Streptozotocin에 의한 lipid peroxidation 측정시 TBA법의 적합성에 관한 연구 (Drect Interaction of Streptozotocin with TBA (thiobarbituric acid) in Lipid Peroxidation Analysis)

  • Streptozotocin에 의한 적혈구의 막 lipid peroxidation 측정시 TBA법의 적합성에 관하여 검토하기 위하여 Streptozotocin와 TBA의 직접반응 여부를 연구하였다. 여러 농도의 Streptozotocin 와 20% RBC suspension을 함께 배양한 후 lipid peroxidation을 TBA법으로 측정했을 때, Streptozotocin의 농도에 따라 흡광도가 증가하다가 100mM부터는 plateau를 이루었다. 그런데 적혈구를 제거한 대조실험에서도 발색단이 관찰되었다. 시험관에서 5mM의 Streptozotocin와 1% TBA를 산성조건에서 가열 직접 반응시킨 후 spectrum을 찍은 결과 TBA법 측정파장인 532nm에서 새로운 peak가 생겼다. 또 이 파장에서의 흡광도는 Streptozotocin 농도에 따라 증가하다가 25mM부터는 plateau가 되었다. 이러한 결과들은 적혈구와 Streptozotocin를 함께 배양하여 lipid peroxidation을 측정할 때, cell 내로 유입된 Streptozotocin 가 TBA와 직접반응을 할 수 있음을 제시한다. 그리고 다른 측정법인 lipid conjugation은 233nm에서의 흡수를 측정하는데. Streptozotocin 자체가 200-300nm에서 흡수를 나타내므로 이 방법으로도 lipid peroxidation 측정을 방해할 수 있다. 따라서 Streptozotocin에 의한 적혈구막의 lipid peroxidation은 TBA method 및 lipid conjugation 방법으로 측정하기는 부적합한 것으로 사료된다.

t-Butyl Alcohol 동결건조법을 이용한 흰쥐 간장의 주사전자현미경적 관찰 (Tissue Preparation with t-Butyl Alcohol Freeze-drying Method for Scanning Electron Microscopy: Application for Rat Liver)

  • T-butyl alcohol (TBA) freeze-drying method originally designed by Inoue and Osadake (1989) was adopted to dry specimens for scanning electron microscopy and the results were compared with those dried using critical point dryer (CPD). Small pieces $(1\times1\times3mm)$ of liver of Sprague-Dawley rats were cut and fixed in 2% glutaraldehyde in 0.1 M sodium cacodylate buffer after anesthesia, and processed for scanning electron microscopy by several modifications of TBA freeze-drying methods and by the standard preparation method using CPD. The bile canaliculi and sinusoidal endothelial surface were observed. Tissue dehydrated with TBA before TBA freeze-drying preserved the structures best comparable to those prepared with CPD. This result suggests that combination of dehydration with TBA and TBA freeze-drying is a superior method to the original TBA freeze-drying method dehydrated with ethanol.

  • 혐기적 조건밑에서 $FeCl_{3}$에 의해 촉진된 TBA 반응을 이용하여 혈청중 과산화물 측정을 위한 새로운미량 분석법을 설정하였다. 즉. 혈청단백침전물이 현탁된 TBA반응흔합액에 혈청 $10{\mu}l$$1.0{\mu}mole$$FeCl_{3}$를 첨가한 다음 질소기류밑에서 50분간 끊는 물에 중탕한다. 이 분석법의 반응 예민도는 현행 일반 TBA법보다 40여배나 증대되었으며 따라서 미량의 시료$(10{\sim}20{\mu}l)$로도 TBA반응생성물의 농도 측정이 비색법으로도 가능하였다. 그리고 TBA반응생성물의 butanol 추출액을 $(NH_{4})_{2}SO_{4}$염석으로 부분말수시킴으로써 예민성 및 재현성이 개선될 수 있었다. 이 분석법을 이용하여 측정한 건강인들의 혈청 TBA같은 높은 연령군일수록 증가하는 경향을 보였다.

