SO42- Uptake and Assimilation in Forage Rape (Brassica napus) (유채의 SO42- 흡수 및 동화에 관한 연구)

  • To investigate the sulfate utilization efficiency that has been examined in rape (Brassica napus L.) cultivars (cv. Akela and Colosse). During 72h of treatment, in two cultivars, ${SO_4}^{2-}$ uptake was significantly higher in complete S condition (2.0 mM ${SO_4}^{2-}$) than that of the S-deficient condition (2.0 mM ${SO_4}^{2-}$). In young leaves, ATP sulfurylase activity increased after S-deficient treatment. However, in old leaves, ATP sulfurylase activity was not significantly changed in Akela. Glutathione concentration in young leaves significantly increased in all cultivars examined under S-deficient condition. The rate of increase in glutathigne concentration caused by S-deficiency treatment was higher in Akela. However, in old leaves, the glutathione concentration in Akela significantly decreased. The results suggest that ${SO_4}^{2-}$ uptake and ATP sulfurylase in rape plants were species specific, and that ${SO_4}^{2-}$ uptake was highly related to its assimilation into S containing compound such as glutathione.

  • A sclerosing stromal tumor of the ovary is an extremely rare benign tumor; it usually is found during the second and third decades of life. Patients present with pelvic pain or a palpable abdominal mass. Hormonal effects such as masculinization are uncommon. Here, an 11-year old premenarchal girl presented with deepening of the voice. In addition, clitoromegaly and hirsutism with a male suprapubic hair pattern were observed. The laboratory findings showed that the testosterone level was elevated to 3.67 ng/ml, andostenedione to above 10 ng/ml, dehydroepiandrosterone-sulfate to 346 ${\mu}g$/dl and 17-hydroxy progesterone (17-OHP) to 11.28 ng/ml. The chromosome evaluation revealed a 46,XX female karyotype. An adrenocorticotropic hormone stimulation test was performed. The 17-OHP to cortisol ratio in 30 minutes was 0.045, which suggested a heterozygote for the 21-hydroxylase deficiency. However, the CYP21A2 gene encoding steroid 21-hydroxylase showed normal. The pelvic ultrasound showed a heterogeneous mass consisting of predominantly solid tissue in the pelvic cavity. The pelvic magnetic resonance imaging revealed an $8.9{\times}6.2{\times}6.6$ cm mass of the left ovary. A left oophrectomy was performed and microscopic examination confirmed a sclerosing stromal tumor. Immunohistochemical studies showed that the tumor was positive for smooth muscle actin and vimentin, but negative for S-100 protein and cytokeratin. Following surgery, the hormone levels returned to the normal range and the hirsutism resolved.

The effect of lipopolysaccharide from uropathogenic Escherichia coli on the immune system, testis tissue, and spermatozoa of BALB/c mice

  • Objective: Uropathogenic Escherichia coli is known to cause urinary tract infections, and the endotoxin (lipopolysaccharide [LPS]) of this bacterium may cause deficiencies of sperm quality and morphology. In the present study, the effects of LPS on mouse sperm were studied, and the levels of interleukin (IL)-17A and possible changes in testis tissue were evaluated. Methods: LPS of uropathogenic E. coli was extracted using the methanol-chloroform method, followed confirmation using sodium dodecyl sulfate-polyacrylamide electrophoresis. Purified LPS (100 ㎍/kg) or phosphate-buffered saline was injected intraperitoneally into BALB/c mice for 7 days consecutively in the test and control groups, mice were sacrificed on days 3, 7, and 42 after the first injection. Blood was tested for levels of IL-17A using the enzyme-linked immunosorbent assay method. Testis tissue and sperm were collected from each mouse and were studied according to standard protocols. Results: The mean sperm count and motility significantly decreased (p=0.03) at 3, 7, and 42 days after the injections. The level of IL-17A in the test groups increased, but not significantly (p=0.8, p=0.11, and p=0.15, respectively). Microscopic studies showed no obvious changes in the morphology of the testis tissue; however, significant changes were observed in the cellular parenchyma on day 42. Conclusion: LPS can stimulate the immune system to produce proinflammatory cytokines, resulting in an immune response in the testis and ultimately leading to deficiency in sperm parameters and testis tissue damage. In addition, the presence of LPS could significantly impair sperm parameters, as shown by the finding of decreased motility.

