다양한 계통의 옥수수 미성숙배로부터 캘러스 유도와 식물체 재분화 (Callus induction and plant regeneration from immature zygotic embryos of various maize genotypes (Zea mays L .))

  • 옥수수의 최적 조직 배양 조건을 확립하기 위하여 옥수수 국내 5 계통과 국외 11 계통 총 16 계통을 포트와 포장 재배하여 미성숙 배를 분리하여 배발생 캘러스 유도 및 식물체 재분화율을 조사하였다. MS 배지에 auxin으로 1.5 mg/L Dicamba와 0.5 mg/L 2,4-D 가 첨가된 배지에서 캘러스 형성은 본 실험에 사용된 옥수수 계통 모두에서 높은 빈도로 유도되었으며, 캘러스로부터 식물체 재분화는 5mg/L zeatin이 첨가된 재분화 배지에서 높은 재분화율을 보였다. 또한 포장에서 재배된 옥수수로부터 미성숙 배를 분리하여 사용하였을 때 캘러스 유기 및 식물체 재분화 효율이 높았던 것으로 보아 미성숙 배를 분리하기 위한 옥수수 상태 및 genotype이 중요한 영향을 준다는 것을 알 수 있었다. 본 실험을 통하여 배 발생 캘러스 형성 및 식물체 재분화 효율이 조사된 옥수수 계통들은 생명공학 기술을 활용한 신품종 개발을 위한 형질전환 시스템 개발에 유전자원으로 활용될 수 있는 정보를 제공할 것으로 사료된다.

금강하구역에서 영양염 거동에 대한 조석 및 담수유출의 영향 (The Effect of Tidal Cycle and River Runoff on the Dynamic of Nutrients in Keum river estuary)

  • This study was to evaluate the impact of river runoff and salt intrusion by tide on nutrient balance of estuary during a complete tidal cycle. 24 hours time series survey was carried out during a spring tide July 2001 on a tidal estuary in the Keum river. Three stations(A,B,C) were set along a transect line of about 10km, which linked the lower part of estuary dyke to the subtidal zone. Surface water was sampled simultaneously at each station every hours f3r the determination of nutrients. Water temperature, pH and dissolved oxygen were measured in situ. Riverine input of silicate and nitrate during ebb tide significantly increased the concentration of all stations. Conversely, during high tide, nutrient concentration were lowered by the mixing of fresh water with sea water Ammonium nitrogen concentration were higher at intertidal zone(Stn.B) due to sewage inflow to Kyeongpo stream and ammonium release under anaerobic conditions. Also, these results was discussed as a biological component that influences the processes of nutrient regeneration within the estuary. Best correlations were found at lower part of estuary dyke(Stn.A) for salinity against DIN(Y=0.121 Sal.+4.97, r2=0.956) and silicate(Y=0.040 Sal.+2.62, r2=0.785). But no significant correlation was found between salinity and ammonium. Unbalanced elemental ratio(N/P, Si/N and Si/P) depended significantly on the import of nutrients (silicate & nitrate nitrogen) from river and stream. The effect of the tidal cycle and river runoff is important that in determining the extend of the variations in nutrient concentrations at all station.

  • This study was based on the results of the study of 'Cheonyeon and Chunghyun Taste Alley Pilgrimage- Introducing Hidden Restaurants in Our Town', which was adopted as a project to revitalize urban regeneration as part of the Cheonyeon and Chunghyun Urban Regeneration New Deal project. This study was conducted in total of two stages, as a first step, the commercial district of Seodaemun Station was analyzed by analyzing the hashtag (#) mentioned along with the "Seodamun Station Restaurant" on Instagram from 2015 to 2020. As a result of the analysis, it was found to be an office commercial district related to "office workers", and it was found to be a commercial district with the characteristics of "small but certain happiness" where you can find hidden restaurants in front of your house. Based on the characteristics of these commercial districts, five stores utilizing the characteristics of the region were selected and cooking classes were conducted for students of Kyonggi University, who are local residents. The purpose of this study was to revitalize the aging Seoul city and contribute to the formation of positive relationships between local residents and merchants through cooking classes. In addition, the process was produced as digital media content and used as local promotional materials.

