Effects of Soil Moisture Control and Truss Limited High Density Culture on the Growth and Yield of Tomato(Lycopersicon esculentum Mill. cv. Momotaroyork) (토양수분조절과 저단밀식재배가 토마토의 생육 및 수량에 미치는 영향)

  • This study aims at investigating the effect of soil moisture control and truss limited high density culture on the growth, fruit yield and quality of tomato. To minimize of loss yield of tomato, flower cluster in number was limited to two and three truss and planting density was raised. Soil moisture control was started from 40 days after anthesis and irrigation point was set in -30kPa and -50kPa, which were compared with -10kPa For high density culture, the planting number of truss limited high density culture was planted twice as many as control. Soil moisture repression reduced the growth of stem diameter, leaf and plant height. Leaf chlorophyll content was higher in -50kPa and -30kPa than control. No significant differences, however, shows in -10kPa. The occurrence rate of bloom-end rot and cracking was increased by growing of irrigation repression. Pinching three fruit truss was higher than pinching two fruit truss in the occurrence rate of them. Soil moisture repression resulted in the reduction of fruit weight and in special, truss limited high density was distinctly decreased in -50kPa. The number of fruit was not affected by soil moisture control, but 3rd flower cluster was lower than 2nd flower cluster in the number of fruits and 2nd one was lower than 1st one. Under irrigation repression, rate of dry matter tended to grow in -30kPa, -50kPa compared with control and pinching three fruit truss was higher than two truss. Marketable yield dropped to 36.7%m 46.3 in -30kPa, -50kPa on pinching two fruit truss and dropped to 27.3%, 32.3% in 3rd flower cluster compared with control.

Effects of Foliar Application of Chitosan and Wood Extraction on Rooting and Tuber Formation of Plug Seedlings in Potatoes (Chitosan과 목초액 엽면살포에 의한 감자 플러그 삽목표의 발근 및 괴경형성효율)

  • To do mass multiplication of plug seedlings in potatoes, apical stem cuttings originated from virus-free microtubers were cut to one-two internodes and transplanted into the plug-tray. After 10days, we applied Chitosan and Wood Extraction on rooting and tuber formation of plug seedlings. To improve field adaptability of plug seedlings, rooted cuttings with a height of 20cm after 20days of cutting were transplanted ito the fields, We applied 500~2000ppm Chitosan on growth characteristics and tuber formation of that. The above and underground growths, such as plant height and number of leaves were significantly more vigorous after treatment with 500~1,000ppm Chitosan and 2,000ppm Wood Extraction, the spray treatment was carried out five times at intervals of four days after ten days of transplanting. T-N, K, P, Mg and Na, were higher as the concentrations of chitosan and Wood Extraction were higher. The growth and tuber yield in plug seedlings planting plot and seed potatoes planting plot were effectively highter as foliar application of Chitosan(500~2,000ppm) was done after planting the plot. T-N content in leaves and tuber was higher as the concentration of Chitosan was high. A similar tendency was shown in K, P and Mg. In the small tuber(under 30g), the number of tubers and tuber yield were relatively increased in the seed potatoes planting plot, but the large tubers(over 80g) yield was higher in the plug seedlings planting plot, and in order to increase tuber yield in plug seedlings it was necessary to add plant density to the field.

  • Macrophages are re-educated and polarized in response to myocardial infarction (MI). The M2 anti-inflammatory phenotype is a known dominator of late stage MI. Mesenchymal stem cells (MSCs) represent a promising tool for cell therapy, particularly heart related diseases. In general, MSCs induce alteration of the macrophage subtype from M1 to M2, both in vitro and in vivo. We conjectured that hypoxic conditions can promote secretome productivity of MSCs. Hypoxia induces TGF-β1 expression, and TGF-β1 mediates M2 macrophage polarization for anti-inflammation and angiogenesis in infarcted areas. We hypothesized that macrophages undergo advanced M2 polarization after exposure to MSCs in hypoxia. Treatment of MSCs derived hypoxic conditioned medium (hypo-CM) promoted M2 phenotype and neovascularization through the TGF-β1/Smad3 pathway. In addition, hypo-CM derived from MSCs improved restoration of ischemic heart, such as attenuating cell apoptosis and fibrosis, and ameliorating microvessel density. Based on our results, we propose a new therapeutic method for effective MI treatment using regulation of macrophage polarization.

