수소화물 발생법-원자흡수 분광계를 이용한 비소의 분석에서 금속이온의 은폐제로 사용되는 L-cysteine, thiourea, KI/ascorbic acid 등의 예비 환원제와 염산, 질산, 황산 및 이들의 1:1 혼산의 종류 및 농도가 비소의 분석에 미치는 영향을 비교 연구하였다. 대부분의 예비 환원제의 경우 3-5M 이상의 고농도 산 용액에서 비소에 대한 높은 흡괌도를 나타냈으나, 0.1M 이하의 낮은 산 용액에서는 L-cysteine을 제외하고는 극히 저조한 흡광도를 나타내었다. 질산 산성용액에서는 흡광도 신호의 불안정성이 심하였다. 방해물질로서 $Cr^{6+}$, $Br^{5+}$ 이온의 방해영향이 크게 관찰되었지만, 이때 예비 환원제는 수소화물 생성을 크게 증가시켰다.
고온내성을 가진 효모 균주, Saccharomyces cerevisiae KNU5377을 황산, 질산 그리고 염산에 노출시켰다. 스트레스 원으로써 무기산은 물에서 쉽게 해리되어 외부 산도를 떨어뜨린다. 여러 가지 무기산이 첨가된 조건에서 배양한 결과 KNU5377은 0.4%의 황산, 질산 농도에서 생육이 가능한 반면 대조 균주인 S. cerevisiae ATCC24858은 이 보다 낮은 농도인 0.3%가 생육의 한계였다. 더욱이 KNU5377은 0.35%의 염산에서 90분 이상, 0.6%의 황산에서는 30분 이상 생존이 가능한 높은 내성을 나타내었다. 반면에 두 균주 모두 0.45%의 질산에서는 생존하지 못하였다. 0.3%의 황산이 첨가된 조건에서 알코올 발효 시 KNU5377은 이틀 후 3%의 알코올을 생산하였다. 더욱이 0.2%의 황산 첨가와 동시에 $40^{\circ}C$ 고온에서도 4.5%의 높은 알코올 생산이 관찰되었다. 또한 황산 0.2%에 한 시간동안 노출시킨 뒤 세포내에 축적되는 trehalose의 농도를 측정한 결과, KNU5377에서는 30분내에 효과적으로 축적되었으며 동일한 스트레스 조건에서 전자현미경(TEM)을 통한 세포의 형태의 관찰 시 어떠한 변화도 나타나지 않았다.
Strong promoter로 밝혀진 alginate lyase 유전자의 promoter 부위에 대한 특성을 검토하기 위해 alginate lyase 유전자의 46개 N-terminal amino acid가 포함된 promoter 부분과, 같은 균으로부터 분리한 $\beta$-agarase의 유전자를 연결시켜 agarase의 activity를 평판배지상에서 보다 쉽게 확인하는 방법으로 promoter의 활성을 측정한 결과 alginate lyase 유전자 promoter에 의해서 $\beta$-agarase 유전자의 대량발현이 유도되고 있었으며 glucose의 존재하에서 $\beta$-agarase 유전자 발현이 일어나지 않는 catabolite repression 양상을 나타내고 있다. PCR로써 alginate lyase의 46개 N-terminal amino acid 부분이 순차적으로 제거된 plasmid를 제조하여 대량발현을 조사한 결과 46개의 아미노산이 제거된 후에도 $\beta$-agarase의 활성에는 변화가 없어 46개의 N-말단이 정상적인 상태에서 발현에는 영향을 미치고 있지 않음을 확인할 수 있었다. 또한 alginate lyase 유전자의 promoter region에 존재하는 가능한 2개의 promoter consensus sequence PI, PII를 subcloning한 결과 promoter PII만이 존재할 때도 대량발현이 유도되고 있음을 확인할 수 있었으며 동시에 glucose가 존재할 때 catabolite repression이 역시 나타나고 있어 이 부분이 발현 및 glucose에 의한 regulation에 매우 중요하게 작용하는 부분이라는 것을 확인할 수 있었다.

