로 나타났다. Geraniin를 20-100 $\mu\textrm{g}$/ml의 농도를 조절하여 전기 영동에 의한 이동상을 조사한 결과 100 $\mu\textrm{g}$/ml geraniin의 농도에서 거의 완전한 억제 효과를 보였다. 사람 LDL에 C $u^{2+}$로 유도하여 LDL를 산화시킬때 conjugated diene를 보면 geraniin를 100 $\mu\textrm{g}$/ml 첨가하였을 때 억제 효과가 높았다. 또한 geraniin은 동맥의 내피세포에 서도 그 농도에 따라 억제효과를 나타내었다. 그리고 phorbol myristate acetate를 처리한 macrophage 유도활성 산소의 소거 효과는 geraniin의 농도가 100 $\mu\textrm{g}$/ml일때 거의 소거하였다. 이상의 결과로 보아 geraniin는 $\alpha$-tocopherol, ascorbir acid 및 합성 항산화제인 probucol과 거의 비슷한 항산화 활성이 있어 동맥 경화의 예방에 효과적이라는 결론을 얻었다.

사료내 생균제, 일라이트, 활성탄 및 목초액의 첨가가 닭 다리육의 품질 및 저장성에 미치는 영향 (Effect of the Feeding Probiotics, Illite, Activated Carbon, and Hardwood Vinegar on the Meat Quality and Shelf-Life in Chicken Thigh)

  • 본 시험은 육계에게 생균제, 일라이트, 활성탄 및 목초액의 사료적 가치를 평가하기 위하여 시판 생균제 0.2%(T1)와, 일라이트(T2), 활성탄(T3) 및 목초액(T4)을 각각 1.0%를 Arbor Acre Broiler 숫병아리 200수를 35일간 급여하여 도계한 계육의 다리부위 근육을 냉장온도($4{\pm}1^{\circ}C$)에서 10일간 저장하면서 pH, VBN, TBARS, 전단력 및 육색을 분석하였다. pH는 대조구에 비해 T3와 T4에서 높았고, 특히 T4에서 유의적으로 높았으며 저장기간이 경과하면서 모든 처리구에서 증가하였다(p<0.05). VBN은 저장 7일까지 대조구와 T2에 비해 T1, T3및 T4에서 낮았으나(p<0.05), 저장 10일에는 처리구간 유의성이 없었다. TBARS는 대조구, T1 및 T2보다 T3와 T4에서 낮았고, 저장기간이 지남에 따라 모든 처리구에서 급속하게 증가하였다(p<0.05). 전단력은 처리구간에 유의적인 차이가 없었지만, 저장기간이 경과함에 따라 감소하는 경향이었다. 육색중 명도(L)는 대조구보다 처리구에서 높은 경향이었고, 특히 T4에서 전 저장기간 동안 가장 높았다(p<0.05). 황색도는(b) 대조구보다 T3와 T4에서 높았다. 이상의 결과를 종합적으로 고찰해 볼 때 육계사료에 활성탄과 목초액 1%급여는 산화안정성 및 저장성을 향상시키는 효과가 있는 것으로 기대된다.

야콘 부산물과 쑥 분말을 사료에 첨가 급여한 육계의 도체 특성 및 품질에 미치는 영향 (Effects of Dietary Supplementation of Yacon By-products and Mugwort Powder on Carcass Characteristics and Meat Quality of Chicken Thigh Meat)

  • 본 시험은 육계에 야콘 부산물과 쑥 분말을 각각 0.5%와 1.0%를 급여하여 사육한 육계의 생산성과 다리 육의 도체특성, pH, TBARS, WHC, 전단력 및 육색을 조사하였다. 시험구는 야콘 부산물과 쑥 분말을 급여하지 않은 처리구를 대조구, 야콘 부산물 분말 0.5% 급여구는 T1, 야콘 부산물분말 1.0% 급여구는 T2, 쑥 분말 0.5% 급여구를 T3 그리고 쑥 분말 1.0% 급여구를 T4로 나누어 사양하였다. 야콘 부산물과 쑥 분말을 급여한 육계의 증체량, 사료섭취량 및 사료 요구율 등의 생산성의 유의적인 변화는 관찰되지 않았고, 폐사율은 유의하게 감소하였다. 혈액 생화학 성상 분석 결과, 야콘 부산물과 쑥 분말의 첨가 급여는 총콜레스테롤과 LDL-cholesterol 및 triglyceride는 감소하고, HDL-cholesterol은 증가하였다. 계육의 TBARS는 야콘 부산물과 쑥 분말을 급여함에 따라 낮아졌으며(P<0.05), 보수성과 전단력은 처리구간 유의적인 변화는 확인되지 않았다. 계육의 육색은 대조구보다 야콘 부산물과 쑥 분말 급여구에서 CIE $L^*$값은 낮아지고, CIE $a^*$값은 높아져 육색이 어두워지는 경향이었으며(P<0.05), CIE $b^*$값은 야콘 부산물과 쑥 분말의 급여에 의한 처리구 간의 유의적인 변화는 없었다. 결론적으로 야콘 부산물과 쑥 분말을 육계에 급여하면 TBARS 값이 낮아지고, 총 콜레스테롤과 LDL-cholesterol 및 triglyceride는 감소하며, HDL-cholesterol이 증가되어 콜레스테롤 대사에 도움이 될 것으로 사료되며, 이러한 경향은 야콘 부산물보다 쑥 분말 급여구에서 뚜렷한 결과를 나타내고 있다.