영지의 항산화성 물질에 관한 연구 (Studies on Antioxidative Substances of Ganoderma lucidum)

  • 영지 추출액의 항산화 활성을 thiocyanate method, TBA method, weighing method를 이용하여 검색한 결과 n-hexane 추출물, methanol 추출물에서 항산화활성이 나타났으며 그 중에서 n-hexane 추출물의 fraction V와 methanol 추출물의 methanol 불용성 fraction II에서 강한 활성을 나타내었다. 이들 항산화 활성을 나타내는 fraction 5와 fraction II를 rat의 간 균질액을 이용한 항산화 활성 검색에서 과산화 지질 생성억제를 TBA값으로 나타내어 inhibition ratio(I.R.)를 산출하였으며 그 결과 n-hexane 추출물의 fraction V는 l5%, methanol 추출물의 methanol 불용성 fraction II는 54.6%의 억제율을 나타내었다 이는 rat의 간 mitochondria에서 과산화지질 생성 억제율이 영지의 물 추출물은 16%, acetone 추출물은 23%인 것에 비하여 약간 높았으나, microsome에서 과산화지질 생성 억제율이 물 추출물은 44%, acetone 추출물은 75%인 것에 비하여 다소 낮은 활성을 보여주었다.

  • Effect of frozen storage(0, 15, 30, 40 days, -18$^{\circ}C$), cooking methods(frying, microwaving) and reheating on lipid oxidation in chicken meats were evaluated by measuring thiobarbituric acid value (TBA value) and by measuring fluorescence value. TBA values were increased by storage days and were higher in leg meats than breast meats. According to cooking method, TBA values were higher in frying chicken meats. The fluorescence values were also increased by storage days and were higher in breast meats than leg meats.

Analysis of tert-Butanol, Methyl tert-Butyl Ether, Benzene, Toluene, Ethylbenzene and Xylene in Ground Water by Headspace Gas Chromatography-Mass Spectrometry

  • Methyl tert-butyl ether (MTBE) is added to gasoline to enhance the octane number of gasoline, tert-butyl alcohol (TBA) is major degradation intermediate of MTBE in environment, and benzene, toluene, ethyl benzene and xylene (BTEX) are also major constituents of gasoline. In this study, a simplified headspace analysis method was adapted for simultaneous determination of MTBE, TBA and BTEX in ground water samples. The sample 5.0 mL and 2 g NaCl were placed in a 10 mL vial and the solution was spiked with fluorobenzene as an internal standard and sealed with a cap. The vial was placed in a heating block at 85 $^{\circ}C$ for 30 min. The detection limits of the assay were 0.01 ${\mu}$g/L for MTBE and BTEX, and 0.02 ${\mu}$g/L for TBA. The method was used to analyze 110 ground water samples from various regions in Korea, and to survey the their background concentration in ground water in Korea. The samples revealed MTBE concentrations in the range of 0.01 - 0.45 ${\mu}$g/L (detection frequency of 57.3%), TBA concentrations in the range of 0.02 - 0.08 ${\mu}$g/L (detection frequency of 5.5%), and total BTEX concentrations in the range of 0.01 - 2.09 ${\mu}$g/L (detection frequency of 87.3%). The developed method may be used when simultaneously determining the amount of MTBE, TBA and BTEX in water.

포장방법이 동결계육의 저장성 및 미생물의 증식에 미치는 영향 (Effect of Packing Method on Shelf-life and Microbiology of Frozen Chicken)

  • This experiment was carried out to find out the effects of packing methods on shelf-life and hy-giene of breast and thigh meats in chicken. The carcass weights were about 1.6 kg per bird, and chilled by air spray method. The breast and thigh meats were packed by either vacuum or atmosphere packing methods. Samples were analyzed at 1, 4, 8, 12 and 16 wk after quick freezing at -45˚C for 35 min. Regardless of packing method. The thiobarbituric acid(TBA) values of thigh meats after 1 wk of storage were higher than those of breast meats. The TBA values of both treatments were increased as storage period extended. In addition, the TBA values of vacuum packed meats were lower than those of atmosphere packed meats. The VBN(volatile basic nitrogen) values, regardless of packing method, at 1 wk of storage were higher than those of breast meats. The VBN contents of breast and thigh meats after 16 wk of storage increased markedly, but failed to show significant difference between the two packaging methods. Total plate counts of breast and thigh meats were increased gradually as storage period extended, and the total plate count of breast meat was higher than that of thigh meat. The coliform bacteria were not detected until 16 wk after storage.