Background: The demand for healthy, lean and consistent meat products containing low saturated fatty acid content and high quality polyunsaturated fatty acids (PUFA), especially long-chain (${\geq}C_{20}$) omega-3 PUFA, has increased in recent times. Fat deposition is altered by both the genetic background and dietary supplements, and this study aimed to assess the effect of dietary Spirulina supplementation levels on the mRNA expression patterns of genes controlling lipid metabolism in the subcutaneous adipose tissue (SAT) and Longissimus dorsi (ld) muscle of Australian crossbred sheep. Methods: Twenty-four weaned lambs belonging to four breeds under the same management conditions were maintained on ryegrass pasture and fed three levels of Spirulina supplement (control, low and high). In terms of nutrient composition, Spirulina is a nutrient-rich supplement that contains all essential amino acids, vitamins and minerals. It also is a rich source of carotenoids and fatty acids, especially gamma-linolenic acid (GLA) that infer health benefits. After slaughter, subcutaneous adipose tissue (SAT) and ld samples were subjected to mRNA extraction and reverse transcription using quantitative polymerase chain reaction (RT-qPCR) to assess the mRNA expression levels of the Aralkylamine N-acetyltransferase (AANAT), Adrenergic beta-3 receptor (ADRB3), B-cell translocation gene 2 (BTG2) and Fatty acid synthase (FASN) genes, which are associated with lipid metabolism. Results: Both low and high Spirulina supplementation levels strongly up-regulated the transcription of all the selected genes in both SAT and ld tissues (mostly in the subcutaneous adipose), but sheep breed and sex did not influence the gene expression patterns in these tissues. Conclusions: The evidence indicates that high Spirulina supplementation level resulted in a decrease in intramuscular fat content in Australian purebred and crossbred sheep due to the enhanced production of melatonin in sheep muscle tissues and strong up-regulation of mRNA expression of BTG2 in SAT which negatively affected fat deposition. In contrast, low Spirulina supplementation level strongly up-regulated the ADRB3 and FASN genes responsible for fat production. These findings are consistent with the observed phenotypic data suggesting that low Spirulina supplementation level can increase lamb production, with higher long-chain PUFA content.
강한 인산 가용력을 가진 인산 용해 세균인 균주 60-2G를 잔디의 근권에서 분리하였다. GC-FAME구조와 탄소이용형태 및 16S rRNA의 부분 염기서열 분석을 통해 균주 60-2G는 Enterobacter intermedium으로 동정되었다. Hydroxyapatite를 첨가한 배지와 생장 시킨 균주 60-2G는 gluconic acid 와 2-ketogluconic acid 및 소량의 lactic acid를 생성하였다. 균주 60-2G의 생장 기간동안 배지의 pH는 3.8 까지 낮아지는 반면에 배지의 유효 인산 농도는 증가하였다. 배지의 낮은 pH와 유효인산농도의 증가는 역 상관관계이며, 이는 균주 60-2G가 생성하는 유기산에 의한 영향이다. E. intermedium 60-2G 균주는 유기산 생성에 관여하는 glucose dehydrogenase의 co-factor인 PQQ를 생성하였으며, pqq의 부분 염기서열 분석 결과 기존에 보고된 서열과 85% 이상의 상동성을 가지고 있었다.
인천 연안의 심하게 오염된 갯벌로부터 강력한 세포외 알카리성 단백질 분해효소를 생산하는 호알카리성 Bacillus clausii I-52를 분리하였으며, 이 균주로부터 알카리성 단백질 분해효소의 유전자를 cloning하여 서열 분석을 하였다. Chromosome 서열이 완전히 밝혀진 Bacillus subtilis의 서열을 기초로 하여 알카리성 단백질 분해효소 및 promoter를 포함하도록 primer를 고안하여 PCR을 수행하여 2,277 bp의 DNA 단편을 얻었으며 BLAST 분석 결과 29 개의 아미노산으로 이루어진 signal peptide, 77 개의 아미노산으로 이루어진 propeptide 및 275 개의 아미노산을 갖는 활성형의 BCAP으로 구성된 총 381 개의 아미노산을 코딩하는 1,143 bp의 open reading frame을 확인하였다. 활성형 BCAP의 N-말단 아미노산은 Ala이며, 분자량 및 pI 값은 각각 27698.7 Da과 6.30으로 계산되었다. 아미노산 상동성을 분석한 결과, B. subtilis 유래의 nattokinase precursor 및 B. subtilis BSn5 유래의 subtilisin E precursor와 99%의 서열 상동성을 나타내어 B. clausii I-52 유래의 BCAP은 subtilisin 계열의 단백질 분해효소임을 확인하였다. E. coli BL21(DE3)에서 발현한 재조합 BCAP는 N-Suc-Ala-Ala-Pro-Phe-pNA 를 효율적으로 분해하였다. Refolding한 재조합 BCAP은 전형적인 serine protease inhibitor인 PMSF에 의하여 강하게 효소 활성이 억제됨으로써 serine protease 계열의 단백질 분해효소임을 알 수 있었다.
과잉으로 생산되는 복분자의 소비 촉진과 신수요 창출을 위해 복분자 식초를 제조한 후 이화학적 성분분석 및 항산화 효과를 조사하였다. 알코올 발효의 최적 조건은 당도 15 $^{\circ}Brix$, 온도 $30^{\circ}C$, 발효기간 4일이었으며, 초산 발효의 최적 조건은 초기 알코올 함량 6%, 초기 산도 2%, 온도 $30^{\circ}C$, 발효기간은 9일이었다. 최적 조건으로 발효된 복분자 식초의 유리당은 surcose, fructose 및 glucose로 확인되었으며, 주요 유기산은 acetic acid, malic acid, succinic acid 및 malonic acid로 나타났다. 또한 유리아미노산 총 함량은 108.03 ${\mu}g/mL$로 alanine, glutamic acid, ${\gamma}$-amino-N-butyric acid 및 o-phosphoethanolamine가 주된 성분으로 나타났으며, 무기성분은 K, Na, Mg, Ca과 같은 알칼리성 원소를 다량 함유한 것으로 나타났다. 복분자 식초의 총 폴리페놀 및 안토시아닌의 함량은 각각 25.19, 80.71 mg/100 mL이었으며, DPPH, $ABTS^{{\cdot}+}$ 라디칼 소거 활성, 환원력, ${\beta}$-carotene bleaching의 실험에서 복분자 식초는 양성 대조군과 유사한 높은 항산화 활성을 가지는 것으로 나타났다.