  • Mucopolysaccharidoses (MPSs) are a group of rare inherited metabolic disorders caused by specific lysosomal enzyme deficiencies leading to the sequential degradation of glycosaminoglycans, causing substrate accumulation in various cells and tissues and progressive multiple organ dysfunction. The rare disease medical care team at Mackay Memorial Hospital in Taiwan has been dedicated to the study of MPSs for more than 20 years. Since 1999, more than 50 academic papers focusing on MPSs have been published in international medical journals. Topics of research include the following items regarding MPSs: incidence, natural history, clinical manifestations, gene mutation characteristics, cardiac function, bone mineral density, sleep studies, pulmonary function tests, hearing assessments, percutaneous endoscopic gastrostomy, anesthetic experience, imaging analysis, special biochemical tests, laboratory diagnostics, global expert consensus conferences, prenatal diagnosis, new drug clinical trials, newborn screening, and treatment outcomes. Of these published academic research papers, more than half were cross-domain, cross-industry, and international studies with results in cooperation with experts from European, American and other Asian countries. A cross-specialty collaboration platform was established based on high-risk population screening criteria with the acronym "BECARE" (Bone and joints, Eyes, Cardiac and central nervous system, Abdomen and appearance, Respiratory system, and Ear, nose, and throat involvement). Through this platform, orthopedic surgeons, rheumatologists, ophthalmologists, cardiologists, rehabilitation physicians, gastroenterologists, otorhinolaryngologists, and medical geneticists have been educated with regards to awareness of suspected cases of MPSs patients to allow for a further confirmative diagnosis of MPSs. Because of the progressive nature of the disease, an early diagnosis and early multidisciplinary therapeutic interventions including surgery, rehabilitation programs, symptom-based treatments, hematopoietic stem cell transplantation, and enzyme replacement therapy, are very important.

Changes in Quality of Welsh Onion (Allium fistulosum L.) during the Freezing Storage Period under Different Freezing Conditions (냉동조건에 따른 대파의 냉동저장 중 품질변화)

  • Purpose: This study was conducted to find the optimal freezing method and storage conditions for welsh onion. Methods: Cut welsh onions (0.3 cm) were packed in nylon/linear low density polyethylene (LLDPE) film bags, and frozen utilizing still-air freezing at -$20^{\circ}C$ (SAF20) and -$40^{\circ}C$ (SAF40), and immersed-liquid freezing at -$40^{\circ}C$ (ILF40); they were then stored at -$20^{\circ}C$ for 7 months. During storage, quality characteristics were measured monthly. Results: Drip loss was the lowest in the ILF40 packaging. Color difference in the stem (white part) did not differ significantly according to freezing conditions and storage time. Color difference in the leaf (green part) and stem was the lowest in SAF20. pH remained unchanged, while total aerobic bacterial count, pyruvic acid and moisture content decreased during storage. Pyruvic acid content of ILF40 was the highest among the freezing treatments. Fructose and glucose contents increased gradually during storage. Citric acid, malic acid, succinic acid and fumaric acid contents were unaffected, regardless of the freezing conditions. Conclusion: The optimal freezing method for welsh onions with the least quality changes was determined to be immersed liquid freezing, following by preservation up to 7 months by freeze-storing.

Effects of film liners, ethylene scrubber, alcohol releaser and chlorine dioxide on the berry quality during simulated marketing in 'Campbell Early' grapes

  • This study investigated the effects of an ethylene scrubber (ES) with a micro-perforated polypropylene (MP-PP, 30 ㎛) or a high density polyethylene (MP-HDPE, 30 ㎛) film liner for the export carton packaging box in 'Campbell Early' grapes. Rachis browning was highest in the untreated group, followed by MP-PP and MP-HDPE for 14 days of simulated marketing at 20℃. The combination treatment of ES with the film liners showed a partial inhibition of the rachis browning regardless of the film liners. The effects of an alcohol releaser (AR) sachet or chlorine dioxide (CD) diffuser co-packaging were also investigated in the 'Campbell Early' grapes packed with the MP-HDPE (40 × 99 pin hole·m-2) film liner. The CD 1 g treatment showed a very limited weight loss of 1.1%, which was significantly lower than the 4.7% of the untreated control after 14 days of simulation marketing at 20℃. The berry shatter was 0.7% for the MP-HDPE + CD 1 g treatment and 1.8% for the MP-HDPE + CD 5 g treatment on the 10th day of the simulated marketing, which was significantly lower than the 8.9% of the control. The stem browning was significant suppressed until the 10th day of the simulated marketing. In particular, the CD 1 g treatment in combination with the MP-HDPE showed a low rachis and pedicel browning index of 2.0, which is 50% and 40% lower than that of the untreated control and the MP-HDPE single treatment, respectively. In addition, the CD 1 g treatment group showed a higher decay reduction effect than the CD 5 g treatment group, which caused high concentration damage.

