  • In order to study the antioxidative action of the effectient garlic components, peroxidase and superoxide dismutase activity were compared through the in vitro and in vivo experiments. RESULTS : 1. Observing the effects on peroxidase activity of efficient components in vitro, garlic oil, alliin and ethanol fraction showed effective, which was similar to the trend of TBA value, peroxide value and induction time for the first period of lipoperoxide formation in vitro. 2. In vivo experiment with peroxidase activity, the ethanol fraction and garlic oil were effective when intraperitoneally administered as well as orally administered. 3. Considering the superoxide disutase activity in vitro, the garlic oil, alliin and ethanol fraction were effective in efficient components. But non-kaolin fraction inhibited the activity on the contrary. 4. In terms of the efefcts on superoxide dismutase activity in vivo, alliin and garlic oil wee effective in intraperitoneal adminstraton and the ethanol fraction and alliin in oral administration.

  • 1. Paraquat에 대(對)한 식물체(植物體)의 반응(反應)은 상이(相異)하며 감수성(感受性) 정도(程度)에 큰 차(差)가 인정(認定)되었다. injury, discooration, leaf area 및 leaf dry weight 등(等)으로 보아 Kwanggyo, Milyang 등의 콩 품종(品種)은 Hood나 $D_4$에 비(比)하여 내성(耐性)을 보였다. Paraquat에 대(對)한 옥수수품종간(品種間)의 반응폭(反應幅)은 아주 좁았다. 2. 밭에 우점잡초(優占雜草)인 바랭이, 쇠비름, 강아지풀은 paraquat에 감수성(感受性)인데 비(比)하여 마디풀, 명아주, 반하 등(等)은 저항성(抵抗性)을 나타내어 저항성원(抵抗性源)을 제공(提供)하였다. 3. Paraquat의 무독화(無毒化)에 관련하는 peroxidase의 활성(活性)은 Kwanggyo에서 높았으나 무처리(無處理)와 차이(差異)가 작았다. Peroxidase의 동위효소(同位酵素)는 Kwanggyo에 6개(個), Hood에 4개(個)였으며 paraquat을 처리(處理)해도 Kwanggyo의 pattern의 거의 변화(變化)가 없으니 Hood에는 다소(多少) 변화(變化)가 인정(認定)되었다. 4. paraquat에 대(對)한 식물체(植物體) 저항성(抵抗性) mechanism인 superoxide dismutase의 활성(活性)은 paraquat 100ppm 처리후(處理後) 48시간(時間)째 Kwanggyo는 48.6 units/$m\ell$, Hood는 34.2 units/$m\ell$이고 무처리(無處理)는 Kwanggyork 33.0 units/$m\ell$, Hoodsms 34.5 units/$m\ell$로서 SOD의 활성차(活性差)가 감수성(感受性)의 차(差)를 발현(發現)시켰다고 사료(思料)된다. 5. paraquat의 농도(濃度)가 증가(增加)할수록 카루스의 증식(增殖)이 크게 억제(抑制)되었으며 100ppm 처리(處理)에서 Kwanggyo 69.4%, Hood는 92.4%의 억제(抑制)를 보여 Hood가 Kwanggyo보다 약(約) 1.5(培) 감수성(感受性)을 나타내어 superoxide dismutase와 더불어 콩 품종간차(品種間差)를 잘 설명(說明)해 주고 있다.

Isolation and Characterization of the sod2$^{2+}$ Gene Encoding a Putative Mitochondrial Manganese Superoxide Dismutase in Schizosaccharomyces bombe

  • The fission yeast Schizosaccharomyces pombe contains two distinct superoxide dismutase (SOD) activities, one in the cytosol encoded by the $sod2^{+}$ gene and the other in mitochondria. The $sod2^{+}$ gene encoding putative mitochondrial manganese superoxide dismutase (MnSOD) was isolated from the S. pombe genomic library using a PCR fragment as the probe. The nucleotide sequence of the $sod2^{+}$ gene and its flanking region (4051 bp HindIII fragment) was determined. An intron of 123 nt in size was predicted and confirmed by sequencing the cDNA following reverse transcription PCR. The predicted Sod2p consists of 218 amino acid residues with a molecular mass of 24,346 Da. The deduced amino acid sequence showed a high degree of homology with other MnSODs, especially in the metal binding residues at the active site and their relative positions. The transcriptional start site was mapped by primer extension at 231 at upstream from the ATG codon. A putative TATA box(TATAAAA) was located 58 nt upstream from the transcriptional start site and putative polyadenylation sites were located at 1000, 1062, and 1074 nt downstream from the ATG start codon.

