전자선 조사를 이용한 히알루론산의 특성 조절 (Modulation of Hyaluronic Acid Properties by Electron Beam Irradiation)

  • A variety of natural polymers have been used as tissue engineering scaffolds, drug delivery system, and cosmetic materials due to their higher biocompatibility and water uptake. As a major component of extracellular matrix, hyaluronic acid consisting of D-glucuronic acid and N-acetylglucosamine has been popularly used as a hydrogel material. Even though it has good properties to be used in the tissue engineering and cosmetic industry, its higher viscosity has limited a potential use in a variety of applications; only low content should be applied in preparing above products. In the present study, we investigated the effect of electron beam irradiation on the properties of hyaluronic acid. Hyaluronic acid paste containing low contents of water changed to solution after electron beam irradiation ranging from 1 to 10 kGy, which didn't exhibit any alteration of surface properties and morphological change after freeze-drying. However, its viscosity was significantly decreased as absorbed dose increased, which was approximately one by hundred in comparison with the viscosity of original hyaluronic acid solution with same concentration. In addition, it can still interact with positive charged chitosan generating polyelectrolyte complex. Therefore, only viscosity was decreased after electron beam irradiation, whereas other properties of hyaluronic acid maintained. Consequently, these hyaluronic acids with lower viscosities can be used in a variety of applications in tissue engineering, drug delivery, and cosmetic industry.

LED 조명계를 결합한 캡슐내시경용 초소형 반사굴절식 전방위 광학계의 설계 (Optical Design of a Subminiature Catadioptric Omnidirectional Optical System with an LED Illumination System for a Capsule Endoscope)

  • 측면 360° 조명을 위한 6개의 LED (light emitting diode)로 구성된 조명계가 부착된 캡슐내시경용 초소형 반사굴절식 전방위 광학계를 설계하고 성능을 평가하였다. 이 광학계의 총 길이, 전장길이, 반화각(HFOV: half field of view), F-수는 각각 14.3 mm, 8.93 mm, 51°~120°, 3.5로 설계되었다. 이 전방위 광학계는 0.1 megapixels의 CMOS (complementary metal-oxide-semiconductor) 센서를 사용하였으며, LED 조명계는 한 개당 0.25 lm의 LED 6개를 사용하여 광축을 따라 놓여 있는 내장의 내벽을 조명하도록 하였다. 이 결과, 최적화 설계된 광학계의 변조전달함수 0.3에서의 공간주파수, 나이퀴스트 주파수인 44 lp/mm와 0.3의 변조전달함수에서 공차에 따른 누적확률, 초점심도는 각각 130 lp/mm, 95%, -0.097 mm~+0.076 mm이다. 또한 광학계의 시야각 범위 내에서 LED 조명계에 의한 광축으로부터 물체거리가 15 mm인 내장의 내벽에서의 시뮬레이션된 조도는 최소 315 lx~최대 725 lx로 내장의 내벽을 촬영할 수 있는 조도와 가시도임을 알 수 있다.

무반사 면을 갖는 DFB 레이저의 빔 분포 시뮬레이션과 검정 (Simulation and Examination for Beam Profile of DFB Laser with an Anti-reflection Coated Mirror)

  • 광대역 광통신 시스템에 사용되는 레이저는 우수한 주파수 선택성과 모드 안정성을 가져야한다. DFB(Distributed Feedback) 레이저는 고주파로 전류 변조를 하더라도 발진 주파수의 변화가 적다. 본 연구에서는 1.55um의 파장을 갖는 DFB 레이저에서 굴절률 격자와 이득 격자가 동시에 존재할 때, 오른쪽 거울 면에 반사가 일어나지 않도록 유전막 코팅을 하여 ρr=0 이 되도록 하였다. 문턱에서 최소 이득을 필요로 하는 제 1모드에 대하여, 종 방향으로의 발진 모드의 빔 분포와 방사전력비 Pl/Pr를 ρl의 위상=π인 경우와 ρl의 위상=π/2인 경우에 대하여 비교 검증했다. ρl의 위상=π인 경우, 낮은 문턱 전류와 높은 주파수 안정성을 얻기 위해서는, κL이 8보다 커야 한다. ρl의 위상=π/2인 경우, 낮은 문턱 전류를 위해서는 κL=1.0이 되도록 해야 하고, 이때 발진 주파수는 격자 주파수와 일치한다. 반사 방지 코팅을 하지 않은, 두 개의 거울 면을 가진 1.55um의 파장을 갖는 DFB 레이저보다, 한쪽 거울 면에 무반사 코팅을 한 경우에 모드 선별성이 훨씬 크다.

