Dammarane-type triterpene oligoglycosides from the leaves and stems of Panax notoginseng and their antiinflammatory activities

  • Background: Inflammation is widespread in the clinical pathology and closely associated to the progress of many diseases. Triterpenoid saponins as a key group of active ingredients in Panax notoginseng (Burk.) F.H. Chen were demonstrated to show antiinflammatory effects. However, the chemical structures of saponins in the leaves and stems of Panax notoginseng (PNLS) are still not fully clear. Herein, the isolation, purification and further evaluation of the antiinflammatory activity of dammarane-type triterpenoid saponins from PNLS were conducted. Methods: Silica gel and reversed-phase C8 column chromatography were used. Furthermore, preparative HPLC was used as a final purification technique to obtain minor saponins with high purities. MS, NMR experiments, and chemical methods were used in the structural identifications. The antiinflammatory activities of the isolated saponins were assessed by measuring the nitric oxide production in RAW 264.7 cells stimulated by lipopolysaccharides. Real-time reverse transcription polymerase chain reaction was used to measure the gene expressions of inflammation-related gene. Results: Eight new minor dammarane-type triterpene oligoglycosides, namely notoginsenosides LK1-LK8 (1-8) were obtained from PNLS, along with seven known ones. Among the isolated saponins, gypenoside IX significantly suppressed the nitric oxide production and inflammatory cytokines including tumor necrosis $factor-{\alpha}$, interleukin 10, interferon-inducible protein 10 and $interleukin-1{\beta}$. Conclusion: The eight saponins may enrich and expand the chemical library of saponins in Panax genus. Moreover, it is reported for the first time that gypenoside IX showed moderate antiinflammatory activity.

  • Recently, Japan has showed strong demand for protection of consumers against misleading specification provided by producers, establishing the committee for JIS and ISO. To protect domestic market and overcome potential subordinate relationship in overseas market, several universities and institutes in Korea have performed collaborative works on the standardization of test method f3r photocatalytic activity concerning the selected application fields. This article deals with the process and the result for the method of testing photocatalytic sol or suspended powder using optical fiber. Currently, this method is Intended for TiO2, but can be applied for various photocatalytic raw materials that can be activated by visible light and solar irradiation.

Study on Antioxidant Activity and Standardization of FDY003 (FDY003의 항산화활성 및 표준화 연구)

  • Objectives: FDY003 is a raw material for medicine consisting of a natural product that is expected to have the advantages of low side effects and high efficacy. In this study, we predict the efficacy and the standardization of the drug by method validation of anticipated index compounds and the measurement of antioxidant activity. Methods: FDY003 is prepared by extracting and purifying 70% of ethyl alcohol (EtOH). The method validation of cordycepin and chlorogenic acid was determined by high-performance liquid chromatography-photo diode array (HPLC-PDA) and the content of FDY003 was calculated. In order to monitor the biological activity of FDY003, antioxidant activity was measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric-reducing antioxidant power (FRAP). The equivalent values of antioxidants such as trolox, ascorbic acid, gallic acid, and caffeic acid were measured by ABTS and FRAP. Results: Chlorogenic acid and cordycepin were both found suitable for method validation in HPLC and FDY003 containing 9.92±0.50 and 17.97±0.27 ㎍/g, respectively. In DPPH, the electron donating ability (EDA) value of FDY003 was increased in a concentration dependent manner. FDY003 confirmed antioxidant activity by ABTS and FRAP. Conclusions: FDY003 contains certain components including cordycepin and chlorogenic acid and has antioxidant ability by various mechanisms. Therefore, it is expected that FDY003 is capable of various physiological activities including anti-cancer activity.

  • Standardization of drug deals with confirmation of drug identity and determination of drug quality and purity. Unani herbal formulations are used in traditional medicine for the treatment of various diseases. Cancer is a disease which causes abnormal, uncontrolled growth of body tissue or cells, which tend to proliferate in an uncontrolled way. Spread of cancer from site of origin to other organs of the body is called metastasis. It is a hyper proliferative disorder involving, transformation, dysregulation of apoptosis, invasion and angiogenesis. The present study aimed to standardize a classical Unani formulation (CUF) described as anticancer properties. The CUF has been used for anti-cancerous activity (Dāfi'-i-saraṭān) in human population by Unani physicians for centuries. The standardization parameters carried out for classical Unani formulation are pharmacognostical studies, physicochemical parameters, high-performance thin layer chromatography (HPTLC), microbial load, aflatoxins, and heavy metals revealing specific identities and to evaluate Pharmacopoeial standards. Experiment and the data obtained established the Pharmacopoeial standards for this formulation for identification and quality control purpose. The CUF has been successfully standardized and standard operating procedures (SOPs) for its preparation has been laid down which may serve as a standard reference in future. The standardization data of this formulation may be used as a standard guideline for preparation of the formulation in future.

A Study on Standardization Guidelines for BIM-based Maintenance and Management of Civil Infrastructures (BIM기반 사회기반시설물 유지관리를 위한 표준화 가이드라인 연구)

  • This paper provides core standardization items and method for the building information modeling (BIM) based maintenance and management (M&M) of civil infrastructure. Value chain of the M&M and characteristics of BIM technology has been analyzed and compared. As a result, it has been revealed that the initial activity recording damage information is very similar to the BIM-based information operation. The core standardization items for generation and operation of BIM data were identified according to the M&M value chain, The identified items were categorized into common standard item and specialized items for M&M of civil infrastructure, and proper government funded organizations for each standardization items were introduced on the basis of the roles in the laws.

  • The standardization of Intelligent Transport Systems(ITS) technique should secure a interoperability among the different softwares or hardwares on ITS service. The domestic standard research takes the advantage of a preoccupation on the world ITS market through the international standard(IS) reflecting the domestic technique. The objective of this research focused on the database technology for ITS is to introduce the domestic standard activity for adopting ISs and the current interior corresponding situation against ISs. The first step, we analyze the international standardization action through gathering and investigating the up-to-date issues and documentations focused on ISO/TC204 WG3. And then, we look into the domestic situation about TTA ITS PG 310 and ITS Korea. consequently, a synthetic view, we have analyzied, how interior standard corresponds with the international standards, and how the standard establishing process is being done. The expectation of this research is to help understanding the up-to-date domestic/international standardization and the situation of the standard action related with the database technology for ITS. Also, we take advantage of this result to encourage the international competitiveness on ITS market.

  • Domestic road type-and period-specific driving pattern measurement was required as Korea's participation in developing "Worldwide harmonized light-duty vehicle emission test procedure (WLTP/DHC)" studied by UN WP29. This study measured road driving data reflecting road and traffic conditions of Korea, and analyzed seven types of representative road type-and period-specific driving patterns with driving pattern standardization methodology proposed by WP29. PAMS (Portable Activity Monitoring Systems) equipment was used to collect enormous (35,410km) road driving data. There are significant difference among seven derived driving patterns.
