Spatial and Temporal Distribution of a Biocontrol Bacterium Bacillus licheniformis N1 on the Strawberry Plants

  • Spatial and temporal distribution of Bacillus licheniformis N1 was investigated over time on the leaves, petioles and crowns of the strawberry plants. Bacterial population on the strawberry plants was quantified over time by selective plating. Bacterial population of N1 containing a plasmid pWH43G carrying green fluorescent protein (GFP) declined relatively faster on the plant surface as compared to the Strain N1 itself. However, this result was found to be enough to utilize the strain to visualize bacterial colonization on the plant surface. When B. licheniformis N1 was treated together with Silwet L-77 at 0.03%, the bacterial population on plant surface persisted for up to 7 days. B. licheniformis N1 (pWH43G) containing Silwet L-77 was applied on the strawberry plants and the GFP expressing bacteria were visualized by confocal laser scanning microscopy. Bacterial persistence was also investigated in a growth chamber and in a plastic house after N1 bioformulation treatment on the strawberry plant. The Strain N1 colonized three different tissues well and persisted over 3 to 5 days on the strawberry plants. They formed bacterial aggregates on plant surfaces for at least 3 days, resulting in a biofilm to resist fluctuating plant surface environment. However, the bacterial persistence dramatically declined after 7 days in all tested tissues in a plastic house. This study suggest that B. licheniformis N1 colonizes the strawberry plant surface and persists for a long time in a controlled growth chamber, while it can not persist over 7 days on the plant surface in a plastic house.

Crown Rot of Strawberry (Fragaria ananassa) Caused by Phytophthora cactorum (Phytophthora cactorum에 의한 딸기 역병)

  • A severely wilting of strawberry caused by a Phytophthora sp. has occurred houses after planting in vinyl-houses field at Ssanglim and Anlim areas Kyungbuk in Korea from October in 1997. Phytophthora sp. isolated from diseased tissues of the crown of strawberry. Browning rot of inner crown and root resulted in wilt and eventual death of the plant. The causal fungus was identified as Phytophthora cactorum. Sporangia were ovoid, conspicuously papillate, caducous and measured 30.0~56.6$\times$23.8~35.2 (av. 39.3~29.9) ${\mu}{\textrm}{m}$. Sexuality of the fungus was homothallic. Oogonia were sperical and 23.3~32.3 (av. 29.0) ${\mu}{\textrm}{m}$ in size. Most ahteridia were paragynous and measured av. 10.2~12.2 ${\mu}{\textrm}{m}$. Cardinal temperature for growth at minimum, optium, maximum were recorded at 7, 20~25, and 32$^{\circ}C$, respectively. The fungus show strong pathogenicity to strawberry. This is the first report of strawberry caused by Phytophthora cactorum in Korea.

Effects of Saline Irrigation Water on Crop Growth in Strawberry and Red Radish (딸기 및 적환무의 관개용수 염도수준에 따른 생육영향 분석)

  • Since the salinity of irrigation water is a critical constraint to the production of certain vegetable crops, salinity was considered as one of the most important factors of irrigation water. The purpose of this study were to monitor and assess the effects of saline irrigation water on strawberry and red radish growth in protected cultivation. One control and three treatments, which were differentiated according to the level of salinity in irrigated water, were employed for each vegetable to assess the effects of the irrigation with saline water. Monitoring has shown that using irrigation water with salinity above a certain level causes excessive accumulation of sodium (Na+) in both strawberry and red radish. Increased Na+ content was analyzed to be able to decrease the sugar content in strawberry. In addition, the salinity higher than the threshold level of irrigation water was found to reduce the growth and yield of strawberry and red radish. This study could contribute to suggest criteria for safe use of saline water in protected cultivation, although long-term monitoring is needed to get more representative results.

Mycological Characteristics of Phytophthora nicotianae var. nicotianae Causing Phytophthora Rot of Strawberry and Resistance of Strawberry Cultivars to the Pathogen (딸기 역병균 Phytophthora nicotianae var. nicotianae의 균학적 특성 및 딸기 품종간 저항성)

  • Mycological characteristics of Phytophthora nicotianae var. nicotianae SPC10 (A1 type) causing Phytophthora rot of strawberry and the resistances of 11 strawberry cultivars against the pathogen were examined. Optimum temperature for the mycelial growth of the pathogen was obtained in the range of 30~35$^{\circ}C$, and the growth was completely stopped under 13$^{\circ}C$ or over 42$^{\circ}C$. Aerial mycelia were abundant on oatmeal agar (OMA), V-8 juice agar (V8A) and lima bean agar (LBA) medium, although there were slight differences, however, on cornmeal agar (CMA) medium, it was a shape of stellate without aerial mycelia. The colony shape on potato dextrose agar (PDA) medium was rough and irregular whereas the mycelial growth was slow, and some aerial mycelia were only produced in the middle of PDA medium. Optimum temperature for sporangial formation was 3$0^{\circ}C$, and zoospores were mostly released at $25^{\circ}C$ from the sporangia. Sporangia were more produced in C/Z solution with pH 5. 0~6.$0^{\circ}C$ than sterilized distilled water (DSW) and distilled water (DW), and zoospores were also released much more than other solutions. Eleven strawberry cultivars such as Reiko, Hokowase, Eyeberry, Akaneko, Sistakara, Toyonoka, Nyoho, Sulhong, Suhong, Myhong and Wonkyo #3104 revealed the disease incidence up to 88.9~100% by the leaf inoculation with mycelial disk. However, Nyoho and Suhong showed higher level of resistance against the pathogen by root inoculation.