  • In order to increase the production of ginseng, a cultural experiment was carried out under different types of natural forest condition. Seedlings were transplanted with three spacing (70,90 and plants per 1.62m2) under the broad leaved, needle and mixed forest. The obtained results are as follows. 1 Growth of aerial part of ginseng plant. 1) Vegetative growth under forest condition of very poor as compared with ordinary cultivation, but there was no significant in number of leaf and teasel. 2) Stem diameter and stem length under the different forest types were a little difference. However petiole length and number of leafet showed an increasing trend in broad leaved forest as compared with other forest types. 3) The withering date of aerial part of ginseng plant in the needle forest was later than of others. 2. Fresh weight of ginseng root per plant was decreasing in the order of broad leaved forest, needle and mixed forest, needle and sized forest respectively. However the root weight was much smaller than that of ordinary cultivated one. 3. No big difference was observed in the growth of both aerial and root among the planting density 4. Nitrogen content in ginseng root under forest was lower, but calcium content In root was higher than that of ordinary one. 5. Fat and fiber content of ginseng root under forest showed higher than that of ordinary one. 6. The saponin content of ginseng root grown under forest condition was higher than that of ordinary ginseng root. According to high performance liquid chromatogram of saponin, only difference from ordinary cultivated ginseng root was that ginsenoside Re showed higher peak than ginsenoside Rg1.

  • The adhesion and proliferation of mammalian cells on polymeric biomaterials depend on the surface characteristics such as wettability, chemistry, charge and roughness. In order to recognize the correlation between the adhesion and proliferation of human bone marrow derived stem cells (BMSCs) and surface property, radio frequency generated plasma treatment on low density polyethylene (LDPE) has been carried out. The modified LDPE surfaces were characterized by measuring the static water contact angle. The adhesion and proliferation of cells on LDPE films were characterized by cell counting and reverse transcription-polymerase chain reaction (RT-PCR). The water contact angle of the film surface decreased with plasma treatment time. Proto-oncogenes (c-myc, c-fos) and tumor suppressor gene (p153) showed maximum expression with contact angle of 60 ∼ 70$^{\circ}$ range of LDPE film. By cell counting, we confirmed that the rate of cell proliferation appeared the higher on the film surface of the contact angle of 60∼70$^{\circ}$ We concluded that the surface wettability is an important role for the growth and differentiation of BMSCs.

Derivation of endothelial cells from porcine induced pluripotent stem cells by optimized single layer culture system

  • Regenerative therapy holds great promise in the development of cures of some untreatable diseases such as cardiovascular diseases, and pluripotent stem cells (PSCs) including induced PSCs (iPSCs) are the most important regenerative seed cells. Recently, differentiation of human PSCs into functional tissues and cells in vitro has been widely reported. However, although porcine reports are rare they are quite essential, as the pig is an important animal model for the in vitro generation of human organs. In this study, we reprogramed porcine embryonic fibroblasts into porcine iPSCs (piPSCs), and differentiated them into cluster of differentiation 31 (CD31)-positive endothelial cells (ECs) (piPSC-derived ECs, piPS-ECs) using an optimized single-layer culture method. During differentiation, we observed that a combination of GSK3β inhibitor (CHIR99021) and bone morphogenetic protein 4 (BMP4) promoted mesodermal differentiation, resulting in higher proportions of CD31-positive cells than those from separate CHIR99021 or BMP4 treatment. Importantly, the piPS-ECs showed comparable morphological and functional properties to immortalized porcine aortic ECs, which are capable of taking up low-density lipoprotein and forming network structures on Matrigel. Our study, which is the first trial on a species other than human and mouse, has provided an optimized single-layer culture method for obtaining ECs from porcine PSCs. Our approach can be beneficial when evaluating autologous EC transplantation in pig models.

Relationship between Lodging and Agronomic Characters of Soybean Plant (대두의 도복 양상 및 관련 형질 연구)

  • This study was carried out to investigate the lodging patterns and the direct and indirect effects of several agronomic characters upon lodging of soybean plants. Most of the lodged plants had bent main stem above first stem node at higher plant density. Lodging was significantly correlated with plant height, internode length and shoot fresh weight in two soybean cultivars, Paldalkong and Jangyeobkong. Root fresh weight was also significantly correlated with lodging in Paldalkong, Jangyeobkong and Kwangkyo. Length of internode showed the greatest direct effect on lodging in Paldalkong and Jangyeobkong.
