Effect of Culture Broth of Cordyceps militaris on Recovery of Mice Hepatic Damage Caused by Benzo($\alpha$)pyrene-Treatment (벤조피렌으로 유발된 흰쥐 간독성에 대한 번데기동충하초 배양액의 회복효과)

  • The hepatoprotective effect of Cordyceps militaris culture broth was determined using HaM/ICR strain mice. Compared to control, the intra-peritoneal injection of benzo($\alpha$)pyrene (B($\alpha$)P) remarkably increased the activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in the serum and the level of lipid peroxide (LPO) in liver tissue, which mean the liver was damaged by B($\alpha$)P. However, compared to B($\alpha$)P, oral administration of C. militaris culture broth showed decrement of AST, ALT, and LPO activities and increment GST activity and GSH level in liver tissue. These suggest that C. militaris culture broth recovered hepatic damage induced by B($\alpha$)P.

Effects of Fly Maggot Extracts on the Liver and Plasma Lipid in Rat Fed High-Fat Diets (고지방식이 유도된 흰쥐의 혈액지질 및 간에 관한 파리유충 추출물의 효과)

  • The bioactive effects of ethanol extracts from fly maggot (ME) on reduction of plasma lipids levels in rats fed high-fat diets (Expt. Ⅰ), and on liver function recovery of hepatotoxicity rats by intraperitoneal injection of carbon tetrachloride ($CCl_4$) or by orally administration of alcohol (Expt. II) were investigated. In expt. I, twenty seven, male rat SDS(sprague dawley strain) were randomly assigned to three treated groups, including normal control group, HF (group with high fat diets which have no extracts) and HFE (HF plus orally administered doses of ME extract at 5.0 mg/100g of body weight). In expt. II, forty five, male rats (SDS) were randomly assigned to each of the five groups: T1 (control), T2 (intraperitoneal injection of $CCl_4$), T3 (intraperitoneal injection of $CCl_4$ after orally administered with ME), T4 (orally administered with combination of ME and alcohol), T5 (orally administration of ME after orally administered with alcohol). There were significant decreases in plasma (TAG), (TC), (LDL-C) in the HFE group with orally administered doses of ME at 5.0 mg/100g of body weight, respectively, however, the (HDL-C) were significantly increased in HFE group as compared to HF group with high fat diets which have no extracts (p<0.05). The levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), ${\gamma}$-glutamyl transferse(${\gamma}$-GTP) and bilirubin were highest in T2 or T3, and high in order T4 or T5, and lowest in T1 except for bilirubin which has same with T4, T5 (p<0.05). The high recovery of liver damage by $CCl_4$ from the light microscopic appearance was observed in rats (T3) with extracts, and also high in T4 than T5 by orally administrated with alcohol. In conclusion, the ethanol extracts from fly maggot may have a bioactive effects to prevent for human lipids disorder and alcoholic disease.

  • Postoperative imaging of the rotator cuff may be performed routinely, even if pain or disability develops after surgery or if there are no symptoms. Postoperative images are obtained through MRI or US, and the purpose is to confirm the integrity of the restored tendon in general. Postoperative MRI has a relatively poor diagnostic accuracy compared to that of preoperative images because various materials used in surgeries deteriorate the image quality. US can dynamically check the condition of the restored tendon and avoid artifacts from the surgical instruments used for recovery. Although imaging findings are not always consistent with the clinical symptoms or prognosis, sub-deltoid fluid retention is more important for pain and functional recovery than the thickness of the reconstructed tendon. Strain elastography can also be a useful method for predicting the prognosis.

Production Conditions and Properties of Glucose Isomerase from Streptomyces griseolus (Streptomyces griseolus기원의 포도당 이성화효소의 생성 조건과 성질)

  • Cultural characteristics of Strptomyces griseolus isolated from the soil were investigated. This strain was disclosed to utilize D-xylose, and D-glactose in preference order as a carbon source with the formation of glucose isomerase. The addition of sweet potato starch also proved effective promoting the total enzyme activity measured at 29% higher than the control. Corn cob, one of waste agricultural resources, was hydrolyzed in 2~3% $H_2SO_4$ solution at $100^{\circ}C$, 3~5 hours to produce a xylose syrup which gave rise to the recovery of 19.9% in a batch system and 28.2% in a repeated system. By the addition of both 2% of xylose syrup(Be'28) prepared by and us 65% of corn steep liquor (total nitrogen 1.2%), enzyme induction was maximized. The enzyme activity was stimulated by the xylose and the cell growth by the C.S.L. Also, remarkable increase of enzyme activity was noticed by the addition of protein acid hydrolysate 86.2% higher than the control. $QO_2$ of the biomass cultured in 30L capacity jarfermentor recorded low oxygen requirement of 251.2 1/hr. Maximum activity of glucose isomerase was observed noted at the 9th hour after inoculation which is 2 hours faster than the stationery was observed noted at the 9th hour after inoculation which is 2 hours faster than the stationery phase of the biomass growth. Glucose isomerase from the strain was activated by adding the $Co^{++}\;and\;Mg^{++}$ with optimum temperature of $73^{\circ}C$ and pH of 7.2. Conversion ratio of 60% glucose to frutose was 42.5% after 70 hours reaction.

