This study was performed to investigate the effect of ginseng saponin from Korean red ginseng on the learning and memory. Total (50, 100 mg/kg, bw) and panaxadiol saponin (15, 30 mg/kg, bw) treated groups did not show the difference of the time score and the number of error in comparison with control group. Panaxatriol saponin (15, 30 mg/kg, bw) significantly decreased both the time score and the number of error in water maze test. These results indicate that panaxatriol saponin from Korean red ginseng may improve the learning ability of rat in water multiple T-maze.
The aim of this study is to investigate whether alcohol alters learning and memory processes pertaining to emotional and spatial factors using the active avoidance and T-maze task in zebrafish. In the active avoidance task, zebrafish were trained to escape from one compartment to another to avoid electric shocks (unconditioned stimulus) following a conditioned light signal. Acquisition of active avoidance task appeared to be normal in zebrafish that were treated with 1% alcohol for 30 min for 17 days until the end of the behavioral test, and retention ability of learned behavior, tested 2 days later, was the same as control group. In the T-maze task, the time to find a reservoir was compared. While the latency was similar during the 1 st training session between control and alcohol-treated zebrafish, it was significantly longer in alcohol-treated zebrafish during retention test 24 h later. Furthermore, when alcohol was treated 30 min after 2nd session without prior treatment, zebrafish demonstrated similar retention ability compared to control. These results suggest that chronic alcohol treatment alters spatial learning of zebrafish, but not emotional learning.
Objectives : The purpose of this study was to characterize the effect of the fraction of Taraxacum officinale and T.coreanum on the learning and memory impairments induced by scopolamine. Methods : The cognition-enhancing effect of Taraxacum officinale and T.coreanum was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The results showed that the Aug harvested T.offiicinale extract-treated group (200 mg/kg, p.o.) and the tacrine-treated group (10 mg/kg, p.o.) significantly ameliorated scopolamine-induced amnesia based on the Passive avoidance Y-maze test and Watermaze test. And these results are same manner in DPPH radical scavenger effect and Acetylcholineseterase inhibition effect. Conclusions : These results suggest that Taraxacum officinale extract maybe a useful cognitive impairment treatment, and its beneficial effects are depending on the collecting time and origin plants. As a result, Taraxacum officinale harvested in August improve memory most.
This study was carried out to investigate; the alpha-fetoprotein, urea and free fatty acid in amniotic fluids and water filled multiple T-maze test of fetuses affected by Ethylenethiourea. The S.P.F. Sprague-Dawley female rats(10 weeks) were used in this study and these animals were divided into four groups; control group I (10mg/kg/day), group II(30mg/kg/day), group III(50mg/kg/day). The results obtained were summarized as follows; 1. In the water filled multiple T-maze test of F1 male rats, The time and errors from start point to goal point of 30mg/kg group are significantly(p<0.001) increased from control group in 3rd day of test. In F1 female rats of 2nd day and 3rd day of water filled multiple T-maze test, The time and some errors in 30mg/kg group are significantly (p<0.05) increased from control group. 2. Alpha-fetoprotein values of all treated groups(10mg/kg, 30mg/kg and 50mg/kg) were significantly(p<0.001) decreased from control values in the amniotic fluids. 3. Urea values of ammniotic fluids in 50mg/kg group are significantly increased from control group. 4. Free fatty acid values of ammniotic fluids in 50mg/kg group are significantly increased from control group.
The roots of the plant Korean ginseng have been extensively used in the traditional Chinese herbal medicine. The effects of chronic administration of Korean ginseng extract (KGE) were investigated on two different anxiety models: the elevated T-maze (for inhibitory avoidance and escape measurements) and the open field test (OFT). Diazepam (1 mg/kg), KGE (10, 30 and 100 mg/kg) were administered orally for 15 days. On the 14th day, mice were previously exposed for 30 min to one of the open arms of the T-maze, 24 h before the test. On 15th day, mice had two exposures to the enclosed and open arm of the elevated T-maze followed by exposure to the open field apparatus. The number of line crossings in the apparatus was used to assess locomotor changes. Cumulative Concentration Response Curve of 5-HT was plotted using rat fundus which were pre-treated in a similar way. Treatment with Diazepam (1 mg/kg) and KGE (10, 30 and 100 mg/kg) significantly (P < 0.05) impaired inhibitory avoidance performance but did not impair escape latency. In OFT, diazepam facilitated locomotion as compared to vehicle and other treatment groups. KGE at any of the selected doses did not impair locomotion. Concentration response curve of 5-HT was shifted towards the right with suppression of maxima in rats treated with KGE. The results suggest that KGE exerts anxiolytic like behaviour in a specific subset of defensive behaviour, particularly those related to generalized anxiety disorder.
