Effects of Flushing, Preservation and Reperfusion in the Canine Transplanted Lung Tissue (관류, 보존 및 재관류 과정이 이식된 개의 폐조직에 미치는 영향)

  • Background: Due to the paucity of suitable donor organs for lung allotransplantation, a number of techniques have been developed to improve the lung preservation. Ultrastructural studies of the morphologic changes of the flushing, preservation and reperfusion injury in donor lungs have rarely been reported. Methods: Adult dogs (n=46) were matched as donors and recipients for the single lung transplantation. The donor lungs were preserved after flushing with preservation solution and transplanted after 20-hours of preservation at $10^{\circ}C$. Ultrastructural features of the lung were examined after flushing, preservation and 2 hours after lung transplantation (reperfusion) respectively. Results: Electron microscopy after flushing showed focal alveolar collapse and mild swelling of type I epithelial cells. After preservation both type I epithelial cells and endothelial cells were swollen and destroyed focally. The endothelial cells showed protrusion of tactile-like structures into the lumina, blebs or vacuoles of the cytoplasm After reperfusion the lung tissue showed fibrin material in the alveoli, prominent type I epithelial cell swelling with fragmented cytoplasmic debris and marked endothelial cell swelling with vacuoles or tactile-like projections. The alveolar macrophages showed active phagocytosis. Scanning electron microscopic examination of the pulmonary parenchyma showed focally alveolar collapse and focal consolidation after the preservation and more prominent changes after the reperfusion procedure. The lungs preserved with low potassium dextran glucose solution, with additional prostaglandin $E_1(PGE_1)$ and verapamil(VP) showed relatively well preserved ultrastructures compared with those which were preserved with modified Euro-Collins or University of Wisconsin, and with additional $PGE_1$ and/or VP. Conclusion: The ultrastructural changes associated with flushing were mild in severity, the donor lungs were injured during the preservation, and further damage was occurred during the reperfusion. The reperfusion injury resulted in prominent pulmonary parenchymal alterations with a pattern of acute lung injury.

  • A 19-year-old man was referred to Seoul National University Dental Hospital for evaluation of a large painless swelling of the left mandibular angle area in August, 1999. The growth had been first noted 6 years ago. He had visited other hospital in 1997. In spite of the treatment given at the hospital, the mass continued to grow rapidly. Conventional radiographs in 1999 showed an expansile, lobulated, and destructive lesion of the left mandibular body. CT scan demonstrated an expansile mass with a corticated margin. Bony septa were seen within the lesion. Internal calcification noted on the bone-setting CT image, and corresponded to the hypointense area in T1-weighted MRI image. MRI clearly delineated the extent of the lesion which had heterogenous intermediate signal intensity in T1-weighted images and heterogenous hyperintense signal intensity in T2-weighted images. The lesion was well-enhanced. Histopathologically, the lesion was well demarcated. Multinucleated giant cells were presented in a fibrous background, demonstrating a storiform pattern. Areas of osteoid rimmed by a few osteoblasts were scattered throughout the lesion. Inflammatory cells, blood vessels, and hemosiderin deposition were also shown. CGCG may show lots of internal calcification foci on the CT, and varied signal intensity in MRI. More cases will be needed to understand the features of the CT & MR finding of CGCG.

  • The physicochemical properties of damaged hair by irradiation of ultraviolet-B (UV-B) have been investigated by using transmission electron microscope and scanning electron microscope. The range of irradiation of hair irradiated for expectative 6 hours, 12 hours, 24 hours and 48 hours with stimulated ultraviolet ray. The treated hairs showed characteristic morphological damage pattern in the cornified cell of matrix and the cuticle following time past. The various sized vacuoles in the endocuticle of the cuticular cells was formed. The statistically significant differences in diameter of cuticular cell were observed in terms of tranverse swelling by formation of vacuoles. The hair cortex and matrix undergo long term exposure to UV-B radiation. The macrofibrils of cortex appeared to be affected most by UV-B, although the morphology and volume of melanin granule was not changed. The physicochemical destruction of hair matrix and cuticular cells is largely accelerated by long term irradiation of UV-B.

