한국산 홍조식물 보라색 우무속(Symphyocladia)직물의 분류 (Taxonomy of the Genus Symphyocladia (Rhodophyta))

  • Four species of Symphyocladia growing along the coast of Korea were investigated in taxonomic point of view. Characters available for identification of the species were; difference in gross morphology, degree of cortication in vegetative axis, presence or absence of vegetative trichoblasts, and developmental pattern of stichidia. Vegetative structure and reproductive organs in developmental anatomy were similar among species. Branching pattern and occurrence of vegetative trichoblasts adopted for systematic criteria between Pterosiphonieae and Polysiphonieae, and between Symphyocladia and Pterosiphonia were no more confidential. The degree of congenital fusion of laterals with the parent axes was considered as only main character to distinguish Symphyocladia from Pterosiphonia.

A novel species Symphyocladia glabra sp. nov. (Rhodomelaceae, Rhodophyta) from Korea based on morphological and molecular analyses

  • Six species of the genus Symphyocladia are currently recognized worldwide, all of which are reported to grow on the Korean coast. We described a new species Symphyocladia glabra sp. nov. based on morphological and molecular evidence. The new species is characterized by mostly an erect and broad thallus, the parallel arrangement of numerous apical cells, completely fused congenital cells, a corticated basal portion of faint midrib with six to eight pericentral cells, rhizoids cutting off from pericentral cells, and absent vegetative trichoblasts. The new species has morphological similarity to S. marchantioides and S. jejuinsula. However, S. glabra sp. nov. is distinguished from S. marchantioides by the corticated lower portion of the thallus and the absence of vegetative trichoblasts, and S. jejuinsula by number of pericentral cells and the fact that the thallus does not taper upward. The phylogeny of rbcL sequences indicated that S. glabra sp. nov. is definitely a separate entity within the genus Symphyocladia.

해조류 추출물의 라디칼 소거활성 검색과 보라우무의 항산화활성 성분

  • The antioxidant activities of methanol and dichloromethane/acetone extracts of twenty three seaweeds were tested by using 1,1-Diphenyl-2-picryl-hydrazyl(DPPH) at a $100{\mu}g/ml$ concentrations. The dichloromethane/acetone extracts of three seaweeds(Symphyocladia latiuscula, Gloiopoltis furcata, Sagassum thunbergii), were found to be most effective in DPPH radical scavenging activity. The DPPH radical scavenging effect of these seaweeds was Symphyocladia latiuscula(85.82%), Gloiopoltis furcata(82.83%), Sagassum thunbergii(74.05%) in order. These seaweeds were evaluated using the pyrogallol UV-VIS spectrophotometeric method to generate superoxide anion. Among them, the methanol extracts of six seaweeds were showed weak superoxide dismutase-like activities. The dichloromethane/acetone extracts obtained from Symphyocladia latiuscula was fractionated with $CH_2Cl_2$, n-hexane, 15% aq.MeOH, n-BuOH, $H_2O$. The 15% aq.MeOH soluble fraction exhibiting the strongest antioxidative activities was further purified by C18 column flash chromatography and reversed HPLC. The two active principles of Symphyocladia latiuscula were isolated and characterized as 2,3,6-tribromo-4,5-dihydroxybenzyl alcohol(1), 2,3,6-tribromo-4,5-dihydroxybenzyl methyl ether(2).

  • 홍조 참보라색우무(Symphyocladia latiuscula (Harvey) Yamada)의 형태적 식별형질들을 야외 및 실내배양 재료를 통해 검토하고 생활사를 밝혔다. 본 종은 엽체의 체장이 5-15 cm이고, 정단을 제외한 전 엽체에 피층이 발달하며, 주축과 측지의 유합정도는 5-7 절편에 달한다는 점에서 Symphyocladia속의 다른 종들과 구분된다. 생식기관의 형태와 발달과정은 속내의 다른 종들과 구분된다. 생식기관의 형태와 발달과정은 속내의 다른 종들과 유사하나 소측지의 변형에 의해 장상의 사분포자탁을 형성하는 것이 특징적이다. 본 종은 야외 및 실내배양 재료 모두에서 영양모상엽이 형성되어 S. pennata와 구분되었고, 따라서 지금까지 polysiphonieae족과 Pterosiphonieae족을 구별하기 위한 기본 식별형질로서 인식되어 왔던 영양모상엽의 존재 유무는 부류학적인 의의가 없음을 확인되었다. 실내배양에서 본 종의 Polysiphonia형 생활사를 반복하나 배우체의 체장이 포자체보다 상재적으로 작아 세대간 크기의 차이가 현저한 이형현상이 관찰되었다. 본 종은 한국 전 해안의 조간대 중부 이하의 지역에서 전 계절에 걸쳐 흔히 채집되며, 하계에 배우체의 출현비율이 높았다.

Antioxidant Effects of 2,3,6-tribromo-4,5-dihydroxybenzyl Methyl Ether (TDB) from the Red Alga, Symphyocladia latiuscula

  • 2,3,6-Tribromo-4,5-dihydroxybenzyl methyl ether (TDB) from the methanolic extract of the red alga Symphyocladia latiuscula exhibits major antioxidant activity. In this study, the activity of TDB against oxidative damage in deoxyribose and DNA was investigated in vitro for potential applications in preventing mutagenesis caused by DNA damage. TDB inhibited the oxidation of deoxyribose at concentrations of up to $1{\mu}g$/mL in the presence of $Fe^{+3}$-EDTA/$H_2O_2$. Furthermore, TDB showed no pro oxidant activity as determined by absence of the reduction of bleomycin-$Fe^{+3}$ to bleomycin-$Fe^{+2}$, which leads to DNA damage. Based on these results, TDB demonstrated considerable antioxidant activity without prooxidant properties.

