4,4'-Biphenol 유도체의 합성 및 충치균 Streptococcus mutans OMZ 176에 대한 항균작용 (Synthesis of 4,4'-Biphenol Derivatives and Antibacterial Activities against a Cariogenic Bacterium, Streptococcus mutans OMZ 176)

  • In a continuous study for the developing of the anticariogenic agents, 3,3'-diacyl-4,4'-biphenol derivatives (Fig. 2, 2 and 3) and 3,3'-bis-(1-hydroxyalkyl)-4,4'-biphenol (Fig. 3, 4 and 5) derivatives are synthesized successively from 4,4'-diphenol (Fig. 2, 1). The synthesized compounds are tested for their antibacterial activity against a cariogenic bacterium, Streptococcus mutans OMZ 176. The acyl derivatives, 2 and 3, do not show antibacterial activity, but the hydroxyalkyl derivatives, 4 and 5, reduced from the acyl group of 2 and 3, show the activity. The antibacterial activity of 2 and 3 may be inhibited due to intramolecular hydrogen bonding between the acyl group and the hydroxyl one (Fig. 4).

淸熱消毒飮 및 구성약물의 항균활성에 관한 실험적 연구 (The Antimicrobial Activity of Chungyulsodokeum and Its Composition Oriental Medicines)

  • Antimicrobial activities against Streptococcus mutans, Candida albicans, Escherichia coli, Staphylococcus epidermidis, Staphylococcus aureus were assayed through the hot water extracts and the ethanol extracts from Chungyulsodokeum and its composition oriental medicines. The stains were incubated on culture medium and activated. We observed the size of inhibition zone on the strains that is incubated in strilized filter paper disc with various concentration extracts of Chungyulsodokeum and its composition 1. The extracts of Chungyulsodokeum, Coptidis Rhizoma, Paeoniae Radix, Glycyrrhizae Radix, Gardeniae Fructus showed antimicrobial activities against Staphylococcus aureus. 2. The extracts of Chungyulsodokeum, Coptidis Rhizoma, Paeoniae Radix, Forsythiae Fructus Ghycyrrhizae Radix, showed antimiaobial activities against Staphylococcus epidermidis. 3. The extracts of Chungyulsodokeum, Coptidis Rhizoma, Paeoniae Radix, Glycyrrhizae Radix, showed antimicrobial activities against Streptococcus mutans. 4. The extract of Coptidis Rhizoma showed antimicrobial activities against Candida albicans. 5. None of the extracts showed antimicrobial activities against Escherichia coli. We observed antimicrobial activities of Chungyulsodokeum and its composition against Streptococcus mutans, Staphylococcus epidermidis, Staphylococcus aureus.

충치균, Streptococcus mutans OMZ176에 대한 황금의 항균활성성분 (Antibacterial Constituents from Scutellariae Radix against Streptococcus mutans OMZ176)

  • To develop anticariogenic agents, Scutellariae Radix was tested for its antibacterial activity against Streptococcus mutans OMZ176 which is known as a strong cariogenic bacterium. The antibacterial activity was evaluated with disk plate method and minimal inhibitory concentration (MIC). The ethyl acetate fraction showed significant activity. By means of activity-guided fractionation, baicalein and ganhuangenin were isolated as the active principles. The MICs of these compounds were $100{\mu}g/ml$ and $200{\mu}g/ml$, respectively.

  • In order to develop natural anticariogenic agents, we investigated inhibitory effects of Areca catechu L. extracts on the acid production of Streptococcus mutans JC-2 and its cytotoxicity on NIH 3T3 fibroblasts were also examined. The results are as follows : 1. Major organic acid produced by Streptococcus mutans JC-2 were lactic acid and acetic acid, and their productions were decreased by additions of Areca catechu L. extracts. 2. Areca catechu L. extracts were showed noncytotoxicity on NIH 3T3 fibroblasts.