  • Adenomatoid odontogenic tumors represent 3 to 7 percent of all odontogenic tumors. These tumors are more common in the maxilla than the mandible and usually include the anterior region. Clinically, the most common symptom is painless swelling and the tumor is associated with an unerupted tooth, typically a maxillary or mandibular cuspid. The adenomatoid odontogenic tumor appears radiographically as a unilocular radiolucency around the crown of an impacted tooth, resembling a dentigerous cyst. More often, it contains fine calcifications. Histopathologically, there is a thick wall cystic structure with a prominent intraluminal proliferation of the odontogenic epithelium. The most striking pattern is varying-sized solid nodules of spindle-shaped or cuboidal epithelial cells forming nests or rosette-like structures with minimal stromal connective tissues. Conspicuous within the cellular areas are structures of tubular or duct-like appearance. The duct-like spaces are lined with a single row of cuboidal or low columnar epithelial cells, of which the ovoid nuclei are polarized away from the luminal surface. Small foci of calcification may also be scattered throughout the tumor. These have been interpreted as abortive enamel formations. In some adenomatoid odontogenic tumors, the material has been interpreted as dentoid or cementum.

Characteristics of Sweet Potato Powders from Eight Korean Varieties (한국산 8 품종 고구마분말의 특성)

  • Sweet potato powders made from eight Korean varieties, including purple-fleshed, orange-fleshed, and commercial dry type sweet potatoes, were investigated for physicochemical and pasting properties to develop processed food. Crude protein and lipid contents of Shinjami and Borami were higher than those of other varieties. The lightness value of raw sweet potato flesh was the highest value in Shinchunmi, and the lowest in Shinjami. Using the color difference (${\Delta}E$), color similarities compared to the white plate occurred in the following order; purple-fleshed > orange-fleshed > commercial dry type sweet potatoes. Total and damaged starch contents were significantly different (p<0.05). Total starch content of sweet potatoes was higher in commercial dry sweet potatoes (61.89-70.46%), particularly Shinchunmi (70.46%) but lower in orange-fleshed sweet potato (48.87 and 49.53%, respectively). Water binding capacity of Yeonwhangmi, swelling power and solubility of Shinyulmi were the highest values (174.70, 25.54 and 87.49%, respectively) among them (p<0.05). But oil absorptions of Shinyulmi and Shinchunmi showed lower values (97.08 and 97.54%, respectively). All sweet potato powders had an A type x-ray diffraction pattern. The initial pasting temperatures of sweet potato powders ranged from 69.50 to $75.95^{\circ}C$ and the amylolytic enzyme in sweet potato powder lowered pasting viscosity.

Studies on the Properties of lntracelluar Arsenic Binding Substances in the Rat Liver (흰쥐 간조직의 세포내 비소결합물질의 특성에 관한 연구)

  • Sodium arsenite (As) aqueous solution containing 4 ppm wss given to male rats for 15 days as drinking water. Electrophoretic pattern of liver cytosol from As-fed rats appeared to be significantly different from that of the control rats. Although the normal protein content of the cytosol fraction of As-fed rat liver was decreased, 8 stress proteins were increased. In liver cytosol fractions of As-treated rat, one kind of arsenic-binding substance (AsBS) was observed. Molecular weight of AsBS was identified to be 500 D and composition of amino acid was glycine, glutamic acid and cysteine. Glutathione (GSH) appeared to bind to arsenic and GSH-As complex showed the same mobility as AsBS on gel filtration chromatography. GSH conjugated As prevented As from inhibiting respiration, conformational change and swelling-contration of mitochondria. According to the above results. it is concluded that in vfuo treated arsenic stimulated synthesis of stress protein, and arsenic-binding substance might be glutathione and have a protective role against arsenic toxicity.

Premenstrual Discomforts and Coping in University Students (여대생의 월경전 불편감과 대처에 관한 연구)

  • Purpose: This study was carried out to identify premenstrual discomforts and coping patterns and their effects. Method: The participants of this study were 297 female university students in C area. The Menstrual Discomfort Questionaire (MDQ) and coping method lists were used as measurement tools. Results: There were significant differences premenstrual discomforts according to age (F=5.76, p=.003) and according to health condition (F=3.43, p=.034). The mean scores of the sub-categorical factors of premenstrual discomfort were 2.35 points for pain, 2.29 for instability and 2.25 for water retention. The worst symptoms among the subcategories of premenstrual discomfort were as follows: backache (M=2.68) in the pain subcategory, irritability (M=2.53) in the instability subcategory and swelling(M=2.40) in the water retention subcategory. Their common coping patterns were 'coping according to menstrual period', 'active behavioral coping' and 'evasional coping'. Frequently used coping methods were 'taking a rest and sleep (99.3%)' and 'taking a warm shower (86.2%)'. Effective coping methods were 'taking a rest and sleep (89.5%)' and 'taking a warm shower (87.1%)'. Conclusion: Most participants have their own coping pattern. Some methods were very effective to PMS but some were not effective to PMS. To manage PMS, effective coping methods should be encouraged for female university students. To keep up with effective coping, education and counseling should be continued. It is considered necessary to make the same research with a larger number of samples and more specified assessment.

