Escherichia Coli ornithine transcarbamylase is the enzyme which catalyzes the L-citrulline biosynthesis from L-ornithine and carbamyl phosphate. To facilitate the purification of enzyme which will be used for many biochemical studies such as structure and function relationships and catalytic mechanisms, the cloning and expression of E. coli argI gene for ornithine transcarbamylase was conducted. argI was amplified from genomic DNA of E. coli strain of $DH5{\alpha}$, by polymerization chain reaction (PCR) method. The amplified argI gene was ligated to the prokaryotic expression vector pKK223-3 and used for transformation of E. coli TB2 which was deficient of ornithine transcarbamylase. The over-produced enzyme by the tnansformant was purified by ammonium sulfate fractionation, heat denaturation and affinity chromatography. The result of SDS denaturation gel electrophoresis for the purified enzyme showed a single band of about 38 kDa of ornithine transcarbamylase. Kinetic data for the expressed enzyme gave almost the s?????? values as those of the wild type enzyme. The $k_{cat}$, of the enzyme was $1.0{\times}10^5min^{-1}$, and $K_ms$ for ornithine and carbamyl phosphate were 0.35 mM and 0.06 mM, respectively.

Fiber Reinforced Plastic (FRP) sheets have been widely used to retrofit and rehabilitate RC structures, while in case of retrofitting steel structures, there are no codes and researches. It stems from configuration of member and characteristics of bonding behavior. This study focused on the static behavior of steel beams reinforcement by AFRP sheets. The main objective of the experimental programme was the evaluation of the force transfer mechanism, the increment of the beam load carrying capacity and the bending stiffness. A bending test was conducted on a H-shaped steel beam, with aramid FRP sheets bonded to its flanges. The mid-span deflection and the strain from three points along AFRP sheets were recorded Test results exhibit that the increment of the load-carrying capacity with reference to a mid-span deflection level of 15 mm(1/125mm of the clear span) was equal to 9.4% and for the two layers case, an elastic stiffness increment is slightly higher than one layer case.
A bacterial strain antagonistic to some fungal phytopathogens was isolated from the stem of a Persimmon tree in Yeongam, Korea. This bacterium was identified as Bacillus subtilis by 16S rRNA gene sequencing and designated as B. subtilis GDYA-1. In in vivo experiment, the fermentation broth exhibited antifungal activities against Magnaporthe oryzae on rice plants, Phytophthora infestans on tomato plants, and Puccinia recondita on wheat plants. We isolated one antifungal compound and its chemical structure was determined by mass and $^1H$-NMR spectral data. The antifungal substance was identified as benzoic acid. It inhibited mycelial growth of M. oryzae, Rhizoctonia solani, Sclerotinia sclerotiorum, and P. capsici with minimum inhibition concentration (MIC) values, ranging from 62.5 to 125 ${\mu}g/ml$. Moreover, the substance effectively suppressed Phytophthora blight of red pepper caused by P. capsici in a pot experiment. To the author's knowledge, this is the first report on the antifungal activity of benzoic acid against phytopathogenic fungi. Benzoic acid and B. subtilis GDYA-1 may contribute to environmental-friendly protect crops from phytopathogenic fungi.

In this study, a U-shape composite beam was developed to be effectively used for a steel parking lot which is 8m or lower in height. When the U-shape composite beam was applied to a steel parking lot, essential considerations were story-height and long-span. In addition, due to the mixed structural system with reinforced concrete and steel material, the U-shape composite beam needed to have a structural integrity and reliable performance over demand capacity. The main objective of this study was to investigate the performance of the structure consisting of the reinforced concrete (RC) slab and U-shape beam. A U-shape composite beam generally used at a parking lot served as a control specimen. Four specimens were tested under four-point bending. To calculate theoretical values, strain gauges were attached to rebar, steel plate, and concrete surface in the middle of the specimens. As the results, initial yielding strength of the control specimen occurred at the bottom of the U-shaped steel. After yielding, the specimen reached the maximum strength and the RC slab concrete was finally failed by concrete crush due to compressive stress. The structural performance such as flexural strength and ductility of the specimen with the increased beam depth was significantly improved in comparison with the control specimen. Furthermore, the design of the U-shape composite beam with the consideration of flexural strength and ductility was effective since the structural performance by a negative loading was relatively decreased but the ductile behavior was evidently improved.
