Trypsin Inhibitor from Streptomyces sp. ( Part 1) Isolation of microorganism and purification of the inhibitor (Streptomyces 속 균주가 생성하는 Trypsin Inhibitor (제1보) 균의 분리 및 저해물질의 정제)

  • One strain of Streptomyces sp. (AS-707) isolated from soil was found to produce a biologically active substance that showed a strong inhibitory activity against proteolytic enzymes viz. trypsin, papain, $\alpha$-chymotrypsin, Azotobacter protease, and Bacillus pretense. The substance was separated from culture filtrate by ion exchange column chromatography using Amberlite IRC-50 and CM-cellulose column chromatography. It was found that the recovery yield was 26% as activity basis. The substance was stable in wide pH range from 2.0 to 12.0 at 37$^{\circ}C$, but it was unstable in alkaline pH values at 6$0^{\circ}C$. The activity was thermostable to give 90% activity compared to the intact sample when it was treated at pH5.6 at 10$0^{\circ}C$ for 2 hours.

Selection of Antagonistic Soil Actinomycetes Against Both Colletotrichum orbiculare and Botrytis cinerea in Cucumber Plants (오이 탄저병원균과 잿빛곰팡이병원균에 억제 효과가 있는 방선균 선발)

  • The purpose of this study is to select actinomycetes with antifungal activity against Botrytis cinerea and Colletotrichum orbiculare, which are airborne pathogens in cucumber plants. In 2020, 560 species of actinomycetes were isolated from rhizome soils of various vegetables in Korea; 7 strains showing simultaneous antifungal activity against two pathogens were selected. Finally, strain S20-465 was selected through dual culture and plant assay. This strain was identified as Streptomyces sp. based on 16S rRNA analysis. The culture filtrate of strain S20-465 inhibited mycelial growth of both pathogens by more than 60%. In addition, when cucumber plants were treated with 20-fold and 40-fold diluted culture filtrates of S20-465, lesions caused by B. cinerea and C. orbiculare on cucumber leaves were significantly reduced compared to the control. This results suggest that strain S20-465 produces specific secondary metabolites with antifungal activity against both pathogens.

  • The two isolates of Streptomyces antagonistic to Phytophthora nicotianae var. parasitica and Fusarium oxysporum f. sp. vasinfectum were identified as based on the morphological, cultural and physiological characteristics on various culture media. Spore chains of St-11 isolate was rectus-flexibilis(RF), whereas the other isolate, St-20, was shown rectinaculum-apertum(RA). Spore surface of St-11 isolate was smooth, while St-20 was spiny. Aerial mycelia of the two isolates were all gray color and growing conditions on media were good as a whole. Any soluble pigment was not shown in cultivation of the two isolates. Stoll isolate showed negative response on starch hydrolysis and gelation liquefaction, whereas St-20 isolate was positive on starch hydrolysis and a negative on gelatin reaction. Stoll isolate was identified as Streptomyces bikiniensis and St-20 Streptomyces echinoruber, respectively.

Screening and Identification of Alkaline Protease Inhibitor-Producing Marine-derived Actinomyces. (Alkaline protease inhibitor를 생산하는 해양유래 방선균의 탐색 및 동정)

  • In this study, we screened and identified the bacterial strain showing high alkaline pretense inhibitor activity from marine environment. Nine bacterial strains with alkaline pretense inhibitor activity were isolated from marine sediments. Among them, strain C12 had the highest alkaline pretense inhibitor activity and was selected for further taxonomical study. On the basis of morphological and physiological characteristics, strain C12 was identified as the genus Streptomyces. A phylogenetic analysis of the 165 rDNA showed that the isolated strain was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyces thermocarboxydus. Morphological characteristics showed cylindrical spore chain and smooth spore surface by scanning electron microscope. Strain C12 was grown on all media except for ISP 9 agar. This strain could be grown in the medium containing up to 9% NaCl.

Purification of Alginate Lyase from Streptomyces violaceoruber and the Growth Activity of Intestinal Bacteria by Degree of Polymerization of Alginate Hydrolysates (Streptomyces violaceoruber 유래 Alginate Lyase의 정제 및 Sodium Alginate 가수분해 올리고당의 중합도별 Bifidobacterium spp.과 Lactobacillus spp.에 대한 생육활성)

  • Alginate lyase from Streptomyces violaceoruber was purified by DEAE sephacel chromatography and SP sepharose chromatography. The specific activity of the purified enzyme was 14.6 units/mg protein, representing a 40.6-fold purification of the crude extract. The final preparation thus obtained showed a single band on Tricine-SDS polyacrylamide gel electrophoresis whose molecular weight was determined to be 23.3 kDa. The polyMG block of sodium alginate was hydrolyzed by the purified alginate lyase and then separated by activated carbon column chromatography and bio gel P-2 gel filtration. The main hydrolysates were composed of hetero type M/G-oligosaccharides with the degrees of polymerization (D.P.) being 6 and 8. To investigate the effects of hetero type M/G-oligosaccharides from the sodium alginate on the growth of some intestinal bacteria, cells were cultivated individually on the modified-MRS medium containing D.P. 6 and 8 M/G-oligosaccharides. B. longumgrew 4.25-fold and 6.44-fold more effectively by the treatment of D.P. 6 and 8 M/G-oligosaccharides compared with those of standard MRS medium. In addition, B. bifidumgrew 3.3-fold and 5.4-fold more effectively by the treatment of D.P. 6 and 8 M/G-oligosaccharides. In conclusion, D.P. 8 was more effective than D.P. 6 hetero M/G-oligosaccharides as regards the growth of Bifidobacteriumspp. and Lactobacillus spp.