산약(山藥)의 기내배양(器內培養)에 미치는 배지(培地) 및 생장조절물질(生長調節物質)과 활성탄(活性炭)의 영향(影響) (Effect of Medium, Growth Regulators and Activated Charcoal on Plant Regeneration from in vitro Culture of Dioscorea japonica TUNBERG)

  • 마산약(山藥) 경절편(莖節片)의 기내조직(器內組織) 배양(培養)을 위해서 적정배지(適正培地)가 선발(選拔)됨에 따라 이에 적합시(適合時) 되는 생장조절물질(生長調節物質) 활성탄(活性炭)의 처리농도(處理濃度)를 구명(究明)함으로서 기내조직(器內組織) 배양(培養)에 의(依)한 증식율(增殖率)를 높이코져 무기강기성(無機강基性)이 낮은 1/8MS, White에 IAA와 NAA를 $1{\sim}4mg\;/\;\ell$ 로 하고 활성탄(活性炭)을 $0,\;1,\;3,\;6g\;/\;\ell$ 수준(水準)으로 실험(實驗)을 실시(實施)하였던 바 다음과 같은 결과(結果)를 얻었다. 1. 1/8MS 배지(培地)의 경우 $1{\sim}4mg\;/\;\ell$, Kinetin $2mg\;/\;\ell$ 그리고 활성탄(活性炭)을 $1{\sim}3g\;/\;\ell$ 첨가(添加) 하므로서 Shoot 유기(誘起)와 식물체(植物體)의 재분화률(再分化率)이 모두 100%에 달하는 효과(效果)를 보였으며 이러한 효과(效果)는 NAA처리시(處理時)에도 같은 경향(傾向)이 있었다. 2. White 배지(培地)의 경우 IAA $1mg\;/\;\ell,\;Kinetin\;2mg\;/\;\ell$ 그리고 활성탄(活性炭) $6g\;/\;\ell$ 첨가처리(添加處理)와 NAA $4mg\;/\;\ell,\;Kinetin\;2mg\;/\;\ell$ 그리고 활성탄(活性炭)을 $1g\;/\;\ell$ 첨가처리(添加處理) 만이 식물체(植物體)의 재분화율(再分化率)이 l00%에 달하였으나 그 외(外) 처리(處理)에서는 대부분(大部分) 저조(低調)하였다. 3. 발육상태(發育狀態)를 보면 1/8M /S 배지(培地)의 경우 IAA $2mg\;/\;\ell,\;Kinetin\;2mg\;/\;\ell$ 그리고 활성탄(活性炭)을 $1g\;/\;\ell$ 첨가처리(添油處理)와 NAA $4mg\;/\;\ell,\;Kinetin\;2mg\;/\;\ell$ 활성탄(活性炭)을 $1g\;/\;\ell$ 첨가처리(添加處理) 하므로서 모두 주당(株當) 본수(本數)가 $2.3{\sim}2.7$개(個)이고 발육상태(發育狀態)도 활성탄(活性炭)$0g\;/\;\ell,\;3g\;/\;\ell,\;6g\;/\;\ell$ 처리(處理)보다 양호(良好)하였다. 4. 그러나 White 배지(培地)의 경우 IAA $1{\sim}4mg\;/\;\ell,\;NAA\;1{\sim}mg\;/\;\ell$ 그리고 $Kinetin\;2mg\;/\;\ell$ 처리(處理)의 대부분(大部分)이 주당(株當) 본수(本數)가 거의 1개체(個體) 정도(程度)이도 발육상태(發育狀態)도 불량(不良)하였으나 NAA $4mg\;/\;\ell\;Kinetin\;2mg\;/\;\ell$ 그리고 활성탄(活性炭) $1g\;/\;\ell$ 첨가처리(添加處理)에서 만이 주당(株當) 본수(本數)가 2.3개이고 개체당(個體當) 발근수(發根數)가 월등(越等)히 많았다. 5. 실험결과(實驗結果) 마 경절편(莖節片) 배양(培養)의 Shoot유가(誘起)에 가장 효과적(效果的)인 배지(培地)는 1/8MS+1AA $2mg\;/\;\ell+Kinetin\;2mg\;/\;\ell$+활성탄(活性炭) $1g\;/\;\ell$ 이였으며, White+NAA $4mg\;/\;\ell+Kinetin\;2mg\;/\;\ell$+활성탄(活性炭)$1g\;/\;\ell$ 로 처리(處理)하는 것이 바람직한 결과(結果)를 얻었다.

