Phylogenetic Relationship of Ganoderma Species with the Polyporaceae Based on RFLP Analysis of the Nuclear ITS Region

  • Restriction-polymorphic patterns of nuclear-ITS were examined for the genetic relationships among 12 bisidiomycetous mushrooms to Aphyllophorales and Agaricales. The taxonomic affinity of Ganoderma species with the family Polyporaceae also was examined. With 13 restriction endonucleases, 159 restriction characters were generated form the 12 species examined. UPGMA and neighbor-joining analyses separated the 12 species into two genetically distinct groups that correspond to orders (Agaricales and Aphyllophorales) where each species is included. This result indicates that there is clear genetic demarcation between Agaricales and Aphyllophorales. Dendrograms constructed by several data analyses showed that even though Ganoderma species are somewhat in intermediate taxonomic position between the Polyporaceae and families of the Agaricales, they are genetically more related to the Polyporaceae. These results are consistent with morphological characters observed in those mushrooms. However, it is premature to conclude taxonomic status Ganoderma species in the present study employing small sample size.

Taxonomic notes and distribution of Gueldenstaedtia (Chesneyinae, Fabaceae) in Mongolia

  • The genus Gueldenstaedtia Fisch. is small genus of Fabaceae that includes four species worldwide. Among these, G. monophylla Fisch and G. verna (Georgi) Boriss. are currently recognized as occurring in Mongolia. Here, we present our findings on G. verna from our recent field surveys in eastern Mongolia in 2020. Gueldenstaedtia monophylla is mostly distributed in the western region, whereas G. verna occurs in eastern Mongolia. The regional conservation status of both species was assessed as near threatened based on criterion B in the country. We provided descriptions, grid distribution maps, taxonomic notes and photographic illustrations of the two species based on our newly collected samples.

The Status of Distribution for Native Freshwater Turtles in Korea, with Remarks on Taxonomic Position

  • Korean freshwater turtles are divided into native turtles, Chinemys reevesii in Geoemydidae, Pelodiscus sinensis in Trionychidae, and imported turtles, Trachemys scripta elegans, T. s. scripta, Pseudemys rubriventris and Ocadia sinensis. In this study, we determined the distribution of Korean freshwater turtles based on a field study and literature study, and considered the taxonomic position of unidentified native turtles. The study was carried out between May 2010 and November 2011 during the day time, and the capturing tool used to collect turtles consisted of net and metal frame. C. reevesii and P. sinensis were found in 16 and 11 places, respectively. C. reevesii, which inhabits the Korean peninsula, is a taxonomically controversial species, but it is presumed that the turtle belongs to Mauremys. Moreover recent taxonomic studies of the soft-shell turtle have shown that the Korean native species is P. maackii, and P. sinensis was imported from abroad and has been in the natural habitats of Korea since 1970s. The exterior shape and skeletal form of P. maackii and P. sinensis are nearly similar. However, the skin color and yellow spots on the shell could be used to distinguish the two species.

  • $L{\acute{e}}veill{\acute{e}}$가 상당수의 한국산 신분류군을 기재하였음에도 불구하고, 이들 분류군에 대한 종합적인 분류학적 검토가 아직까지 국내에서 수행된 바가 없다. 본 논문에서는 $L{\acute{e}}veill{\acute{e}}$가 Decades plantarum novarum이라는 연속 논문에서 한국에서 채집된 표본을 근거로 신분류군으로 발표한 것들의 실체를 파악하기 위하여, 지금까지의 연구 결과를 비교 검토하였다. 그 결과 $L{\acute{e}}veill{\acute{e}}$의 의해 총 275개의 신분류군이 발표되었음이 확인되었다. 이들 가운데에는 Hydrolirion $H. L{\acute{e}}v.$이라는 신속도 포함되어 있으나, 이는 현재 Blyxa Noronha ex Thouars와 같은 분류군으로 간주되고 있다. 나머지 분류군들은 종 수준에서 231개 분류군, 변종 수준에서 43개 분류군이 포함되는 것으로 확인되었다. 종 수준으로 보고된 분류군 중, 현재까지 종 또는 변종 수준에서 인정되는 분류군은 17개였고, 학자에 따라 종 또는 이명으로 간주되는 분류군은 52개로 파악되었으며, 151개 분류군은 다른 분류군과 동일한 것으로 처리되었다. 한편, 11개 분류군은 국내 학자들에 의해 소개되지 않은 것으로 확인되었다.

더위지기 및 근연분류군의 분류학적 재검토 (Taxonomic revision of the Artemisia sacrorum group)

  • 더위지기 및 근연분류군은 분류학적 위치 및 학명에 대하여 많은 이견이 있어왔다. 이에 본 연구는더위지기 및 근연분류군인 털산쑥, 흰산쑥의 외부형태학적 연구를 바탕으로 정확한 학명을 부여하고 분류학적 위치를 재검토하기 위하여 실시되었다. 원기재와 기준표본을 비교 및 분석한 결과, 털산쑥(A. freyniana f. discolor)과 흰산쑥(A. freyniana f. vestita)은 잎에서 털의 분포 양상, 지리적 분포, 체세포염색체 수 및 화분의 자상돌기 기부의 연결 유무 등의 특징에 의해 A. sacrorum var. sacrorum와 A. sacrorum var. incana으로 각각 이명 처리하였으며, A. gmelinii의 학명으로 잘못 사용되어온 더위지기(A. iwayomogi)의 학명은 A. sacrorum그룹과의 형태학적 유사성을 바탕으로 신조합명인 A. sacrorum var. iwayomogi (Kitam.) M. S. Park & G. Y. Chung으로 처리하였다. 또한, 이들의 기재, 도해 및 검색표를 제시하였다.

