Susceptibility of Two Potato Cultivars to Various Environmental Stresses (다양한 환경스트레스에 대한 감자 2품종의 감수성 분석)

  • Environmental stress is the major limiting factor in plant productivity. In order to evaluate the stress tolerance of potato plants, leaf discs of two potato cultivars, Atlantic and Superior, were subjected to various stress conditions of high temperature, methyl viologen, H2O2, or $H_2O$$_2$. When potato leaf discs were exposed to high temperature at 37$^{\circ}C$ for 84 hr, Atlantic plants, a cultivar with high sensitivity to heat stress, showed about 20% higher membrane damage than Superior plants. When exposed to 2$\mu$M methyl violgen (MV), a superoxide generating non-selective herbicide, for 36 hr, Atlantic plants also showed about 38% higher membrane damage than Superior plants, and were more susceptible up to 10$\mu$M MV concentration tested. On treatment with 0.75M NaCl, Atlantic plants also had about 45% less chlorophyll contents in leaf discs than Superior plants. There was, however, no difference in chlorophyll content of two cultivars at higher NaCl concentrations. The effect of $H_2O$$_2$ on the two cultivars was mixed. At low $H_2O$$_2$ concentration (25 mM) , Superior plants were more susceptible to $H_2O$$_2$stress after 36 hr. However, at high $H_2O$$_2$ concentration (100 mM), Atlantic plants exhibited higher susceptibility after 36 hr. The results indicate that in vitro leaf discs reflecting the whole plants in this study will be useful for selection and characterization of elite transgenic potato plants with enhanced tolerance to environmental stress.

Induction of Anthocyanin and Betaine by Salinity Stress in Germinating Seeds (발아중인 종자로부터 Salinity Stress에 의해 유도되는 Anthocyanin과 Betaine에 관안 연구)

  • The effect of salinity stress of Brassica olearacea and Capsicum annuum were studied at various levels of salinity conditions(Na-gluconate, K-gluconate, NaCl, KCl). The effects of salinity stress were measured by seedling growth rates and secondary metabolites contents of the stressed plants. Each seedling studied on the response of different salinity stress. Seedling growth of Capsicum annuum was inhibited up to 200 mM salt tolerance and Brassica olearacea was inhibited up to 400 mM salt tolerance. The produced anthocyanin was separated to high value from 200 mM NaCl in case of Brassica olearana and 50 mM K-gluconate in case of Capsicum annuum. The BADH activity was very high in Brassica olearacea seedlings treated with 200 mM NaCl and in Capsicum annuum seedlings treated with 100 mM K-gluconate. The BADH activities were increased during the early culture days, it induced betaine synthesis. The salinity stress promoted BADH activiy, subsequently endogenous betaine contents were increased, and it seemed to be secure seedling from salinity stress. The salinity concentration of 200 mM was effective on the inhibition of seed germination and on the increase of proline accumulation in tissue. The inhibition of seedling growth and accumulation of secondary metabolites in seedling were caused osmotic hypersensitivity against salinity stress.

Effect of Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 Inoculation on Seed Germination and Early Growth of Maize and Sorghum-sudangrass hybrid Seedling under Different Salinity Levels

  • Salinity is one of the most relevant abiotic factor limiting crop yield and its net primary productivity. In addition, salinity induces an increased stress ethylene synthesis in plants which, in turn, exacerbate the responses to the stressor. Bacterial single or co-inoculation effect was tested using previously characterized plant growth promoting (PGP) bacteria Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 on maize and sorghum-sudan grass hybrid under different concentrations of NaCl. Non-inoculated maize and sorghum-sudangrass hybrid showed 33.4% and 20.0% reduction in seed germination under highest NaCl (150 mM) level tested. However, under the same NaCl concentration, co-inoculation with B. iodinum RS16 and M. oryzae CBMB20 PGP strains increased the seed germination in maize (16.7%) and sorghum-sudangrass hybrid (4.4%). In Gnotobiotic growth pouch experiments conducted for maize and sorghum-sudangrass hybrid, co-inoculation of PGP B. iodinum RS16 and M. oryzae CBMB20 mitigated the salinity stress and promoted root length by 22.9% and 29.7%, respectively. Thus the results of this study could help in development of potential bioinoculants that may be suitable for crop production under saline conditions.

