Experimental Study on the Radiosensitivity and Chemosensitivity of A-431 Cell Line (A-431 세포주의 방사선 및 항암제의 감수성에 관한 실험적 연구)

  • Objectives: The purpose of this study was to aid in the prediction of tumor cell tolerance to radiotherapy and/or chemotherapy. Material and Methods: Human epidermoid carcinoma A-431 cell lines were irradiated by 2, 4, 6, 8, 10Gy at a dose rate of 210cGy/min using /sup 60/Co Irradiator ALDORADO 8 and then were exposed to bleomycin or cisplatin at concentration of 2㎍/㎖ for 1 hour. The viable cells were determined for each radiation dose and/or each drug at the 4th day and cell surviving curves were obtained using semiautomated MTT assay. Results: The surviving fraction after irradiation of 2Gy was 0.99, and there was not significant difference of surviving fraction in comparison with the control group on A-431 cell line(P>0.05). But there were significant differences of surviving fractions at doses of 4, 6, 8, 10Gy in comparison with the control group(P<0.05). The cytotoxicity of bleomycin or cisplatin was significantly different in comparison with the control group on A-43l cell line (P<0.05). And the cytotoxicity of cisplatin was greater than that of bleomycin on A-431 cell line (P<0.05). There were significant differences of surviving fractions after irradiation of 2, 4, 6, 8, 10Gy with bleomycin or cisplatin in comparison with each group of irradiation only on A-431 cellline(P<0.05). There were significant differences of surviving fractions between the groups of irradiation with bleomycin and cisplatin at doses of 2, 4Gy(P<0.05), but there were not significant differences of surviving fractions at doses of 6, 8, 10Gy on A-431 cell line (P>0.05).

  • Potentially lethal damage repair (PLDR) in HFL-I was investigated by delayed plating experiments. The surviving fraction data were fitted to the linear Quadratic equation ($LogSn=-n{\gamma}({\alpha}d+{\beta}d^2$) where ${\gamma}=1$ for immediate plating). And a repair factor ${\gamma}$ was developed to compare survival for immediate and delayed plating. When we only took into account the repair factor of PLDR ${\gamma}$ which was derived from the delay assay, the cell survival response th fractionated carbon ion irradiation was not fully matched. This gap suggested that consideration of another repair process is necessary. So this suggests that the various repair process plays an important role in the fractionated irradiations.

  • The purpose of this study is to investigate whether gold nanoparticles had radiosensitization when combined with gamma and x ray beam. Cytotoxicity was mearsured with comparing survival fraction after incubated time 6,12,18 and 24 hours. Clonogenic assay was employed to assess survival fraction of cells with and without gold nanoparticles treatment following gamma ray irradiation. The most of gold nanoparticles were distributed in the cytoplasm. And the toxicity of gold nanoparticles used this study were found to be non-cytotoxic. And we also observed enhancement by about 40% in RBE value for gamma ray irradiation of cells treated with gold nanoparticles. Dose reduction of about half for gamma ray irradiation is demonstrated for gold nanoparticles treated cells as compared to untreated cells. In cells with exposed to gamma ray, DNA damage was increased when compared to only radiation exposed cells. The study revealed a significant reduction in radiation dose for killing the cells with internalized gold nanoparticles as compared to the cells without gold nanoparticles. The gold nanoparticles treatment resulted in enhancement of radiation effect as evident from increase in relative biological effectiveness values for photon irradiated cells.

  • We obtained Surviving Fraction (SF) after irradiation carbon beam to compare the applicability of the Linear-Quadratic model, Incomplete Repair model, Marchese model. Mathematica software(ver 9.0) used to calcurate parameters and compared result. LQ model could not explain the entire response of fractionated carbon beam irradiation. It becomes necessary to construct models that extend the LQ model of conventional radiotherapy for the carbon beam therapy. By combining both Potentially Lethal Damage Repair (PLDR) and Sublethal Damage Repair (SLDR) a new LQ model can develop that aptly modeled the cellular response to fractionated irradiation.