고속철도역 신설과 도시 재구조화 연계 계획의 특성 - JR큐슈 주요 역을 중심으로 (A Study on the Characteristics of Urban Re-Organization regarding as an Establishment of New High-Speed Railway Stations focused on JR Kyushu's Main Stations)

  • 본 연구는 일본의 큐슈지역에서 고속철도가 추가로 부설되면서, 기존의 구 철도역사에 이를 수용하기 위한 기능을 도입하기 위해 새로운 고속철도역사를 추가로 건립하고, 건립과 동시에 철도역사를 중심으로 한 인접역세권 개발을 추진하고 있는 사례들에 주목한 연구이다. 일본 큐슈 지역의 고속철도역사를 중심으로 한 구도시의 재구조화 방식 및 기법 및 특성을 분석하여 그 계획적 의미 및 특성을 도출하고, 국내 역세권 개발에 적용 가능한 도시계획기법을 연구하는데 그 목적이 있다. 연구대상은 일본 큐슈 지역에 신칸센이 연장되면서 최근 5년 이내 신축되었거나, 개발 중인 하카타역, 구마모토역, 가고시마 중앙역의 3개 역사를 사례로 하였다. 연구의 결과 큐슈 북부, 중부, 남부에 위치한 각 해당 철도역은 큐슈 전 지역 내에서 각 역의 지리적 위치에 의한 지역적 관계, 즉 지리적 입지와 철도네트워크 내에서의 각 역 기능의 적극적 연계, 도심과 역과의 관계 재설정, 관광지 개발과 밀접한 연계체계 확립, 낙후될 수밖에 없었던 선로 반대측으로의 개발을 확장시키는 고속철도역사 병설방식, 고밀 복합용도 개발을 통한 상주인구 확보 등과 같은 전략을 통해 역을 중심으로 지역계획과 통합하여 도심재생을 추진하였음을 확인하였다.

Molecular Characterization of Intergeneric Hybrids between Trichoderma harzianum and Gliocladium virens

  • Nuclei were isolated from the protoplasts of Trichoderma harzianum T95 and treated with colchicine, a polyploid inducer. The nuclei were transferred into the protoplast of multi-auxotrophic Gliocladium virens G88 which cannot grow in minimal medium. The protoplast of G. virens G88 carrying the transferred nuclei were regenerated in a regeneration minimal medium containing $17{\mu}g/ml$ of chloroneb as a haploid inducer. Six intergeneric hybrids between G. virens and T. harzianum were isolated from the regeneration minimal medium. The hybrids could be classified into three types according to morphology, those with an isozyme pattern, those with an protein band and those with an randomly amplified polymorphic DNA(RAPD) pattern produced by random primers and repetitive sequences. The first group was identified to be a haploid recombinant, the second group a heterokaryon, and the third appeared to be petite.

