Sulfhydryl-Related and Phenylpropanoid-Synthesizing Enzymes in Arabidopsis thaliana Leaves after Treatments with Hydrogen Peroxide, Heavy Metals, and Glyphosate

  • Three-week grown Arabidopsis thaliana leaves were wounded by cutting whole leaves with a razor blade into pieces (about$3\;mm\;{\times}\;3\;mm$) submerged in various solutions, and incubated in a growth chamber for 24 h. We measured and compared activities of several enzymes such as phenylalanine ammonia-lyase (PAL), tyrosine ammonia-lyase (TAL), thioredoxin, thioredoxin reductase, thioltransferase, glutathione reductase, and $NADP^+$ -malate dehydrogenase. PAL activity was decreased in $HgCl_2$-, $CdCl_2$-, and glyphosate-treated leaf slices, and could not be detected after treatment with $CdCl_2$. TAL activity was found to be maximal in the $CdCl_2$-treated leaf slices. Activity of thioredoxin, a small protein known as a cofactor of ribonucleotide reductase and a regulator of photosynthesis, was significantly increased in the $CdCl_2$-treated leaf slices, while thioredoxin reductase activity was maximal in the $HgCl_2$-treated leaf slices. Thioltransferase and glutathione reductase activities were significantly decreased in the $HgCl_2$-treated leaf slices. $NADP^+$ -malate dehydrogenase activity remained relatively constant after the chemical treatments. Our results strongly indicate that sulfhydryl-related and phenylpropanoid-synthesizing enzyme activities are affected by chemical treatments such as hydrogen peroxide, heavy metals, and glyphosate.

LDB2 regulates the expression of DLL4 through the formation of oligomeric complexes in endothelial cells

  • Delta-like ligand 4 (DLL4) expression in endothelial cells is intimately associated with angiogenic sprouting and vascular remodeling, but the precise mechanism of transcriptional regulation of DLL4 remains incompletely understood. Here, we showed that LIM-domain binding protein 2 (LDB2) plays an important role in regulating basal DLL4 and VEGF-induced DLL4 expression. Knockdown of LDB2 using siRNA enhanced endothelial sprouting and tubular network formation in vitro. Injection of ldb2-morpholino resulted in defective development of intersegmental vessels in zebrafish. Reduction or over-expression of LDB2 in endothelial cells decreased or increased DLL4 expression. LDB2 regulated DLL4 promoter activity by binding to its promoter region and the same promoter region was occupied and regulated by the LMO2/TAL1/GATA2 complex. Interestingly, LDB2 also mediated VEGF-induced DLL4 expression in endothelial cells. The regulation of DLL4 by the LDB2 complex provides a novel mechanism of DLL4 transcriptional control that may be exploited to develop therapeutics for aberrant vascular remodeling.

Anti-oxidant and Hepatoprotective Activities of Alcoholic Extract of Terminalia arjuna

  • Alcoholic extract of Terminalia arjuna [TA] was evaluated far its hepatoprotective activity against carbon tetrachloride (CCl$_4$)-induced hepatic damage in rats. The hepatoprotective activity of TA was evaluated by measuring levels of serum marker enzymes like serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), serum alkaline phosphatase (SALP). The serum levels of total proteins(TP), total albumins (TAL) and bilirubin (BILN) were also estimated. The histological studies were also carried out to support the above parameters. Silymarin (SM) was used as standard drug. Administration of TA (250 and 500 mg/kg/po) markedly prevented CCl$_4$-induced elevation of levels of SGOT, SGPT, SALP, TP, TAL and BILN. These biochemical observations were supplemented by histopathological examination of liver sections. Alcoholic extract of TA also shown significant in-vitro free radical scavenging activity against 1, 1-diphenyl-2-picryl hydrazyl (DPPH) and nitric oxide (NO) radicals. Thus, the present study provides a scientific rationale for the traditional use of this plant in the management of liver diseases.

Steam Reforming of Toluene Over Ni/Coal Ash Catalysts: Effect of Coal Ash Composition

  • The development of a low cost catalyst with high performance and small amount of carbon deposition on catalyst from toluene steam reforming were investigated by using coal ash as a support material. Ni-loaded coal ash catalyst showed similar catalytic activity for toluene steam reforming compared with the Ni/Al2O3. At 800 ℃, the toluene conversion was 77% for Ni/TAL, 68% for Ni/KPU and 78% for Ni/Al2O3. Ni/TAL showed similar toluene conversion to Ni/Al2O3. However, Ni/KPU produced higher hydrogen yield at relatively lower toluene conversion. Ni/KPU catalyst showed a remarkable ability of suppressing the carbon deposition. The difference in coke deposition and hydrogen yield is due to the composition of KPU ash (Ca and Fe) which increase coke resistance and water gas shift reaction. This study suggests that coal ash catalysts have great potential for the application in the steam reforming of biomass tar.

  • Fractals which represent many of the sets in various scien-tific fields as well as in nature is geometrically too complicate. Then we usually use Hausdorff dimension to estimate their geometrical proper-ties. But to explain the fractals from the hausdorff dimension induced by the Euclidan metric are not too sufficient. For example in digi-tal communication while encoding or decoding the fractal images we must consider not only their geometric sizes but also many other fac-tors such as colours densities and energies etc. So in this paper we define the dimension matrix of the sets by redefining the new metric.