For this study, cell surviving curves were obtained for mouse lymphoma YAC-1 cell line using semi automated MTT assay. 2, 4,6, 8, 10Gy were irradiated at a dose rate of 210cGy/rnin using /sup 60/Co Irradiator ALDORADO 8. After irradiation, YAC-1 cell lines(3×10⁴cells/ml) were exposed to bleomycin or cisplatin for 1 hour. The viable cells were determined for each radiation dose and/or each concentration of drug at the 4th day. And they were compared to control values. The obtained results were as follows : 1. The surviving curve with gentle slope was obtained after irradiation of 2, 4, 6, 8, 10 Gy on YAC-1 cell line. 2. The cytotoxicity of bleomycin or cisplatin was increased significantly at all concentration of 0.2㎍/ml, 2㎍/ml and 20㎍/ml on YAC-1 cell line (P<0.01). And the cytotoxicity of cisplatin was greater than that of bleomycin at all concentration on YAC-1 cell line (P<0.01). 3. There were no significant differences of surviving fractions among 4Gy, 6Gy and 8Gy after irradiation of each radiation dose with 2㎍/ml of bleomycin compared with irradiation only on YAC-1 cell line. 4. There was significant difference of surviving fraction between 2Gy and 10Gy after irradiation of each radiation dose with 2㎍/ml of cisplatin compared with irradiation only on YAC-1 cell line(P<0.05). 5. There were significant differences of surviving fractions between the groups of irradiation only and the groups of irradiation with 2㎍/ml of bleomycin or cisplatin at all doses of 2, 4, 6, 8 and 10Gy on YAC-1 cell line(P<0.05).

For this study, cell surviving curves were obtained for human squamous cell carcinoma KB cell line after radiation exposure and/or administration of antitumor drugs. 2, 4, 6, 8, 10Gy were irradiated at a dose rate of 210cGy/min using /sup 60/Co Irradiator ALDORADO 8. After irradiation, KB cell lines (3×104cells/ml) were exposed to 2㎍/ml of bleomycin or cisplatin for 1 hour. The viable cells were determined by MTT assay for each radiation dose and/or each drug at the 4th day. And they were compared to control values. The obtained results were as follows : 1. The slope of the surviving curve after irradiation of 2, 4, 6, 8, 10Gy on KB cell line was relatively steep. 2. There was no significant difference between the cytotoxicity of bleomycin compared to control group. But, there was significant difference between the cytotoxicity of cisplatin compared to control group. And the cytotoxicity of cisplatin was greater than that of bleomycin on KB cell line. 3. There were significant differences of surviving fractions after irradiation of 2Gy and 10Gy with 2㎍/ml of bleomycin compared with the groups of irradiation only on KB cell line. 4. There were significant differences of surviving fractions after irradiation of 2, 4, 6, 8, 10Gy with 2㎍/ml of cisplatin compared with the groups of irradiation only on KB cell line. 5. There was significant difference of surviving fraction between groups after irradiation of 10Gy with 2㎍/ml of bleomycin and cisplatin.

  • The effect of heat treatment on membrane permeabilities of yeast's cells was studied, the amounts of efflux out of yeast cells were put to analysis, and fraction survival was also counted by viable plate counting method. Effects of nutritional substances on thermally injured yeast cells were also investigated under the highlight of reabsorption mechanism, then the relationship between permeability and surviving action in injured yeast cells are discussed.

For this study, cell surviving curves were obtained for murine melanoma Bl6 cell line using semiautomated M1T assay. 2,4,6,8, 10Gy were irradiated at a dose rate of 210cGy/min using /sup 60/Co Irradiator ALOORADO 8. After irradiatior, B16 cell lines(2.5×10⁴ cells/ml) were exposed to bleomycin and cisplatin at concentration of 0.2㎍/㎖, 2㎍/㎖ and 20㎍/㎖ for I hour respectively. The viable cells were determined for each radiation dose and/or each concentration of drug. And they were compared to control values. The obtained results were as follows : 1. There was significant difference of surviving fraction at 4, 6, 8, 10Gy on B16 cell line(P<0.05). 2. There was significant difference of cytotoxicity between bleomycin and cisplatin at concentration of 0.2㎍/㎖ and 2㎍/㎖(P<0.05) on B16 cell line, but there was no significant difference of cytotoxicity at concentration of 20㎍/㎖ on B16 cell line. 3. There was significant difference of cytotoxicity of bleomycin after irradiation of 2Gy and 10Gy on B16 cell line(P<0.01). 4. There was significant difference of cytotoxicity of cisplatin at concentration of 20㎍/㎖ after irradiation on B16 cell line.