폭발물 감지 시스템 개발을 위한 TNT 분자 흡착에 대한 WSe2 소자의 전기적 반응 특성 평가 (Electrical response of tungsten diselenide to the adsorption of trinitrotoluene molecules)

  • 분자 단위의 폭발물질을 탐지하기 위하여, 고감도 응답성 센서의 개발이 요구되고 있다. 2차원 반도체는 얇은 적층형 구조를 가져 전하 캐리어가 축적될 수 있어, 전하 캐리어의 급격한 신호 변조 특성을 기대할 수 있다. WSe2 반도체 소재의 TNT(Trinitrotoluene) 폭발물질에 대한 탐지 효용성을 연구하기 위해, CVD(Chemical Vapor Deposition) 공정을 이용해 WSe2 박막을 합성하여 FET(Field Effect Transistors)을 제작하였다. 라만 분석과 FT-IR(Fourier-transform infrared) 분광 결과는 TNT 분자의 흡착과 WSe2 결정질의 구조적 전이 분석 정보를 나타내었다. 또한, WSe2 표면의 TNT 분자 흡착 전후의 전기적 특성을 비교하였다. TNT 도포 전, WSe2 FET에 백 게이트 바이어스로 -50 V를 인가함에 따라 0.02 μA의 최대 전류 값이 관측되었고, 0.6%(w/v) TNT 용액을 도포하였을 때 Drain 전류는 p-type 거동을 보이면서 0.41 μA의 최대 전류 값을 기록하였다. 이후 On/Of f Ratio 및 캐리어 이동도, 히스테리시스를 추가적으로 평가하였다. 본 연구에서는 WSe2의 TNT 분자에 대한 고감도와 신속한 응답성을 통해 폭발물질 탐지 센서 소재로서의 가능성을 제시하였다.

Hypoxia-dependent mitochondrial fission regulates endothelial progenitor cell migration, invasion, and tube formation

  • Tumor undergo uncontrolled, excessive proliferation leads to hypoxic microenvironment. To fulfill their demand for nutrient, and oxygen, tumor angiogenesis is required. Endothelial progenitor cells (EPCs) have been known to the main source of angiogenesis because of their potential to differentiation into endothelial cells. Therefore, understanding the mechanism of EPC-mediated angiogenesis in hypoxia is critical for development of cancer therapy. Recently, mitochondrial dynamics has emerged as a critical mechanism for cellular function and differentiation under hypoxic conditions. However, the role of mitochondrial dynamics in hypoxia-induced angiogenesis remains to be elucidated. In this study, we demonstrated that hypoxia-induced mitochondrial fission accelerates EPCs bioactivities. We first investigated the effect of hypoxia on EPC-mediated angiogenesis. Cell migration, invasion, and tube formation was significantly increased under hypoxic conditions; expression of EPC surface markers was unchanged. And mitochondrial fission was induced by hypoxia time-dependent manner. We found that hypoxia-induced mitochondrial fission was triggered by dynamin-related protein Drp1, specifically, phosphorylated DRP1 at Ser637, a suppression marker for mitochondrial fission, was impaired in hypoxia time-dependent manner. To confirm the role of DRP1 in EPC-mediated angiogenesis, we analyzed cell bioactivities using Mdivi-1, a selective DRP1 inhibitor, and DRP1 siRNA. DRP1 silencing or Mdivi-1 treatment dramatically reduced cell migration, invasion, and tube formation in EPCs, but the expression of EPC surface markers was unchanged. In conclusion, we uncovered a novel role of mitochondrial fission in hypoxia-induced angiogenesis. Therefore, we suggest that specific modulation of DRP1-mediated mitochondrial dynamics may be a potential therapeutic strategy in EPC-mediated tumor angiogenesis.