  • 산지가 많은 부산시의 지형적 특성상 가용토지가 부족한 상황에서 경부선 철도까지 도심을 가로지르고 있어 생활권의 단절과 도심발전을 저해하고 있다. 이에 토지 확보를 위하여 입체개발의 필요성이 제기되고 있으며, 최근에는 경부선 철도의 지하화로 양분화 된 도시를 연결하고 토지를 효율적으로 사용할 수 있도록 하는데 관심이 증가하고 있다. 본 연구에서는 토지의 입체개발의 사례를 통해 개발유형을 분석하고, 부산시내 경부선 철도의 지하화 사업 시 메인이 되는 4개 권역(사상역, 구포역, 가야역, 부산진역)의 입체개발로 발생하는 가용토지에 효과적인 활용을 위한 시민인식조사 결과를 바탕으로 입체개발 유형별 개발방향 설정하였다. 또한 대상지 지역이 가지고 있는 지역적 특성과 현재 진행되고 있는 도시재생사업을 반영하여 철도에 의해 양분된 지역생활권의 단절 해소 및 철도시설 부지의 효율적 토지이용을 통한 입체적인 개발을 통해 철도 부지의 수평적, 수직적 공간활용의 설계방안을 제시하고자 한다.

Reuses Of Wash Water Effluents Of The Ion-Exchanger Units Of Water Demineralization Plant For Economic And Environmental Benefits

  • In industrial field, a large volume of regenerants (acid and caustic soda) and their washing effluents are regularly disposed off from the water demineralization plant during regeneration of the ion-exchanger units. Of these waste effluents, a part of the wash water discharged from the single bed Anion and Mixed Bed units can be utilized at a certain stage of their washing cycles when its conductivity is fallen down and becomes considerably less than that of the input raw water. The main aim of this specific waste effluent utilization is to dilute the TDS concentration of the input raw water (fed into the single bed ion-exchanger units) by blending. The achievement is the increase of the longevity of the production cycles of the I.E. units along with the improvement of the production quality and decrease of the regeneration frequencies. As a result, regenerant consumption would be saved because of the reduction of ionic load in feed water which will ultimately reduce the water purification cost. At the same time, the environment pollution will also be protected to a considerable extent. This operational measure is quite effective and useful specially where high TDS water is demineralized only by single bed ion-exchangers. In such case, the water treatment plant is very often found to suffer from both production quality and quantity in addition to carrying out of random and restless regenerations. Proper reuses of the aforesaid wash water effluents of the Anion and MB units excellently minimizes the difficulties experienced in practice. This paper contains the utilities and techniques of reuses of the different kinds of waste effluents of the industrial water treatment plant in addition to the specific reuses of the post-regeneration wash waters of the Anion and MB ion-exchanger units.

Natural Regeneration Potential of the Soil Seed Bank of Land Use Types in Ecosystems of Ogun River Watershed

  • Soil seed banks as natural storage of plant seeds play an important role in the maintenance and regeneration of watershed. Natural regeneration potential of the soil seed bank of Land use types (LUTs) in Ogun River watershed (ORW) was investigated. ORW was stratified using proportionate sampling technique into Guinea Savannah (GS), Rainforest (RF) and Swamp Forest (SF) Ecological Zones (EZs). Three LUTs: Natural Forest (NF), Disturbed Forest (DF) and Farmland (FL) were purposively selected in GS: GSNF, GSDF, GSFL; RF: RFNF, RFDF, RFFL and SF: SFNF, SFDF, SFFL, respectively. Systematic line transects was used in the laying of the sample plots. Sample plots of 25 m×25 m were established in alternate positions. Ten 1 m×1 m quadrats were randomly laid for soil core sampling from previously randomly selected ten plots. The core samples (10) were pooled per plot in each LUT and placed in individual trays. Ten trays with sterilized soil were used as control. The trays were watered regularly and checked for seedlings emergence fortnightly for 18 months. The experimental design used was 3×3 factorial experiments. ANOVA, Diversity index (H') and Similarity index (SI) were used to analyze the data. There was significant difference in seedling emergence among ecological zones and land use types (p<0.05). A total of 4,400 seedlings emerged from the soil samples. All species were distributed among 32 families. FL in the RF had the highest number of germinated seeds (705±37.33 seedlings) followed by DF in the RF (701±49.6 seedlings). The lowest emergence was in NF of the SF (199±28.41 seedlings). DF in the RF had highest number of species (34) distributed among 22 families. Emergence from soil seed bank of NF in ORW was generally with more of tree species than herbs that were predominant in FL and DF.