A study on the stability analysis for double deck tunnel branch geometry (복층터널 분기 기하구조에 따른 안정성 검토에 관한 연구)

  • The tunnel can be planned to connect to underground roadway and surface road. The large tunnel and branch section are made when the ramp tunnel access to the main tunnel. In the branch section, stress concentration can be assigned and it can be very important for the stability of the tunnel. This study assessed the behavior of rock pillar in double deck tunnel diverging area by using a two dimensional numerical analysis. This study evaluated different safety factors according to pillar width and the ramp tunnel position in branch. By the assessment of the strength-stress ratio, tunnel pillar width is suggested in order to secure the safety factor 1.5.

Flaw Assessment on an Offshore Structure using Engineering Criticality Analysis (ECA 기법을 이용한 해양구조물의 결함 평가)

  • Offshore structure may be considerably vulnerable to fatigue failure while initial flaw propagates under cyclic loading, so crack propagation analysis/fracture/yield assessments about initial flaw detected by NDT are necessarily required. In this paper, case studies have been conducted by flaw assessment program using engineering criticality analysis (ECA) approach. Variables such as flaw geometry, flaw size, structure geometry, dynamic stress, static stress, toughness, crack growth rate, stress concentration factor (SCF) affected by weld are considered as analysis conditions. As a result, the safety of structure was examined during fatigue loading life. Also, critical initial flaw size was calculated by sensitivity module in the developed program. The flaw assessments analysis using ECA approach can be very useful in offshore industries owing to the increasing demand on the engineering criticality analysis of potential initial flaws.

Stress analysis of the effect of debonding of cement-femoral stem interface to the bone-cement interface -A three-dimensional Finite Element Analysis- (시멘트-대퇴Stem 경계면 해리가 골-시멘트 경계면에 미치는 응력 분석 -3차원 비선형 Finite Element Analysis-)

  • Debonding of cement-femoral stem interface has been suggested as a initial focus of loosening mechanism in many previous studies of cemented total hip replacement. The purpose of this study was to investigate the effect of debonding of cement-femoral stem interface to the bone-cement interface by using three-dimensional non-liner finite element analysis. Three cases of partial debonded, full debonded, full bonded cement-bone interface were modelled with partial bonding of distal 70mm from the tip of femoral stem. Each situation was studied under loading stimulating one-leg stanced gait of 68kg patient. The results showed that under partial and full debonded cement-stem interface condition the peak von Mises stress(3.1 MPa) were observed at the cement of bone-cement interface just under the calcar of proximal medial of femur, and sudden high peak stresses(3.5MPa) were developed at the distal tip of femoral stem at the lateral bone-cement interface in all 3 cases of bonding. The stresses were transfered very little to the cement of upper lateral bone-cement interface in partial and full debonded cases. Thus, once partial or full debonded cement-femoral stem interface occured, 3 times higher stress concentration were developed on the cement of proximal medial bone-cement interface than full bonded interface, and these could cause loosening of cemented total hip replacement. Clinically, preservation of more rigid cement-femoral stem interface may be important factor to prevent loosening of femoral stem.

Structural Improvement for Crack of Integrated Circuit in Single Board Computer by Structure Analysis (단일보드컴퓨터 구조해석을 통한 집적회로 균열현상의 구조적 개선)

  • In this study, we aim to derive a solution from the structural analysis for electrical failure of single board computers for computing navigation information. By analyzing the characteristic factor, we identify that crack occur on the central processing unit board due to a certain structural problem, and that the physical effect by the crack make communication function be impossible to perform, which it causes booting error. In order to find the location of excessive stress causing the crack, structural analysis for the single board computer is done. From the structural analysis, the areas where stress concentration occurs are identified, and improvement methods changing the structures are developed. As a result, we shows that stresses are reduced entirely on the stress distribution for the improved structure. In addition, heat analysis shows that changing the structure to reduce stresses is not affect to the heat radiation, and the thermal resistance of the actual equipment is verified by measuring the temperature of the heat sink applied with the improved structure.