There was significant difference of surviving fraction at 4, 6, 8, 10Gy on MG-63 cell line(p<0.05). 2. There was significant difference of cytotoxicity of bleomycin or cisplatin at all concentration of 0.2, 2, 20㎍/㎖ (p<0.05) on MG-63 cell line. The cytotoxicity of cisplatin was more effective than bleomycin at concentration of 20㎍/㎖ on MG-63 cell line. 3. There was significant difference of cytotoxicity of bleomycin or cisplatin at all concentration after irradiation of 2Gy on MG-63 cell line. 4. There was significant difference of cytotoxicity of bloemycin or cisplatin at concentration of 20㎍/㎖ after irradiation than that of irradiation alone(p<0.01). But there was no significant difference of cytotoxicity of bleomycin at concentration of 20㎍/㎖ after irradiation of l0Gy than that of irradiation alone.

Relationship between Radiosensitivity and Repair Capacity in Human Epithelial Cancer Cell Lines (인체 상피암 세포주에서 방사선감수성과 손상회복의 상관관계에 관한 연구)

  • To investigate the relationship between radiosensitivity and postirradiation recovery in human cancer cells, a study was performed using human cancer cell lines-A549, CaSki, SNU-C5 and PCI-13. For the study of radiosensitivity, single doses of 2, 4, 6, 8, 10, 12, and 14 Gy were given and for postirradiation recovery, two fractions of 4 Gy were separated with a time interval of 0, 0.5, 1, 1.5, 2, 2.5, 3, 4, 5, or 6 hours. Surviving fraction was estimated using colony forming ability. Surviving fractions at 2 Gy (SF2) were 0.496 (0.570-0.412) for A549, 0.496 (0.660-0.332) for CaSki, 0.386 (0.576-0.216) for SNU-C5, and 0.185 (0.247-0.123) for PCI-13. By statistical analysis the SF2 of PCI-13 was lower significantly than those of others (p<0.05). This difference was also observed at 4, 6 and 8 Gy dose levels. At 6 and 8 Gy the surviving fractions of SNU-C5 were also lower significantly than A549 and CaSki (p<0.05). By the analysis with linear quadratic model, the values of ${\alpha}$ for A549, CaSki, SNU-C5 and PCI-13 were 0.3016, 0.3212, 0.4327 and 0.8423, respectively, and those of ${\beta}$ were 0.02429, 0.02009, 0.03349 and 0.00059, respectively. So, the value of ${\alpha}$ showed increasing tendency with decreasing SF2. By the multitarget single hit model the values of Do for A549, CaSki, SUN-C5 and PCI-13 were 1.97, 1.97, 1.46 and 0.81, respectively, and those of n were 1.53, 1.50, 1.56 and 2.28, respectively. So, the value of Do decreased with decreasing SF2. Post-irradiation recovery reached plateau at around 2 hours. Recovery ratio at plateau phase ranged from 1.2 to 4.2; the value were 1.2 for PCI-13, 3.2 for CaSki, 3.3 for SNU-C5, and 4.2 for A549. Recovery rate well correlated with SF2, and increased with increasing Do and decreasing ${\alpha}$. According to above results, the intrinsic radiosensitivity was quite different among the tested cell lines; PCI-13 was the most sensitive and A549 and CaSki was similar. This difference of radiosensitivity is thought to be partly due to the difference in amount of postirradiation recovery. By linear quadratic model the difference of ${\alpha}$ values was very high, and by multitarget single hit model the difference of Do value was significantly high among four cell lines.
