Association Between Genetic Polymorphism of XRCC1 Gene and Risk of Glioma in а Chinese Population

  • Background: Gliomas are the most common type of primary brain tumor in adults, and the X-ray repair complementing group 1 gene (XRCC1) is an important candidate gene influencing its risk. The objective of this study was to detect the influence of XRCC1 genetic polymorphisms on glioma risk. Materials and Methods: A total of 629 glioma patients and 641 cancer-free subjects were enrolled in this case-control study. The genotypes of the c.1471G>A genetic polymorphism were determined by created restriction site-polymerase chain reaction (CRS-PCR) and DNA sequencing methods. The influence of the XRCC1 genetic polymorphism on glioma risk was evaluated by association analysis. Results: Our data indicated that the alleles/genotype of this genetic variant was statistically associated with glioma risk. The AA genotype was statistically associated with the increased risk of glioma compared to the GG wild genotype (odds ratios (OR) = 1.89, 95% CI 1.25-2.87, P = 0.003). The allele-A may contribute to increased the susceptibility to glioma (OR = 1.23, 95% CI 1.04-1.46, P = 0.017). Conclusions: These preliminary findings indicate that the c.1471G>A genetic polymorphism of XRCC1 has the potential to influence glioma susceptibility, and might be used as molecular marker for assessing glioma risk.

딸기 흰가루병 저항성 계통 선발을 위한 분자마커 개발 (Development of Cleaved Amplified Polymorphic Sequence (CAPS) Marker for Selecting Powdery Mildew-Resistance Line in Strawberry (Fragaria×ananassa Duchesne))

  • 딸기 흰가루병은 Podosphaera aphanis에 의해 발병되며 수확기에 가장 큰 피해를 주는 병으로 현재 유황, 농약으로 주로 방제 되고 있는 실정이다. 본 연구에서는 딸기 흰가루병 저항성 품종 육성을 위한 흰가루병 저항성 특이마커 개발로 내병성 육종효율을 높이고자 하였다. 흰가루병 저항성 계통 선발을 위한 분자마커를 개발하기 위해 아키히메${\times}$설향 집단을 대상으로 자가수분을 통해 후대 양성 후 병저항성을 검정하였다. 마커분석은 RAPD primer 200 세트 중 OPE10 331bp에서부터 흰가루병 저항성 특이 마커 선발하였다. 흰가루병 저항성 특이밴드만 선발하기 위하여 클로닝 후 유전자정보 분석하여 SP1F/R의 Primer를 제작하였다. 그러나 SP1F/R을 이용하여 PCR한 결과 저항성, 감수성간에 다형성이 확인되지 않아 염기서열을 정렬한 후 SNP, In/del의 다형성 유무를 확인한 결과 6개의 SNP를 확인하였다. 이들 PCR 산물을 해당 사이트와 연관된 제한효소로 절단한 결과 그 중 Eae I(Y/GGCCR)의 절단으로 231bp 위치에서 저항성과 감수성간의 다형성을 확인함으로써 흰가루병 저항성 계통선발을 위한 분자마커를 선발하였다. 이러한 과정을 통해 딸기 흰가루병 저항성 품종 육성을 위한 MAS(marker assisted selection) 체계 확립으로 내병성 육종효율 증진에 기여를 할 수 있을 것으로 기대된다.

Monitoring antimalarial drug-resistance markers in Somalia

  • The use of an effective antimalarial drug is the cornerstone of malaria control. However, the development and spread of resistant Plasmodium falciparum strains have placed the global eradication of malaria in serious jeopardy. Molecular marker analysis constitutes the hallmark of the monitoring of Plasmodium drug-resistance. This study included 96 P. falciparum PCR-positive samples from southern Somalia. The P. falciparum chloroquine resistance transporter gene had high frequencies of K76T, A220S, Q271E, N326S, and R371I point mutations. The N86Y and Y184F mutant alleles of the P. falciparum multidrug resistance 1 gene were present in 84.7 and 62.4% of the isolates, respectively. No mutation was found in the P. falciparum Kelch-13 gene. This study revealed that chloroquine resistance markers are present at high frequencies, while the parasite remains sensitive to artemisinin (ART). The continuous monitoring of ART-resistant markers and in vitro susceptibility testing are strongly recommended to track resistant strains in real time.