Objective: This study was conducted to find out the effect of saruktanggamibang(SRT) on memory and learning of rats. Methods: The mon·is water maze was used. It was performed Acquisition test, Retention test of water maze. After Behavioral test, it was investigated AChE cell numbers of CAI and CA3 on hippocampus. Results: of water maze revealed that acqusitive abilities of SAP+SRT group significantly improved on 3, 4, 5th day compared with SAP+Saline group. Retention test of water maze didn't reveal that retentive abilities of SAP+SRT group significantly improved comparing with another group. ChAT cell numbers of medial septum of SAP+SRT group significantly increased compared with SAP+Saline group. AChE cell numbers of CAl and CA3 on hippocampus of Sham+SRT group significantly increased compared with Sham group. And AChE cell numbers of CAl and CA3 on hippocampus of SAP+SRT group significantly increased compared with SAP+Saline group. Conclusion: Saruktanggamibang(SRT) has an effect on memory and learning of rats.
Marine organisms are sources of many bioactive compounds, such as essential fatty acids, essential amino acids, vitamins, and minerals, making them useful candidates for the production of safe bioactive substances. They also synthesize glutamic acid, which can be used to produce γ-aminobutyric acid (GABA), the major inhibitory neurotransmitter in the central nervous system (CNS), via fermentation with Lactobacillus brevis BJ-20. This study investigated the degree to which fermented sea tangle (FST) inhibits enzymes such as acetylcholine esterase (AChE) and prolyl endopeptidase (PEP) and affects memory of normal mice using the T-maze test. FST inhibited more than 90% of AChE at 1 mg/mL and 50% of PEP at 8 mg/mL. Oral FST (100 mg/kg) significantly improved performance of normal mice on the T-maze. Therefore, sea tangle fermented with L. brevis BJ20 effectively contributes to memory improvement and might be a useful functional food ingredient.
We investigated the effects of chronic administration of different extracts of ginger rhizome [pet ether extract (PE); toluene fraction (TF) of pet ether extract] on anxiety models: the elevated T-maze (ETM) (for inhibitory avoidance and escape measurements) and the open field test. Ondansetron (1 mg/kg), FE (10, 30 &100 mg/kg) and TF (10 & 30 mg/kg) were administered orally for 15 days. On the $14^{th}$ day mice were previously exposed for 30 min to one of the open arms of the T-maze, 24 h before the test. On $15^{th}$ day mice had two exposures to the enclosed and open arm of the ETM followed by exposure to the open field apparatus. The number of line crossings in the apparatus was used to assess locomotor changes. Cumulative Concentration Response Curve of 5-HT was plotted using rat fundus which were pretreated in a similar way. Treatment with Ondansetron (1 mg/kg), PE (100 mg/kg), TF (10 mg/kg) and TF (30 mg/kg) significantly (P<0.05) impaired inhibitory avoidance performance but did not impair escape latency. Concentration response curve of 5-HT was shifted towards the right with suppression of maxima in rats treated with PE and TF. The results suggest that PE and TF of Ginger rhizome exerts anxiolytic like behaviour in a specific subset of defensive behaviour, particularly those related to generalized anxiety disorder.
Long-term working memory (LTWM) is a subdivision concept of working memory and indicates the enhancement of performance in a working memory task. LTWM has been shown in humans who have been engaged in a specific task requiring working memory over a long time. However, there is very little understanding of the exact mechanism of LTWM because of limitations of experimental methods in human studies. We have modified the standard T-maze task, which is used to test working memory in mice, to demonstrate LTWM in an animal model. We observed an enhancement of performance by repeated experience with the same working memory load in mice, which can be regarded as an LTWM. This effect seems to depend on the condition wherein a delay was given. This task may be a good experimental protocol to assess LTWM in animal studies.

This study is intended to examine the tDCS and Morris Water maze training in Alzheimer's disease(AD) rats on Tau protein expression. Experiment groups were divided into four groups and assigned 16 rats to each group. Group I was a control group(AD induced by scopolamine); Group II was a experimental control group(AD injured by scopolamine and treatment tacrine); Group III was a group of tDCS application after AD injured by scopolamine; Group IV was a group of morris water maze training after AD injured by scopolamine. In cognition test, the outcome of group II was significantly lower than the groups(p<.001). and group III, IV were significantly low result at 14 days(p<.05). In histological finding, the experimental groups were destroy of micro vessels and finding of cell atropy and swelling. Group III, IV were decreased in degeneration of liver and kidney cells. In immuno- histochemistric response of BDNF and tau protein in hippocampus, BDNF expression of Group II was more increase than the other groups. and increase of BDNF expression was III, IV were higher than group I at 21 days. Tau protein expression of Group II was more decrease than the other groups. and decrease of Tau protein expression was III, IV were lower than group I at 21 days. These result suggest that improved tDCS and morris water maze training after scopolamine induced is associated with dynamically altered expression of BDNF and Tau protein in hippocampus and that is related with cognitive function.
