Improvement of Motor Behavior of Parkinson′s Disease Animal Model by Nurr1-Transfected Human Embryonic Stem Cells.

  • The purpose of this study is to evaluate an efficacy of in vitro differentiated human embryonic stem (hES, MB03) cells expressing Nurr1 in relief of symptomatic motor behavior of Parkinson's disease (PD) animal models MB03 was genetically modified to express Nurr1 protein and was induced to differentiate according to 2-/4+ protocol using retinoic acid and ascorbic acid. The differentiation-induced cells were selected for 10 to 20 days thereafter in N2 medium. Upon selection, cells expressing GFAP, TH, or NF200 were 38.8%, 11%, and 20.5%, respectively. in order to examine therapeutic effects of the differentiated cells in PD animal model, rats were unilaterally lesioned by administration of 6-kydroxydopamine HCI (6-OHDA) into medial forebrain region (MFB, AP -4.4 mm, ML 1.2 mm, DV 78 mm with incision bar set at -2.4 mm), as a reference to bregma and the surface of the skull. Confirmation of successful lesion by apomorphine-induced rotational behavior, differentiated cells were transplanted into the striatum (AP 1.0, ML 3.5, DV -5.0; AP 0.6, ML 2.5, DV -4.5). Improvements of asymmetric motor behavior by the transplantation were examined every two weeks after the surgery. In two weeks, numbers of rotation by the experimental rats were $-14.8 \pm 33.9%$ (P<0.05) of the number before transplantation, however, the ratio increased slightly to $13.6 \pm 56.3%$ in six weeks. In contrast, the ratio of sham-grafted animals ranged from 112.3+8.5% to 139.2+28.9% during the examination. Immunohistochemical studies further confirmed the presence, survival, migration, and expression of TH of the transplanted human cells.

  • The purpose of this study was to develop a practical caries activity test using laser fluorescence. The subjects of study were 30 children of $7\sim10$ years old. Fluorescence from initial carious lesion of teeth illuminated by an argon laser(488nm) was observed through barrier alter. For evaluation of accuracy and propriety of this method fer caries activity test, teeth with initial caries lesion on buccal or labial surface of children was examined with visual inspection and laser fluorescence. Visual examination for the dDfFtT and the $Cariescreen^{(R)}$ test were also done. The results from the present study can be summarized as follows: 1. Laser fluorescence test could differentiate initial caries lesions more easily than visiual inspection. 3. There was highest correlation(= 0.73) between laser fluorescent test and $Cariescreen^{(R)}$ test and. And also apparent correlation(= 0.66) exists between laser fluorescent test and caries experience measured by dDfFtT.

  • This paper represents a basic statistical examination on the navigability of ocean-going ship from the point of estimating the time lasting period when propeller racing occurred by using the basic probability theory and the statistics. The propeller racing is one of the most important seakeeping qualities in relation to the safety of the main engine and shafting system. The trend of the racing has been mainly investigated in order to estimate allowable maximum propeller diameter, operation of ocean-going ships, etc.. In those studies, the propeller racing generally and mainly means the situation (propeller exposed) in which the relative motion amplitude between ship hull and wave surface would exceed a depth of point in rotary disk propeller. Therefore, it seems that the magnitude of the amplitude and its exceeding frequency of propeller racing have been examined as a principal subject of study as usual. However, the time during which the amplitude exceeds the depth of point, that is, the propeller exposes in the air, must be also one of most important factor affecting the trend of propeller racing. Then, this paper proposes a new practical method for estimating the time lasting of exposed propeller related to propeller racing in rough-confused seas on the basis of the linear strip theory and the statistics. And, numerical examples of estimating the propeller racing probability are given for four wide ship forms. Finally the usefulness of the proposed method for predicting propeller racing based on the time lasting period is discussed.

  • This study was undertaken to observe the effects of the antineoplastic agent, 5-Fluorouracil(5 FU) on the hair in Sprague-Dawley white rats. Twenty four sprague-Dawley strain white rats, each weighing about 150-200 grams were used and divided into control and experimental groups. In the experimental group, eighteen rats were injected intraperitonially with 60 mg of 5-FU per killogram body weight with one time per two days, Six rats were injected with 0.5 cc of normal saline solution intraperitoneally as a placebo on this control group. Rats were serially sacrificed on the first, third, fifth, seventh, tenth and fourteenth day after 2 times of injection of 5-FU and saline. The hair were obtained and observed SEM. After examination and comparision of all specimens, the results of this study were as follows: 1. In the control group, the scale and cuticle of hair was observed smooth surface and equal interval 2. In the experimental group, the first day, scale change was seen from body of hair and crack was seen. from fifth day, and irregular scale and cuticle of hair was seen from 10, 14 days 3. The apperance of root of hair was not almost change From above results, 5-Fluorouracil was more effective on the hair body. The change was begun from first day and crack of scale was seen from fifth day and irregular scale and cuticle of hair was seen from 10,14 days. The.

