In this study we purified a fibrinolytic enzyme from Cordyceps militaris using a combination of ion-exchange chromatography on a DEAE Sephadex A-50 column, gel filtration chromatography on a Sephadex G-75 column, and FPLC on a HiLoad 16/60 Superdex 75 column. This purification protocol resulted in a 191.8-fold purification of the enzyme and a final yield of 12.9 %. The molecular mass of the purified enzyme was estimated to be 52 kDa by SDS-PAGE, fibrin-zymography, and gel filtration chromatography. The first 19 amino acid residues of the N-terminal sequence were ALTTQSNV THGLATISLRQ, which is similar to the subtilisin-like serine protease PR1J from Metarhizium anisopliae var. anisopliase. This enzyme is a neutral protease with an optimal reaction pH and temperature of 7.4 and $37^{\circ}C$, respectively. Results for the fibrinolysis pattern showed that the enzyme rapidly hydrolyzed the fibrin $\alpha$-chain followed by the $\gamma$-$\gamma$ chains. It also hydrolyzed the $\beta$-chain, but more slowly. The A$\alpha$, B$\beta$, and $\gamma$ chains of fibrinogen were also cleaved very rapidly. We found that enzyme activity was inhibited by $Cu^{2+}$ and $Co^{2+}$, but enhanced by the additions of $Ca^{2+}$ and $Mg^{2+}$ ions. Furthermore, fibrinolytic enzyme activity was potently inhibited by PMSF and APMSF. This enzyme exhibited a high specificity for the chymotrypsin substrate S-2586 indicating it's a chymotrypsin-like serine protease. The data we present suggest that the fibrinolytic enzyme derived from the edible and medicinal mushroom Cordyceps militaris has fibrin binding activity, which allows for the local activation of the fibrin degradation pathway.
In this study, we have cloned a novel cDNA encoding for a papain-family cysteine protease from the Uni-ZAP XR cDNA library of the polychaete, Periserrula leucophryna. This gene was expressed in Escherichia coli using the T7 promoter system, and the protease was characterized after partial purification. First, the partial DNA fragment (498 bp) was amplified from the total RNA via RT-PCR using degenerated primers derived from the conserved region of cysteine protease. The full-length cDNA of cysteine protease (PLCP) was prepared via the screening of the Uni-ZAP XR cDNA library using the $^{32}P-labeled$ partial DNA fragment. As a result, the PLCP gene was determined to consist of a 2591 bp nucleotide sequence (CDS: 173-1024 bp) which encodes for a 283-amino acid polypeptide, which is itself composed of an 59-residue signal sequence, a 6-residue propeptide, a 218-residue mature protein, and a long 3'-noncoding region encompassing 1564 bp. The predicted molecular weights of the preproprotein and the mature protein were calculated as 31.8 kDa and 25 kDa, respectively. The results of sequence analysis and alignment revealed a significant degree of sequence similarity with other eukaryotic cysteine proteases, including the conserved catalytic triad of the $Cys^{90},\;His^{226},\;and\;Asn^{250}$ residues which characterize the C1 family of papain-like cysteine protease. The nucleotide and amino acid sequences of the novel gene were deposited into the GenBank database under the accession numbers, AY390282 and AAR27011, respectively. The results of Northern blot analysis revealed the 2.5 kb size of the transcript and ubiquitous expression throughout the entirety of the body, head, gut, and skin, which suggested that the PLCP may be grouped within the cathepsin F-like proteases. The region encoding for the mature form of the protease was then subcloned into the pT7-7 expression vector following PCR amplification using the designed primers, including the initiation and termination codons. The recombinant cysteine proteases were generated in a range of 6.3 % to 12.5 % of the total cell proteins in the E. coli BL21(DE3) strain for 8 transformants. The results of SDS-PAGE and Western blot analysis indicated that a cysteine protease of approximately 25 kDa (mature form) was generated. The optimal pH and temperature of the enzyme were determined to be approximately 9.5 and $35^{\circ}C$, respectively, thereby indicating that the cysteine protease is a member of the alkaline protease group. The evaluation of substrate specificity indicated that the purified protease was more active towards Arg-X or Lys-X and did not efficiently cleave the substrates with non-polar amino acids at the P1 site. The PLCP evidenced fibrinolytic activity on the plasminogen-free fibrin plate test.