표고버섯 영양요구성 변이주의 전기영동법에 의한 Isozyme 비교 (Electrophoretic Patterns of Isozymes from the Mycelia of the Auxotrophs of Lentinula edodes)

  • 표고버섯(Lentinula edodes)의 동위효소 양상을 표고 연구의 기초 연구의 일환으로 실시하였다. 균사체의 Tris-HCl 완충액에 용해되는 세포내 효소를 nondenaturing polyacrylamide gel 전기영동 법으로 분리후, peroxidase, esterase, superoxide dismutase, acid phosphatase, alkaline phosphatase, alcohol dehydrogenase, ${\alpha}-amylase$ 효소 활성을 측정하였다. 표고는 peroxidase, esterase, superoxide dismutase, acid phosphatase 활성이 검출되었으며, 본 연구에서 사용한 방법으로 alkaline phosphatase, alcohol dehydrogenase, ${\alpha}-amylase$ 효소 활성은 검출되지 않았다. 표고버섯의 peroxidase, esterase band는 배양 기간에 따라 큰 차이가 없이 안정하였으며, 동위효소는 표고의 유전적 연구 및 원형질체 융합 후 융합체의 특성 연구에 중요하다.

Molecular Cloning and Expression of Sequence Variants of Manganese Superoxide Dismutase Genes from Wheat

  • Reactive oxygen species (ROS) are very harmful to living organisms due to the potential oxidation of membrane lipids, DNA, proteins, and carbohydrates. transformed E.coli strain QC 871, superoxide dismutase (SOD) double-mutant, with three sequence variant MnSOD1, MnSOD2, and MnSOD3 manganese superoxide dismutase (MnSOD) gene isolated from wheat. Although all QC 871 transformants grown at $37^{\circ}C$ expressed mRNA of MnSOD variants, only MnSOD2 transformant had functional SOD activity. MnSOD3 expressed active protein when grown at $22^{\circ}C$, however, MnSOD1 did not express functional protein at any growing and induction conditions. The sequence comparison of the wheat MnSOD variants revealed that the only amino acid difference between the sequence MnSOD2 and sequences MnSOD1 and 3 is phenylalanine/serine at position 58 amino acid. We made MnSOD2S58F gene, which was made by altering the phenylalaine to serine at position 58 in MnSOD2. The expressed MnSOD2S58F protein had functional SOD activity, even at higher levels than the original MnSOD2 at all observed temperatures. These data suggest that amino acid variation can result in highly active forms of MnSOD and the MnSOD2S58F gene can be an ideal target used for transforming crops to increase tolerance to environmental stresses.

Iron Superoxide Dismutase( Fe-SOD)를 생산하는 미생물의 선발 및 배양 (Selection and Cultivation of Microorganism Producing Iron Superoxide Dismutase(Fe-SOD))

  • 비교적 연구가 미비한 Fe-SOD의 효소화학적 특성 및 그 생리적 기능을 검토하기 위해 여러 종의 세균을 대상으로하여 Fe-SOD의 고생산균주를 screening하였다. 그 결과 Fe-SOD를 대량 세포내에 생성하는 Pseudomonas polycolor를 선발하여, 이 균주의 효소생산 최적 배양조건을 설정하였다. 본 균주가 생산하는 효소는 특이적인 저해제의 작용양식에 의해 Fe을 cofactor로 요구하는 Fe-SOD임이 밝혀졌다. SOD 생성을 위한 최적배지조성은 glycerin 3%, polypeptone 1%, meat extract 0.5%, KCI 0.2%이었고, 최적 초발 pH는 9.0이었으며, 이 조건에서 500ml용 shaking flask에 배지 100ml를 넣어 15시간 전 후 배양했을 경우가 효소생산량은 최대가 되었다.

