Multiphasic Analysis of Growth Curve of Body Weight in Mice

  • The present study describes the analysis of the multiphasic growth function (MGF) to body weight in laboratory and wild mice. Three genetic groups of laboratory mice (Mus musculus domesticus) designated $CF_{{\sharp}1}$, C3H/HeNCrj and C57BL/6NCrj, and a genetic group of Yonakuni wild mice (Mus musculus molossinus yonakuni, Yk) were used. Mean body weights of each genetic group-sex subclass from birth to 69 days of age taken at 3-day intervals were analyzed by a monophasic, diphasic and triphasic functions for describing growth patterns. A comparison among the three functions of the MGF was based on the goodness-of-fit criteria: residual standard deviation (RSD), adjusted R-square (Adj $R^2$) and Akaike's information criterion (AIC). Result of this study indicated that body weight averaged heavier for males than for females. Among the four genetic groups within both sexes, $CF_{{\sharp}1}$ showed the highest, subsequent followed by C3H/HeNCrj, C57BL/6NCrj and Yk. Comparison among the three functions revealed that the triphasic function was the best fit to growth data, with the lowest RSD, the highest Adj $R^2$ and the lowest AIC, for the four genetic groups. For the triphasic function, RSD within each genetic group-sex subclass was similar for males and females. Adj $R^2$ was 0.999 for all genetic group-sex subclasses. AIC for laboratory mice males and females ranged from -70.48 to 66.50 and from -92.81 to -68.64, respectively; whereas for Yk wild mice males was -74.29 and females -78.42.

  • The present study was undertaken to investigate the metabolic problems of high carbohydrate and cellulose diets of Korean. Forty males and same number of females of Albino rats, aged $45{\pm}5$days were divided into 95% high carbohydrate (H. CHO)group, 83.8% medium carbohydrate (M. CHO)group, 50% low carbohydrate (L. CHO) group and standard (Stand). group containing 72.2% sugar. Each group was divided into two again-1.55% cellulose group and non-cellulose group, 10 rats each of eight groups in both sexes. Cellulose was added to each of non-cellulose diets in the forms of spinach powder and rice bran. After 14 weeks the rats were sacrificed for chemical analysis and the results were elucidated as follows. (1) H. CHO+Cell. group showed the lowest value in body weight gained and shrinkage of almost all organs, in contrast with this group the L.CHO group showed higher body weight gained than Stand. group. M. CHO+Cell, group showed much the same body weight gained curve as Stand. group. (2) It was observed that cellulose group showed lower F.E.R and P.E.R value than non-cellulose group comparatively. (3) Total nitrogen retention and retention rate were decreaced in H. CHP groups compared with M. CHO or L. CHO groups. (4) The amount of feces was increased due to addition of cellulose to experimental diets and in accordance with the increasing total fecal excretion of nitrogen and glucose was also increased, especially noticeable in fecal glucose excretion. (5) It was noteworthy that serum cholesterol level was decreased due to addition of cellulose in H. CHO group and L. CHO group. (6) M. CHO+Cell. group was designed to reflect the average survey data of Korean diets and there was no significant differences on body weight gained, F.E.R, P.E.R, total nitrogen retention and hematology between M. CHO+Cell. and Stand. group. Total glucose excretion was increased due to dietary cellulose in M. CHO+Cell. group, but it seemed to be no metabolic problems in this group.

A Study on the Factors Causing Analytical Errors through the Estimation of Uncertainty for Cadmium and Lead Analysis in Tomato Paste (불확도 추정을 통한 토마토 페이스트에서 카드뮴 및 납 분석의 오차 발생 요인 규명)

  • BACKGROUND: This study aimed to estimate the measurement uncertainty associated with determination of cadmium and lead from tomato paste by ICP/MS. The sources of measurement uncertainty (i.e. sample weight, final volume, standard weight, purity, molecular weight, working standard solution, calibration curve, recovery and repeatability) in associated with the analysis of cadmium and lead were evaluated. METHODS AND RESULTS: The guide to the expression of uncertainty was used for the GUM (Guide to the expression of Uncertainty in Measurement) and Draft EURACHEM/CITAC (EURACHEM: A network of organization for analytical chemistry in Europe/Co-Operation on International Traceability in Analytical Chemistry) Guide with mathematical calculation and statistical analysis. The uncertainty components were evaluated by either Type A or Type B methods and the combined standard uncertainty were calculated by statistical analysis using several factors. Expected uncertainty of cadmium and lead was $0.106{\pm}0.015$ mg/kg (k=2.09) and $0.302{\pm}0.029$ mg/kg (k=2.16), on basis of 95% confidence of Certified Reference Material (CRM) which was within certification range of $0.112{\pm}0.007$ mg/kg for cadmium (k=2.03) and $0.316{\pm}0.021$ mg/kg for lead (k=2.01), respectively. CONCLUSION(s): The most influential components in the uncertainty of heavy metals analysis were confirmed as recovery, standard calibration curve and standard solution were identified as the most influential components causing uncertainty of heavy metal analysis. Therefore, more careful consideration is required in these steps to reduce uncertainty of heavy metals analysis in tomato paste.