소포체는 세포막의 합성뿐만 아니라 세포막에 존재하거나 세포외로 분비되어져야 할 단백질을 합성하는 세포내 소기관이다. 소포체에서 단백질이 합성되어질 경우 이황화결합이 형성되고 glycosylation 등의 수식이 일어나며, 이와 동시에 folding과 assembly과정을 거쳐 삼차원적 구조로 성숙이 되는데 이 과정은 folding enzyme과 molecular chaperone의 도움을 받아 이루어진다. 소포체 내에 존재하는 molecular chaperone 중 가장 잘 알려진 것으로 BiP이 있다. BiP의 기능은 N-terminus의 ATPase domain에 의해 조절되고 ATPase domain은 이것과 선택적으로 결합하는 조절인자에 의해 ATPase의 활성이 영향을 받는다. BiP의 핵산치환조절인자로서 발견된 BAP은 ATPase domain에 결합된 ADP를 ATP로 치환하는 것으로 기능이 알려져 있다. 이 BAP의 핵산치환기능이 BiP의 샤페론 작용에 어떤 영향을 미치는지를 in vitro에서 항체 heavy chain을 이용하여 알아보았다. BAP은 ATP보다 ADP가 결합되어 있는 BiP과 더 잘 결합을 하며, in vitro에서 BiP과 결합하고 있는 unfolded 단백질을 BAP은 BiP으로부터 해리하였다. 또한 소포체내에 존재하는 Hsp70 homologue chaperone인 BiP과 Grp170에 대한 BAP의 결합특이성을 anti-Grp170과 anti-BAP 항체로 co-immunoprecipitation을 하여 확인한 결과 BAP은 Grp170과 결합을 하지 않았다. 따라서 BAP은 ER내에 존재하는 동일한 family group에 속하는 Grp170과 BiP에 대하여 BiP에만 특이성을 갖는 것으로 나타났다.
본 연구에서는 된장으로부터 혈전용해효소(subtilisin CP-1) 생산 균주를 단리하여 16S rRNA 유전자 분석과 생화학적 분석을 통하여 동정하여 Bacillus sp. CP-1로 명명하였으며, TSB와 LB 배지를 이용하여 혈전용해효소의 생산에 적합한 Bacillus sp. CP-1 배양 배지에 대하여 검토하였다. 그 결과 균주 생육과 균주 유래 전체 단백질의 생산에는 LB 배지가 더욱더 효과적임에 반해 높은 혈전용해효소 활성은 TSB 배지에서 Bacillus sp. CP-1 배양하였을 때 얻어졌다. Bacillus sp. CP-1의 배양 상층액의 fibrin zymography에 의한 분석에서 gel상의 상단 부분에 하나의 명확한 혈전용해 활성을 확인하였으며, 분자량은 약 29-30 kDa으로 추정되며, pH 9.0와 $45^{\circ}C$에서 최적의 효소활성을 보였다. 그리고, 기질 특이성 검토에 있어서는 chymotrypsin에 대한 특이적 기질인 Meo-Suc-Arg-Pro-Tyr-pNA (S-2586)에 대하여 가장 높은 기질 특이성을 나타내었다. Subtilisin CP-1단백질의 N-말단 염기 서열을 분석한 결과 처음 8개가 AQSVPYGI로 분석되었으며 이 배열은 subtilisin NAT및 E와 동일하였다.
고온, 알칼리성 Bacillus sp. F204의 CMCase 유전자를 pUC19의 HindIII부위에 연결하여 전이된 E.coli 형질전환체 중 2개의 재조합 플라스미드 즉, pBC191과 pBC192를 선발하였는데, 이들은4.6 Kb와 5.8 Kb HindIII 절편을 각각 함유하고 있었다. pBC191의 4.6 Kb HindIII 절편을 BamHI, EcoRI, KpnI, PvuII 부위가 각각 1개씩 존재하였다. Dioxigenin-labeled deoxyuridin-triphosphate에 4.6 Kb 절편을 표식한 것을 probe로 하여 상동성을 검정한 결과 모균주와 강한상동성이 없었고, 면역학적 실험에서도 Bacillus sp. F204 유래임이 인정되었다. pBC191의 4.6 Kb 절편을 E.coli의 발현 벡타인 pKK223-3과 Bacillus vector인 pGR71에 연결시켜 구축한 pKC231과 pGC711은 각각 pBC191에 비하여 효소활성이 3.2배와 2.8배정도 증가되었으며, 그리고 E.coli에서는 대부분 세포내와 periplasmic 분획에서 검출되었다. 기질 특이성을 조사한 결과에 의하면 pBC191과 pBC192는 CMCase gene을 코딩하고 있는 것으로 나타났다.
단위 동물의 사료 내 phytate 인의 효율적 이용으로 배출되는 인의 양을 줄여 환경오염을 감소시키기 위해 phytate의 분해활성이 뛰어난 phytase 효소 분비 미생물을 탐색하기 위하며 우분으로부터 phytate 분해활성이 뛰어난 phytase를 생산하는 균주를 분리하였다. 분리균주를 동정한 결과, API 50 CHB test에 의해 B. circulans로 분류되었고, 16S rRNA sequencing 결과, B. subtilis로 동정되어 Bacillus sp. CF 5-26으로 명명하였다. 효소생산을 위한 최적배지 조성은 10% rice bran extract, 0.1%, whey protein powder, $0.01%\;CaCl_{2},\;0.01%\;KH_{2}PO_4$ 이었다. Bacillus sp. CF 5-26이 생산하는 phytase는 ethanol 침전, Sephadex G-100, CM Sepharose CL-6B, Sephacryl S-100-HR column chromatography를 통하여 정제도 20.3배, 수율 5.6% 정제되었고, SDS-PAGE에서 분자량 66 kDa의 단일 band를 확인하였다. 정제된 phytase는 pH 5.0, 7.0, 11.0에서 안정하였으며, $100^{\circ}C$에서 1시간 처리하였을 때 50%의 잔존활성을 보였다. 기질 특이성은 inositol polyphosphate인 sodium phytate 분해활성이 뛰어났으며, tripolyphosphate와 pyrophosphate에도 약간의 활성을 보였다. Bacillus sp. CF 5-26이 생산하는 phytase의 sodium phytate에 대한 Km은 0.64 mM 이었고, Vmax는 $4.41{\mu}mol/min$ 이었다.
