Chlorophyll, Mineral Contents and SOD-like Activities of Leeks Harvested at Different Times (부추의 수확시기에 따른 클로로필, 무기질 및 superoxide dismutase 유사활성의 변화)

  • This study was performed to determine the contents of mineral and bioactive components in leek samples harvested at different times. Analysis of chlorophyll contents of leek harvested at different times showed the latest one (5th sample) had the highest level among samples. The leek harvested at the earliest (1st) had the highest amount of Fe, f and Cu while 5th sample was highest in Ca, Mn, P, Zn and Na contents. Lead (Pb) was not detected in any leek sample harvested at different times. SOD (superoxide dismutase)-like activity was the highest in leek harvested at the earliest.

Effect of Myricetin on mRNA Expression of Different Antioxidant Enzymes in B16F10 Murine Melanoma Cells (B16F10 Murine Melanoma Cell에서 Myricetin이 항산화효소의 m-RNA 발현에 미치는 영향)

  • Flavonoids are class of polyphenolic compounds widely distributed in the plant kingdom, which display a variety of biological activities, including antiviral, antithrombotic, antiinflammatory, antihistaminic, antioxidant and free-radica 1 scavenging abilities. The antioxidant enzyme (AOE) system plays an important role in the defense against oxidative stress insults. To determine whether flavonoid, myricetin can exert antioxidative effects not only directly by modulating the AOE system but also scavenging free radical, we investigated the influence of the flavonoid myricetin on cell viability, different antioxidant enzyme activities, ROS level and the expression of different antioxidant emzyme in B16F10 murine melanoma cells. Myricetin in a concentration range from 6.25 to $50\;{\mu}M$ decreased superoxide dismutase (SOD) and glutathione peroxidase (GPx) enzyme activities, but catalase (CAT) activity was increased. In the myricetin-treated group, ROS levels were decreased dose-dependently. Antioxidant enzyme expression was measured by RT-PCR. Myricetin treatment of B16F10 cells increased catalase expression. Expression levels of copper zinc superoxide dismutase (CuZn SOD) were not affected by exposure of myricetin. Manganese superoxide dismutase (Mn SOD) and GPx expression levels decreased slightly after myricetin treatment. In conclusion, the antioxidant capacity of myricetin was due to CAT and free-radical scavenging.

The Relationship between Structural Denaturation of Antioxidative Enzymes and Their Enzyme Activity due to Repeated Exposure to UV-A (UV-A 반복노출로 인한 항산화효소의 구조변성과 효소활성의 상관관계)

  • Purpose: The present study was conducted to investigate whether the changes in structure and activity of antioxidative enzymes, superoxide dismutase(SOD) and catalase(CAT) present in the eyes appeared when they were repeatedly exposed to UV-A, and reveal the correlation of these changes. Methods: Each enzyme solution was prepared from the standardized SOD and CAT, and repeatedly exposed to UV-A of 365 min under the condition of 30 minutes, 1 hour and 2 hours a day over 1, 2, 3, 4 and 5 days. Structural denaturation of SOD and CAT induced by repeat UV-A irradiation was confirmed by the electrophoretic analysis, and their enzyme activity was determined by the colorimetric assay using the proper assay kit. Results: SOD exposed repeatedly to UV-A showed the polymerization pattern through the electrophoretic analysis when it was repeatedly exposed under the condition of at least 1 hour a day however, the change of its activity was found to be less than 12%. On the other hand, CAT repeatedly exposed to UV-A showed reduced size of the electrophoretic band which indicated a structure denaturation and its activity was significantly decreased. In the case of that the repeat exposure time was longer, CAT activity was completely lost even though some enzyme band was shown in the electrphoretic analysis. Conclusions: From these results, it was revealed that the degree and pattern in structural denaturation of antioxidative enzymes differently appeared according to the type of enzyme, and the degree of structural denaturation was not always consistent with the reduction in enzyme activity.