A Study on a Morphological Identification of Cnidium Rhizome and Ligusticum Rhizome (천궁(川芎)의 형태(形態)에 관한 연구(硏究))

  • Objectives : In order to distinguish Cnidii Rhizoma and Ligustici Chuanxiong Rhizoma (cultivated in Korea and China), their external and internal states were observed via microscopic examination. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Results : Cnidii Rhizoma, Ligustici Chuanxiong Rhizoma (from Korea), and Ligustici Chuanxiong Rhizoma (from China) all demonstrated similar patterns as following : 1. external form 1) showing irregular nodular form with $2{\sim}7cm$ diameter. 2) having several trochites in parallel, and nodulose root trace on surface. 3) in addition to yellow-white or yellow-gray intersections, yellow-brown oil sacs were distributed, with cambium resembling a form of annulus. 2. internal form 1) its root and rhizome is composed of 10 layers of cork, with narrow cortical layers. 2) its phloem is wide, and oval light yellow-brown oil sacs are distributed around. The closer to the cambium, the smaller it is. 3) cambium is a form of a wave-like ring or an irregular polygon. 4) xylem and vessel are rare, most of which are arranged in a 'V' format, and sometimes xylem fiber is found. 5) pith is composed of parenchymatous cell, and parenchyma is full of starch grains. Conclusions : In conclusion, C officinale, L. chuanxiong (from Korea), and L. chuanxiong (from China) have rhizomes in a similar form with no special difference. Therefore, it is concluded that C. officinale, L. chuanxiong (from Korea), and L. chuanxiong (from China) are categorized into the same species.

  • Background: Lung cancer is usually diagnosed at an advanced stage, resulting in a poor prognosis. The detection of these lesions at an earlier stage would be a clear benefit to patients. However, it is extremely difficult to detect carcinomatous lesions in the bronchial mucosal sites during a routine bronchoscopy. Methods: This study employed a novel optical technique, known as narrowband imaging (NBI), which allows noninvasive visualization of the microvascular structure of an organ's surface using reflected light. Results: Narrow band imaging was performed on 10 patients who were radiologically suspicious or had a high risk of lung cancer. The median age of the patients was 57.5 years (range, 44~81 years), and 80% of the patients were male. All lesions showed a microvascular proliferation pattern (dotted, tortuous and abruptly ending vessel) on the magnified NBI. Two lesions were confirmed histologically to be adenocarcinoma and the remaining lesions were squamous cell carcinomas. Two lesions were confirmed histologically to be a carcinoma in situ. Conclusion: NBI is a promising and potentially powerful tool for identifying carcinomas at an earlier stage or a central lesion during a routine bronchoscopy examination.

Use of Protease Produced by Bacillus sp. SJ-121 for Improvement of Dyeing Quality in Wool and Silk

  • In this study, a microorganism-produced protease was used to improve the quality of fabrics. First, the protease-producing bacteria were isolated from soils, and one of them was selected and identified as Bacillus sp. SJ-121. The optimal medium composition for its growth and protease production was determined to be as follows: glucose 1g/L, soybean meal 0.5g/L, soy peptone 0.5, $K_2HPO_4\;0.2,\;MgSO_4\cdot7H_2O\; 0.002,\;NaCl\;0.002,\;and\;Na_2CO_3g/L$. Also, the optimal temperature for the production of the protease by Bacillus sp. SJ-121 was about $40^{\circ}C$ at pH 7. The wool and silk were treated with the protease from Bacillus sp. SJ-121. Following the protease treatment, changes in the surface of a single yarn of the fabrics were observed by both an optical microscope and a scanning electron microscope (SEM). Changes in the K/S value of the wool and silk were measured by spectrophotometric analysis, in order to determine the amount of dye uptake in the fabrics. We also performed a tensile strength examination in order to determine the degree and nature of mechanical changes in single yarns of the wool and silk fabrics. By increasing the protease treatment time to 48 h, the dyeing characteristics of the fabrics were enhanced, and the surfaces of the single yarns of the fabrics became smoother, due to the removal of soil and scale in them. However, no mechanical changes were detected in the fabrics. Therefore, we suggest that proper treatment of the protease produced by Bacillus sp. can improve the quality of silk and wool.