An ester-carbonate copolymer was synthesized, in which carbonate was inserted into a biodegradable aliphatic polyester, poly(butylene succinate) (PBS), to modify its mechanical properties. The synthesis was carried out by condensation reactions in two steps. In the first step, oligo(butylene succinate) was prepared by the reaction of succinic acid with 1,4-butanediol (BD). In the second step, it was reacted with oligohexamethylenecarbonate diol (OHMCG) to prepare the ester-carbonate copolymer. Titanium(IV) isopropoxide (TIP) was used as a catalyst for the reaction. The structure of the copolymer was confirmed by FT-IR and $^1H$-NMR and the thermal behavior and mechanical properties were investigated by differential scanning calorimetry (DSC) and universal testing machine (UTM), respectively. It was found that optimum amount of the catalyst for the formation of high molecular weight copolymer was 1wt% for succinic acid. When the BD:OHMCG is in the range 149:1~249:1, the copolymer with high viscosity was obtained. As the OHMCG content was increased, melting temperature ($T_m$) of the copolymer was decreased. When BD:OHMCG is 149:1, the copolymer showed a increase in ultimate strain by two times and the slight decrease in modulus compared to those of PBS.

Recently, there has been substantial interest in flexible and wearable devices whose properties and performances are close to conventional devices on hard substrates. Despite the advancement on flexible devices with organic semiconductors or carbon nanotube films, their performances are limited by the carrier scattering at the molecular to molecular or nanotube-to-nanotube junctions. Here in this study, we demonstrate on the vertical semiconductor crystal array embedded in flexible polymer matrix. Such structures can relieve the strain effectively, thereby accommodating large flexural deformation. To achieve such structure, we first established a low-temperature solution-phase synthesis of single crystalline 3D architectures consisting of epitaxially grown ZnO constituent crystals by position and growth direction controlled growth strategy. The ZnO vertical crystal array was integrated into a piece of polydimethylsiloxane (PDMS) substrate, which was then mechanically detached from the hard substrate to achieve the freestanding ZnO-polymer composite. In addition, the characteristics of transferred ZnO were confirmed by additional structural and photoluminescent measurements. The ZnO vertical crystal array embedded in PDMS was further employed as pressure sensor that exhibited an active response to the external pressure, by piezoelectric effect of ZnO crystal.
Identification was performed in March 2008 for the 76 Enterococcus strains isolated from the Han-river, which is used as water supply for Seoul citizens. The antibiotic susceptibility, antibiotic resistant structural analysis, trans-conjugation, pulsed-field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST) were also carried out for the isolated strains. Among the isolated strains, 25 strains were E. casseliflavus, 4 strains were E. faecalis and 1 strain was E. hirae. Investigation of antibiotic susceptibility indicated that 15 strains demonstrated tolerance against vancomycin, and that 11 strains of E. faecium and 4 strains of E. casseliflavus were VRE. The vanA gene detection of the VRE strains revealed that 6 E. faecium strains were vancomycin-resistant Enterococcus faecium (VREF) possessing vanA. Analyses of transposon Tn1546 structure containing vanA demonstrated that Km36 and Km37 belonged to Tn1 type, Km20 and Km38 was Tn2 type, and Km39 and Km40 was Tn3 type. PFGE disclosed that among the 6 VREF strains, Km36 and Km37 exhibited equivalent subtype, while the rest 4 strains showed subtypes different to each other. MLST for the 6 VREF strains disclosed that 3 strains were ST78, while the rest 3 strains were ST18, ST192 and ST230, respectively. All these clonal complexes were derived from CC17 which has been isolated from clinical sources. 4 strains belonged to CC78, while the rest 2 strains were CC18 and CC192, respectively.
