Synthesis and Antimicrobial Activity of Phenazine Derivatives -Synthesis and Antimicrobial Activity of 7- Substituted-2,3-dihydroxyphenazine-5,10-dioxides- (페나진 유도체의 합성과 항균성 -7-치환-2,3-디히드록시페나진-5,10-디옥시드류의 합성 및 항균성-)

  • 7-Substituted 2,3-dihydroxyphenazine 5,10-dioxides were synthesized by the reaction of 1,2,4-trihydroxybenzene with 6-substituted benzofuroxan derivatives which had been obtained from aniline derivativies bearing methoxy, methyl, acetyl and nitro group at the para position. 2,3-Dihydroxyphenazine 5,10-dioxide was also prepared by the reaction of 1,2,4-trihydroxybenzene with benzofuroxan. The antimicrobial activities of these phenazine dioxide were investigated in terms of minimum inhibitory concentration by the common twofold dilution technique. It was observed that the antimicrobial activity of the phenazine dioxides bearing electron releasing substituents was stronger than that of those bearing electron withdrawing substitutents. From this result, it was concluded that the antimicrobial activity of phenazine dioxide derivatives has a direct relationship with the electronic effect of the substituents.

DFT/B3LYP Study of the Substituent Effects on the Reaction Enthalpies of the Antioxidant Mechanisms of Magnolol Derivatives in the Gas-Phase and Water

  • In this paper, the study of various ortho- and meta-substituted Magnolol derivatives is presented. The reaction enthalpies related to three antioxidant action mechanisms HAT, SET-PT and SPLET for substituted Magnolols have been calculated using DFT/B3LYP method in gas-phase and water. Calculated results show that electron-withdrawing substituents increase the bond dissociation enthalpy (BDE), ionization potential (IP) and oxidation/reduction enthalpy (O/RE), while electron-donating ones cause a rise in the proton dissociation enthalpy (PDE) and proton affinity (PA). In ortho- position, substituents show larger effect on reaction enthalpies than in meta-position. In comparison to gas-phase, water attenuates the substituent effect on all reaction enthalpies. In gas-phase, BDEs are lower than PAs and IPs, i.e. HAT represents the thermodynamically preferred pathway. On the other hand, SPLET mechanism represents the thermodynamically favored process in water. Results show that calculated enthalpies can be successfully correlated with Hammett constants (${\sigma}_m$) of the substituted Magnolols. Furthermore, calculated IP and PA values for substituted Magnolols show linear dependence on the energy of the highest occupied molecular orbital ($E_{HOMO}$).

  • The relationship between chemical structure and antifungal activity of benzene ring substituents of 1-phenylpyrrolidine-2,5-diones is studied by testing of model compounds and use of conformational data. The analysis revealed a number of structural features as essential for the antifungal effect: (1) the presence of an intact-NCO group and benzene moiety; (2) the distance between para substituents and the N atom within the 5.318∼5.320 ${\AA}$ range; (3) the distance between substituent X3 and X5 in the interval 5.437∼6.072 ${\AA}$ for the active analogues. And other parameters were discussed.

  • The thiocyanate-selective PVC membrane electrodes based on 5,10,15,20-tetrakis(2,4,6-trimethylphenyl)-porphyrinatomanganese(III) chloride [Mn(TMP)Cl] and 5,10,15,20-tetrakis(2,6-dichlorophenyl)porphyrinatomanganese(III) chloride $[Mn(Cl_8TPP)Cl]$ as ion carriers were investigated. The effect of ionophores, membrane compositions, plasticizers, and solution pHs on the response characteristics were studied. The Mn(TMP)Cl as an ionophore shows the best potentiometric sensitivity with a slope of -58.7 mV/decade and a detection limit of $log[SCN^-]$ = -6.90, and selectivity for thiocyanate over strong hydrophobic interfering anions such as ${ClO_4}^-$ and salicylate. The potentiometric response is affected by the electronic effect of the substituents and solution pHs. The presence of substituents with electron donating and more liphophilic characters around the ligated metal center produces an improved response toward $SCN^-$.

  • The reactions of mercury(II) chloride with O3S2-donor macrocyclic ligands with (L1) and without (L2) dibenzosubunit afforded respective exo- (1) and endo-coordinated (2) complexes depending on the ring rigidity of the ligands. From the X-ray crystal structures and comparative NMR studies for the complexes 1 and 2, it is confirmed that the resulting species with different coordination modes exist not only in solid state but also in solution state.