강유전체 기억소자 응용을 위한 하부전극 최적화 연구 (Bottom electrode optimization for the applications of ferroelectric memory device)

  • 본 논문은 PZT 박막의 기억소자 응용을 위한 Pt 그리고 RuO2 박막을 조사하였다. 초고주파 마그네트론 스퍼터링 방법을 이용하여 하부전극을 성장하였으며, 조사된 실험변수는 기판온도, 가스 부분압, RF 전력 그리고 후열처리 등이다. 기판온도는 Pt, $RuO_2$박막의 결정구조 뿐만 아니라 표면구조 및 비저항 성분에 크게 영향을 주었다. Pt 박막의 XRD 분석으로 기판온도가 상온에서 $200^{\circ}C$까지는 (111) 그리고 (200) 면이 혼재하는 결과를 보였으나 $300^{\circ}C$에서는 (111) 면으로 우선 방위 성장 특성을 보였다. XRD와 AFM 해석으로부터 Pt 박막 성장시 기판온도 $300^{\circ}C$, RF 전력 80W가 추천된다. 산소 분압비를 0~50%까지 가변하여 조사한 결과 산소가 5% 미만으로 공급되면 Ru 금속이 성장되고, 산소 분압비가 10 ~40%까지는 Ru와 $RuO_2$ 상이 공존하였으며 산소 분압비가 50%에서는 순수한 $RuO_2$상만이 검축되었다. 이 결과로부터 RuO2/Ru 이층 구조의 하부전극 형성이 산소 가스 부분압을 조절하여 한번의 공정으로 성장 가능하며, 이런 구조를 이용하면 금속의 낮은 비저항을 유지하면서도 PZT 박막의 산소 결핍에 의한 기억소자의 피로도 문제를 완화할 것으로 사료된다. 후 열처리 온도를 상온에서부터 $700^{\circ}C$까지 증가할 때 Pt와 $RuO_2$의 비저항 성분은 선형적 감소 추세를 보였다. 본 논문은 강유전체 기억소자 응용을 위한 최적화된 하부전극 제적조건을 제시한다.

비소세포폐암에서 MUC1과 CD44s의 발현 (The Expression of MUC1 and CD44s in Non-small Cell Lung Cancer)

  • 배 경 : MUG1은 고분자량의 당화 당단백으로 정상적으로 선상피의 선단부에서 발현되나, 종양세포에서는 비정상적인 발현 양상을 보인다. CD44는 세포-세포간 부착, 세포-기질간 부착에 관여하는 당단백이다. 일부 종양에서 MUC1 과 CD44는 종양의 침습과 전이에 관계한다고 알려져 있다. 본 연구에서는 비소세포폐암에서의 MUC1과 CD44의 standard form (CD44s)의 발현과 조직형, TNM-병기와의 상관성을 조사하였다. 대상 및 방법 : 수술적 방법으로 얻은 비소세포폐암 80예의 파라핀포매 조직절편을 대상으로 하여 MUC1과 CD44s에 대한 면역조직화학염색을 시행하였다. 결 과 : MUC1 은 편평상피암종의 12/43예 (27.9%), 선암종의 12/37예 (32.4%)에서 양성으로 조직형에 따른 차이는 보이지 않았다(p=0.660). CD44s는 편평상 피암종의 36/43예(83.7%), 선암종의 14/37예 (37.8%)에서 양성으로 선암종에 비해 편평상피암종에서 발현이 유의하게 높았다(p<0.001). 편평상피암종에서 TNM-병기에 따른 MUC1의 양성율은 통계적으로 유의한 차이를 보이지 않았다. 선암종에서 MUC1 양성은 N0에서 4/22예(18.2%), N1-2에서 8/15예 (53.3%)로 림프절 침범과 관련되었다(p=0.036). 편평상피암종에서 CD44s의 발현은 T-병기에 따른 의미있는 차이는 없었으나, N0기에서 16/21예 (72.7%), N1-2기에서 9/22예 (38.1%)로 CD44s의 발현의 감소는 림프절 침범과 관련되었다(p=0.031). 편평상피암종의 TNM-병기 I기에서 15/18예(83.3%), II-III기에서 10/25예 (40.0%)에서 CD44s 양성으로 CD44s 발현의 감소는 TNM-병기의 진행과 관련되었다(p=0.006). 결 론 : 비소세포폐암에서 MUC1과 CD44s는 종양의 조직형에 따라 다른 발현 양상을 보이고, 각각은 종양의 침습과 전이에 관여하는 것으로 보인다.