  • In this study, biomonitoring methods were developed to measure BTEXs exposure level in the air, metabolites of benzene and toluene in human urine, individual susceptibility markers in human blood for evaluation of the health effects about environmental pollution. We have also performed a small-scaled molecular epidemiology study on residents in Chuncheon and workers in workplace for these method applications. The workers in workplace were surveyed as study areas, and the residents in Chuncheon which is in the suburban area were surveyed as comparative areas in this study. Actually, 31 workers in as target group and 33 residences in as control group this epidemiological study. The results obtained from this study were as follows: 1. Benzene is a well-known carcinogen, it's median concentrations were 0.00024∼0.02057ppm at suburban area and 0.002∼00.654ppm at work place, These benzene concentrations were not exceed the OSHA(Occupational Safety and Health Administration) threshold benzene level of 1ppm in the states. 2. Metabolites product of benzene(t,t-Muconic Acid) and toluene(Hippuric Acid) were not significant both in suburban and workplace area. The median concentration of t,t-MA and HA were 0.0122, 1.44277g/g creatinine, respectively. 3. In the case of individual susceptibility markers as CYPlAl, 41.8% of them has homozygous wild type(W) and who has heterozygous variant type(H) was 35.4% and 22.8% of homozygous variant type(M) genetic type. In the case of CYP2E1, 62.82% of them has homozygous wild type(D) type, 34.62% of each has heterozygous variant type (DC) and 2.56% of them has homozygous variant type (CC). Who doesn't have GSTM1 gene was 46.25% and who has GSTM1 gene was 53.75%. Who doesn't have GSTT1 gene was 40.0% in study groups and who has GSTT1 gene was 60.0%. Who has W genetic type, which is homozygous wild type of GSTP1, was 69.18% and H genetic type, which is heterozygous variant type was 28.4%. M genetic type which is homozygous variant type was 2.4%. 4. Concentration differences of metabolites such as t,t-MA and HA in urine, which is generated by individual susceptibility marker of GSTM1, GSTT1, GSTP1 gene of Phase I and CYP1A1, CYP2E1 gene of Phase II, was examined. As a result, GSTP1 and GSTM1 indicate slight differences depend on the amount of metabolites in urine, it was not statistically significant.

사상체질에 따른 류마티스 관절염의 위해요인 및 TNF-${\alpha}$(G308A) 유전자 다형성간의 연관성 연구 (A Study of TNF-${\alpha}$(G308A) Genetic Polymorphism and Risk Factors Associated with Rheumatoid Arthritis by Sasang Constitution)

  • Objectives: The purpose of this study is to examine the risk factors and the genetic polymorphism of TNF-alpha associated with rheumatoid arthritis by Sasang constitution Methods : This study was planned to detect the susceptibility of the patients diagnosed by rheumatoid arthritis to Sasang Constitution and to examine the risk factor such as life style and environmental stress (smoking, environmental tobacco smoke, alcohol intake and so on). The genetic polymorphism of TNF-alpha (G308A) were analyzed by PCR-RFLP in rheumatoid arthritis patients and controls. Rheumatoid arthritis patients and matched controls are assessed with QSCCII question for Sasang Typology. Then the genetic polymorphism of patients by Sasang constitution are compared to those of control, which are statistically analyzed and adjusted by age, sex, smoking status, alcohol intake, BMI, and econocmic status. Results: Differential effect of passive smoking on the association between Sasang constitution and rheumatoid arthritis risk was found. This study showed that the genetic polymorphism (TNF-${\alpha}$(G308A)) of rheumatoid arthritis patients and controls associated with the susceptibility to rheumatoid arthritis by sasang constitution was analyzed. Differential effects of TNF-${\alpha}$(G308) genetic polymorphism on the association between rheumatoid arthritis risk and Sasang constitution were found. Conclusion : It is suggested that the genetic polymorphism correlated with susceptibility to rheumatoid arthritis by specific sasang constitution used as its susceptibility marker and further as basic data to prevent the risk factors for rheumatoid arthritis. But larger studies will be needed to confirm these preliminary findings.