Antioxidant Activity of 2,3,6-Tribromo-4,5-dihydroxy benzyl methyl ether from Symphyocladia latiuscula

  • Antioxidant activity of a methanol extract of Symphyocladia latiuscula was evaluated by the thiocyanate method in the linoleic acid system. The methanol extract inhibited the peroxidation of linoleic acid in a dose-dependent manner. The MeOH extract was then sequentially partitioned with n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH and H20. The antioxidant activity of the fractions increased in order of $CH_2Cl_2$, n-hexane, EtOAc, and n-BuOH. There was no activity found in $H_2O$ partitoned fraction by the thiocyanate method. Especially, the activities of the fractions of n-hexane and $CH_2Cl_2$ were comparable to that of 2,6-di-tert­butylhydroxytoluene (BHT). Column chromatography of the $CH_2Cl_2$ fraction over silica gel yielded cholesterol (1) and 2,3,6-tribromo 4,5-dihydroxybenzyl methyl ether (2) which were identified by instrumental analysis of MS and $^1H-$ and $^{13}C-NMR$. The latter (2) demonstrated significant antioxidant activity.

Nitrite Scavenging Activity of Bromophenol Congeners from Symphyocladia latiuscula

Column chromatography of the most active $CH_2Cl_2$ fraction over silica gel yielded three active bromophenol congeners (1-3) which were identified as (2R)-2-(2,3,6-tribromo 4,5-dihydro­xybenzyl) cyclohexanone (1), 2,3,6-tribromo 4,5-dihydroxybenzyl methyl ether (2), and 2,3,6­tribromo 4,5-dihydroxybenzyl alcohol (3) respectively.

보라우무 (Symphyocladia latiuscula) 추출물의 항산화활성 (The Antioxidant Activity in Extracts of Symphyocladia latiuscula)

  • 일본, 따렌 등에 분포되어 있고 우리나라에서 동해안, 서해안, 남해에서 흔히 볼 수 있는 해조류로서, 조간대의 바위위에서 서식하는 빨강 검둥이과 홍조류인 보라우무 (Symphyocladia latiuscula)를 대상으로 하여 안정한 free radical인 1,1-diphenyl-2-picrylhydrazyl (DPPH)를 사용하여 그 소거능을 측정하고, 또한 생체내의 과산화지질을 평가하는 방법으로 과산화지질 분해산물인 malondialdehyde를 thiobarbituric acid(TBA)로 비색정량하는 방법과 Hepatocyte에 의해 생성되는 free radical 생성량 측정 등의 다양한 방법을 통하여 항산화능을 측정하였다. DPPH radical을 이용한 항산화활성 검색에서 강력한 radical 소거 효과를 나타내었다. 각 용매 분획별로 항산화활성을 살펴본 결과 수층을 제외한 모든 분획들에서 농도 의존적으로 항산화활성이 나타났다. 특히 dichloromethane과 hexane 가용성 분획의 항산화활성이 가장 크게 나타났다. 지질과산화 억제 효과에서도 유사한 결과를 보였으며 dichloromethane 가용성 분획의 항산화활성이 가장 높게 나타났으며 L-ascorbic acid의 항산화활성과 비교하여 더 높은 활성을 나타내었다. Free radical 생성 억제 효과에서도 dichloromethane 가용성 분획의 경우 control과 비교하여 0.1 mg/ml에서 $50\%$ 이상의 저해효과를 나타내었다.

Antioxidant, anti-inflammatory, and antibacterial activities of a 70% ethanol-Symphyocladia linearis extract

  • Research on the potential biological activity of red alga Symphyocladia spp. has been limited to Symphyocladia latiuscula, which is widely used as a food ingredient in Korea. Here, we examined the biological activity of another species, Symphyocladia linearis, which is found in Korea and was reported as a new species in 2013. The aim of this study was to evaluate the antioxidant, anti-inflammatory, and antibacterial properties of a 70% ethanol extract of S. linearis. Antioxidant activity, which was evaluated using radical scavenging assays, revealed half maximal inhibitory concentration values for 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) of 34.57 and 11.70 ㎍/mL algal extract, respectively. Anti-inflammatory activity of the S. linearis ethanolic extract was evaluated using RAW 264.7 cells by measuring the inhibition of lipopolysaccharide-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production. The potential cytotoxicity of NO and PGE2 was first examined, confirming no toxicity at concentrations ranging from 10-100 ㎍/mL. NO production was inhibited 61.1% and 78.0% at 50 and 100 ㎍/mL S. linearis extract, respectively; and PGE2 production was inhibited 69.1%, 83.2%, and 94.8% at 25, 50, and 100 ㎍/mL S. linearis extract, respectively. Thus, the S. linearis extract showed very strong efficacy against PGE2 production. The cellular production of reactive oxygen species, measured using 2',7'-dichlorofluorescin diacetate fluorescence, was inhibited 48.8% by the addition of 100 ㎍/mL S. linearis extract. Antibacterial activity was evaluated using the disc diffusion method and minimum inhibitory concentration (MIC). S. linearis was effective only against gram-positive bacteria, exhibiting antibacterial activity against Staphylococcus aureus with a MIC of 256 ㎍/mL extract and against Bacillus cereus with a MIC of 1,024 ㎍/mL extract. Based on these results, we infer that a 70% ethanolic extract of S. linearis possesses strong anti-inflammatory properties, and therefore has the potential to be used in the prevention and treatment of inflammatory and immune diseases.
