Isolation of Acylated Sterylglycosides from the Legumes of Albizzia julibrissin (자귀나무 꼬투리로부터 Acylated Sterylglycoside의 분리)

  • From the legumes of Albizzia julibrissin Durazzini (Leguminosae), a mixture of long-chain alcohols, ${\alpha}-spinasterol$, a mixture of acylated sterol glycosides and ${\alpha}-spinasteryl$ $3-O-{\beta}-D-glucoside$ were isolated. Two mixtures of long-chain alcohols and acylated sterol glycosides were characterized as 1-hexacosanol (major) and 1-tetracosanol, and $3-O-[6'-O-palmitoyl-{\beta}-D-glucosyl]-{\alpha}-spinasterol$ (major) and its 6'-O-stearoyl ester. All compounds were identified on the basis of spectral data and chemical reactions.

  • To elucidate chemical structure of the wood extracitives, softwood Larix leplolepis Gorden) metal was extracted with 95% ethanol at room temperature for 72 hours. The extract was fractionated with organic solvents such as n-hexane, chloroform. ether, and ethylacetate. From the n-hexane soluble fraction of the extratives, flavonoid and sterol compounds were isolated and identified as taxifolin(5,7,3',4'-tetrahydroxyflavanonol)(I) and ${\beta}$-sitosterol(II) by UV, IR, $^1$H-NMR spectroscopy and Mass spectrometry.

  • Effects of the extraction-methods and solvents on some minor components in crude rapeseed-oil and the autooxidation of the oils were examined in this study. The contents of unsaponifiable matters and sterols were not significantly variied depending on the extraction-methods and solvents. Chlorophyll-contents were elevated with the increasing molecular weights of extractionsolvents. Any significant effect of the extraction-temperature on the yield of chlorophyll was not observed. Autoxidation-tendency of the oils was found to be dependent on the boiling points of extraction-solvents.

  • "Ha-Neul-Ta-Ri" (Trichosanthes kirilowii Max., Cucurbitaceae) cultivated in Korea and used in popular medicine, was investigated for sterol composition of the seed oil. This seed oil contains unusual sterols having ${\delta}^{7, 22}_-$, ${\delta}^{7, 24}_-$ and ${\delta}^{7, 22, 25}_-$ di and trienols. ${\delta}^{7, 22}_-$ Stigmastadienol and ${\delta}^{7, 22, 25}_-$ Stigmastatrienol were identified as the major sterols in the seed oil of Trichosanthes kirilowii by gas loiquid chromatographic analysis. The results showed that contents of the seed sterols were campesterol 1.0-1.2%, stigmasterol 1.5-1.7%, ${\delta}^{7}_-$ campstenol 0.6-0.8%, sitosterol 10.6-11.4%, ${\delta}^{7, 22}_-$ stigmastadienol 39.1-41.8%, ${\delta}^{7, 22, 25}_-$ stigmastatrienol 28.9-29.3%, ${\delta}^{7, 25}_-$ stigmastadienol 14.9-15.5% and ${\delta}^{7, 24}_-$ stigmastadienol 0.7-1.0%.

  • From the carpophores of Coriolus sanguineus Fr., a bright-red wood-rotting fungus, which were collected at Suwon, an antibiotic component was isolated as red crystal and identified as cinnabarin by UV and IR spectra. The antibiotic activities against five bacteria and three fungi were determined. It showed considerably high activity against Bacillus subtilis ATCC 6633 and Bacillus maceraus and also against all the three fungi tested. And from this fungus a sterol was isolated and identified as erogosterol by cochromatographing with four authentic sterols.

Variation in Lipid Composition during the Growing Period of the Prawn I. Comparative Studies on the Flesh Lipid Composition of the Wild and Cultured Prawn (보리새우 성장중의 지질성분 변화에 관한 연구 1. 천연 및 양식산 보리새우의 근육 지질성분의 비교)