꿀풀 추출물과 그 활성 화합물인 카페인산의 Na+/H+ exchanger 1 (NHE1) 발현 조절을 통한 피부 pH 개선 효과에 대한 연구 (Study on Skin pH Improvement Effect through Regulation of Na+/H+ Exchanger 1 (NHE1) Expression of Prunella vulgaris Extract and Its Active Compound, Caffeic Acid)

  • 본 연구는 인간 각질형성 세포주를 이용하여 피부 표면의 산성화를 조절하는 물질을 발굴하고, 이를 통해 각질층의 보습 능력 및 피부 장벽 기능에 미치는 영향을 조사하기 위해 수행되었다. 꿀풀은 아프리카 북서부 및 북미에 널리 분포하는 허브 중 하나로 세포사멸, 항산화 및 항염에 대한 효능이 연구되어 왔다. 하지만 NHE1의 발현 조절 및 피부 장벽기능 회복에 대한 연구는 진행되지 않았다. 꿀풀의 피부 pH 조절 효과를 확인하기 위하여 꿀풀의 활성 성분 분석 결과 로즈마린산과 카페인산이 검출되었다. 꿀풀과 이것으로부터 검출된 성분인 카페인산은 인간 각질형성 세포주인 HaCaT 세포에 고농도(100 ㎍/mL 또는 100 µM)를 처리한 결과 독성을 보이지 않았다. 노화된 피부에서는 각질층의 산성 pH를 유지하기 위한 나트륨-수소 이온 교환 펌프(NHE1)의 발현감소가 나타나는 것으로 알려져 있으며, 이러한 이유로 노화 피부에서 나타나는 피부 장벽 기능 회복의 저하에 중요한 원인으로 작용할 것으로 추측된다. 꿀풀 추출물 및 카페인산은 각질형성 세포에서 NHE1의 발현을 증가시켰으며, 자연 보습 인자 전구체인 필라그린과 세라마이드 합성 효소인 serine plmitoyl transferase (SPT)의 발현을 증가시켰다. 또한, 직접적인 pH 조절 효과를 확인하기 위해 각질형성 세포 내/외 pH 수치를 측정한 결과 꿀풀과 카페인산은 세포 외부 pH를 감소시켰다. 위 결과들을 종합해 볼 때, 꿀풀과 카페인산은 NHE1 조절을 통해 피부 pH를 조절할 수 있고, 자연 보습 인자(NMF)와 세라마이드 합성을 증가시켜 피부 장벽 기능 회복을 도울 수 있을 것으로 추측된다. 이러한 결과는 꿀풀과 카페인산이 피부 건강에 긍정적인 영향을 미칠 수 있다는 가능성을 보여주며, 이를 활용한 새로운 피부 보호 제품 개발에 대한 토대가 될 수 있다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. In this paper, a simple reflective infrared optic system with a 2-dimensional optic scanner is proposed for the realization of medical infrared thermography system.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction.