The Protective Effect of Selenium on Radiation in Rat (흰쥐에 있어 방사선에 대한 셀레늄의 보호효과)

  • In this study, the radiation protection effects of selenium, known as a powerful antioxidant, were studied. After 14 days of oral injection of selenium into the rat, a full body irradiation of 10 Gy was carried out. And after 1 day, 3.5 day, 7 day and 21 day, we observed blood cell components and superoxide dismutase(SOD) and small intestine changes. Compared to irradiation groups, there was a significant protection effect of reducing the hematopoietic immune system damage in the irradiated group after selenium administration(p<0.05). Then, selenium is a valid ingredient that increases the activity of the superoxide Dismutase(SOD), and it is confirmed that it has an effect of inhibiting apoptosis expression of small intestinal cells by irradiation. Based on these results, selenium is considered to be an essential ingredient for protecting living things from radiation.

Micronutrients and superoxide dismutase in postmenopausal women with chronic periodontitis: a pilot interventional study

  • Purpose: The study was aimed at investigating changes in periodontal parameters and superoxide dismutase activity triggered by root surface debridement with and without micronutrient supplementation in postmenopausal women. Methods: Forty-three postmenopausal chronic periodontitis patients were divided into two groups: group 1 (n=22) were provided periodontal treatment in the form of scaling and root planing (SRP) and group 2 (n=21) patients received SRP along with systemic administration of micronutrient antioxidants. Patients in both groups were subjected to root surface debridement. Group 2 patients also received adjunctive micronutrient antioxidant supplementation. Serum and salivary superoxide dismutase (SOD) activity along with periodontal parameters were recorded at baseline and 3 months after therapy. Results: Salivary and serum SOD values significantly (P<0.05) improved with periodontal treatment. Improvement in systemic enzymatic antioxidant status along with reduction in gingival inflammation and bleeding on probing (%) sites was significantly greater in group 2 as compared to group 1. Conclusions: Adjunctive micronutrient supplements reduce periodontal inflammation and improve the status of systemic enzymatic antioxidants in postmenopausal women.

Resistance Function of Woody Landscape Plants to Air Pollutants(II) - POD Activity - (조경수목(造景樹木)의 대기오염물질(大氣汚染物質)에 대한 방어기능(防禦機能)(II) - POD 활성(活性)을 중심으로 -)

  • This study was conducted to determine the toxic effects of air pollutants on landscaping tress, Pinus densiflora, Pinus koraiensis, Ginkgo biloba, Liriodendron tulipifera, Platanus occidentalis and their resistance to the pollutant toxicity in urban and industrial regions of Seoul and Taejon, Korea. Total sulfur contents and enzyme activities such as superoxide dismutase and peroxidase were analyzed in tree foliage of Pinus densiflora, Pinus koraiensis, Ginkgo biloba, Liriodendron tulipifera, Platanus occidentalis. In addition, POD activity was analyzed in the foliage on tree seedlings, i.e. Pinus densiflora, Pinus koraiensis, Ginkgo biloba, Lirioderdron tulipifera, with the fumigation of $SO_2$ in gas chamber 4 hours a day for six days. In Ginkgo biloba total sulfur content and POD activity had a negative correlation while other species had a positive relationship in total sulfur content and enzyme activity. Air pollutants accumulated in tree tissues were supposed to enhance the enzyme activity like POD providing the resistance mechanisms. Especially Pinus koraiensis and Platanus occidentalis had higher POD activity than other species. The increase of temporary POD activity against environmental stress appeared in sensitive trees and prolonged increase of POD activity played an important role in resistance mechanism. SOD and POD activities in all species had a positive correlation except Ginkgo biloba. Changes of SOD and POD activities were different between species and in most species SOD as well as POD seemed to participate in resistance mechanism.

Change in Functional Properties by Extraction Condition of Roasted Pleurotus eryngii (볶음 처리 새송이버섯의 추출조건에 따른 생리기능적 특성 변화)

  • This study was performed to investigate the functional properties of roasted Pleurotus eryngii by the extraction conditions. Total phenolic compound content and electron donating ability (EDA) were high at 50% ethanol concentration. Superoxide dismutase (SOD)-like activity was high at 75% ethanol concentration. Nitrite scavenging ability increased as ethanol concentration in extracting solvent decreased. EDA decreased, SOD-like activity and nitrite scavenging ability increased as the extraction time increased. With the increase in extraction temperature, EDA and SOD-like activity decreased. But extraction temperature did not significantly affect the nitrite-scavenging ability With the increase in ratio of sample content to solvent, EDA, SOD-like activity and nitrite scavenging ability decreased. The results would be useful for understanding the extraction condition of roasted Pleurotus eryngii.