Effect of Natural Additives on In Vitro Growth Medium of Strawberry 'Seolhyang' (배지내 천연유기물 첨가가 딸기 "설향" 배양묘 기내 생육에 미치는 영향)

  • Strawberry which is the genus Fragaria under family Rosaceae is one of the most important fruit plants for both fresh consumption and food processing in the temperate and subtropical countries. Propagation of strawberry is achieved either through runners or by in vitro micropropagation. Meristem tips, generally obtained from runners of virus-free plants, are commonly used to establish in vitro cultures, which are employed for mass propagation or as a source of plant material for regeneration and transformation experiments. This study was conducted to determine the optimal natural additives strength to improve sprouting shoot rate of apical meristem of strawberry 'Seolhyang'. Strawberry apical meristem at size (0.2 mm to 0.3 mm) with leaf primordials were cultured on the 1/3MS(Murashige & Skoog) medium supplemented with five natural additives such as coconut milk, maple sap, banana powder and peptone. The sprouting ratio and growth characteristics were evaluated after eight weeks after in vitro culture. Shoot ratio of 'Seolhyang' apical meristem was 72.9% in 1/3MS medium supplemented with maple sap. On the other hand, the low shoot ratio was observed 47.7% in 1/3MS medium supplemented with banana powder. Shoot length was different as natural additives but numbers of leaf was not significantiy different among the natural additives. As a result, the sprouting ratio and plant growth were enhanced effectively in 1/3MS medium with maple sap compared to the others.

Characteristics, Stability and Reisolation of nit Mutant of Fusarium oxysporum from Strawberry (딸기로부터 분리된 Fusarium oxysporum nit 변이주의 특성과 안정성 및 재분리)

  • This study was conducted to investigate the potential of nitrate-nonutilizing mutants (nit mutants) in ecological studies of Fusarium disease of strawberry. Nit mutants of Fusarium oxysporum from strawberry were easily formed on chlorate-containing media. Nit mutants were assigned to three phenotypic classes, nit1, nit3, and NitM, on the basis of their growth on media containing one of the following five different nitrogen sources ; nitrate, nitrite, hypoxanthine, ammonium and uric acid. Frequency of nit mutation and proportion of three phenotypes of nit mutants depended on the isolate. Mutation rate was 45.6% and ranged from 15.0% to 95.0%. The frequency of nit1 mutants was higher than that of nit3 or NitM. The complementary reaction between nit1 and NitM was higher than that of other combination. There has been no complementary response observed between nit3 and nit3. The nit mutants showed similar growth pattern as the that of wild type isolate on potato sucrose agar and potato sucrose liquid media. Most of the mutants retained pathogenicity, and maintained their phenotypes even after two year preservation through subculture on slanted PSA at room temperature. Nit mutants were selctively isolated from infested soil and infected plants on the selective medium (MMCPA) containing potassium chlorate with their original phenotypes, while naturally occurring isolates of Fusarium oxysporum were not grow on the medium. On the contrary, nit mutants showed very slight growth on the medium (MMPA) containing nitrate as a sole nitrogen source, and therefore could be distinguished from wild type isolate.

Effect of Root Zone Temperature on Root and Shoot Growth of Strawberry (딸기의 뿌리 및 지상부 생육에 미치는 근부온도의 영향)

  • The experiment was investigated the effects of lower root zone temperature on shoot and root growth of 'Akihime' strawberries in aeroponics in cold season. Root growth was decreased with decrease of root zone temperature, especially in lateral roots. Elongation of main roots was highest in $18^{\circ}C$ of root zone temperature. Number of lateral roots and length of lateral roots were lowest in $8^{\circ}C$ of root zone temperature. Lower root zone temperature resulted significantly in decrease of fresh weight of root and shoot and leaf area of strawberry. But there were no significant statistical differences in shoot fresh weight and leaf area in $8^{\circ}C$ and $13^{\circ}C$ of root zone temperature. Leaf length, leaf width and number of leaves of strawberry were decreased in lower root zone temperature. The results of this experiment will be utilized in the winter season cultivation for strawberry in hydroponics.

Case Study of N Deficiency Symptom of Strawberry in the Soil Applied with Sea Deposit Compost (바다부유물질로 만든 퇴비를 시용한 토양에서 발생한 딸기의 질소결핍증 해결 사례연구)

  • Nitrogen content in soil is a major factor for the crop growth. Ammonium nitrogen in soils is volatilized when soil pH is high. The growth and development problem of strawberry such as color of leaves turning into purple was found when soils were treated with the compost prepared from suspended solids of high pH from the sea. From in-situ analysis it was found that soil pH was 8.33 and nitrate, phosphorous, and potassium contents were relatively low. Nitric acid was added to adjust pH of irrigating water as 1.7, 1.9, and 2.3, then $KNO_3$ $0.25g\;L^{-1}$ and $KH_2PO_4$ $0.25g\;L^{-1}$ were added. It was resulted that soils with pH 1.7 produced the most developed strawberries. Strawberry was recovered by irrigation containing the same solution. From the results, the growth and development problem of the strawberry resulted from low nitrate absorption rate. It was concluded that the growth and development of strawberries were recovered by the reduced soil pH using nitric acid.