New Fracture Toughness Test Method of Zircaloy-4 Nuclear Fuel Cladding (Zircaloy-4 핵연료 피복관의 신파괴인성 시험법)

  • To define the causes of cladding degradation which can take place during the operation of nuclear power plants, it is required to develop the new fracture toughness test of spent fuel cladding. The fracture toughness of Zircaloy-4 cladding was estimated using the recently developed KAERI embedded Charpy (KEC) specimen. Axially notched KEC specimens cut directly from unirradiated fuel claddings, were tested in a way similar to the standard toughness test method of a Single Edge Bending (SEB) specimen. The results of KEC fracture toughness test at room temperatures were discussed and compared with those of the previous other studies. In conclusions, even though the KEC fracture toughness test of nuclear fuel claddings was easier and more reliable than those developed earlier, the results from the cladding fracture tests were not the material characteristics but the specific fracture parameters which were deeply related to the specification of claddings. In addition, the phenomenon of a thickness yielding was not observed from the fracture surface. It was closely related to the fact that the plane strain condition of the KEC specimen was changed to the plane stress condition during crack advancing. It was also supported by the fractographic evidence that the formation of ductile dimples at the crack initiation became the similar appearance such as a quasi-cleavage after the sufficient crack advancing.

Polyurethane-Coated Breast Implants Revisited: A 30-Year Follow-Up

  • Background Polyurethane coating of breast implants has been shown to reduce capsular contracture in short-term follow-up studies. This 30-year study is the longest examination of the use of polyurethane-coated implants and their correlation with capsular contracture. Methods This study evaluates the senior surgeon's (F.D.P.) experience with the use of polyurethane-coated implants in aesthetic breast augmentation in 382 patients over 30 years. Follow-up evaluations were conducted for six months after surgery. After the six-month follow-up period, 76 patients returned for reoperation. The gross findings, histology, and associated capsular contracture were noted at the time of explantation. Results No patient during the six-month follow-up period demonstrated capsular contracture. For those who underwent reoperation for capsular contracture, Baker II/III contractures were noted nine to 10 years after surgery and Baker IV contractures were noted 12 to 21 years after surgery. None of the explanted implants had macroscopic evidence of polyurethane, which was only found during the first five years after surgery. The microscopic presence of polyurethane was noted in all capsules up to 30 years after the original operation. Conclusions An inverse correlation was found between the amount of polyurethane coating on the implant and the occurrence of capsular contracture. Increasingly severe capsular contracture was associated with a decreased amount of polyurethane coating on the surface of the implants. No contracture occurred in patients whose implants showed incomplete biodegradation of polyurethane, as indicated by the visible presence of polyurethane coating. We recommend research to find a non-toxic, non-biodegradable synthetic material as an alternative to polyurethane.

Western blot diagnosis of vivax malaria with multiple stage-specific antigens of the parasite

  • Western blot analysis was performed to diagnose vivax malaria using stage-specific recombinant antigens. Genomic DNA from the whole blood of a malaria patient was used as templates to amplify the coding regions for the antigenic domains of circumsporozoite protein (CSP-1), meroxoite surface protein (MSP-1), apical merozoite antigen (AMA- 1), serine repeat antigen (SERA), and exported antigen (EXP- 1) of Plasmodium vivax. Each amplified DNA fragment was inserted into a pGEX-4T plasmid to induce the expression of GST fusion protein in Escherichia coli by IPTG. The bacterial cell extracts were separated on 10% SDS-PAGE followed by western blot analysis with patient sera which was confirmed by blood smear examination. When applied with patient sera, 147 (91.9%) out of 160 vivax malaria, 12 (92.3%) out of 13 falciparum malaria, and all 9 vivax/falciparum mixed malaria reacted with at least one antigen, while no reactions occurred with 20 normal uninfected sera. In the case of vivax malaria, CSP-1 reacted with 128 (80.0%) sera, MSP-1 with 102 (63.8%), AMA-1 with 128 (80.0%), SERA with 115 (71.9%), and EXP-1 with 89 (55.6%), respectively. We obtained higher detection rates when using S antigens (91.9%) rather than using each antigen solely (55.6 - 80%), a combination of 2 (76.3 - 87.5%), 3 (85.6 - 90.6%), or 4 antigens (89.4 - 91.3%). This method can be applied to serological diagnosis, mass screening in endemic regions, or safety test in transfusion of prevalent vivax malaria.