The effects of thermal cycling on residual stresses in both inorganic passivation/insulating layer that is deposited by plasma enhanced chemical vapor deposition (PECVD) and organic thin film that is used as a bonding adhesive are evaluated by 4 point bending method and wafer curvature method. $SiO_2/SiN_x$ and BCB (Benzocyclobutene) are used as inorganic and organic layers, respectively. A model about the effect of thermal cycling on residual stress and bond strength (Strain energy release rate), $G_c$, at the interface between inorganic thin film and organic adhesive is developed. In thermal cycling experiments conducted between $25^{\circ}C$ and either $350^{\circ}C$ or $400^{\circ}C$, $G_c$ at the interface between BCB and PECVD $ SiN_x $ decreases after the first cycle. This trend in $G_c$ agreed well with the prediction based on our model that the increase in residual tensile stress within the $SiN_x$ layer after thermal cycling leads to the decrease in $G_c$. This result is compared with that obtained for the interface between BCB and PECVD $SiO_2$, where the relaxation in residual compressive stress within the $SiO_2$ induces an increase in $G_c$. These opposite trends in $G_cs$ of the structures including either PECVD $ SiN_x $ or PECVD $SiO_2$ are caused by reactions in the hydrogen-bonded chemical structure of the PECVD layers, followed by desorption of water.
In this study, we investigated the necessary mechanical properties of conductive multifilament yarns for fabricating the electrodes of biosignal measurement pressure and stretch textile sensors using embroidery. When electrodes and circuits for smart wearable products are produced through the embroidery process using conductive multifilament yarns, unnecessary material loss is minimized, and complex electrode shapes or circuit designs can be produced without additional processes using a computer embroidering machine. However, because ordinary missionary threads cannot overcome the stress in the embroidery process and yarn cutting occurs, herein, we analyzed the S-S curve, thickness, and twist structure, which are three types of silver-coated multifilament yarns, and measured the stress in the thread of the embroidery simultaneously. Thus, the required mechanical properties of the yarns in the embroidery process were analyzed. In the actual sample production, cutting occurred in silver-coated multifilament rather than silver-coated polyamide/polyester, which showed the lowest S-S curve. In the embroidery process, the twist was unwound through repetitive vertical movement. Further, we fabricated a piezoresistive pressure/tension sensor to measure gauge factor, which is an index for measuring biological signals. We confirmed that the sensor can be applied to the fabrication of embroidery electrodes, which is an important process in the mass production of smart wearable products.

Nonlinear analysis of RC containment structure under thermal load and pressure is presented to trace the behaviour after an assumed LOCA. The temperature distribution varying with time through the wall thickness is determined by transient finite element analysis with the two time level scheme in time domain. The layered shell finite elements are used to represent the containment structures in nuclear power plants. Both geometric and material nonlinearities are taken into account in the finite element formulation. The constitutive relation of concrete is modeled according to Drucker-Prager yield criteria in compression. Tension stiffening model is used to represent the tensile behaviour of concrete including bond effect. The reinforcing bars are modeled by smeared layer at the location of reinforcements accounting elasto-plastic axial behaviors. The steel liner model under Von Mises yield criteria is adopted to represent elastic-perfect plastic behaviour. Geometric nonlinearity is formulated to consider the large displacement effect. Thermal stress components are determined by the initial strain concept during each time step. The temperature differential between any two consecutive time steps is considered as a load incremental. The numerical results from this study reveal that nonlinear temperature gradient based on transient thermal analysis will produces excessive large displacement. Nonlinear behavior of containment structures up to ultimate stage can be traced reallistically. The present study allows more realistic analysis of concrete containment structures in nuclear power plants.
