To understand the effect of water temperature on growth pattern of Stephanodiscus sp., we weekly or biweekly investigated in the lower part of the Nakdong River from 1994 to 2006 and performed a laboratory experiment. Stephanodiscus was the most dominant species among phytoplankton in winter when low flow persisted and the high abundances of the species were maintained from December to February. Three strains of Stephanodiscus sp. were isolated for the in vitro experiment from the Nakdong River in January 2005. Over the water temperature range of $4^{\circ}C$ to $20^{\circ}C$, the growth patterns of Stephanodiscus sp. were different in the short-term batch culture. The maximum cell density of Stephanodiscus sp. was observed at approximately $5^{\circ}C$ in the river systems, but the optimum water temperature of Stephanodiscus sp. was $10^{\circ}C$ for the growth in the laboratory experiment. However, the proliferation of Stephanodiscus sp. was related to low water temperature in the Nakdong River.
DNA-based discrimination of species is a powerful way for morphologically otherwise similar species, like centric diatoms. Here, the author sequenced long-range nuclear ribosomal DNAs, spanning from the 18S to the D5 region of the 28S rDNA, of Stephanodiscus, particularly including a Korean isolate. By comparisons, high DNA similarities were detected from the rDNAs of nine Stephanodiscus (>99.4% in 18S rDNA, >98.0% in 28S rDNA). Their genetic distances, however, were significantly different (Kruskal-Wallis test, p < 0.01) compared to two related genera, namely Cyclotella and Discostella. In addition, genetic distances of 18S rDNAs were significantly different (Student’s t-test, p = 0.000) against those of the 28S rDNAs according to individual genera (Cyclotella, Discostella, and Stephanodiscus). Phylogenetic analyses showed that Stephanodiscus and Discostella showed a sister taxon relationship, and their clade was separated from a cluster of Cyclotella (1.00 PP, 100% BP). This suggests that Stephanodiscus has highly conserved sequences of both 18S and 28S rDNA; however, Stephanodiscus is well-separated from other freshwater centric diatoms, such as Cyclotella and Discostella, at the generic level.
We conducted the weekly monitoring (December 2012~April 2013) to evaluate the temporal variation and identification of Stephanodiscus spp. that are generally dominant during winter in the Yeongsan River. Phytoplankton species were identified and counted using the optical microscope and scanning electron microscope (SEM). Phytoplankton in the river were grouped into 6 classes (bacillariophyceae, chlorophyceae, cryptophyceae, cyanophyceae, dinophyceae, euglenophyceae), 30 genus and 41 species. Phytoplankton composition showed high abundance of diatoms in winter and Aulacoseira sp., Cyclotella sp. and Stephanodiscus spp. were dominant. Among the species, Stephanodiscus spp. was relatively abundant compared to other diatom species. Stephanodiscus spp. appeared from December 2012 to April 2013 and their abundance peaked in January. Abundance of diatoms especially peaked ($21,080cells\;mL^{-1}$) in January 15, 2013 when Stephanodiscus spp. also bloomed ($20,560cells\;mL^{-1}$). The abundances of Stephanodiscus spp. were gradually decreased from March and reached as low as $60cells\;mL^{-1}$ in April 26. Cyclostephanos (C. invisitatus), Cyclotella (C. meneghiniana), Discostella (D. pseudostelligera, D. woltereckii) and Stephanodiscus (S. hantzschii, S. minutulus, S. parvus) were classified in the circular diatoms. Abundance of S. hantzschii was extremely high compared to S. minutulus and S. parvus.
Blooms of the small centric diatom Stephanodiscus is quite occasional in winter season in temperate freshwater ecosystems. Often, it leads to degradation of water quality and affects quality of supplied drinking water. In previous studies, naphthoquinone (NQ) compounds have been shown to be effective and selective for controlling winter bloom species Stephanodiscus hantzschii. We conducted a 5 ton scale mesocosm experiment to investigate the effects of NQ on native Stephanodiscus sp. collected from Nakdonggang River in water. After treatment with NQ 4-6 compound (0.2 μM), the cell density of Stephanodiscus sp. was rapidly reduced from 5 × 103 cells mL-1 to 0.2 × 103 cells mL-1 for 10 days. Additionally, NQ 4-6 compound did not affect physicochemical factors (water temperature, dissolved oxygen, pH, conductivity, nutrients) and biological factors (bacteria, heterotrophic nanoflagellates, zooplankton). Therefore, these findings suggest that the NQ 4-6 compound has potential as an alternative algicidal substances to effectively mitigate natural Stephanodiscus sp. blooms, and the application of NQ 4-6 compound will restore the healthy aquatic ecosystems.
To examine the algicidal effect of co-treatment of biological control agent on centric diatom Stephanodiscus hantzschii in the filtered water, an endemic algicidal bacterium (Pseudomonas putida) and a heterotrich ciliate (Stentor roselli) were iso-lated from Pal'tang reservoir, Korea. Bacterial isolate and ciliate removed 98% and 80% of the diatom for 7 days of cultivation. Co-treatments of these two agents perfectly inhibited the diatom growth, compared to the single treatment of each agent. This synergistic interaction of the bacterium and ciliate could provide an effective tool in the biomanipulation to control the diatom bloom in freshwater lakes and streams.
