초임계수 산화는 난분해성 유기화합물의 분해를 위한 공정으로 각광받고 있다. 본 연구에서는 연속반응기에서 온도 $387-500^{\circ}C$, 압력 250 bar의 초임계수 조건하에서(EDTA 분해효율) 체류시간 15.9-88.9초의 범위의 EDTA 분해효율을 측정하였다. 이때 산화제로는 과산화수소($H_2O_2$)를 사용하였다. EDTA의 분해효율은 온도 및 산화제투입량의 증가에 따라 상승하였으며, 반응물 도입유속의 감소 즉, 체류시간의 증가에 따라서 상승하였다. 본 연구결과 온도 $500^{\circ}C$, 압력 250 bar, 산화제 투입량 400%의 조건에서 최대 99.6%의 분해효율을 나타내었다. 분해효율에 미치는 온도의 영향이 산화제 투입량 증가의 영향보다 컸으며, 5,000 mg/L의 EDTA($COD_{Cr}$로서 3,063 mg/L)의 99% 이상 분해효율은 온도 $500^{\circ}C$와 압력 250 bar의 초임계수 산화조건에서 산화제투입량 200% 이상 및 체류시간 40.1초 이상에서 얻을 수 있었다.
본 연구는 로페라마이드로 처리된 흰쥐의 결장점막에 1,2-dimethylhydrazine (DMH)로 유도시 aberrant crypt foci(ACF)에 대한 길항 작용 효과를 갖는 초임계 열처리된 무 복합추출물의 화학적 암 예방 효과를 알아보고자 하였다. 초임계 열처리된 무 복합물추출물을 DMH유발 발암의 개시(12주 동안) 또는 개시 후 단계(9주 동안)에 식수(3.2 mg/ml)에 투여 하였다. 12주 동안 DMH와 함께 초임계 열처리된 무 복합추출물 투여군은 비정상적인 선와(전신경병변)의 총 수와 ACF의 수를 각각 35 %와 30 %로 크게 줄였다. 12주 동안 초임계 열처리된 무 복합추출물을 투여한 흰쥐의 결장에서 4가지 이상의 이상선와소(ACF)가 있는 ACF의 수가 현저히 감소하였다. 9주 동안 개시후 초임계 열처리된 무 복합추출물을 투여한 그룹에서, 4 이상의 이상선와소(ACF)의 수만이 현저히 감소하였다. 12주 동안 단독으로 투여한 초임계 열처리된 무 복합추출물은 ACF의 발생을 유발하지 않았다. 이러한 결과는 초임계 열처리된 무 복합추출물이 화학적으로 유도된 흰쥐의 결장 발암에 화학적 암예방 효과를 가지고 있음을 시사한다.

Having proving the physiologic functions of green tea, high consumption of coffee is shifting to green tea. Because of the carcinogenic effect of the organic solvents, decaffeine processing with supercritical carbon dioxide has industrialized and have an advantage in environment-friendly and minimized flavor loss. Decaffeined green tea using supercritical carbon dioxide is considered to be safe but there are not enough study, We investigated the chromosome aberration test with mammalian cell line, CHL. When the cells were treated with 5000, 2000, 1000 ${\mu}g/ml$ and compared with the negative controls, there were no significant (P>0.05) increased chromosome aberration. Same results was observed when adding S9 mixture or not. As a result, water extract of decaffeined green tea using supercritical carbon dioxide does not induce chromosome aberration.