Seismic behavior of properly designed CBFs equipped with NiTi SMA braces

  • Shape memory alloys (SMA) exhibit superelasticity which refers to the capability of entirely recovering large deformation upon removal of applied forces and dissipating input energy during the cyclic loading reversals when the environment is above the austenite finish temperature. This property is increasingly favored by the earthquake engineering community, which is currently developing resilient structures with prompt recovery and affordable repair cost after earthquakes. Compared with the other SMAs, NiTi SMAs are widely deemed as the most promising candidate in earthquake engineering. This paper contributes to evaluate the seismic performance of properly designed concentrically braced frames (CBFs) equipped with NiTi SMA braces under earthquake ground motions corresponding to frequently-occurred, design-basis and maximum-considered earthquakes. An ad hoc seismic design approach that was previously developed for structures with idealized SMAs was introduced to size the building members, by explicitly considering the strain hardening characteristics of NiTi SMA particularly. The design procedure was conducted to compliant with a suite of ground motions associated with the hazard level of design-basis earthquake. A total of four six-story CBFs were designed by setting different ductility demands for SMA braces while designating with a same interstory drift target for the structural systems. The analytical results show that all the designed frames successfully met the prescribed seismic performance objectives, including targeted maximum interstory drift, uniform deformation demand over building height, eliminated residual deformation, controlled floor acceleration, and slight damage in the main frame. In addition, this study indicates that the strain hardening behavior does not necessarily impose undesirable impact on the global seismic performance of CBFs with SMA braces.

A Study on High Temperature Deformation Behavior of Spray-Formed High Speed Steels (분무주조 고속도공구강의 고온변형 거동에 관한 연구)

  • In the present study, the mechanical behavior of the spray-formed high speed steel was investigated employing the internal variable theory of inelastic deformation. Special attention was focused on the effect of the microstructure evolution during the hot working process, such as the distribution of carbides to provide a basic database for the production condition of high speed steels with excellent properties. The billets of high speed steel ASP30TM were fabricated by a spray forming, and the subsequently hot-rolled and heat-treated process to obtain uniformly distributed carbide structure. As noted the spray-formed high speed steel showed relatively coarser carbides than hot-rolled and heat-treated one with fine and uniformly distributed carbide structure. The step strain rate tests and high temperature tensile tests were carried out on both the spray-formed and the hot-rolled specimens, to elucidate their high temperature deformation behavior. The spray-formed high speed steel showed much higher flow stress and lower elongation than the hot-rolled and heat-treated steel. During the tensile test at $900^{\circ}C$, the interruption of the deformation for 100 seconds was conducted to reveal that the recovery was a main dynamic deformation mechanism of spray formed high speed steel. The internal variable theory of the inelastic deformation was used to analyze data from the step strain rate tests, revealing that the activation energies for hot deformation of as-spray-formed and hot-worked steels, which were 157.1 and 278.9 kJ/mol, and which were corresponding to the dislocation core and lattice diffusions of ${\gamma}-Fe$, respectively.

Chloroplast-type Ferredoxin Involved in Reactivation of Catechol 2,3-Dioxygenase from Pseudomonas sp.S-47

  • Pseudomonas sp. S-47 is capable of degrading catechol and 4-chlorocatechol via the meta-cleavage pathway. XyITE products catalyze the dioxygenation of the aromatics. The sylT of the strain S-47 is located just upstream of the xylE gene. XylT of the strain S-47 is located just upstream of the xylE gene. XyIT is typical chloroplast-type ferredoxin, which is characterized by 4 cystein residues that are located at positions 41, 46, 49, and 81. The chloroplast-type ferredoxin of Pseudomonas sp. S-47 exhibited a 98% identity with that of P. putida mt-2(TOL plasmid) in the amino acid sequence, but only about a 40 to 60% identity with the corresponding enzymes from other organisms. We constructed two recombinant plasmids (pRES1 containing xylTE and pRES101 containing xylE without xylT) in order to examine the function of XyIT for the reactivation of the catechol 2,3-dioxygenase (XyIE) that is oxidized with hydrogen peroxide was recovered in the catechol 2,3-dioxygenase (C23O) activity about 4 mimutes after incubation, but the pRES101 showed no recovery. That means that the typical chloroplast-type ferredoxin (XyIT) of Pseudomonas sp. S-47 is involved in the reactivation of the oxidized C23O in the dioxygenolytic cleavage of aromatic compounds.

