Novel Heterogeneous Carbohydrase Reaction Systems for the Direct Conversion of Insoluble Carbohydrates: Reaction Characteristics and their Applications

  • Most carbohydrates exist in nature in an insoluble state, which reduces their susceptibility towards various carbohydrases. Accordingly, they require intensive pretreatment for structural modification to enhance an enzyme reaction. The direct conversion of insoluble carbohydrates has distinct advantages for special types of reaction, especially exo-type carbohydrase; however, its application is limited due to structural constraints. This paper introduces two novel heterogeneous enzyme reaction systems for direct conversion of insoluble carbohydrates; one is an attrition coupled enzyme reaction system containing attrition-milling media for enhancing the enzyme reaction, and the other is a heterogeneous enzyme reaction system using extruded starch as an insoluble substrate. The direct conversion of typically insoluble carbohydrates, including cellulose, starch, and chitin with their corresponding carbohydrases, including cellulase, amylase, chitinase, and cyclodextrin glucanotransferase, was carried out using two proposed enzyme reaction systems. The conceptual features of the systems, their reaction characteristics and mechanism, and the industrial applications of the various carbohydrates are analyzed in this review.

Carbohydrate and lipid spectroscopic molecular structures of different alfalfa hay and their relationship with nutrient availability in ruminants

  • Objective: This study was conducted to determine molecular structures related to carbohydrates and lipid in alfalfa hay cut at early bud, late bud and early flower and in the afternoon and next morning using Fourier transform infrared spectroscopy (FT/IR) and to determine their relationship with alfalfa hay nutrient profile and availability in ruminants. Methods: Chemical composition analysis, carbohydrate fractionation, in situ ruminal degradability, and DVE/OEB model were used to measure nutrient profile and availability of alfalfa hay. Univariate analysis, hierarchical cluster analysis (CLA) and principal components analysis (PCA) were conducted to identify FT/IR spectra differences. Results: The FT/IR non-structural carbohydrate (NSCHO) to total carbohydrates and NSCHO to structural carbohydrate ratios decreased (p<0.05), while lignin to NSCHO and lipid CH3 symmetric to CH2 symmetric ratios increased with advancing maturity (p<0.05). The FT/IR spectra related to structural carbohydrates, lignin and lipids were distinguished for alfalfa hay at three maturities by PCA and CLA, while FT/IR molecular structures related to carbohydrates and lipids were similar between alfalfa hay cut in the morning and afternoon when analyzed by PCA and CLA analysis. Positive correlations were found for FT/IR NSCHO to total carbohydrate and NSCHO to structural carbohydrate ratios with non-fiber carbohydrate (by wet chemistry), ruminal fast and intermediately degradable carbohydrate fractions and total ruminal degradability of carbohydrates and predicted intestinal nutrient availability in dairy cows ($r{\geq}0.60$; p<0.05) whereas FT/IR lignin to NSCHO and CH3 to CH2 symmetric stretching ratio had negative correlation with predicted ruminal and intestinal nutrient availability of alfalfa hay in dairy cows ($r{\geq}-0.60$; p<0.05). Conclusion: FT/IR carbohydrate and lipid molecular structures in alfalfa hay changed with advancing maturity from early bud to early flower, but not during the day, and these molecular structures correlated with predicted nutrient supply of alfalfa hay in ruminants.

옥수수 및 Sorghum에 있어서 탄수화물과 NEL 축적에 관한 연구. I. Fructosan , Mono 및 Disaccharose의 합성 및 축적형태 (Studies on Reserved Carbohydrates and NEL ( Net energy Lactation ) in Corn and Sorghum I. Synthesis and Accumulation Pattern of Fructosan, Mono-and Disaccharose)