Quality and Lactic Acid Bacteria Diversity of Pork Salami Containing Kimchi Powder

  • We investigated the quality properties and bacteria diversity of pork salami containing homemade kimchi powder (KP). Pork salamis were manufactured with commercial starter culture (control), and 1% KP (KP1), 3% KP (KP3), and 5% KP (KP5). The salami was fermented and aged for 2 days and 56 days, respectively. The pH and $A_W$ values of salamis with KP were significantly lower than that of the control (p<0.05). The 2-thiobarbituric acid reactive substance values of all salamis with KP increased but were below 0.2 mg MDA/kg. Salamis with KP, decreased the $L^*$ value, but increased the $a^*$ and $b^*$ values (p<0.05). The Lactobacillus count was significantly higher in the salamis with KP than in the control by day 14 (p<0.05). The number of Leuconostoc was higher in salamis with KP than in the control and was the highest in salamis in KP1. The KP1 protected lipid oxidation and showed low TBARS value of pork salami compared to the control. This study demonstrates that KP can improve pork salami properties and can serve as a potential natural compounds for fermented meat prodcuts like Metzgerei.

Antioxidant Effects of Gamma-oryzanol on Human Prostate Cancer Cells

  • Background: To assess the antioxidant effects of gamma-oryzanol on human prostate cancer cells. Materials and Methods: Cytotoxic activity of gamma-oryzanol on human DU145 and PC3 prostate cancer cells was determined by proliferation assay using 3-(4, 5-dimethylthiazol, 2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) reagent. mRNA levels of genes involved in the intracellular antioxidant system, superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX) and glutathione reductase (GSR) were determined by reverse transcription-polymerase chain reaction (RT-PCR). Cancer cell lysates were used to measure lipid peroxidation using thiobarbituric acid reactive substance (TBARS). Glutathione contents of the cell lysates were estimated by the reaction between sulfhydryl group of 5, 5'-dithio (bis) nitrobenzoic acid (DTNB) to produce a yellow-color of 5-thio-2-nitrobenzoic acid using colorimetric assay. Catalase activity was also analysed by examining peroxidative function. Protein concentration was estimated by Bradford's assay. Results: All concentrations of gamma-oryzanol, 0.1-2.0mg/ml, significantly inhibited cell growth in a dose- and time-dependent fashion in both prostate cancer cell lines, DU145 and PC3. Gene expression of catalase in DU145 and PC3 exposed to gamma-orizanol at 0.5mg/ml for 14 days was down regulated, while mRNA of GPX was also down regulated in PC3. The MDA and glutathione levels including catalase activity in the cell lysates of DU145 and PC3 treated with gamma-oryzanol 0.1 and 0.5mg/ml were generally decreased. Conclusions: This study highlighted effects of gamma-oryzanol via the down-regulation of antioxidant genes, catalase and GPX, not cytotoxic roles. This might be interesting for adjuvant chemotherapy to make prostate cancer cells more sensitive to free radicals. It might be useful for the reduction of cytotoxic agents and cancer chemoprevention.

산사(山査)가 노화유발(老化誘發) 흰쥐의 항산화능(抗酸化能)에 미치는 영향(影響) (Effect of CRATAEGII FRUCTUS(山査) on Antioxidant Capacity in D-galactose induced Aging Rats)

  • Objectives : CRATAEGII FRUCTUS(山査) is known as the substance which delays aging by the antioxidant effects. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on antioxidant enzyme activities such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group (D-galactose 50mg/kg and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 6 weeks, SS Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, neutral fat and cholesterol were measured in rat erythrocytes and blood plasma. Results : There was no significant difference in the level of TBARS in the blood plasma between each experimental group. Red blood cell SOD activities were significantly different in each group. They were significantly decreased in the Control group compared to those of Normal and there was an increasing phenomenon in the SS group compared to the Control group. There was a significant difference in the activities of the red blood cell - glutathione peroxides in each group. There was a significant increase in the SS group compared to the Control group. Red blood cell catalase activities was no significant difference in each group. Plasma total lipid concentration was significantly different in each group. It was significantly increased in the Control group compared with the Normal group and it was decreasing in the SS group compared to the control group. Plasma triglyceride was not significantly different in each group. Plasma total cholesterol and Plasma HDL -cholesterol concentrations in the blood plasma were not significantly different in each group. Conclusions : According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in inhibiting lipid peroxides and increasing the activities of antioxidative(anti aging) enzyme in D-galactose induced aging rat.