  • Taurine is a type of sulfur-containing amino acid having a sulfate functional group, that is biosynthesized from cysteine. It is mainly distributed in high concentrations in animal tissues and is known to have various effects such as osmotic pressure control, calcium control, anti-inflammatory, antioxidant, and hepatocellular protection. Also, taurine deficiency causes a variety of symptoms, including visual impairment. In particular, in the case of cats, taurine is not biosynthesized and must be supplied through food, so it is classified as an essential amino acid. In this study, an analysis method using mass spectrometry was developed instead of the commonly used derivatization method to quickly, environmentally, and precisely analyze taurine in various animal feeds. The developed analytical method showed good linearity (R2 > 0.99), accuracy (81.97-105.78%), and precision (0.07-12.37%). In addition, the developed method was further verified through quantitative comparison with the derivatization method. This developed method was used in the determination of taurine in 20 animal feed samples obtained from South Korea. The levels of taurine found ranged from 81.53 to 6,743.53 mg/kg. The developed analysis method will be used for the detection and quantification of taurine in domestic feed.

The Mechanism of Iron Transport after Intratracheal Instillation of Iron in Rats (랏트의 기관내 Fe 노출후 Fe 이동에 대한 연구)

  • Objectives : Iron (Fe) is an essential element in biological processes; however excessive Fe is harmful to human health. Some air pollutants contain a high level of Fe, and the human lung could therefore be over-exposed to Fe through inhaled air pollutants. This study was performed to investigate the role of metal transporters (divalent metal transporter 1, DMT1, and metal transporter protein 1, MTP1) in the lung under the environments of Fe deficiency in the body and Fe over-exposure in the lung. Methods : Rats were fed Fe deficient (FeD, 2-6 mg Fe/kg) or Fe supplemented (FeS, 120 mg Fe/kg) diet for 4 weeks, followed by a single intratracheal instillation of ferrous sulfate at low (10 mg/kg) or high (20 mg/kg) dose. Fe concentration was analyzed in the serum, lung and liver, and histopathological findings were observed in the lung at 24 hours after Fe administration. The level of DMT1 and MTP1 expression in the lung was analyzed by RT-PCR. Also, the effect of Fe deficiency in the body was evaluated on the level of Fe concentration and metal transporters compared to FeS-diet fed rats at the end of 4-week FeD or FeS diet. Results : The 4-week FeD diet in rats induced an Fe deficiency anemia with decreased serum total Fe, increased unsaturated Fe binding capacity and hypochromic microcytic red blood cells. The concentration of Fe in the lung and liver was lower in the FeD-diet fed rats than in the FeS-diet fed rats. The level of metal transporters mRNA expression was higher in the FeD-diet fed rats than in the FeS-diet. The concentration of Fe in the lung was increased in a dose-dependent pattern after intratracheal instillation of Fe into the rats, while the level of Fe in the serum and liver was not increased in the low-dose Fe administered rats. Therefore, DMT1 and MTP1 mRNA was highly expressed in both FeD-diet and FeS-diet fed rats, after intratracheal instillation of Fe. Conclusions : DMT1 and MTP1 mRNA were more highly expressed in FeD-diet fed rats than in FeS-diet fed rats. The over-exposure of Fe intratracheally induced high expression of metal transporters and increased Fe deposition in the lung in both FeD-diet and FeS-diet fed rats, but did not increase the Fe level of the serum and liver in low-dose Fe administered rats. These results suggest that the role of metal transporters in the lung might be different in a part from the duodenum under the environment of over-exposure to Fe.

  • Using tracer technique of $^{65}Zn$, a field experiment has been carried out to evaluate the efficiency of zinc fertilizer by rice plant grown under flooded conditions. The treatments include zinc slufate mixed throughout the soil with and without organic matter, combined urea-zinc fertilizer (N: 37.7%, Zn: 3.1%), and surface application at transplaning and two weeks after transplanting at the rate of 5kg Zn/ha respectively. Other treatments were zinc sulfate mixed throughout the soil at the rate of 10 kg and 20 kg Zn/ha respectively. Root dipping in 2 % ZnO suspension, only organic matter added, and control were also included. There was not much difference in rough grain yield between zinc levels and different application methods, but the highest yield was obtained from the treatment of the root dipping in 2% ZnO suspension. Among the 5kg Zn/ha treatments, the highest total zinc yield was observed from the zinc mixed throughout the soil. The organic matter treatment seemed to reduce the zinc fertilizer efficiency. In case of the zinc fertilizer levels, 5kg Zn/ha mixed throughout the soil showed the highest zinc fertilizer efficiency as compared with 10kg and 20kg Zn/ha treatments.