  • The R- and Z-substituent effects for the gas-phase thermal decompositions of carbamates, R2NC(=O)- OCH2CH2Z, have been investigated theoretically at B3LYP level with 6-31G(d) and 6-31++G(d,p) basis sets. Both the Z- and R-substituent effects on reactivity (ΔH≠) were well consistent with experimental results, although the R-substituent effect was underestimated theoretically. No correlations were found between activation enthalpies and reaction enthalpies. The substituent effects on reactivity seemed to be complicated at a glance, but were understandable by concurrent electronic and steric factors. Variations of bond lengths at TS structures were well correlated with the Taft's σ* values and the TS structures became tighter as the Zsubstituent became a stronger electron-acceptor (δσ* > 0). However the effects of R-substituents on the TS structures were much smaller when compared to those of Z-substituents.

Substituent Effects on the Leaving Groups in Benzyl Arenesulfonates (Benzyl Arenesulfonate의 離脫基의 置換基效果에 關한 硏究 (第 1 報))

  • Determination has been made of the kinetics of the reaction of benzyl arenesulfonates with pyridine in acetone. The substituent effects of the leaving groups in benzyl arenesulfonates are correlated by Hammett equations, with the exception of p-MeO and $p-NO_2$ groups, where the electron attracting substituents in the benzyl arenesulfonate increase the rate. The substituent effects of the leaving groups are as expected due to the nucleophilic attack of amine on the benzyl carbon atom. This can be understood in terms of changes in bond formation (C-N) and bond breaking (C-O) in the transition state with charges in electron-attracting ability of the substituents. The predicted substituent effects may indicate a small increase in bond formation and thus a tighter transition state, in benzyl p-bromobenzene sulfonate than in benzyl p-nitrobenzenesulfonate. Predicting made by Thornton concerning the substituent effects on $S_N2$ transition state structures agrees with the changes in bond formation and bond breaking.

  • The kinetics of the reaction of m-or p-substituted benzyl bromides with thiourea in methanol at 35.0$^{\circ}$were determined by an electric conductivity method. According to a plot of log k against the Hammett substituent constants, C-Br bond cleavage in benzyl bromide is postulated to be a rate determining step at the SN2 reaction of benzyl bromide with thiourea. Both electron-donating substituents and electron-withdrawing substituents quantitatively affected the rate of reaction, but each in a different manner. A mechanistic possibility was proposed to account for the results. Some activation parameters were also calculated.

Ab Initio Studies on Proton Affinities of Substituted Furans (치환 퓨란의 양성자 친화도에 대한 Ab Initio 연구)

  • The geometry of furan, relevant to the binding of bis-furan lexitropsin that contains this ring to the base pair of minor groove of DNA, is optimized by semiempirical (MNDO) and ab initio (Hartree-Fock) methods. The proton affinity and electronic structure are evaluated at the 6-31G and $6-31G^{\ast}$ level of theory for the optimized geometry. The proton affinities are also studied for various substituted furans with the electrondonating and -withdrawing groups to estimate the substituent effect on the proton affinity of furans. It has been found that the electron-donating substituents increase the proton affinity of furan, whereas the electron-withdrawing substituents decrease it. This result can be explained with atomic charge and electron density at oxygen of substituted furans.

Effects of newly synthesized benzimidazole derivatives on gastric H^+/K^+$ ATPase

  • The effects of various synthetic benzimidazole derivatives on gastric H^+/K^+$ATPase activity in vitro were examined. The results showed that the effects of substituents on the benzimidazole ring were not significant. However, replacement of sulfoxide connecting two ring systems to sulfide resulted in a completely inactive compound in vitro, suggesting the essential role of sulfoxide group in the inhibition. In addition, compounds with 5 or 6-membered oxacyclic substituents attached to the pyridine ring displayed the most effective inhibitory activity. Among these derivatives, AU-47 was the most potent, and detailed mechanistic studies with the compound were carried out. AU-47 inhibited gastricH^+/K^+$ATPase in a concentration and time dependent manner with 50% inhibition at $6\muM$. The presence of sulfhydryl reducing agents or substrate analogue protected H^+/K^+$ATPase from the inactivation. The inhibition by AU-47 was potentiated by acid pretreatment of the compound, suggesting the structural conversion of AU-47 into a more active intermediate which was favored in acidic condition. Consistent with in vitro results, AU-47 inhibited in vivo gastric acid secretion. The results suggest that AU47 is a relevant candidate for the development of new antiulcer agent.