  • The present investigation was undertaken to find out the anticancer activity of monoterpene compounds. Monoterpenes showed generally the inhibitory effect on the proliferation o f L1210 cancer cells (cytotoxicity). Geraniol was found to exibit the most potent cytotoxic effect on L1210 cells with an IC50 values of $0.67{\mu}g/ml$. On the other hand, geraniol proved to be capable of stimulating the macrophage proliferation (135% of control). When the life prolonging activity of geraniol by daily oral administration of 0.1~10${\mu}g/10{\mu}l/20$ g body weight to Sarcoma 180 bearing ICR mouse was examined, there was also a significant elevation of survival (best result of 134% of control). The contradictory effects of geraniol on the proliferation of L1210 cells and macrophages proved to be accompanied by the coincident alterations of RNS (reactive nitrogen species) related enzymes activities such as inducible nitric oxide synthase (Inos) in macrophages and ROS (reactive oxygen species) related enzymes activities such as superoxide dismutase (SOD) in L1210 cells, respectively.

사료 내 분말 청국장이 돌돔, parrootfish, Oplegnathus fasciatus의 간 내 superoxide dismutase 활성에 미치는 영향 (Effects of Dietary Cheongkukjang on Liver Superoxide Dismutase Activity of Parrotfish Oplegnathus fasciatus)

  • A four-week feeding trial was conducted to investigate the effects of dietary soybean meal (SBM) and powdered Cheongkukjang (CKJ) on non-specific immune responses of parrotfish Oplegnathus fasciatus. Three isonitrogenous (42% crude protein) and isocaloric (17.1 MJ/kg) diets were formulated to replace fish meal by 0, 25% SBM or 25% CKJ (designated as FM, 25SBM and 25CKJ, respectively). Ninety fish (initial body weight 122 g) were randomly allotted into nine 150 L tanks. One of the three experimental diets was fed to triplicate groups of fish for 4 weeks. After the feeding trial, no differences were observed in growth performances and feed utilization among fish groups. Liver superoxide dismutase activity of the fish fed CKJ containing diet was significantly higher than that of the control groups. DPPH radical scavenging and $Fe^{2+}-chelating$ activities of the experimental diets containing SBM or powdered CKJ were significantly higher than that of the control diet. The results of the present study suggest that dietary inclusion of powdered 25CKJ significantly increased liver superoxide dismutase activity and did not affect the growth performances, feed utilization, morphological parameters, as well as hematological values of parrotfish.

일산화탄소 폭로후 고압산소 투여가 흰쥐 신장에서의 malondialdehyde 함량과 catalase 및 superoxide dismutase 활성에 미치는 영향 (Effects of Hyperbaric Oxygen Treatment on the Malondialdehyde Level and Activities of Catalase and Superoxide Dismutase in the Kidney of the Rats Exposed to Carbon Monoxide)

  • In an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen free radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde (MDA) level and activities of catalase and superoxide dismutase in the kidney of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260 gm were used. Experimental groups consist of Control group (=breathing with air), HBO group (=exposed to hyperbaric oxygen 〔HBO, 3ATA, 100%〕 after air breath), CO group (=exposed to CO〔3,970 ppm〕after air breath), CO-Air group (=exposed to CO after air breath followed by air breath) and CO-HBO group (=exposed to CO after air breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of CO-induced nephrotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the kidney of the rats exposed to carbon monoxide.

오이 추출물에 존재하는 Superoxide Dismutase의 열안정성 (Thermostability of Superoxide Dismutase from Cucumber(Cucumis sativa))

  • The superoxide dismutase(SOD) in peeled pericarp of cucumber was most stable at pH 8.0 and relatively stabe between pH 5.0 and 9.0. The enzyme was stable up to 6$0^{\circ}C$ and retained 12% by heat treatment at 10$0^{\circ}C$ for 5 min. At pH 2.0, the peeled pericarp enzyme activity was decreased to 10% by incubation for 3 hrs. However, the enzyme activity was increased above 25% after incubating the enzyme at pH 7.0 for 6 hrs. Retention of SOD activity in cucumber by various heating methods was also measured. The residual SOD activities of peeled pericarp and whole cucumber was estimated to be 25% and 27% after blanching(2 min), respectively. The skin enzyme retained 53% of its activity after steaming (3 min). When the peeled pericarp enzyme was incubated at 4$^{\circ}C$ for 20 days, the enzyme activity remained about 81%. However, when the enzyme incubated at 3$0^{\circ}C$ for 20 days, the peeled pericarp enzyme activity decreased to 17% of its original activity. The enzyme activity of peeled pericarp cucumber was not changed after exhaustive dialysis for 3 days, which indicated that the SOD activity in cucumber seems to have molecular weight above 12,000.