A Study on The Oriental-medical Understanding of Depression (우울증(憂鬱症)의 한방적(韓方的) 이해(理解)에 관한 문헌고찰(文獻考察))

  • According to ICD-10, The typical Depression are characterized by dejected mood, loss of interest and pleasure, feeling of helplessness. Also, other symptoms such as loss of attentiveness and concentration, guilty conscience, pessimistic attitude for the future, sleeping disorder, lack of appetite, thinking of or doing a self-injury or suicidal act may characterize Depression. These Symptoms of Depression are similar to Wool-Zeng(鬱證), Zeon-Zeng(癲證), Her-ro(虛勞), Bul-Sa-Sik(不思食), Tal-Young-Sil-Zeong(脫營失精), Bul-Myeon(不眠), Ki-Myeon(嗜眠) in Oriental Medicine. In general, Depression is classified into Wool-Zeng type and Zeon-Zeng type. The former is similar to the neurotic type of depression, the latter is similar to the psychopathic type of depression. The clear causes of Depression are not known yet. But psychodynamic and biological factors are assumed to cause Depression. Psychodynamic factors may correspond to Chil-Zeong(七情) and biological factors to Tae-Byeong(胎病) and Dam(痰) in Oriental medicine. In Occidental medicine, Depression is treated by psychotherapy and medication. Electrically-induced spasm therapy and phototherapy also used for the treatment of Depression. In Oriental medicine, Depression is treated by more various therapy such as herb medication, acupuncture and moxibustion, oriental psychotherapy and dirigation(Gi-Gong, 氣功). Depression is not easy to cure. When the treatments of Occidental medicine and Oriental medicine are mutually cooperated, more effective medical care might be expected. Therefore, futher study to treat depression by mutually cooperated therapy is necessary.

A Simple Embryonic Stem Cell-Based in vitro Differentiation System That Recapitulates Early Erythropoietic Events in the Mouse Embryo (생쥐 배아에서의 초기 적혈구 분화를 재현 할 수 있는 배아주 세포에 기초한 간단한 시험관내 분화체계)

  • An embryonic stem (ES) cell-based in vitro model system was examined to determine whether a simple differentiation of embryoid bodies (EB) in the suspension medium is useful to dissect early erythropoiesis. Characteristics of the differentiating EBs were monitored for their differentiation potential to generate hematopoietic cell types by general morphology, benzidine staining and two-step colony assays, and expressivity of several erythroid marker genes by the RT-PCR analysis for total cellular RNA prepared from the differentiating EBs. Every ematopoietic lineage cells were generated from the differentiating EBs with reproducible frequencies, similar to the other sophisticated differentiation protocols. Furthermore, the globin gene switching in differentiating ES cells paralleled the sequence of events found in the mouse embryo, and such that their expression was activated by at least 12 hrs later than those of erythroid-specific transcription factors, GATA-1 and Tal-1 The erythropoietic differentiation program initiated reproducibly and efficiently in this simple differentiation system in a suspension culture, such that this system may be useful for dissection of the molecular events of early erythropoiesis.

The Study of Literature on Meridians and Acupoints about Acupncture Treatment of Alopecia (탈모(脫毛)의 침구치료(鍼灸治療)에 대한 경락(經絡), 경혈적고찰(經穴的考察))

  • Objectives: This study was designed to investigate acupuncture treatment of alopecia patients by researching literature and to standardize acupuncture treatment. Methods: We extracted the parts about acupuncture treatment of alopecia which are called 'Tal Bal(說髮), 'Ban Dok'(斑禿), 'Tal Mo'(脫毛), 'Bal Rak'(髮洛), 'Wonhyung Talmo'(圓形脫老) from 2 ancient and 49 modern oriental medicine literal sources. We surveyed the frequency and characteristics of the acupoints used for treatment of alopecia, and the acupoint was classified according to its meridian or demonstration. The results of this study were recorded as follows: 1. The most frequently used acupoints were GB20(風紙), GV20(百會), BL13(膈兪), SP6(三陰交), ST36(足三里), BL23(腎兪), SP10(血海), LI11(曲池), in that order. 2. The most frequently used meridians were the urinary bladder meridian (足太湯膀胱經), the Governor Vessel(督豚), the spleen meridian(足少陰脾經), and the gall bladder meridian(足少陰膽經), in that order 3. The most frequently used regions were the head, under the knee, and the back, in that order. 4. The most frequently used Jang organs and Bu organs were the spleen(脾), the stomach(胃), the liver(肝), the gall bladder(擔), the lungs(肺), and the large intestines(大腸), in that order. 5. Common methods of differentiation of alopecia are Hyulyulsaengpung(血熱生風), Gichehyutea(氣滯血瘀), Gihyulyanghea(氣血兩瘀), and Gansinbugok(肝腎不足). Conclusions : For treatment of alopecia, all the patient's symptoms, including alopecia, must taken into consideration and demonstrated.
