클로탈라리아 시들음병을 일으키는 Fusarium udum의 특성 (Characterization of Fusarium udum Causing Fusarium Wilt of Sunn Hemp in Korea)

  • 클로탈라리아는 국내에서 토양개량을 위한 질소고정, 토양침식 감소, 잡초 및 선충 억제를 위해 풋거름작물로 이용하고 있다. 2014년, 클로탈라리아를 풋거름작물로 재배하는 완주 지역의 포장에서 시들음 증상이 관찰되었다. 감염된 식물체의 잎은 아래잎부터 황화 되면서 시들기 시작하였고, 위쪽 잎도 황화 되었으며, 결국 식물체는 완전히 고사하였다. 감염된 식물체 줄기에서 어두운 색의 자낭각이 다수 관찰되었고, 이 자낭각으로부터 자낭포자를 단포자 분리하여 6개 균주를 분리하였다. 균학적 특성에 의해 분리균은 Fusarium udum(완전세대: Gibberella indica)로 동정되었다. 대형포자의 정단세포는 대부분 갈고리 모양으로 굽어 있고, 소형포자는 단경자에서 false head 상으로 형성되었다. 후벽포자는 균사에서 단일 혹은 무리지어 풍부하게 형성되었다. 이와 같은 동정 결과는 translation elongation factor 1 alpha(TEF), calmodulin (CAL), histone 3 (HIS3) 유전자 염기서열 분석으로 재확인되었다. 그 결과, 분리 균주는 NCBI GanBank에 등록된 F. udum과 TEF 유전자는 94.4~96.2%, CAL 유전자는 99.7%, HIS3 유전자는 99.6~99.8%의 상동성을 보였다. 클로탈라리아와 두 품종의 콩을 대상으로 포자현탁액을 토양에 관주 접종하여 병원성 검정을 수행한 결과, 접종 14~21일 이내에 접종한 클로탈라리아와 태광콩에서 병징이 관찰되었다. 따라서 이 병을 F. udum에 의한 클로탈라리아 시들음병으로 명명하고자 제안하며, 국내에서 처음으로 보고한다.

  • The purpose of this study was to trace and identify termilogies recorded im kei lim yu sa. ma(麻), kyun(絹), po(布), je(苧), and je-po(苧布) were included in this study. For the analysis of this research comparative analytical methods was employed. Some significant findings and results of this study can be summarized as follows : 1. Text : hemp is called san(麻曰三). The word san of Koryo was derived from Indian sann, san, sana, sunn, 2. Text : Kyun is called jyi or kib(絹曰及). The word jyi(及) of Koryo was from jyi or kib() of Silla. 3. Text : Hemp cloth is called bey(bei), pey(pei), bai(布曰背). Be (베) has two meanings of hemp cloth and cloth in Korean. In the text it is interpreted as hemp cloth. 4. Text : Ramie or China grass is called mar, maw, mao, mu (毛). (苧曰毛). It is considered that the word mau(mo in Korean) (毛) in text is correct and word mau(毛) meaning China grass of Koryho is derived from mah or mak that is an old Egyptian name for flax or linen. The word mau(mo) (毛) of Koryo seems to be a transliteration of mah(or mak) in Korean or a variety of it. 5. Raime cloth or China grass cloth is clalled mau shy (苧布曰毛施). Silmar words to mau shy(毛施) of Koryo were found in Chinese, Manchurian and japanese as well as in Ancient Hebrew.

Cloning and Characterization of a new tobamovirus infecting Hibiscus rosa-sinensis

  • A near full-length sequence of a new tobamovirus infecting Hibiscus rosa-sinensis L. was determined. The genome consists of 58 nucleotides (nt) 5' UTR, followed by a 4.9 kb ORF which methyl transferase helicase domain (128 kDa), readthrough protein RNA dependent RNA polymerase (RdRp) 185 kDa and a 52 kDa protein. The 128 kDa protein had a maximum homology of 51.4 % to TMGMV and amino acids (an) were 54.3 % identical to TMV- vulgare strain. The 185 kDa RdRp had a maximum homology of 53.5% to TMV-Ob and KGMMV-Y and a 59.6% homology at the an level to CGMMV-SH. The MP gene encodes 282 aa and its theoretical molecular weight is 30.4 kDa. The nt and an sequence identities of MP ranged from 38.8% to 43.9% and 30.9% to 37.9%, respectively. The CP gene encodes 163 residues and with a theoretical molecular weight of 18.2 kDa The (nt) and aa sequences of the CP were 46.9 % to 51.6% and 45.3% to 57.1% identical to other tobamoviruses, respectively. The predicted virion origin of assembly (OAS) was located in the CP gene. Phylogenetic trees generated based on the nt and as sequences of RdRp, MP and CP genes indicated that this new virus clustered with subgroup II tobamoviruses. Although the CP ORF of this virus shared a high nt and aa sequence identity with Sunn-hemp mosaic virus (SHMV), Western analysis showed that it is serologically unrelated to SHMV. We propose the name Hibiscus virus S (HVS) for this Singapore isolate. This is the first report on a near full-length sequence of a Tobamovirus that infects hibiscus.