Study on the pathogenesis of the piglets experimentally infected with Korean isolate of Aujeszky′s disease virus I. Histopathologic and electron microscopic observation (Aujeszky's disease virus 국내분리주 접종자돈의 병리발생에 관한 연구 I. 병리학적 및 전자현미경적 관찰)

  • This study was conducted to elucidate the pathogenesis of Aujeszky's disease virus(ADV) by histopathologic examination. The first Korean ADV Isolate, which was isolated from piglets with clinical signs of Aujeszky's disease in Yangsan(YS) county, Kyungnam province, was inoculated into 32 days old piglets with a dose of $10^{5.9}$$TCID_{50}/ml$ through intranasal or intramuscular route. These piglets were sacrificed at intervals of every 24hrs for 8 days. The virulence of YS strain was determined by the observation of clinical signs, gross findings, and histopathologic changes in tissues. The virus recovery test was performed from brain, spleen, lung and tonsil in cell culture. The pathogenesis of YS strain was determined by the observation of histopathologlc lesions in CNS and neuronal tracts. The major clinical signs were fever, anorexia, dyspnea, constipation, tremor, ataxia, circling movement, hindleg paralysis and salivation. The clinical signs were more severe in piglets of the group inoculated intranasally than those of the intramuscularly inoculated gorup. Lymphocytopenia was detected on day 5 to day 6 postinoculation (PI). The ADV was recovered from the tissue homogenates of tonsil, lung, spleen and cerebrum in cell culture. The highest virus titer was detected from tonsil between day 6 and day 7 PI. Reddish sublobar consolidation foci were scattered in the apical and cardiac lobes of lung. Although yellowish necrotic foci were detected in tonsil and liver, hemorrhagic lesions were mainly observed in heart, kidney and lymph nodes. Histopathologically, degeneration and necrosis of nerve cells, nonsuppurative meningoe-ncephalitis, nodular gliosis and perivascular cuffings were observed in CNS. Multifocal fibronecrotic foci were observed in lung, liver, lymph nodes and spleen. The major pathologic changes were detected in the midbrain, pons and medulla oblongata. Eosinophilic intranuclear inclusion bodies were mainly observed in epithelia and /or macrophages of tonsil, liver, lung, spleen and submandibular lymph nodes, and neurons of brain, respectively. Observation of viral particles at various stages of replication were possible from the endothelial cells of the alveolar capillaries and tonsillar crypt epithelia by transmission electron microscope.

Three-Point Bending Fatigue Properties of Austenitic 304 Stainless Steel Sheets for Membrane (Membrane용 오스테나이트계 304 스테인리스강 판재의 3점 굽힘피로 특성)

  • Three-point bending fatigue properties of austenitic 304 stainless steel sheets were investigated at room temperature and LNG temperature($-162^{\circ}C$) in the strain range from 0.43 to $1.7\%$. The fatigue properties at $-162^{\circ}C$ were superior to those at room temperature due to the higher volume fractions of deformation-induced martensite. The cyclic hardening behavior owing to the deformation- induced martensite transformation was detected in both specimens. In room temperature testing, the mean load amplitude increased steadily with cycles, meaning that cumulative plastic incubation strain was required for martensite transformation. On the contrary, in $-162^{\circ}C$ tested specimen, the mean load amplitude increased rapidly within a few cycles due to the rapid transformation of martensite, and slightly decreased after the maximum is reached probably due to dynamic recovery.

Characterization of Alpha Amylase Producing Thielaviopsis ethacetica and Its Raw Starch Hydrolyzing Ability on Different Agricultural Substrates

  • The present study reports the morphological and molecular characterization of the fungal strain, CMSS06 and evaluates its raw starch hydrolyzing ability in four different agricultural substrates (rice bran, banana peel, cassava tubers, and coconut water). The potential use of each agricultural substrate to replace the expensive fermentation media was evaluated with six different fermentation media: rice bran (RB), banana peel (BP), cassava starch (CS), cassava in coconut water (CSCW), cassava in modified coconut water (CMCW), and pure Coconut water (CW). The fungal strain CMSS06 was identified as Thielaviopsis ethacetica by the analysis of the ITS sequences. The T. ethacetica alpha amylase enzyme exhibited maximum alpha amylase activity at 72 h, pH 7.0, and $40^{\circ}C$ on soluble starch. This species resulted in the highest enzyme activity (mU/ml) of 26.06, 10.89, 58.82, 14.2, and 54.67 with the RB, BP, CS, CSCW, and CMCW fermentation media, respectively. The results indicate that CS can be used as a carbon substrate and CMCW can be used to accelerate the fermentation by T. ethacetica. The enzyme was partially purified by 40-60% ammonium sulphate fraction, and it showed total enzyme activity, total protein content, specific activity, purification fold, and a recovery of 2400 mU, 30 mg, 80 mU/mg, 2.7, and 71.1%, respectively. The molecular mass of the T. ethacetica alpha amylase was estimated on SDS-PAGE, and two bands around 50 kDa and 70 kDa were identified. The present study implies that T. ethacetica can produce alpha amylase, and it can be used to hydrolyze raw starch during the fermentation processes.