Objective: Unlike mammals, goose fatty liver shows a strong tolerance to fatty acids without obvious injury. Stearyl-coenzyme A desaturase 1 (SCD1) serves crucial role in desaturation of saturated fatty acids (SAFs), but its role in the SAFs tolerance of goose hepatocytes has not been reported. This study was conducted to explore the role of SCD1 in regulating palmitic acid (PA) tolerance of goose primary hepatocytes. Methods: 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide was examined to reflect the effect of PA on hepatocytes viability, and quantitative polymerase chain reaction was used to detect the mRNA levels of several genes related to endoplasmic reticulum (ER) stress, inflammation, and apoptosis, and the role of SCD1 in PA tolerance of goose hepatocytes was explored using RNA interfere. Results: Our results indicated that goose hepatocytes exhibited a higher tolerant capacity to PA than human hepatic cell line (LO2 cells). In goose primary hepatocytes, the mRNA levels of fatty acid desaturation-related genes (SCD1 and fatty acid desaturase 2) and fatty acid elongate enzyme-related gene (elongase of very long chain fatty acids 6) were significantly upregulated with 0.6 mM PA treatment. However, in LO2 cells, expression of ER stress-related genes (x box-binding protein, binding immunoglobulin protein, and activating transcription factor 6), inflammatory response-related genes (interleukin-6 [IL-6], interleukin-1β [IL-1β], and interferon-γ) and apoptosis-related genes (bcl-2-associated X protein, b-cell lymphoma 2, Caspase-3, and Caspase-9) was significantly enhanced with 0.6 mM PA treatment. Additionally, small interfering RNA (siRNA) mediated downregulation of SCD1 significantly reduced the PA tolerance of goose primary hepatocytes under the treatment of 0.6 mM PA; meanwhile, the mRNA levels of inflammatory-related genes (IL-6 and IL-1β) and several key genes involved in the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT), forkhead box O1 (FoxO1), mammalian target of rapamycin and AMPK pathways (AKT1, AKT2, FoxO1, and sirtuin 1), as well as the protein expression of cytochrome C and the apoptosis rate were upregulated. Conclusion: In conclusion, our data suggested that SCD1 was involved in enhancing the PA tolerance of goose primary hepatocytes by regulating inflammation- and apoptosis-related genes expression.
The volatile flavor components were obtained from the aerial parts of Houttuynia cordata by a simultaneous distillation-extraction(SDE) method and bactericidal effects of the volatile flavor components on some strains were examined. Strong antibacterial activities were observed against Bacillus cereus, Bacillus subtilis, Vibrio cholerae 0-1 and Vibrio parahaemolyticus. To further elucidate the effective components in the extract, SDE extract was analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry(GC/MS). A total of 98 volatile compound were detected. Of these, 90 were confirmed including 6 hydrocarbons(0.34%), 12 alcohols(1.31%), 13 aldehydes(33.81%), 1 acetal(0.01%), 6 esters(1.16%), 2 acids(3.10%), 5 ketones(5.87%), 2 furans(0.06%), 1 phenol(0.18%), 41 terpenes(53.23%)and 3 miscellaneous compound(0.93%). Major components were determined to be $\beta$-mycene, decanal, cis-ocimene and 2-undecanone.

The electric transport of the mixture of hydrochloric and phosphoric acids through strong base (Neosepta ACM) and weak base (Selemion AAV) anion-exchange membranes was investigated. The instantaneous efficiency of HCl removal from the cathode solution, $CE_{Cl}$, with and without $H_3PO_4$ was determined. It was found that $CE_{Cl}$ was 0.8-0.9 if the number of moles of elementary charge passed through the system, $n_F$, did not exceed ca. 80% of the initial number of HCl moles in the cathode solution, $n_{Cl,ca,0}$. The retention efficiency of $H_3PO_4$ in that range was close to one. The transport of acid mixtures was satisfactorily described by a model based on the extended Nernst-Planck and Donnan equations for $n_F$ not exceeding $n_{Cl,ca,0}$. Among the tested model parameters, most important were: concentration of fixed charges, the porosity-tortuosity coefficient, and the partition coefficient of an undissociated form of $H_3PO_4$. For the both membranes, the obtained optimal values of fixed charge concentration, $\bar{c}_m$, were up to 40% lower than the literature values of $\bar{c}_m$ obtained from the equilibrium measurements. Regarding the $H_3PO_4$ equilibria, it was sufficient to consider $H_3PO_4$ as a monoprotic acid.