토마토 황화잎말림바이러스(TYLCV) 저항성 품종 선발 및 원예특성 분석 (Selection and Characterization of Horticultural Traits of Tomato leaf curl virus (TYLCV)-resistant Tomato Cultivars)

  • 본 연구는 외국 종자 회사의 토마토 $F_1$ 품종들을 대상으로 토마토황화잎말림바이러스(TYLCV) 저항성 분자마커 분석과 포장병리검정을 통해 각 품종의 저항성 유전자형과 내병성 수준을 분석하고, 원예형질 특성평가를 통해 TYLCV 저항성 품종보급과 분리육종에 적절한 품종선발을 위해 수행되었다. 40개 공시품종의 분자마커 및 병리검정 결과, 대부분 저항성으로 표기된 품종들이 TYLCV 저항성 유전자인 Ty-1, Ty-3, 또는 Ty-3a유전자를 이형접합 또는 동형접합으로 지니고 있으며, 표현형에서도 매우 낮은 발병률을 보였다. 반면, 중간 저항성으로 표기된 4 품종 중 3 품종은 18.1-33.3%의 발병률을 보였으며, 마커형이 이병성이었다. 내병성이 확인된 $F_1$품종들을 대상으로 원예형질 특성조사 결과, 유럽형 TYLCV 저항성 대과종 품종은 국내 선호 대비종보다 비교적 수량이 높고 당도 및 당산도도 크게 떨어지지 않아 국내용 품종 육성재료로 적합하였지만, 높은 과실경도의 문제점이 있었다. 반면, 소과종 품종들은 수량, 당도 등에서 국내 선호 품종보다 낮고 절간장도 길어 이들을 활용한 고품질 TYLCV 저항성 품종 육성을 위해서는 대과종에 비해 많은 시간과 노력이 소요될 것으로 판단되었다.

멜론 흰가루병의 race 분화 및 저항성 계통 선발을 위한 분자마커 개발 (Development of molecular marker to select resistant lines and to differentiate the races related to powdery mildew in melon (Cucumis melo L.))

  • Powdery mildew (Podosphaera xanthii) commonly occurs in cultivated fields of melon (Cucumis melo L.). It inflicts a lot of damages. Therefore, breeding resistant lines is essential. Development of a resistant line by integrating resistance gene takes a long time. In addition, break down of developed resistance by generating new virulent fungus strains increases disease susceptibility. This phenomenon was related to races of powdery mildew. Therefore, it is important to develop a DNA marker to genetically analyze race-specific resistance genes of melon powdery mildew to breed resistant lines. To date, a total of 28 races of Podosphaera xanthii have been reported in the literature. In Japan, 10 races have been reported in the Ibaraki region. We developed a system to characterize the races of Podosphaera xanthii and confirmed eight out of those 10 races in the Ibaraki region. In Korea, only one race has been characterized to date. However, some different races were detected. Through genetic analysis of resistant lines and susceptible lines of powdery mildew, resistance genes of race1 (Pm-X, PXB, and Pm-R 1), race N1 (PXA), race 2 (Pm-w and Pm-R 2), race 3 (Pm-X3), and race 5 (Pm-X5 and Pm-R5) were identified in melon. These related genes of race 1, 3, N1, 5, and race 1, 2, 5 were located at linkage group II and V, respectively. In race 1, resistance gene was located in the linkage group XII. In addition, each race-specific marker related to specific resistance gene was developed. Using race information and race selection system obtained in this study, resistant line can be bred to develop resistant cultivar for several areas. Furthermore, this will make it more easily and economically to breed resistant lines by using selected markers.

완도해역 해수에서 분리한 장염비브리오(Vibrio parahaemolyticus)의 항균제 내성 및 병원성 유전자의 특징 (Antimicrobial-resistance Profiles and Virulence Genes of Vibrio parahaemolyticus Isolated from Seawater in the Wando Area)

  • Sixty-seven Vibrio parahaemolyticus isolates from surface seawater from the Wando area, on the southern coast of Korea, were analyzed for their susceptibility to 15 different antimicrobials and the presence of virulence genes. According to the disk diffusion susceptibility test, all of the strains studied were resistant to ampicillin and oxacillin, while decreasing percentages were resistant to vancomycin (64.2%), streptomycin (56.7%), amikacin (31.3%), kanamycin (22.3%), cephalothin (20.9%), erythromycin (10.4%), ciprofloxacin (4.5%), and tetracycline (3.0%). All of the strains were susceptible to five antimicrobials: chloramphenicol, gentamycin, nalidixic acid, sulfamethoxazole/trimethoprim, and trimethoprim. Fifty-nine isolates (88.1%) were resistant to three or more classes of antimicrobial and defined as multidrug resistant, and two strains were resistant to seven antimicrobial agents. The minimum inhibitory concentration (MIC) of the 67 V. parahaemolyticus isolates to ampicillin and oxacillin ranged from 512-2,048 and $64-512{\mu}g/mL$, respectively. All 67 isolates were also examined for the presence of the tdh and trh virulence genes using the polymerase chain reaction (PCR). However, no isolates possessed either tdh or trh. The VPA0477 (${\beta}$-lactamase) gene, present in all of the tested strains, was validated as a new specific marker gene in PCR assays for the accurate detection and identification of V. parahaemolyticus.