The Phylogenetic Relationship of Several Oscillatorian Cyanobacteria, Forming Blooms at Daecheong Reservoirs, Based on Partial 16S rRNA Gene Sequences

  • The partial 16S gene sequences of six filamentous cyanobacterial strains, Oscillatoria lmosa KCTC AG10168, Oscillatoria princeps KCTC AG10153, Oscillatoria sp. KCTC AG 10184, Phormidium tenue KCTC AG10158, Phormidium parchydematicum KCTC AG10164, and Lyngbya hieronymusii KCTC AG10199, which were isolated in the late summer at Daecheong Reservoirs, were determined and assigned their phylogenetic and taxonomic position among taxa of order Ocillatoriales whose partial 16S rRNA gene sequences aligned in this suty, were very heterogeneously clustered with other taxa. The two strains, Oscillatoria limosa KCTC AG10168 and O. princeps KCTC AG10153, formed a cluster with O. sancta PCC7515, which supported 64% of the bootstrap trees with high similarity (19-96.15%). Strain Oscillatoria sp. KCTC AG10184, that was known to produce a nasty substance, was closely related to the toxic Oscillatoria group. The study on morphological variation in various environments and toxin production will confirm the taxonomic status of these species. Phormidium tenus KCTC AG10158 and Phormidium parchydematicum KCTC AG10164 made a cluster with other oscillatorian species of Phormidium, Oscillatoria, and Leptolynbya, which supproted 100% of the bootstrap trees with a very high sequence smilarity (96.8-99.8%) in thsi study. The sequence analysis in this study also supported that taxa of oscillatoriales are not monophyletic. Some of the fractures, such as the presence or absence of sheath and cell shape, which were used to define them, would be inadequate and should be reconfirmed. We suggest that sequences of partial 16S rRNA gene fragments aligned in this study should be more useful than morphological features in the identification and reconfirmation of the taxonomic status of these oscillactorian cyanobacteria.

한국 완도에 서식하고 있는 등줄쥐(포유강: 설치목)의 분류학적 위치 (Taxonomic Stautus of Striped Field Mice(Mammalia: Rodenita) from Wando Island, Korea)

  • 완도산 등줄쥐의 분류학적 위치를 재검토하기 위해, 한국의 8개 지역에서 채집한 등줄쥐(Apodemus agrarius)의 31개 형태적 형질들을 다변량분석방법으로 분석하였다. 두 군으로 나누어 졌는데, 6개 지역(태백산, 월악산, 청주, 지리산, 군산, 진도)의 작은 형과 2개 지역(완도, 제주도)의 큰 형이었다. 6개 지역의 작은 형은 Thomas(1908)가 밝힌 대로 A. a. coreae이고, 제주도 지역의 큰 형은 Johnson과 Jones(1955)이 이 기재한 대로 A. a. chejuensis임이 확이되었다. 뿐만 아니라, 완도의 큰형은 Joh(1989)가 제안한대로 A. a. chejuensis임도 확인되었다.

Phylogenetic rind Taxonomic Status of the Phytoplasmas Associated with Water Dropwort (Oenanthe javanica DC) Disease in Korea and Japan

  • To evaluate the phylogenetic and taxonomic status of the phytoplasmas associated with water dropwort (Oenanthe javanica DC) disease in Korea and Japan, their 16S rDNA was analyzed. DNAs extracted from water dropworts collected in Korea (Kyongnam province) and Japan (Chiba prefecture) affected by witches' broom and yellows were subjected to PCR using phytoplasma-specific primers, which amplified a 1.4-kbp fragment that included the 16S rDNA. Phytoplasmas were characterized by RFLP analysis using AluI, HaeIII, HhaI, KpnI, MseI, and RsaI restriction enzymes and by sequence analysis of the PCR products. The mater dropwort witches'broom (WDWB) and water dropwort yellows (WDY) 16S rDNA sequences were identical and closely related to onion yellows (OY, 99.9% identity), which belong to the aster yellows (AY) 16S-subgroup. However, the KpnI RFLP analyses clearly distinguished the WDY and WDWB phytoplasmas from the OY phytoplasma. The phylogenetic analysis based on 16S rDNA showed that WDWE and WDY phytoplasmas are members of a relatively homogeneous group that evolved from a common ancestor.

Taxonomic Status of Siberian Flying Squirrel from Korea (Pteromys volans aluco Thomas 1907)

  • Sequences of mitochondrial DNA (mtDNA) cytochrome b gene (1,140 bp) and control region (803 bp) of Siberian flying squirrels from Korea (Pteromys volans aluco) and Mt. Changbai of northeast China (P. v. arsenjevi) were obtained to reexamine the taxonomic status of the Korean subspecies. In the cytochrome b gene, six haplotypes of P. v. aluco formed a clade with six haplotypes of P. v. arsenjevi, and in control region, seven haplotypes of P. v. aluco formed a clade with six haplotypes of P. v. arsenjevi. Furthermore, six haplotypes of cytochrome b gene of P. v. aluco from this study formed a clade with four haplotypes of P. v. arsenjevi in far-east Russia obtained from GenBank. We also investigated the research papers previously published that reported the length of tail vertebrae of P. volans, and found that the length was not sufficiently large as to be a key character of P. v. aluco. This result is not consistent with morphological description for its haplotype. Therefore, we conclude that P. v. aluco from Korea might possibly be a synonym of P. v. arsenjevi from northeast China and nearby Russia.
