토양에서 분리한 Streptomyces nigrifaciens GMT-497호무터 G(+) bacteria에만 강한 항균활성을 갖는 항생물질 MT-497을 용매추출, silica column chromatography와 재결정화를 통하여 분리 정제하였다. MT-497의 UV, 융점, 원소분석, IR spectrum, $^1H-NMR$과 구성아미노산 분석을 통해 actinocin chromophore와 threonine, proline, methyl valine, sarcosine, aspartic acid로 구성된 peptide을 갖는 actinomvcin계열의 항생물질로 동정하였다.

우리나라 토양중의 Streptomyces를 분류, 동정하고자 토양으로부터 총 826 균주이 Streptomyces를 분리하여 1차 배양하고 형태적, 생리적 특성이 다른 65 균주를 선발하여 I.S.P 방법에 따라 총 46 종의 Streptomyces를 동정하였으며 그 중 국내미기록종은 38종으로서 마음과 같다. 1) melanin 색소형성균 S. lavendulae, S. tuirus, S. purpurascens, S. violarus, S. lateritius, S. olivochromogenes, S. purpeofuscus, S. fulvoviolaceus, S. gallilaeus, S. naganishii, S. hygroscopicus subsp. ossamyceticus, S. nevagawaensis, S. anandii, S. phaeopurpureus, S. mirabilis, S. arenae, S. massasporeus, S. eurythermus. 2) Melanin 색소 비형성군 S. fellus, S. flavidovirens, S. exfoliatus, S. fumanus, S. termitum, S. glomeroaurantiacus, S. luteofluorescens, S. prunicolor, S. fradiae, S. tauricus, S. griseolus, S. misakiensis, S. poonensis, S. antimycoticus, S. diastaticus subsp. ardesiacus, S. nigrifaciens, S. tendae, S. narbonensis subsp. josamvceticus, S. atroolivaceus, S. chrysomallus subsp. fumigatus.

Identification of the Actinomycetes isolate strain No. 497 producing an actinomycin antibiotic MT-497 was performed by ISP and chemotaxonomic methods. The strain Nu. 497 formed various shapes of sclerotia and smooth surface spore. Menaquinone MK-9 ($H_6, H_8$) and iso-, anteiso-branched $C_{15}C_{17}$ fatty acids were detected from whole cell extract. The wall chemotype of stram No. 497 was decided as wall chemotype I from the analysis of DAP isomer, peptidoglycan type and sugar pattern. From these morphological, chemotaxonomic characteristics and analysis of various physiological characteristics. the strain No. 497 was identified as Streptomyces nigrifaciens.