  • 本試驗은 옥수수 및 Sorghum類에 싫어서 形態的特性과 環境溫度가 Fructosan Mono- 및 Disaccharose의 合成, 移動 및 蓄績形態와 NEL價値이 미치는 影響을 究明코저 옥수수의 Blizzard와 Sorghum의 Sioux 및 Pioneer931을 供試品種으로 하여 圃場 및 Phytotron試驗으로 實施하였다. Phytotron의 晝/夜間 室內溫度는 30/25, 25/20, 28/18및 18/8$^{\circ}C$로 하였으며 日照는 30.000~35.000 Lux로 13時間 照謝하였다. 1978~’81年까지 얻어진 結果를 要約하면 다음과 같다. 1. 옥수수 및 Sorghum類의 non-structural carbohydrate는 主로 Mono- 및 nisaccharose形態로 蓄績된다. Mono- 및 Disaccharose의 蓄績은 幼穗가 分化된以後 本格的으로 이루어져 乳熟期初期에 各各 옥수수 27.8~29.1 % 및 Sorghum類 16.8-20.4%로 最高濃度水準에 달한다. 2 Fructosan含量은 옥수수와 수수 各作物 共히 1.5~2.5%內外로서 全生育期間을 通해 큰 變化가 없다. 3. Mono- 및 nisaccharose는 主로 줄기部位에 蓄績되나 出糖期以後 穀實이 形成됨에 따라 이들 T財C의 大部分은 이삭部位로 移動되어 Starch 形態로 貯藏된다. Starch의 合成 및 蓄績은 Mono- 및 Disaccharose의 減少와 灰比例 한다. 4. 옥수수 및 Sorghum의 同化能力은 溫度가 上昇됨에 따라 增大되나 溫度 30$^{\circ}C$以上의 高溫下에서는 植物體內의 Mono- 및 Disaccharose蓄績이 적게 일어난다. 한편 溫度 18/8$^{\circ}C$以下의 低溫에서는 合成된 Saccharose가 移動되지 못하고 줄기 및 葉에 과다 蓄績되어 葉의 同化能力이 減少되고 物質生産이 中止된다. 5 NEL 및 Starch value는 non- structural carbohydrate의 合成 및 蓄續形態에 따라 影響을 받는다. 植物體內의 NEL 蓄績은 Mono- 및 Disaccharose의 蓄積이 本格的으로 進行되는 幼穗分化期以後부터 서서히 增加하여 完熟初期에 各各 옥수수 6.6~6.9MJ/kg 및 Sorghum類 5.8~6.0MJ- NEL/kg으로 가장 높은 水準을 나타낸다.

Evaluation of Structural Carbohydrates Losses and Digestibility in Alfalfa and Orchardgrass during Ensiling 1

  • The evaluation of structural carbohydrate losses and its effect on silages digestibility in alfalfa (Medicago sativa L.) and orchardgrass (Dactylis glomerata L.) was studied during 5, 21 and 56 days ensiling. About 70 and 60 kg fresh matter of the two forages were ensiled in 9 silos of 120 L capacity. The digestion trials were conducted in two phases using the two grasses in two $4{\times}4$ Latin square design according to the four treatments being the grass and the three silages. There were no differences in the DM and CP contents resulting from 5 to 56 days ensiling in both forages. The water-soluble carbohydrates (WSC), hemicelllose, pectin, and energy were slightly reduced and appeared lower in 56 days silage. The ether extract and cellulose contents slightly increased as the ensiling process advanced in the two species. Hemicellulose losses of 29 and 41 g/kg DM were obtained in alfalfa and orchardgrass, respectively, 56 days after ensiling. While the cellulose losses in both species were very little, compared to that for hemicellulose, the pectin losses, 56 days after ensiling were 15 and 12 g/kg DM in alfalfa and orchardgras respectively. The total structural carbohydrates lost (ie., hemicellulose + cellulose + pectin) in g/kg DM of fresh material forage ensiled, is about four fifths the amount lost by WSC, in alfalfa and about two thirds, in orchardgrass, by 21 days ensiling after the activity of microorganism terminated, indicating that appreciable amount was used as substrate for silage fermentation. Ensiling alfalfa and orchardgrass for 0, 5, 21 and 56 days maintained a decreasing trend of 83.8, 82.5, 79.3 and 78.9% digestibility in alfalfa and 80.5, 77.0, 77.1 and 76.4% digestibility in orchardgrass. While the digestibility of cellulose and ether extract increased in silage in both species, the digestible energy values in silage were reduced from 2.6 to 2.3 and 2.9 to 2.7 Mcal/kg DM respectively in alfalfa and orchard during 5-56 days ensiling.

Ozone Impacts on Soluble Carbohydrates, Antioxidant Activity and Macro-element Concentrations in Rice Seedling

  • The present study describes carbohydrate metabolism, macro-element utilization and antioxidant defenses in response to an ozone dose (100 ppb, 8d) in two rice varieties. Tolerant (cv. Jinpumbyeo) and sensitive (cv. Chucheongbyeo) varieties of rice were grown in growth chamber for 30 days after sowing. Concentrations of chloroplast pigments and non-structural carbohydrates as well as activity of antioxidant enzymes were determined to evaluate the resistance against ozone stress. Ozone caused the decrease in chlorophyll a and carotenoid contents, and also resulted in faster decomposition of non-structural carbohydrate in leaf blade and leaf sheath. The contents of nitrogen and potassium in leaves were visibly decreased in cv. Chucheongbyeo with an increase in ozone exposure, but not in cv. Jinpumbyeo. Enzymatic antioxidants against ROS in both varieties responded in the order of POD, SOD and CAT, and their capacity was stronger in cv. Jinpumbyeo.