In a SCWR (SuperCritical pressure Water cooled Reactor), the coolant temperature initially at below the pseudo-critical temperature at the bottom of a reactor core increases as the coolant flows upward through the sub-channels of the fuel assemblies, and it finally becomes higher than the pseudo-critical temperature when it leaves the reactor core. At certain conditions, heat transfer deterioration occurs near the pseudo-critical temperature and it may cause a drastic rise of the fuel surface temperature resulting a fuel failure. Therefore, an accurate estimation of the heat transfer coefficient is very important for the thermal-hydraulic design of a reactor core. An experiment on heat transfer to the vertically upward flowing $CO_2$ at a supercritical pressure in a circular tube were performed at KAERI. The internal diameter of the test section is 6.32 mm, which corresponds to the hydraulic diameter of a sub-channel in the conceptional design proposed by KAERI. The test range of the mass flux is 285 to 1200 kg/m$^2$s and the maximum heat flux is 170 kW/m$^2$. The inlet pressure is maintained at 8.12 MPa, which is 1.1 times the critical pressure. A new correlation, which covers both the normal and deterioration heat transfer regimes was proposed and compared with the estimations by exiting correlations.
Optimal extraction conditions such as pressures, temperatures, and modifiers on glycyrrhizin extraction from licorice were investigated using supercritical $CO_2\;(SC-CO_2)$ at 3 mL/min flow rate. Morphology of licorice tissue, after glycyrrhizin extraction, was examined by SEM, and absolute density ($g/cm^3$) measurement and glycyrrhizin content were determined by HPLC. Pure $SC-CO_2$ had no effect on glycyrrhizin extraction, but recovery of glycyrrhizin ($32.66{\pm}0.77%$) was enhanced when water was used as modifier. The highest recovery was $97.22{\pm}2.17%$ when 70% (v/v) aqueous methanol was added to 15% (v/v) $SC-CO_2$ at 50 MPa and $60^{\circ}C$. Under optimal extraction conditions, 30 MPa pressure and $60^{\circ}C$ heating temperature, glycyrrhizin recovery reached maximum ($102.67{\pm}1.13%$) within 60 min. Licorice tissue was severely damaged by excessive swelling, and absolute density of licorice residues was highest when aqueous methanol was used as a modifier.
In this paper, the mechanism identification and the avoidance measures on the phenomena of transient acoustic vibration amplified at the water-supply piping system to regulate the steam temperature of the boiler reheater in 500MW class supercritical power plant are presented. The pressure pulsation waves induced by the impeller passing of two feed-water pumps with five blades are coincident with the local acoustic modes of boiler reheater water-supply piping system. There are the phenomena amplified at the peaks of 5X, 10X, 15X and 20X in spectrums of piping vibration, sound pressure, and the feed-water's pressure pulsation waves. The shut-off device is installed in the piping system for the interception of pressure pulsation waves transmitted from two feed-water pumps and the modified design change of the piping layout is applied for the acoustic resonance avoidance. The acoustic natural frequencies are separated from the harmonics of pressure pulsation waves induced by the pump impellers passing through the design change of the span length. The acoustic vibration is gone by resonance avoidance measures. As a result, more than 20 dBA reduction is achieved from 100 dBA to 80 dBA.
본 연구에서는 연속형 초임계수 분해장치를 이용하여 처리온도 별로 현사시목재의 당화 가능성과 현사시목재 각 성분의 분해특성을 분석하였다. 미세하게 분쇄한 현사시목분을 물의 초임계압력(23MPa) 조건하에서 각각 아임계온도$(275^{\circ}C,\;325^{\circ}C)$와 초임계온도$(375^{\circ}C,\;415^{\circ}C)$에서 각각 2분간 처리하였다. 갈색을 띤 현사시목분의 액상분해산물에는 어느 정도의 미분해 고형분이 포함되어 있었다. 초임계수 분해온도가 높을수록 현사시목분의 분해율은 증가하여 초임계조건인 $375^{\circ}C$에서는 현사시목분의 $94\%$까지 분해되었다. 각 처리조건에서 생성된 환원당량을 DNS법으로 측정한 결과, 처리온도가 상승함에 따라 생성된 환원당량은 점차 감소하는 경향을 나타내었다. 이는 반응 초기에 생성된 환원당들이 높은 온도의 반응기를 통과하면서 열분해와 유사한 화학적 반응에 의해 더욱 분해되어 퓨란계 화합물로 전환된 것으로 예측되었다. 리그닌의 분해특성을 살펴보기 위하여 액상의 분해산물을 ethylacetate로 추출하여 유기용매 가용부를 GC-MS로 분석하였다. 리그닌의 분해산물은 대부분 페놀성 유도체인 vanillin, guaiacol, syringaldehyde, 4-prophenyl syringol과 dihydrosinapyl alcohol등으로 확인되었으며, 이들의 농도는 처리온도가 상승함에 따라 증가하는 것으로 나타났다. 이러한 리그닌 분해산물은 리그닌고분자의 주요 결합인 에테르 결합이 높은 온도조건하에서 분해에 의한 것으로 예측되었다.