  • Differences in lipid composition including fatty acid, lipid class, sterol and especially carotenoid between fleshes of wild and cultured prawn, Penaeus japonicus, were studied. Total lipids were extracted from the flesh during the spawning period and fractionated into two lipid classes of polar and nonpolar lipids by silicic acid column chromatography. The fatty acid composition of each lipid classes, total lipid (TL), nonpolar lipid (NL) and polar lipid (PL) were analyzed by gas liquid chromatography. The sterol and carotenoid composition of total lipids were determined by using thin layer chromatography, gas liquid chromatography and column chromatography using MgO-celite 545 and silicic acid-celite 545 as an absorbent, and by UV spectrophotometry. Total lipid contents of both fleshes from the wild and cultured prawn were about $2.0\%$ on average, but the content of the unsaponifiable matters in the cultured prawn (about $16.2\%$ in total lipid) showed a little higher than that of the wild prawn (about $13.9\%$ in total lipid) and the ratio of NL to PL in total lipid was 1:1.7. In the fatty acid composition of TL, the contents of $Cl_{16:0}\;and\;C_{20:3}$ fatty acids were higher in wild prawn than in cultured prawn, while the contents of $Cl_{18:1}\;and\;C_{20:5}$ fatty acids in cultured prawn were higher than those in wild prawn. The cultured prawn contained higher amounts of monoenoic acids and lower amounts of polyenoic acids than the wild prawn. In the fatty acid composition of NL, the wild prawn showed higher levels in $Cl_{18:0}\;and\;C_{20:1}$ fatty acid contents than the cultured prawn, while the cultured prawn contained much amout of $Cl_{16:0}\;and\;C_{18:1}$ fatty acids. On the other hand, the fatty acid composition of PL showed that $Cl_{16:1}\;and\;C_{17:1}$ fatty acid were higher in the wild prawn than in the cultured prawn, but in $Cl_{16:0}\;and\;C_{18:1}$ fatty acids, the levels were reversed. Consequently, the cultured prawn contained higher amount of monoenoic acids, and similar amounts of saturated acids and polyenoic acids to the wild prawn in NL. And the cultured prawn contained lower amount of monoenoic acids, and similar amounts of saturated acids and polyenoic acids to the wild prawn in PL. In sterol composition of both the wild and cultured prawn, the predominant sterol was cholesterol with the proportion of $78.7{\sim}88.9\%$ to the total sterol. In addition to the cholesterol, the other minor sterols such as 24-methylene cholesterol and sitosterol were detected. Total carotenoid content in flesh of the wild prawn was relatively higher than that of the cultured prawn marking 70 mg/100g of lipid in wild prawn and 40 mg/100 g of lipid in cultured prawn, respectively. The main carotenoids of the both prawns were astaxanthin($54.1{\sim}60.8%$), phoenicoxanthin ($16.5{sim}22.9%$),${\bata}-carotene\;(20.0{\sim}22.0%)$.

Genome-Wide Analysis of Hypoxia-Responsive Genes in the Rice Blast Fungus

  • Rice blast fungus, Magnaporthe oryzae, is the most destructive pathogen of rice in the world. This fungus has a biotrophic phase early in infection and switches to a necrotrophic lifestyle after host cell death. During the biotrophic phase, the fungus competes with host for nutrients and oxygen. Continuous uptake of oxygen is essential for successful establishment of blast disease of this pathogen. Here, we report transcriptional responses of the fungus to oxygen limitation. Transcriptome analysis using RNA-Seq identified 1,047 up-regulated genes in response to hypoxia. Those genes were involved in mycelial development, sterol biosynthesis, and metal ion transport based on hierarchical GO terms and well-conserved among three different fungal species. In addition, null mutants of three hypoxia-responsive genes were generated and tested for their roles on fungal development and pathogenicity. The mutants for a sterol regulatory element-binding protein gene, MoSRE1, and C4 methyl sterol oxidase gene, ERG25, exhibited increased sensitivity to hypoxia-mimetic agent, increased conidiation, and delayed invasive growth within host cells, suggesting important roles in fungal development. However, such defects did not cause any significant decrease in disease severity. The other null mutant for alcohol dehydrogenase gene, MoADH1, showed no defect in the hypoxia-mimic condition and fungal development. Taken together, this comprehensive transcriptional profiling in response to a hypoxia condition with experimental validations would provide new insights on fungal development and pathogenicity in plant pathogenic fungi.

Lipid Metabolism of Korean Breast-Fed Formula-Fed Infants (모유영양아와 인공영양아의 지질 대사)

  • The difference in lipid metabolism between breast-fed(BF) and formula-fed(FF) infants were studied in 31 Korean infants at 1, 2 and 3 months postpartum. The formulas had more total lipids(TL), triglycerides(TG) and phospholipids and less cholesterol(CHOL) and free fatty acids(FFA) than the breast milk. The milk consumption of the FF infants was significantly greater than that of the BF infants with a wide individual variation. As a result, the FF infants appeared to consume more TL, TG and PL and less CHOL and FFA than the BF infants during 3 months. The lipid contents of the breast milk tended to decrease in due course of lactation, therefore the intake of lipids of BF infants was reduced during the first three month of lactation. The plasma TG and CHOL levels of infants at 3 month were not significantly different between the BF and FF infants, but the PL level of the BF infants was higher then that of the FF infants. The plasma levels of TG and CHOL were not significantly correlated with the intakes of TL, TG, CHOL, PL and FFA, respectively. The PL level, however, was positively correlated with the intakes of CHOL and FFA, respectively and negatively correlated with PL intake as well as 18 : 2w6 fatty acid content of the breast milk or the formulas. The fecal weight and fecal loss of TL of the FF infants were greater than those of the BF infants during 3 months, however, apparent lipids digestibility was not significantly different between the BF and FF infants. The fecal excretions of CHOL and bile acids of FF infants were substantially higher than those of BF infants during 3 months. New sterol balance showed a particularly large difference between the BF and FF infants. The value of the net sterol balance of the BF infants was negative, but that of the FF infants was positive. This study shows that the consumption pattern of various lipid components of BF and FF infants were different as a result of different lipid compositions between breast milk and formula. The significantly lower sterol balance of the BF infants than the FF infants may have been derived from the unique dietary characteristic of breast milk.

  • Cymbidium goeringii REICHB. fil was extracted with 80% MeOH and solvent-fractionated with EtOAc, n-BuOH and $H_2O$, successively. From EtOAc fraction, three sterols were isolated on repeated silica gel and ODS column chromatographies. The chemical structures were determined as ${\beta}-sitosterol$ (1), daucosterol (2) and ergosterol peroxide (3) by NMR, MS and IR, which is the first to be isolated from Cymbidium goeringii REICHB. fil.