Distribution and Antibiotic Production Characteristics for Streptomyces (Streptomyces의 토양중(土壤中) 분포(分布) 및 항생물질생산(抗生物質生産))

  • Out of 826 isolates of Streptomyces isolated from different soils, their distribution and antibiotic productivity were investigated. Distribution of the organism in the soil was affected by the soil conditions and plants. The highest isolation frequency was occurred from Quercus forest, Robinia forest and grass field, while soils from orchards and cultivating fields showed low density of Streptomyces. More than 49% of the isolates showed antibacterial activity against Bacillus subtilis, Erwinia carotovora subsp. carotovora and Xantomonas campestris pv. oryzae and about 40% of the isolates showed antiyeasty activity against Saccharomyces cerevisiae but only a few isolates showed antibiotic activity against E. coli and Pseudomonas solanacearum. Forty isolates of the Streptomyces showed strong antifungal activity against Pyricularia oryzae. Rate of isolation of Streptomyces was the highest on starch agar among the eight media tested. Antibiotic productivity of the isolates was the highest on potato sucrose agar medium among the 5 media tested.

Production and Purification of Alkaline Protease from Streptomyces sp. (Streptomyces속 균주가 생성하는 Alkaline Protease의 생산 및 정제)

  • An alkaline protease producing microorganism was isolated from soil and identified as Streptomyces griseus HC-1141. The optimum culture condition of Streptomyces griseus HC- 1141 for the production of alkaline protease was as follows; 0.5% casein, 0.05% ammonium chloride, 0.1% ferrous sulfate. 2.0% lactose, pH 8.0 and 84 hrs. The enzyme was purified about 53 folds by ammonium sulfate treatment, DEAE-cellulose ion exchange chromatography and gel filtratioo on Sephadex G-150. The homogeneity of the purified enzyme was verified by polyacrylamide gel electrophoresis. The molecular weight was estimated to be 31,000 on sodium dodecyl sulfate polyacrylamide gel electrophoresis. This enzyme consists of glycine and glutamic acid as major amino acids. The N-terminal and C-terminal residues of the alkaline protease were leucine and histidine respectively.

Zinc-Solubilizing Streptomyces spp. as Bioinoculants for Promoting the Growth of Soybean (Glycine max (L.) Merrill)

  • Zinc-solubilizing bacteria can convert the insoluble form of zinc into soluble forms available to plants. This study was conducted to isolate and screen zinc-solubilizing actinobacteria from rhizosphere soils and to assess their effect on vegetable soybean growth. In total, 200 actinobacteria strains belonging to 10 genera were isolated from rhizosphere soil samples. Among these isolates, four showed zinc solubilization with solubilizing index values ranging from 3.11 to 3.78 on Bunt and Rovira agar supplemented with 0.1% zinc oxide. For the quantitative assay, in broth culture, strains CME34 and EX51 solubilized maximum available zinc contents of 529.71 and 243.58 ㎍/ml. Furthermore, indole-3-acetic acid (IAA) and ammonia were produced by these two strains, the strain CME34 produced the highest amount of IAA 4.62 ㎍/ml and the strain EX51 produced the highest amount of ammonia 361.04 ㎍/ml. In addition, the phosphate-solubilizing abilities in Pikovskaya's medium of CME34 and EX51 were 64.67 and 115.67 ㎍/ml. Based on morphological and biochemical characterization and 16S rDNA sequencing, the strains CME34 and EX51 were closely related to the genus Streptomyces. In a greenhouse experiment, single-strain inoculation of Streptomyces sp. CME34 or EX51 significantly increased the shoot length, root length, plant dry weight, number of pods per plant and number of seeds per plant of vegetable soybean plants compared to the uninoculated control. These findings facilitated the conclusion that the two Streptomyces strains have potential as zinc solubilizers and can be suggested as bioinoculants to promote the growth and yield of soybean.

An Antibiotic from Actinomycetes Becoming Effective for Cephalosporin Resistant Pathogenic Pesudomonas sp. (방선균이 생산하는 Cephalosporin 내성 병원성 Pseudomonas에 유효한 항생물질)

  • We isolated activnmycetes LAM-98-80 as strain producing an effective antibiotic for cephalosporin re-sistant pathogenic PSeudomonas sp. and identified as Streptomyces sp. LAM-98-80 from cultural and phyisological characteristics. We investigated the optimal culture conditions for producation of an anti-biotic becoming effective for cephalsporin-resistant pathogenic Pseudomonas sp. It was found that 1.5% soluble starch and 1.0% yeast extract were good as carbon and nitrogen source respectively. The pro-duction of antibiotic was also activated by 0.04% Mn2+ as 80% degree. The optimum initial pH on pro-ductio of antibiotic was pH 7.0. The culture condition for the maximal productivity of the antibiotic was at 3$0^{\circ}C$ for 5 days. The cephalosporin-resistant pathogenic Pseudomonas sp. as test bacteria was rev-ealed to resist antibiotic of cepha families but revealed to not resist those of $\beta$-lactam families ampicil-lin and amoxicillin. Parital purified antibiotic was stable for the pH from 3 to 9 and was also stable when treated at 70 $^{\circ}C$ for 1 hour, This antbiotic was effective against all gram positive and negative bac-teria but was not effective against molds and yeasts.