본 연구의 목적은 온도조건 별로 밀도요동 초임계 유체를 이용하여 추출한 대마종자 추출물의 수율 측정과 생리활성능을 평가하고, 리포좀 제형으로 난용성 추출물의 용해를 가능하게 하여 경피투과도를 증진시키는 데 있다. 수율 측정 결과, 60℃로 추출한 대마종자가 가장 높게 나타났고, 항산화 실험에서 total polyphenol content assay, DPPH radical scavenging assay, ABTS+ radical scavenging assay 결과, 최고농도에서 45℃ 추출물의 폴리페놀 함량과 radical 소거능이 가장 크게 나타났다. 항균실험 결과, P ropionibacteium acnes 균주에서만 clear zone이 나타났다. 추출물을 3차 정제수에 용해시킨 제형보다 리포좀으로 캡슐화한 제형에서 particle size의 감소와 zeta potential의 절댓값 증가를 확인하여 제형 안정성을 확인하였고, 경피투과도의 증진 또한 확인하였다. 이러한 실험결과를 바탕으로 난용성인 초임계 대마종자 추출물을 화장품의 기능성 천연물 소재로써 활용 가능성을 확인하였다.
Paclitaxel is a taxane diterpene amide, which was first extracted from the stem bark of the western yew, Taxus brevifolia. This natural product has proven to be useful in the treatment of a variety of human neoplastic disorders, including ovarian cancer, breast and lung cancer. Paclitaxel is a highly hydrophobic drug that is poorly soluble in water. It is mainly given by intravenous administration. Therefore, The pharmaceutical formulation of paclitaxel ($Taxol^{(R)}$; Bristol-Myers Squibb) contains 50% $Cremophor^{(R)}$ EL and 50% dehydrated ethanol. However the ethanol/Cremophor EL vehicle required to solubilize paclitaxel in $Taxol^{(R)}$ has a pharmacological and pharmaceutical problems. To overcome these problems, new formulations for paclitaxel that do not require solubilization by $Cremophor^{(R)}$ EL are currently being developed. Therefore this study utilized a supercritical fluid antisolvent (SAS) process for cremophor-free formulation. To select hydrophilic polymers that require solubilization for paclitaxel, we evaluated polymers and the ratio of paclitaxel/polymers. HP-${\beta}$-CD was used as a hydrophilic polymer in the preparation of the paclitaxel solid dispersion. Although solubility of paclitaxel by polymers was increased, physical stability of solution after paclitaxel/polymer powder soluble in saline was unstable. To overcome this problem, we investigated the use of surfactants. At 1/20/40 of paclitaxel/hydrophilic polymer/ surfactant weight ratio, about 10 mg/mL of paclitaxel can be solubilized in this system. Compared with the solubility of paclitaxel in water ($1\;{\mu}g/mL$), the paclitaxel solid dispersion prepared by SAS process increased the solubility of paclitaxel by near 10,000 folds. The physicochemical properties was also evaluated. The particle size distribution, melting point and amophorization and shape of the powder particles were fully characterized by particle size distribution analyzer, DSC, SEM and XRD. In summary, through the SAS process, uniform nano-scale paclitaxel solid dispersion powders were obtained with excellent results compared with $Taxol^{(R)}$ for the physicochemical properties, solubility and pharmacokinetic behavior.
