Lectins from Allomyrina dichotoma (ADL) and Bombyx mori (BML) were partially purified by physiological saline extraction, ammonium sulfate fractionation, anion exchange column chromatography on DEAE Sephadex A-50 and gel filtration column chromatography on Sephadex G-200. An assay for cytokine expression was carried out by using reverse transcription polymerase chain reaction(RT-PCR). mRNA isolated from PBMC(human peripheral blood mononuclear cells) were stimulated with ADL(O.D.=0.2) and BML(O.D.=0.1) for various times(1,4,8,24,48 and 72 h) and various cytokine mRNA assessed by RT-PCR were shown as follows: The patterns of bands for IL-1 mRNA of BML were very similar with those from ADL and these bands were decreased along the increasing reaction times after showing a strong band at 1 h. However mRNA expressions for IL-2, IL-6, $IFN{\gamma}$ and $TNF{\alpha}$ showed different patterns between ADL and BML. With the effect of ADL, the expression of IL-2 and IL-6 mRNA were continuously detected until 72 h with the strongest band of IL-2 mRNA at 24 h. The strong bands of $IFN{\gamma}$ mRNA were observed from 4 to 8 h but the strongest one of $TNF{\alpha}$ was just observed at 1 h. Meanwhile with BML, the bands for IL-2 and $IFN{\gamma}$ were increased along the increasing reaction times until 72 h. The strongest bands were showed from 4 to 8 h with IL-6 and at 8 h with $TNF[\alpha}$. To verify quantitatively ELISA was used for assay of protein secretions of the cytokine gene with IL-2 and $IFN{\gamma}$ expressed markedly different in RT-PCR. The highest cytokine secretion for IL-2 was demonstrated at 48 h. The production of $IFN{\gamma}$ was markedly increased at 24 h and secreted highest at 72 h. These result suggest that ADL and BML, as inducers of cytokines, can elicit detectable cytokine mRNA from PBMC within the first few hours of stimulation and maintain the production of cytokines for a few days by the methods of RT-PCR and ELISA.
Terminalia chebula Retz., which was showed antimicrobial activity against intestinal pathogens through screening herbs related treatments of intestinal diseases, were extracted by methanol and fractionated by n-hexane, ethylether, ethylacetate,and water. Antimicrobial activities of the methanol extract and each fractionates were then investigated under the anaerobic broth system. The methanol extract showed antimicrobial activity against all intestinal pathogens(Eubacterium limosum ATCC 10825, Escherichia coli ATCC 25922 and Staphylococcus aureus KFCC 11764 hardly grew at 2,000$\mu$g/ml of concentration. Especially, Escherichia coli ATCC 25922 and Staphylococcus aureus KFCC 11764 hardly grew at 2,000$\mu$g/ml of concentration. There is no significant difference of antimicrobial activity among each fractionates. Fraction of Terminaliz chebula Retz. ethylacetate fractionate, which were fractionated by Sephadex G-200 and Silica gel column chromatography revealed the strongest antimicrobial activity at 12 to 21 and 22 to 34 of fraction number, respectively.
In order to estimate volume transport by upwelling for single artificial seamount, same shape and size of artificial seamount already deployed was applied to numerical experiment. The result showed that strong upwelling appeared at front while took place downwelling at rear. The strongest upwelling existed at the top of the artificial seamount. Volume transport by upwelling was computed as 785 m3/s. Column arrangement was applied to two artificial seamount in three cases; case 1) no clearance, case 2) sixty-five meters of clearance as half of artificial seamount's length, and case 3) hundred-thirty meters of clearance as an artificial seamount's length. All cases of column arrangements showed more upwelling volume transport than that of single seamount. Particularly, the case of no clearance calculated as 106% and appeared the most upwelling effect comparing to two other cases. Row arrangement was also applied to two artificial seamount in three cases; case 4) no clearance, case 5) forty meters of clearance as an artificial seamount's width, and case 6) eighty meters of clearance as twice of artificial seamount's width. Upwelling volume transport in case 4 increased 48% than the case of single seamount. Other two cases of 5 and 6 were estimated as 97% increased and more effective than case 4. According to the case experiments, column arrangements show more upwelling volume transport than that of row arrangements. In cases of column arrangements, with decreasing clearance between two seamount, the effect increases while showing maximum value at clearance zero. In cases of row arrangements, on the contrary, with decreasing clearance between two seamount, the effect decreases while showing minimum value at clearance zero. Since simple barotropic condition was considered for this study, further study is necessary by considering baroclinic condition to get close to reality. In conclusion, in deploying artificial seamount, optimal arrangement should be well designed to enhance primary and secondary productivity and to increase the diversity of species as well as reducing time and space.