A novel alkaline protease from Streptomyces sp. M30, SapHM, was purified by ammonium sulfate precipitation, hydrophobic interaction chromatography, and DEAE-Sepharose chromatography, with a yield of 15.5% and a specific activity of 29,070 U/mg. Tryptic fragments of the purified SapHM were obtained by electrospray ionization quadrupole time-of-flight mass spectrometry. Nucleotide sequence analysis revealed that the gene sapHM contained 1,179 bp, corresponding to 392 amino acids with conserved Asp156, His187, and Ser339 residues of alkaline protease. The first 24 amino acid residues were predicted to be a signal peptide, and the molecular mass of the mature peptide was 37.1 kDa based on amino acid sequences and mass spectrometry. Pure SapHM was optimally active at 80℃ in 50 mM glycine-NaOH buffer (pH 9.0), and was broadly stable at 0-50℃ and pH 4.0-9.0. The protease relative activity was increased in the presence of Ni2+, Mn2+, and Cu2+ to 112%, 113%, and 147% of control, respectively. Pure SapHM was also activated by dimethylformamide, dimethyl sulfoxide, Tween 80, and urea. The activity of the purified enzyme was completely inhibited by phenylmethylsulfonyl fluoride, indicating that it is a serine-type protease. The Km and Vmax values were estimated to be 35.7 mg/ml, and 5 × 104 U/mg for casein. Substrate specificity analysis showed that SapH was active on casein, bovine serum albumin, and bovine serum fibrin.
한천분해세균 SC-22균을 대전 대청댐부근의 담수에서 분리하였다. 생화학적 분석 및 16S rRNA 염기서열 분석을 통한 계통학적 분류를 통해 SC-22는 Cellvibrio mixtus로 동정되었다. 분리균의 생육 및 agarase 효소 생성능을 검토한 결과, SC-22는 탄소원으로 0.2% agar를 첨가한 배지에서 배양 36시간에 최대 생육을, 배양 48시간에 58.5 units/mL의 최대 효소활성을 나타내었다. 분리균은 세포외 및 세포내 agarase를 생성하였으며, zymogram 실험에 의해 P1, P2 및 P3의 isoenzyme을 분비하는 것으로 확인 되었다. 배양여액으로부터 겔여과법과 이온교환수지법을 단계적으로 이용하여 SC-22 균주로부터 SDS-PAGE에 의해 25 kDa의 효소를 정제하였으며, 정제효소는 zymogram에서 확인된 주 단백질인 P2(29 kDa)과 동일한 단백질임이 확인되었다. 또한 정제한 agarase의 효소학적 성질을 검토한 결과, 효소의 최적 pH와 최적온도는 pH 7.0과 $50^{\circ}C$이었으며, 금속이온 효과의 경우 1 mM 농도의 수준에서도 $Fe^{2+}$, $Na^+$, $Ca^{2+}$ 이온 등은 정제효소의 활성을 10-20% 증가시킨 반면 $Hg^{2+}$, $Mn^{2+}$ 및 $Cu^{2+}$ 이온들은 효소 활성을 크게 저해하였다. 또한 TLC 분석을 통해 정제효소는 한천 분해 올리고당으로 주로 단당류와 이당류를 생성하므로 $\beta$-agarase의 작용특성을 보였다. 기질특이성 실험에서는 정제효소는 agar와 agarose만을 이용 하였고 유사 해조 다당류인 alginate는 물론 다른 다당류를 분해하지 못하였다.

This review suggested that the high rate of glutamine utilization by cells of the immune system serves to maintain a high intra cellular concentration of the intermediates of biosynthetic pathways such that optimal rates of DNA, RNA and protein synthesis can be maintained. In the absence of glutamine, lymphocytes do not proliferate in vitro: proliferation increase greatly as the glutamine concentration increase. Glutamine is synthesized in skeletal muscle. Skeletal muscle and plasma glutamine levels are lowered by sepsis, injury, bums, surgery and endurance exercise and in the overtrained athlete. This study is expected to contribute the area of sports science, medicine, hereafter more effort is required to establish the relation between gene alters and exercise amount.