다양한 토양 환경에서 Methyl tert-Butyl Ether와 그의 대사산물이 노출되었을 때 미생물 군집에 미치는 영향: 논, 밭, 갯벌 시료 비교 (Effect of Methyl tert-butyl Ether and Its Metabolites on the Microbial Population: Comparison of Soil Samples from Rice Field, Leek Patch and Tidal Mud Flat)

  • Toxic effect of methyl tert-butyl ether (MTBE), tert-butyl alcohol (TBA) and formaldehyde (FA) on microbial activity and diversity was compared in rice field, leek patch, and tidal mud flat soil samples. MTBE, TBA and FA with different concentrations were added into microcosms containing these soil samples, and placed at room temperature for 30 days. Then the microbial activities such as dehydrogenase and viable cell numbers and microbial community using a DGGE (Denaturing gradient gel electrophoresis) fingerprinting method were measured. Among the samples, dehydrogenase activity in rice field was inhibited the most by MTBE, TBA and FA. The toxic effect was higher according to the following orders: FA > MTBE > TBA. Dominant species in the microcosms contaminated with MTBE, TBA and FA were Chloroflex, Bacilli, gamma-proteobacteria in the rice field sample, Sphingobacteria, Flavobacteria, Actinobacteria, Bacilli, gamma-proteobacteria in the leek patch sample, and Sphingobacteria, Flavobacteria, delta-proteobacteria, gamma-proteobacteria in the tidal mud flat sample.

냉장방법과 포장방법이 냉장계육의 저장성 및 미생물의 증식에 미치는 영향 (Effects of Packing Methods on Storage and Microbiology of Chilled Chicken Breast and Thigh Meats)

  • This experiment was carried out to investigate the effects on shelf-life of chilling and packing methods of cold chicken breast and thigh meats. Deboned chicken breast and thigh meats were packed by either vacuum or atmosphere packing method, and stored at -2˚C for 1, 3, 7, 11, 15, and 20 days. The thiobarbituric acid(TBA) values of all treatments were significantly increased as the storage period extended(P<0.05), however, TBA values of all treatments were remark-ably decreased at 15 days. TBA values of immersion chilled and vacuum-packed thigh meats were increased, but no difference between atmosphere and vacuum-packing methods was found. The volatile basic nitrogen(VBN) values at 1 day of storage were low, but they were increased as the storage period extended. The VBN values at 20 days after storage were from 12.25~19.28 mg %. Total bacterial counts tended to increase in all treatments, however, no significant difference was found in any treatment. Total plate counts of breast meat were higher than those of thigh meat as the storage period extended.

바질의 항산화 물질 측정에 관한 연구 (Measurement of Antioxidation Substances in Basil)

  • Basil is known to contain six types of polyphenols that engage in physiological activation; protocateuic acid, caffeic acid, chlorogenic acid, courmaric acid, rosmarinic acid and quercetin. In this study, the antioxidants in eight types of basil were evaluated. Specifically, the antioxidative activation of basil was evaluated based on the relationship between active oxygen scavenging (DPPH radical-scavenging), which was used as an index showing the content and functionality of the polyphenol compounds in basil, and $Fe^{2+}$/ascorbate (FTC) and 2-thiobarbituric acid (TBA). The total polyphenol content of the different types basil occurred in the following order: Dark-opal> Lettuce> Bush> Greek> Lemon> Sweet> Geno> Holy. The highest content Dark-opal was 173.3 mg, which was about three times greater than the lowest content Holy, which was 49.85 mg. In addition, DPPH radical-scavenging by Dark-opal 51%, which was the highest scavenging activity occurred in the folling order: Dark-opal> Lettuce> Bush> Greek> Lemon> Sweet> Geno> Holy. The antioxidative activation values measured using the FTC and TBA were the same as the value obtained using the DPPH method. Finally, the level of antioxidative activation measured using FTC, TBA and DPPH methods showed that the higher the content of polyphenol substance was, the stronger the antioxidative activation became.