To confirm the relationship between climate change and Stephanodiscus in Mulgeum station of Nakdong River, Korea, this study was conducted. The temperature in crease by climate change was observed in the study site, where the temperature was gradually increased in most seasons, except for summer season. The mass proliferation of Stephanodiscus constantly appeared in every year, especially between November and March, and when Stephanodiscus abundance was above 90% in phytoplankton biomass. Among this period, phytoplankton biomass was high related with water temperature ($r^2$=0.249, P<0.01) than nutrient factors such as nitrogen and phosphorus in the study site. Finally, temperature by climate change can be regarded as the affecting factor for chl. a variation, because temperature was strongly related with water temperature ($r^2$=0.748, P<0.01). From 1997 to 2010, the annual maximum phytoplankton biomass was recorded in the range of temperature from $4.8^{\circ}C$ to $8.4^{\circ}C$, and the range was regarded as the temperature condition for the optimal growth of Stephanodiscus in the study site. On the optimal growth temperature, the trend of monthly average temperature corresponded to the trend of chl. a variation from November to March. In future, the increase of temperature by climate change can prolong Stephanodiscus blooming period in winter and spring seasons.

The construction of eight large weirs in the Nakdong River, Korea, caused a decrease in the water flow velocity and several physical changes to the water environment. Here, changes in phyto- and zooplankton communities and water quality in the areas near the eight weirs were investigated from 2011 to 2013, and relationships between phytoplankton abundances and environmental factors were analyzed. Special emphasis was given to the succession patterns in algal abundance based on temperature fluctuations. At the eight weirs, 24 dominant species were found. The most abundant phytoplankton species was Stephanodiscus sp. (39.4% of dominant frequency). Cyanobacteria of the genus Microcystis dominated during the summer, with an dominant frequency of 8.5% and cell abundance ratio of 36.6%. Significant correlations were observed between temperature and abundance of eight of the main dominant species; seven species showed positive correlations with temperature. Stephanodiscus sp., however, showed a negative correlation with temperature (r=-0.26, p<0.01). In addition, this species showed a significant negative correlation with the dominant algal species-Aulacoseira granulata and Aphanizomenon flos-aquae, with the zooplankton Copepoda and with Cladocera. On the contrary, seven other dominant species of algae showed significant positive correlations with zooplankton. Thus, we showed that the seasonal succession of plankton communities in the Nakdong River was related to the water temperature changes.
To assess the algicidal effect of a bacterium, Pseudomonas fluorescens HYK0210-SK09 (SK09), attached to activated carbon polyvinyl alcohol (ACPA) and cellulose sponge (CS) carriers against Stephanodiscus hantzschii, the present study was carried out in an indoor-microcosm. As comparing immobilization effects of two carriers, the ACPA carrier allowed for higher packing cell density of SK09 compared to the CS carrier. In the microcosm, immobilized SK09 cells were applied to control S. hantzschii blooms. Immobilized SK09 cells exhibited a species-specific activity towards the diatom, showing an algicidal effect up to 72% attached by ACPA carriers and to 51% attached by CS carriers. In particular, a level of conductivity treated with ACPA carriers was decreased than that of CS carriers. The present study clearly demonstrates that ACPA-immobilized SK09 cells could effectively control S. hantzschii blooms and improve water quality in the microcosm ecosystem.
Environmental parameters and phytoplankton community structure were investigated at four sites of Yeongsan River between April 2010 and December 2011. The standing crops of the phytoplankton ranged from $275cells{\cdot}mL^{-1}$ to $58,600cells{\cdot}mL^{-1}$ with an average of $5,850cells{\cdot}mL^{-1}$. The dominant species were Cyclotella sp., Stephanodiscus sp., Aulacoseira granulata, Scenedesmus quadricauda, Pediastrum biwae, Coelastrum sp., Aphnizomenon sp., and Oscillatoria sp.. The most dominant species was Stephanodiscus sp.. The concentration of chlorophyll-a ranged from $2.3mg{\cdot}m^{-3}$ to $164.2mg{\cdot}m^{-3}$. The phytoplankton community structure of the survey area was influenced by temperature and rainfall.
Ecological characteristics of phytoplankton communities were investigated biweekly at 6 stations from Feb. 2001 to Feb. 2002 at the lower part of the Han River. During the study, a total of 267 taxa which were composed of 114 green algae, 104 diatoms, 22 blue-green algae, 13 chrysophytes and 2 dinoflagellates were observed. The standing crops of phytoplankton communities were ranged from 1.30 ${\times}10^6$ cells ${\cdot}l^{-1}$ to 37.47 ${\times}10^6$ cells ${\cdot}l^{-1}$. The dominant species were 11 taxa including Pediastrum duplex, Scenedesmus quadricauda in green algae, Osillatoria limosa, Microcystis aeruginosa, Merisomorpedia sp., Phormidium sp. in blue-green algae, and Asterionella gracillima, Aulacoseira granulata, Fragilaria crotonensis, Stephanodiscus hantzschii f. tenuis, Synedra acus in diatoms. Asterionella gracillima and Stephanodiscus hantzschii f. tenuis were abundant at every station from winter to spring. Stephanodiscus hantzschii f. tenuis occupied the highest dominant ratio and standing crop. From summer to autumn, dominant species were Aulacoseira granulata at all stations and blue-green algae such as Osillatoria limosa, Microcystis aeruginosa, Merisomorpedia sp. at some stations.