  • A field experiment has been carried out to evaluate the residual effect of zinc fertilizers by rice plant grown under flooded conditions in the field. The results obtained are summarized as fellows ; Residual effect of zinc fertilizers on yields of rough and hulled grains showed slight increases. Effect of zinc application methods on yields of the grains were shown that zinc mixed treatment could be more effectively utilized than treatment of zinc on the soil surface. In case of levels of zinc application, 5 kg zinc per hectare represented high yields of the grains than those obtained from 10kg and 20kg zinc placement per hectare respectively. Regarding the form of zinc fertilizers, the urea-zinc complex showed less effective on yields of the grains than did the zinc sulfate. This phenomenon was consistent with the previous result. Yields of total zinc in rice plant grown on the rice straw added soils (Treatment No. 2 and 8) and the urea-zinc complex treated soil were increased markedly as compared to those data obtained from the previous year. The percentage of zinc derived from fertilizer decreased largely as compared to that of the first year crop. The yield of fertilizer zinc in rice plant decreased slightly in the most zinc treatments but in the case of treatments of zinc mixed with the straw added soil and the urea-zinc complex increased reversely as compared to the previous results. The mixed application of zinc with soil showed higher yield of fertilizer zinc than the soil surface placement. Approximately from 4.6 to 24.3 per cent of zinc taken up by rice plants were derived from the fertilizer zinc. Zinc fertilizer use efficiency ranged from 0.213 to 0.584 per cent when 5 kg zinc per hectare applied.

Effects of Iron from an Amino Acid Complex on the Iron Status of Neonatal and Suckling Piglets

  • This experiment was conducted to investigate the effects of iron from an amino acid complex (Availa-$Fe^{\circledR}$) on the iron status of neonatal and suckling piglets. A total of 24 gestating sows (Landrace${\times}$Large White) were randomly allocated to three dietary treatments. The control diet contained 80 mg $kg^{-1}$ Fe from ferrous sulfate heptahydrate ($FeSO_4$.$7H_2O$), while the two experimental diets were supplemented with an additional 120 mg $kg^{-1}$ Fe from Availa-$Fe^{(R)}$ or $FeSO_4$.$7H_2O$, respectively. The lactating sows remained the same iron treatments as gestating sows, while neonatal piglets of 24 litters born from the above sows were allotted to another three treatments. Piglets from the sows of the control treatment were fed basal diet with no supplemental Fe as control treatment, but were injected with 100 mg Fe as Fe dextran at birth. Piglets from the sows of Availa-$Fe^{(R)}$ or $FeSO_4$.$7H_2O$ treatments were supplemented with 120 mg $kg^{-1}$ iron from Availa-$Fe^{(R)}$ or $FeSO_4$.$7H_2O$, respectively. The total born alive and weaned, and the average piglets weight at birth and at weaning were not significantly affected by the sow' dietary treatments (p>0.05). Iron from Availa-$Fe^{(R)}$ did not demonstrate a statistically significant improvement in hemoglobin concentration, hematocrit and plasma iron of sows on day 90 and 105 of pregnancy and the milk iron of sows during lactation (p>0.05). Neonatal piglets in the Availa-$Fe^{(R)}$ treatment had a significantly higher hemoglobin concentration (p<0.05) and higher hematocrit and plasma iron (p>0.05) than those in the other two treatments, respectively. The hemoglobin of suckling piglets in the Availa-$Fe^{(R)}$ treatment was higher than that of piglets in $FeSO_4$.$7H_2O$ treatment on day 28 (p<0.05). The total iron binding capacity of piglets in Availa-$Fe^{(R)}$ treatment was lower than that of piglets in the control and $FeSO_4$.$7H_2O$ treatment on day 14 (p<0.05), but there was not a statistically significant difference among three treatments on day 28 (p>0.05). However, the hemoglobin and hematocrit of suckling piglets injected with Fe were higher than those of piglets in the other two treatments (p<0.05). This study indicated that the addition of 120 mg $kg^{-1}$ iron from amino acid complex into the diets improved iron status of neonatal and nursing piglets more effectively than the addition of 120 mg $kg^{-1}$ iron from $FeSO_4$.$7H_2O$, however, this improvement of the organic Fe was not sufficient to replace the Fe injection for prevention of iron-deficiency anemia.
