Serratia marcescens에서 글리옥실산이 Prodigiosin 생합성에 미치는 연구 (Studies on the Effect of Glyoxylate on the Biosynthesis of Prodigiosin in Serratia marcescens)

  • 최소 배지에 여러 아미노산과 대사 산물을 첨가하여 혐기성 조건하에서 배양시킨 Serratia marcescens ATCC 25419 세포 추출무에서 prodigiosin의 생합성을 조사한 결과 아미노산과 대사 산물들은 prodigiosin 생합성을 대조군보다 50-80% 정도 감소시켰다. 글리옥실산은 1-3mM에서 prodigiosin의 생합성을 대조군보다 20-40% 정도 증가시켰고, 특히 5mM의 농도에서는 최대 122% 증가시켰으나, 20-30mM에서 prodigiosin의 생합성을 50-90% 정도 감소시켰다. 한편, 혐기성과 호기성 조건의 피루브산과 $\alpha$-케토부티르산은 호기성 조건의 글리옥실산과 함께 조사한 모든 농도(0.5-30mM)에서 prodigiosin이 생성되지 않았으며 또한, 혐기성 조건하에서 높은 농도(20mM 이상)의 글리옥실산은 S. marcescens의 성장을 현저하게 억제시켰다. 이러한 결과들로부터 글리옥실산은 피루브산과 $\alpha$-케토부티르산과는 다르게 낮은 농도(1-5mM)에서는 S. marcescens의 1, 2차 대사물질로 작용하여 prodigiosin 생합성을 증가시키지만, 높은 온도(20mM 이상)에서는 S. marcescens의 성장을 억제시켜 2차 대사물질인 prodigiosin의 생합성도 억제된다고 사료된다.

Human collagen alpha-2 type I stimulates collagen synthesis, wound healing, and elastin production in normal human dermal fibroblasts (HDFs)

  • Skin aging appears to be the result of overlapping intrinsic (including genetic and hormonal factors) and extrinsic (external environment including chronic light exposure, chemicals, and toxins) processes. These factors cause decreases in the synthesis of collagen type I and elastin in fibroblasts and increases in the melanin in melanocytes. Collagen Type I is the most abundant type of collagen and is a major structural protein in human body tissues. In previous studies, many products containing collagen derived from land and marine animals as well as other sources have been used for a wide range of purposes in cosmetics and food. However, to our knowledge, the effects of human collagen-derived peptides on improvements in skin condition have not been investigated. Here we isolate and identify the domain of a human COL1A2-derived protein which promotes fibroblast cell proliferation and collagen type I synthesis. This human COL 1A2-derived peptide enhances wound healing and elastin production. Finally, the human collagen alpha-2 type I-derived peptide (SMM) ameliorates collagen type I synthesis, cell proliferation, cell migration, and elastin synthesis, supporting a significant anti-wrinkle effect. Collectively, these results demonstrate that human collagen alpha-2 type I-derived peptides is practically accessible in both cosmetics and food, with the goal of improving skin condition.

기상의 $NH_3$를 침전체로 사용하는 $BaTiO_{3}$ 분말의 합성 (The synthesis of $NH_3$ powder using gaseous $NH_3$ as precipitator)

  • The synthesis of high purity and ultra-fine $BaTiO_{3}$ by precipitation with gaseous $NH_{3}$ as precipitator was investigated to find an alternative process to solve various problems of present wet methods. This study consisted of two parts ; synthesis of $BaTiO_{3}$ precipitation with gaseous $NH_{3}$ and test of electrical property for the $La_{2}O_{3}$ doped $BaTiO_{3}$. The proper condition for the synthesis of $BaTiO_{3}$ by precipitation with gaseous $NH_{3}$ is as follows. The pH was 9.0. $H_{2}O_{2}$ mole ratio to $TiCl_{4}$ was 10. $NH_{3}$ gas follow rate did not influence the synthesis of $BaTiO_{3}$. The calcination temperature of $BaTiO_{3}$ was $300^{\circ}C$. Also, the synthesis of $La_{2}O_{3}$-doped $BaTiO_{3}$ was tested through the wet process. Under these condition, the shape of prepared $BaTiO_{3}$ powder was spherical type and the size of that was about $0.2{\mu}m$. After the powder was pressed, this green body was sintered at the $1300^{\circ}C$. Under these conditions, the water absorptance and the density of the obtained sintered body were below 0.04 %, 5.2 g/$cm^{3}$, respectively. Also the grain size of that was about $10{\mu}m$ and it was similar to commercial product.

열화학기상증착법에 의한 백금 기판 위의 그래핀 합성 (Graphene Synthesis on Pt Substrate using a Chemical Vapor Deposition Method)

  • Graphene is a carbon-based two dimensional honeycomb lattice with monoatomic thickness and has attracted much attention due to its superior mechanical, electronic, and physical properties. Here, we present a synthesis of high quality graphene on Pt substrate using a chemical vapor deposition (CVD). We optimized synthesis condition with various parameters such as synthesis temperature, time, and cooling rate. Based on the results, we concluded that graphene synthesis is driven by mainly carbon adsorption on surface rather than precipitation of carbon which is dominant in other metal substrate. In addition, Pt substrate can be repeatedly used several times with high quality graphene.