  • 천연항균제의 개발에 대한 관심이 날로 증가하는 가운데, 다수의 연구들에서 Escherichia coli, Salmonella typhimurium, Pseudomonas aeruginosa, Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus subtilis, Aspergillus parasiticus, Helicobacter pylori 등 다양한 세균들에 대한 서양산 고추냉이의 항균 효과가 밝혀진 바 있다. 서양산 고추냉이가 치아우식증의 원인균인 Streptococcus mutans에 대해서도 항균성을 나타낸다는 보고가 있으나, 아직까지 구강내에서 분리된 임상분리균주를 대상으로 한 연구는 미진한 실정이다. 본 연구에서는 사람의 구강내에서 분리 및 동정된 Streptococcus mutans에 대한 서양산 고추냉이 뿌리 추출물의 항균 효과를, 대표적 항균제인 클로르헥시딘과 비교하여 알아보고, Streptococcus mutans 표준균주에 대한 서양산 고추냉이 뿌리 추출물의 효과와 비교하고자 하였다. 항균 효과를 평가하기 위해 최소억제농도와 최소살균농도를 측정하여 다음과 같은 결론을 얻었다. 1. 본 연구에서 Streptococcus mutans 임상 분리균주와 표준균주에 대한 서양산 고추냉이 뿌리 추출물의 최소억제 농도는 각각 평균 $0.083{\sim}0.25%$ ($833.33{\sim}2500 ppm$), 평균 0.25%(2500 ppm)으로 나타나, 두 종류의 균주에 대한 항균 효과에는 뚜렷한 차이가 없는 것으로 나타났다. 2. 서양산 고추냉이 뿌리 추출물은 $0.083{\sim}0.25%$의 농도에서 클로르헥시딘($0.0021{\sim}0.0041%$)과 대등한 항균 효과를 가지는 것으로 나타났다.

  • The effect of topical application on the number of S. mutans was tested in a group of 44 orthodontic patients (mean age, 12Y 3M). They were divided into 5 groups according to the method using NaF and $SnF_2$. The number of S. mutans CFU were counted in stimulated saliva of each subject at baseline, and after one, two, three, and eight weeks. The following results were obtained. 1. In NaF rinsing group, and NaF topical application and NaF rinsing group, the number of S. mutans per ml saliva was not significantly changed. 2. In $SnF_2$ topical application group, and $SnF_2$ topical and NaF rinsing group, the number of S. mutans per ml saliva was significantly reduced. 3. After 8 weeks, there were no significant reduction of the number of S. mutans in comparison with baseline.

Isolation and Characterization of the Streptococcus mutans from Korean Children with Caries

  • Streptococcus mutans, one of a major causal agents of dental caries, is component of the dental plaque and produces various organic acids such as lactic acid as the end-product of glycolysis. In this study, we isolated S. mutans from Korean children with caries and also investigated the expression of protein under acid stress. S. mutans was identified at the species level using a 16S ribosomal DNA sequencing comparison method. The primer specificity was tested on eleven S. mutans strains isolated from Korean children with caries. The data showed that eleven strains are S. mutans. At treatment of concentration of 20 mM lactic acid in the mid-log phage, K-7 exhibited the highest maximum culture OD compared with those of other groups. As a consequence, we examined the expression of protein under 20 mM lactic acid stress using S. mutans K-7. The results of 2D gel electrophoresis by image analysis showed that thirteen proteins are up-regulated under the stress. Further study is being focused on amino acid analysis by mass spectrometry in order to analyze those spots.

  • Streptococcus mutans were isolated from dental plaques of carious lesions of 4 patients on mitis-salivarius agar medium. Three patients known to harbor S. mutans in their dental plaques. Identification of the isolated S. mutans was established by colonial morphology on mitis-salivarius agar medium, the fermentation of mannitol and sorbitol, and confirmed by agglutinating reaction with home made anti-S. mutans NCTC 10449 (serotype c) antiserum. Of the isolated S. mutans, one strain (P2-1) showed strong agglutinating reaction with antiserum, another strain (P1-2) showed weak agglutinating reaction. P2-1 strongly adhered to the wall of the test tube containing 5% sucrose broth, while p1-2 weakly colonized on the wall of the test tube. Biotyping of the isolated S. mutans based on the fermentation of mannitol, sorbitol, raffinose and melibiose, and the production of ammonia from L-arginine, and the inhibition of acid production by bacitracin. Biochemical characteristics of P2-1 strain correlated with the recognized biotype c, pl-2 strain resembled biotype d of S. mutans.