This study was attempted to isolate and identify the anticancer compounds from Eucommia ulmoides leaves using a human colon cancer cell line HCT-116. The petroleum ether extracts with anticancer activity was chromatographed on silica gel TLC and finally anticancer compounds was purified by HPLC. Their chemical structures were roughly elucidate by UV-VIS absorption spectral data HPLC elution pattern and FAM/MS spectroscopy. From this study these compounds were suspected to be pheophytin a formed by the removal of $Mg^{2+}$ from chlorophyll a and pyropheophytina formed by the removal of acetate group from pheophytin a respectively. To confirm the anticancer effects against HCT-116 cancer cell petroleum ether extract fractions of column chromatography and fractions separated on TLC were tested. All samples tested including the extract of petroleum ether fractions of column chromatograph and three bands (0.13,0.19,0.25) of TLC appeared to inhibit the growth of HCT-116 cancer cell however especially 0.19 and 0.25 fractions separated on TLC plate revealed the strongest effect. These results suggest that chlorophyll derivatives in Eucommia ulmoides may be potential anticancer agents against a human colon cancer cell HCT-116.

The main purpose of this paper is to determine the dynamic optimal shapes of simple beam-columns with the constant volume. The parabolic function is chosen as the variable equation for the depth of regular polygon cross-section. The ordinary differential equation including the effect of axial load is applied to calculate the natural frequencies. The Runge-Kutta and Regula-Falsi methods are used to integrate the differential equation and compute the frequencies, respectively. Then the dynamic optimal shape whose lowest natural frequency is highest is determined by reading the critical value of the frequency versus section ratio curve plotted by the frequency data. In the numerical examples, the simple beam-columns are analysed and the numerical results of this study are shown in tables and figures.
As a search for natural antioxidants, antioxidative fractions in pueraria root were extracted and identified using column chromatography, thin layer chromatography and high performance liquid chromatography. Components which have most effective antioxidative activities were futher identified by IR and GC/MS. The strongest antioxidative component of pueraria root methanol extract was identified as puerarin. Puerarin obtained from pueraria root was practically effective as antioxidant at the level of 100 ppm. Antioxidative activity of the puerarin was higher in linoleic acid-water system than in a linoleic acid substrate. Puerarin, daidzin and daidzein contents in pueraria root juice were 0.39%, 0.45% and 0,03%, respectively.

Total ginsenosides were purified and their antioxidant, antibacterial and anticancer activities were measured. The crude extracts of ginseng, which were extracted with 75% ethanol by ultrasonification method, were firstly purified on AB-8 macroporous adsorption column to remove water soluble impurities, and decolored on Amberlite IRA 900 Cl anion-exchange column. Then, they were purified on Amberlite XAD16 adsorption column to delete the non-polar impurities. Total ginsenosides contents of the purified extracts were 79.4%, 71.7% and 72.5% in cultured wild ginseng, red ginseng and white ginseng, which were significantly increased than those of crude extracts. All of the three extracts showed concentration-dependant scavenging activities against DPPH radicals, among which white ginseng showed the most powerful activity. Cultured wild ginseng roots showed strongest effect against both B. subtilis PM 125(Gram-positive) and E. coli D31 (Gram-negative) bacteria, while red ginseng and white ginseng only showed the activity against B. subtilis. According to the result of the MTT assay, ail of the three extracts inhibited the growth of U-937 human hohistiocytic lympma cell, which were significantly different (p < 0.05) when compared to the control.
This study was proposed to measure total polyphenols and antioxidant activities from leaves of Stachys sieboldii MIQ. Solvents such as acetone 60%, petroleum ether, ethylacetate, and water were used for this purpose The fraction by ethyl acetate fraction showed the strongest activity by DPPH method and contained much more polyphenols than those by other solvents.
To confirm the effects of binlang(Areca catechu L.) and tansen(Salvia miltriorrhiza bung) on the mutagenicity induced by hydrogen peroxide, SOS Chromotest with Escherichia coli PQ37 and Ames test with Salmonella typhimurium TA104 were perlormed. Methanol-soluble parts of their water extracts showed high inhibitory effect against the mutagenicity of hydrogen peroxide in two bacterial mutation assays. Step-wise fractionation of methanol-soluble part from tansen was done using ethyl acetate, butanol and water. Among these fraction was further partitioned by Sephadex LH-20 column chromtography, and 6 subfractions were obtained. The fraction III showed the strongest inhibitory effects against the mutagenic activities induced by hydrogen peroxide. The inhibition rates of fraction III at concentration of 500$\mu\textrm{g}$/assay were 28%, 30% and 15% against 4-NQO, MNNG and B(a)P, respectively. But the mutagenic potency of AFB1 was increased.
The cytotoxic activities of methanolic extract and its fractions of Ailanthi Cortex Radicis and column chromatographic eluates of its dichloromethane fraction (DCM fr.) were investigated. DCM fr. Showed the strongest cytotoxicity against hepatoma cells. Furthermore, the active equates 1-3, 8 and 9 were obtained. Effects on free radical generation and the growth of vascular endothelial cells were tested to elucidate the action mechanism of anticancer activity. Eluates 1-3 stimulated free radical generation, while eluates 8 and 9 showed no changes. Especially, eluates 8 and 9 efffectively inhibited the proliferation of vascular endothelial cells in a dose- dependant manner. It is speculated that the anticancer effects of eluates 1-3, 8 and 9 might be due to free radical generation and inhibition of endothelial cell growth, respectively.