사상인(四象人)의 생리적 특성 연구 (Psychological, Physical and Genetic Traits of Sasang Typology)

  • The major concept of Sasang typology is that the disease susceptibility and drug response as well as physiological characteristics are presumed to be different depending on their Sasang types. Although characterizing fundamental basis of their traits are crucial in this research field, only pathological susceptibility and physical appearances were thoroughly studied. We evaluated their physiological characteristics by tapping psychological, physical and genetic traits of each Sasang types. After determining the Sasang type of one hundred three college students based on the Questionnaire for the Sasang Constitution Classification, the psychological, physical and genetic traits of each type were analyzed with the Myers-Briggs Type Indicator (MBTI), Bioelectrical Impedance Analysis and genetic polymorphism test, respectively. Each of the Sasang types showed significantly different profiles (Generalized estimation equation, coef=11.88, z=2.13, p=0.033), and could be distinctively classified based on their MBTI scores (discriminant analysis Wilks Lambda=0.611, df=8, chi-square=36.7, p<0.001). Subjects with the So-Eum type (Introversion and Judging) and the So-Yang type (Extroversion and Perceiving) showed contrasting psychological features, however they had similar anthropometric characteristics. Subjects with the Tae-Eum type showed bigger Body Mass Index ($R^2$=0.22, df=4, 74, F=5.07, p=0.001) and body shape compared to others. Although there were no significant differences in G-protein beta-3 subunit polymorphism, angiotensin-converting enzyme polymorphism and Methylenetetrahydrofolate reductase polymprhisms among groups with Sasang types, it was shown that the dopamine system could be one for genetic marker for Sasang typology. These results demonstrated distinctive and essential traits of Sasang typology using reproducible psychometric, anthropometric and genetic evaluations. We also found that the Sasang typology was a bio-psychological typology which could show trait-specific guideline for individualized medicine.

Line probe assay를 이용한 신속한 rifampicin내성결핵 진단법의 임상적 유용성 (Clinical Usefulness of the Line Probe Assay for Rapid Detection of Rifampicin-resistant Tuberculosis)

  • 배 경 : rpoB 유전자 돌연변이는 rifampicin 내성결핵의 96-98%에서 발견된다. 실험실 검사에서는 rpoB 유전자가 다제내성결핵의 지표로서 빠른 진단에 사용될 수 있음이 보고되었으나 임상적 적용에 대해서는 아직 국내 및 외국에도 보고가 없는 실정이다. 연구 방법 : 서울중앙병원에서 1998 년 6월부터 2000년 7월 까지 LiPA법을 이용하여 rpoB유전자 돌연변이분석이 시행된 33명 환자에서 후향적으로 의무기록을 조사하였다. 환자의 임상상, 약제 감수성, 그리고 LiPA 검사 결과를 비교 분석하였다. 전통적인 약제감수성 결과를 표준으로 하여 LiPA 검사 결과와 비교하였으며 양 검사결과가 불일치하는 경우에 rpoB 유전자 염기서열분석을 시행하였다. 연구결과 : 평균 나이는 $42{\pm}19$세이고, 남녀비는 24 : 9 이었다. rpoB 유전자 검사 요청 시간으로부터 결과 보고까지 평균 시간은 $5.2{\pm}2.6$일 이었다. rpoB 유전자 검사 결과는 약제 감수성 결과보다 평균시간은 $56{\pm}35$일 빠르게 보고되었다. 감수성 결과가 얄려진 33명 중 28(85%)명에서는 유전자 검사와 동일한 결과를 보였고, 5명은(15%) 반대 결과를 보였다. 반대 결과를 보인 5명에서 염기서열 분석을 하였을 때, 3예에서 약물 감수성 결과가 오류였을 가능성이 있고, 나머지 2예는 LiPA 검사결과가 오류였을 가능성이 있다. 치료 약 선택에 도움을 준 경우가 28예(85%) 있었다. 결 론 : LiPA 방법을 이용한 rpoB 유전자 검사는 임상에서도 다수에서 다제내성을 신속하고 정확하게 진단할 수 있었고, 치료약 선택에 도움을 주었다. 하지만 현재의 시점에서 LiPA 방법이 기존의 도말 및 배양검사를 완전히 대치할 수는 없고 임상상, 도말 및 배양검사결과와 종합하여 보조적으로 사용하면 도움이 될 것으로 사료되었다.