A CASE REPORT OF ANEURYSMAL BONE CYST OF THE MANDIBULAR CONDYLE (하악과두에 발생한 동맥류성골낭)

  • The aneurysmal bone cyst is a nonmalignant reactive bone lesion. Developing rarely in the craniofacial region, and more commonly affecting the long bones and the spine, the lesion has variable etiopathogenic characteristics. The authors diagnosed a 33-year-old female as aneurysmal bone cyst after undergoing clinical, radiological and histological examinations. The characteristics were as followed: 1. The patient complained of pain and swelling of the right preauricular area. 2. The conventional radiograms showed a relatively well defined radiolucent lesion with partially scalloping margin. The cortical bone of the right condyle was thinned and expanded by the lesion. 3. Bone scintigraphy with ~c demonstrated ring-like or doughnut-pattern accumulation of radioactivity. 4. On Tl-weighted imaging of MRI, the lesion on the right condyle had middle signal intensity. T2-weighted MRI demonstrated multiple high signal intensities seperated by septa which had low signal intensity. Finger in balloon appearance was seen. 5. Histologically, the lesion was composed of large sinusoidal blood spaces lined by fibroblasts and histiocytes. Its fibrous stroma consisted of fibroblstic element, multinucleated giant cells, extravasated erythrocytes and focal hemosiderin pigmentation. New bone formation was also observed around larger sinusoidal spaces.

Physicochemical Properties of Sweet Potato Starch Reclaimed from Sweet Potato Processing Sludge (고구마 가공 슬러지로부터 회수된 고구마 전분의 물리화학적 특성)

  • The physicochemical properties of sweet potato (SP) starches reclaimed from an SP-processing sludge without freezing (RC/NF) and with freezing (RC/FR) were investigated. Lab-isolated (LI) SP starch, as a control, were prepared from raw SP. RC/NF and RC/FR SP starches were recovered from SP-processing sludges by the repeated sieving and washing procedure. The total starch contents and amylopectin branch-chain distributions did not differ for three SP starches. Relative to LI and RC/NF SP starches (possessing similar physicochemical characteristics), the apparent amylose and phosphorus contents, swelling factor, and pasting viscosity were reduced for RC/FR SP starch. However, the freezing treatment altered X-ray diffraction pattern (at $5.5^{\circ}$, $11-12^{\circ}$, and $24^{\circ}$ $2{\theta}$) of RC/FR SP starch, which likely increased its gelatinization peak and completion temperatures. Its amorphous region in total diffractogram was reduced, resulting in the enhanced relative crystallinity. Overall results suggested SP starches recovered from an SP-processing sludge would have the potential to replace commercial SP starch products.

Physicochemical Characteristics of Starches in Rice Cultivars of Diverse Amylose Contents

  • Through the sampling four rice cultivars with differing amylose contents, the relationship between the structural and gelatinization properties of endosperm starches was analyzed. These rice varieties exhibited different chain length distribution ratio within the amylopectin cluster as well as varing amylose levels. The proportion of amylopectin short chains of in Goami cutlivars was higher than the other varieties, whereas the Goami 2 which shows amylose extender mutant properties in the endosperm showed the highest proportion of long chains. In X-ray diffraction analysis of rice starches, the Goami 2 variety displayed a B-type pattern whereas the other varieties were all A-type. Among the cultivars with high and normal rice starch levels, those with the higher amylose contents showed distinctly lower swelling. Goami 2 rice was found to have the highest onset and peak gelatinization temperature from the differential scanning calorimetry results. The four rice cultivars under analysis also showed different rates of hydrolysis by amyloglucosidase. These findings suggest that the composition and chemical structure of the starch content is a major determinant of both the gelatinization and functional properties of rice.