  • An approach using complex potentials is presented for analysis of composite plate with an elliptical hole or a rectilinear crack. Composite structure is susceptible to encounter impact damages, which lead to considerable decrease in its residual strength. Such impact damages could be modeled as an equivalent elliptical hole or notch-like crack. Even though finite element method is widely used to analyze stresses or fracture mechanics parameters around such damage, it is tedious to make successive FE-modeling for damage tolerance assessment under fatigue loadings. In this point of view, the solutions based on complex potentials are very simple and easy to use. The computed results are also compared and discussed with those from FEA.

Effect of onion (Allium cepa L.) peel extract on natural killer cell and cytokines in a randomized, double-blind, placebo-controlled trial

  • BACKGROUND/OBJECTIVES: Onion, particularly onion peel, is a quercetin-rich food with, anti-inflammatory and immunomodulatory effects. However, the effect of onion peel extract (OPE) in humans is unclear. Thus, the present study aimed to investigate whether OPE improves natural killer (NK) cell activity and cytokine concentration in a randomized double-blind placebo-controlled trial. SUBJECTS/METHODS: Eighty participants aged 19-64 yrs old with a white blood cell count of 4,000-10,000 cells/µL, symptoms of upper respiratory infection at least once within the previous 12 mon, and perceived stress scale (PSS) over 14 were included. Participants were randomly assigned to take either 1,000 mg/day OPE or a placebo for 8 weeks. RESULTS: Compliance were 87.4 ± 8.6% and 86.9 ± 79.0% in OPE and placebo groups. Compared to the placebo, OPE supplementation improved "Hoarseness" (P = 0.038) of the Wisconsin Upper Respiratory Symptom Survey (WURSS)-21 symptom, and stress scores (P = 0.001; 0.021) of PSS. Supplementation of OPE had no significant effect on NK cell activity and concentrations of cytokines such as interleukin (IL)-2, IL-6, IL-12, IL-1β, interferon-γ, and tumor necrosis factor-α. At baseline, the WURSS-21 symptom and PSS score (P = 0.024; 0.026) were higher in the OPE group than the placebo group. Among participants with higher than median WURSS-21 symptom score, OPE supplementation increased NK cell activity (P = 0.038). Supplementation of OPE had no significant effects on safety measurements and adverse events. CONCLUSIONS: The present study suggested that OPE supplementation improves NK cell activity in participants with moderate upper respiratory symptoms without any significant adverse effects.

Improvement of Fatigue Life with Local Reinforcement for Offshore Topside Module during Marine Transportation (해양플랫폼 탑사이드 모듈의 해상 운송 시 국부 보강을 통한 피로 수명 개선에 관한 연구)

  • In this study, finite element analysis was performed to evaluate a method of increasing the fatigue life of the pipe connection structure commonly used in the topside structure of offshore platforms. MSC Patran/Nastran, a commercial analysis program, was used, and the critical structural model was selected from the global analysis. To realize the stress concentration phenomenon according to the load, modeling using 8-node solid elements was implemented. The main loads were considered to be two lateral loads and a tensile load on a diagonal pipe. To check the hotspot stress at the main location, a 0.01 mm dummy shell element was applied. After calculating the main stress at the 0.5-t and 1.5-t locations, the stress generated in the weld was estimated through extrapolation. In some sections, this stress was observed to be below the fatigue life that should be satisfied, and reinforcement was required. For reinforcement, a bracket was added to reduce the stress concentration factor where the fatigue life was insufficient without changing the thickness or diameter of the previously designed pipe. Regarding the tensile load, the stress in the bracket toe increased by 23 %, whereas the stress inside and outside of the pipe, which was a problem, decreased by approximately 8 %. Regarding the flexural load, the stress at the bracket toe increased by 3 %, whereas the stress inside and outside of the pipe, which was also a problem, decreased by approximately 48 %. Owing to the new bracket reinforcement, the stress in the bracket toe increased, but the S-N curve itself was better than that of the pipe joint, so it was not a significant problem. The improvement method of fatigue life is expected to be useful; it can efficiently increase the fatigue life while minimizing changes to the initial design.