  • Nano-sized iron was synthesized using borohydride reduction of $Fe^{3+}$ in aqueous solution. A wide range of synthesis conditions including varying concentrations of reagents, reagent feeding rate, and solution pH was applied in an aqueous system under anaerobic condition. The reactivity of nano-sized iron from each synthesis was evaluated by reacting the iron with TCE in batch systems. Evidence obtained from this study suggest the reactivity of iron is strongly dependent on the synthesis solution pH. The iron reactivity increased as solution pH decreased. More rapid TCE reduction was observed for iron samples synthesized from higher initial $Fe^{3+}$ concentration, which resulted in lower solution pH during the synthesis reaction. Faster feeding of $BH_4^-$ solution to the $Fe^{3+}$ solution resulted in lower synthesis solution pH and the resultant iron samples gave higher TCE reduction rate. Lowering the pH of the solution after completion of the synthesis reaction significantly increased reactivity of iron. It is presumed that the increase in the reactivity of iron synthesized at lower pH is due to less precipitation of iron (hydr)oxides or less surface passivation of iron.

Synthesis of Sphene-pink Pigment under Various Firing Conditions

  • The present research was performed to determine the optimal firing condition and holding time for malayaite crystal, which is responsible for the stable pink-red coloration in glaze at high temperatures, using Cr$Cl_3$ as chromophore for the synthesis of $Cr_2O_3-SnO_2-CaO-SiO_2$ system pigments. The malayaite crystal was influenced by the raw materials used for synthesis, firing temperature, and holding time. Thus there are differences in the crystal phase and in the coloration according to the condition of synthesis. When Cr$Cl_3$ was used as chromophore, the pigment could be synthesized at lower temperatures, because Cr$Cl_3$ melts at $1500{^{\circ}C}$, which is much lower than the temperature at which $Cr_2O_3$ melts (higher than $2435{^{\circ}C}$). And the employed Cr ion showed a change in oxidation state. When a mineralizer was used to improve the employment of malayaite and the Cr ion, and the low temperature was maintained at which the malayaite crystal is produced, the production of malayaite crystal was promoted and the employment of chromophore was also promoted in the oxidation state of Cr (IV). The results of the experiment showed that the optimal firing condition was 18 h of holding time at $800{^{\circ}C}$, using Cr$Cl_3$ as chromophore, followed by 2 h at the raised temperature of $1150{^{\circ}C}$. The change in coloration of the Cr (IV) employed by malayaite showed a very rich color of red. Thus it was possible to effectively synthesize sphene-pink pigments with more red tint at a low temperature.

Brevibacterium flavum의 glutamate dehydrogenase결핍돌연변이주의 분리 및 특성 (Isolation and characterization of glutamate dehydrogenase defective mutant of brevibacterium flavum)

  • In order to understand the regulation of glutamate dehydrogenase(GDH) synthesis in Brevibacterium flavum, we have isolated a mutant lacking NADP-linked GDH activity by ethlmethane sulfonate treatment. The $gdh^-$ mutant was grown on the minimal plate with 1mM ammonium chloride and not that with 300mM ammonium chloride. The cell-free extracts from $gdh^-$ mutant and prototroph were also examined with glutamine synthetase(GS) and glutamate synthase (GOGAT) production by niteogen sources. The growth of $gdh^-$ mutant in presence of 20mM ammonium chloride means that GOGAT synthesis is sufficient to allow growth in this condition. GS production of $gdh^-$ mutant as well as parental strain was induced by 1mM urea and ammonium tartrate, but it was repressed by higher concentration of ammonia, and also induced by 20mM to 50mM glutamate as a substrate. It was special attention that GOGAT synthesis from $gdh^-$ strain was more repressed by higher concentration of ammonia than prototroph as described in E. coli system.

$H_{\infty}$ 제어법을 이용한 부유식 플랫폼의 동위치 제어 (Dynamic Positioning Control of Floating Platform using $H_{\infty}$ Control Method)

  • This paper presents a design method of dynamic positioning control system for floating platform with rotatable and retractable thruster using $H_{\infty}$ control technique. The norm band of uncertainty is captured by multiplicative perturbation between nominal model and reduced order model. A controller robust to the uncertainty is designed applying $H_{\infty}$ synthesis. The control law satisfying robust stabillity and nominal performance condition is determined through the mixed sensitivity approach. The evaluation for the resultant controller obtained by $H_{\infty}$ synthesis is done through simulations of the closed loop system. The results of $H_{\infty}$ synthesis are compared to those of the traditional LQ synthesis method.