The Relationship between the Sugar Preference of Bacterial Pathogens and Virulence on Plants

  • Plant pathogenic bacteria colonize plant surfaces and inner tissues to acquire essential nutrients. Nonstructural sugars hold paramount significance among these nutrients, as they serve as pivotal carbon sources for bacterial sustenance. They obtain sugar from their host by diverting nonstructural carbohydrates en route to the sink or enzymatic breakdown of structural carbohydrates within plant tissues. Despite the prevalence of research in this domain, the area of sugar selectivity and preferences exhibited by plant pathogenic bacteria remains inadequately explored. Within this expository framework, our present review endeavors to elucidate the intricate variations characterizing the distribution of simple sugars within diverse plant tissues, thus influencing the virulence dynamics of plant pathogenic bacteria. Subsequently, we illustrate the apparent significance of comprehending the bacterial preference for specific sugars and sugar alcohols, postulating this insight as a promising avenue to deepen our comprehension of bacterial pathogenicity. This enriched understanding, in turn, stands to catalyze the development of more efficacious strategies for the mitigation of plant diseases instigated by bacterial pathogens.

하이에너지수수의 생물학적 전처리 (BIOLOGICAL PRETREATMENT OF HIGH ENERGY SORGHUM)

  • 식물체 biomass 에너지 생산, 즉 메탄과 에탄올에서 부딪치는 제일 큰 문제는 식물체의 structural components, 즉 lignocellulose가 자연상태에서 발생하는 lactic acid bacteria에 의해 분해되지 않는데 있다. 이에 structural components의 주성분인 lignocellulose를 분해시키기 위해 여러 가지 처리를 가한 High Energy Sorghum에 C. celluluyticum과 B. succinogenes를 주입하여 그 효과를 이 논문에서 규명하였다. 살균한 High Energy Sorghum에 C. celluluyticum을 주입한 경과 cellobiose가 structural components에 생산되었다. 그러나, 살균처리를 안한 샘플에서는 C. celluluyticum가 lactic acid bacteria와 처리범위 내에서는 효과적으로 경쟁하지 못한 것으로 사료된다. 또, B. succinogenes도 High Energy Sorghum의 structural components를 어느 정도 분해시켰다. B. succinogenes에의한 cellobiose의 degradation에서 glucose와 succinte에 의해 inhibition은 없었다.

Influence of dietary nonstructural carbohydrate concentration on growth performance and carcass characteristics of Holstein steers

  • Objective: Since very little information exists about the topic; in this experiment we compare, in a long-term finishing program, the growth-performance responses and carcass characteristics of Holstein steers where non-structural carbohydrate concentration of the diet is reduced from 64% to 51% (dry matter basis). Methods: Sixty Holstein steer calves ($129{\pm}2.2kg$) were blocked by initial weight into five groups and randomly assigned within weight groupings to 10 pens. Calves were fed with a steam-flaked corn-based finishing diets containing 51% higher fiber (HF) or 64% lower fiber (LF) nonstructural carbohydrates. Non-structural carbohydrates concentrations were manipulated substituting dried distiller grain with solubles and alfalfa hay for flaked corn. Cattle were weighed every 112 days and at the end of the experiment (day 308) when the cattle were harvested and carcass characteristics were evaluated. Results: Steers fed the HF diet showed improvement (8.8%) in average daily gain (ADG) during the initial 112-d period. This effect was followed by a numerical trend for greater ADG throughout the remainder of the study so that overall ADG tended to be greater (4.9%, p = 0.06) for the HF than for LF. There were no treatment effects on dry matter intake. Gain efficiency and estimated dietary net energy (NE) were greater 8.3% and 5.2%, respectively for HF during the initial 112-d period. Overall (308-d) gain efficiency and estimated dietary NE were similar for both dietary treatments. However, due to differences in tabular dietary NE, the ratio of observed:expected dietary NE tended to be greater (4.1%, p = 0.06) for the HF vs LF diet. There were no treatment effects on carcass characteristics except for a tendency toward a slightly greater (0.5%, p = 0.09) estimated carcass yield. Conclusion: Reducing the non-structural carbohydrate concentration of a conventional steam-flaked corn-based growing finishing diet for Holstein steers can effectively enhance growth performance, particularly during the early growing and late finishing phases.

Modification of Carbohydrate Metabolism in Transgenic Potato

  • Carbohydrates serve three different principal functions in the metabolism of plants. They are the primary products of energy fixation, they are important transport metabolites, and they are deposited as structural or storage compounds. Modification of carbohydrate metabolism therefore covers approaches to modify yield, to change sink/source relationships and thereby alter the ratio of harvestable material, and to improve the quality of crop plants. The scope of this article is to summarize research done at the Max-Planck-Institute related to the first two fields and to present in some detail what we learned, when we established a new carbohydrate storage form in potato.