Effects of Hyperbaric Oxygen Treatment on the Malondialdehyde Level and Oxygen Free Radical Reactions in the Heart of the Rats Exposed to Carbon Monoxide (일산화탄소 폭로후 고압산소 투여가 흰쥐 심장에서의 malondialdehyde 함량과 산소유리기 반응에 미치는 영향)

  • ln an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen See radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde(MDA) level and activities of catalase and superoxide dismutase in the heart of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260gm were used. Experimental groups consist of Control group (=breathing with air), HBO group(=exposed to hyperbaric oxygen(HBO, 3ATA, 100%) after air breath), CO group(=exposed to CO(3,970 ppm) after air breath), CO-Air group(=exposed to CO after air breath followed by air breath) and CO-HBO group(=exposed to CO after ai. breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of Co-induced cardiotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the heart of the rats exposed to carbon monoxide.

Anti-oxidant and anti-inflammatory effects of Salix Koreensis Andersson in DC. leaf methanol extract in vitro models

  • Oxidative rancidity in foods causes undesirable changes in nutritive value, aroma, flavor, and color. Salix Koreensis Andersson in DC. (SK) has anti-oxidative and anti-inflammatory effects and is traditionally used to treat neuralgia, edema, pain, and inflammatory diseases. However, the regulatory effects of SK on oxidative and inflammatory reactions have not been elucidated. In this context, we scientifically validated the anti-oxidative and anti-inflammatory activities of SK leaf (SKL). The methanol extract of SKL was evaluated for in vitro anti-oxidative activities. SKL showed increased superoxide dismutase (SOD)-like activity and 1, 1-diphenyl-2-picrylhydrazyl radical scavenging activity. The in vitro anti-oxidant and anti-inflammatory activities of SKL were also investigated in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. LPS resulted in decreased SOD activities compared with the unstimulated cells, but SKL significantly increased SOD activities reduced by LPS. In addition, LPS-induced nitric oxide, tumor necrosis factor-${\alpha}$, and interleukin-6 productions were significantly and dose-dependently reduced by SKL in RAW264.7 macrophages without inducing cytotoxicity. In conclusion, these results indicate that SKL will be able to be effectively used as a food additive with anti-oxidative and anti-inflammatory effects.

Effects of Temperature Stress and Paraquat on SOD Activity and Photochemical Efficiency of PSII in Leaves of Araliaceae Plants (두릅나무과 식물의 SOD 활성과 광계II의 광화학적 효율에 미치는 온도 스트레스와 Paraquat의 영향)

  • The effects of temperature stress and paraquat on the superoxide dismutase (SOD) activity and the photochemical efficiency of photosystem II were studied in the leaves of Araliaceae plants. The SOD activity of Acanthopanax koreanum leaf discs increased at 4$^{\circ}C$ and 28$^{\circ}C$, and increased significantly at 4$^{\circ}C$ ,28$^{\circ}C$ and 35$^{\circ}C$ in the presence of paraquat. However, the SOD activity of Dendropanax morbifera leaf discs decreased at 4$^{\circ}C$, 28$^{\circ}C$ and 35$^{\circ}C$ regardless of paraquat treatment. The photochemical efficiency of photosystem II, Fv/Fm, of leaf discs of A. koreanum and D. morbifera fell remarkably at 35$^{\circ}C$. In the presence of paraquat, the Ev/Fm values fell slightly at 4$^{\circ}C$ in A. koreanum leaf discs and at 35$^{\circ}C$, in D. morbifera leaf discs. These results indicate that A. koreanum plants are more resistant to temperature stress or oxidative stress than D. morbifera plants although their photochemical efficiency falls slightly at 4$^{\circ}C$ in the presence of paraquat.