A Comparative Study on Quantifying Uncertainty of Vitamin A Determination in Infant Formula by HPLC (HPLC에 의한 조제분유 중 비타민 A 함량 분석의 측정불확도 비교산정)

  • The purpose of this study was to determine the accurate quantification of vitamin A in infant formula by comparing two different standard stock solutions as well as various sample weights using high performance liquid chromatography. The sources of uncertainty in measurement, such as sample weight, final smaple vloume, and the instrumental results, were identified and used as parameters to determine the combined standard uncertainty based on GUM(guide to the expression of uncertainty in measurement) and the Draft EURACHEM/CITAC Guide. The uncertainty components in measuring were identified as standard weight, purity, molecular weight, dilution of the standard solution, calibration curve, recovery, reproducibility, sample weight, and final sample volume. Each uncertainty component was evaluated for type A and type B and included to calculate the combined uncertainty. The analytical results and combined standard uncertainties of vitamin A according to the two different methods of stock solution preparation were 627 ${\pm}$ 33 ${\mu}$g R.E./100 g for 1,000 mg/L of stock solution, and 627 ${\pm}$ 49 ${\mu}$g R.E./100 g for 100 mg/L of stock solution. The analytical results and combined standard uncertainties of vitamin A according to the various sample weighs were 622 ${\pm}$ 48 ${\mu}$g R.E./100 g, 627 ${\pm}$ 33 ${\mu}$g R.E./100 g, and 491 ${\pm}$ 23 ${\mu}$g R.E./100 g for 1 g, 2 g, and 5 g of sampling, respectively. These data indicate that the preparation method of standard stock solution and the smaple amount were main sources of uncertainty in the analysis results for vitamin A. Preparing 1,000 mg/L of stock solution for standard material sampling rather than 100 mg, and sampling not more than 2 g of infant formula, would be effective for reducing differences in the results as well as uncertainty.

Quantitative Analysis and Qualification of Acrylamide Using LC/ESI-MS (LC/ESI-MS를 이용한 Acrylamide의 정성확인 및 정량분석)

  • Acrylamide, difficult to analyze by GC and GC/MS due to the polarity and low volatility, was analyzed by LC/ESI/MS in the study. Acrylamide its(molecular weight 71amu) showed m/z=72 $(M+H)^+$ and high peak intensity at 22V in SIR mode. The mass spectrum ratios of acrylamide for qualitative identification had m/z=72 in precursor ion and m/z=55 in products ion, respectively. Those ratios at 30V in SIR mode ranged from 1: 1.4 to 1:1.17 despite various acrylamide concentrations. The ion intensity ratios of acrylamide $(m/z=72,\; [M+H]^+)$ to acrylamide isotopes $(m/z=73,\;[M+H]^+)$ ranged from 100 : 3.57 to 100 : 3.92. The results verified theoretical mass spectrum ratio that was 100:3.82. The linearity of standard calibration curve was y : 520.584x + 1815.26 with $r^2=0.99.$ In quality assurance and quality control, the recovery rate ranged from 81.64 percent to 90.97 percent and relative standard deviation was less than $10\%$ with 5 repeated injections at individual standard calibration solutions. The method was applied to analyze acrylamide in food at grocery stores. Snacks made of potatoes showed the highest acrylamide concentration followed by products made of French fries, wheat, and corn.

A Stress-Strain Relationship of Alkali-Activated Slag Concrete (알칼리활성 슬래그 콘크리트의 응력-변형률 관계)

  • The present study summarizes a series of compressive tests on concrete cylinder in order to examine the stressstrain relationship of alkali-activated (AA) slag concrete. The compressive strength and unit weight of concrete tested ranged from 8.6 MPa to 42.2 MPa and from $2,186kg/m^3$ to $2,343kg/m^3$, respectively. A mathematical equation representing the complete stress-strain curve was developed based on test results recorded from 34 concrete specimens. The modulus of elasticity, strain at peak stress, slopes of ascending and descending branches of stress-strain curves were generalized as a function of compressive strength and unit weight of concrete. The mean and standard deviation of the coefficient of variance between measured and predicted curves were 6.9% and 2.6%, respectively. This indicates that the stress-strain relationship of AA slag concrete is represented properly with more accuracy in the proposed model than in some other available models for ordinary portland cement (OPC) concrete.

Cross-sectional Survey for Prevalence Rate of Scoliosis in Primary, Middle and High School Boys in Pusan City (부산시내 남자 초.중.고등학생의 척추측만증 유병률 측정을 위한 단면조사)

  • A cross-sectional survey was conducted to determine the prevalence rate of scoliosis in elementary, middle and high school boys in May 1988. The study population included 1,393 male students in Pusan ; 463 students of 4th grade in two elementary schools, 543 students of 1st grade in one middle school and 387 students of 1st grade in one high school. The students of elementary school were screened with inspection by two physicians at the same time and the students of middle and high schools were screened with both inspection by the same physicians and 70mm chest X-ray. Positive students in any one of the two screening tests were measured for height and body weight and asked for the shoulder side on which he carries the school bag. The angle of curvature on X-ray film was measured by Cobb's method. The number of positive students in any one of the two screening tests were 15(3.2%) in elementary schools, 174(32.0%) in middle school and 92(23.8%) in high school. However, positive rates in both tests were only 2.2% for the middle school students and 2.6% for the high school students and among these students 1.1% out of total middle school students and 2.3% of high school students had a curvature equal to or greater than $5^{\circ}$ of Cobb's angle on 70mm chest X-ray film. There was a statistically significant association between the direction of spinal curve and the shoulder side on which one carries school bag among positive students in both screening 1.sts (p<0.05) Mean height and body weight of 281 positive students in any one of two screening tests were compared with the Korean standard for the same age. Mean weight of elementary school students was nearly the same as the standard weight but the height was slightly shorter than the standard. However, both mean height and weight of the middle school students were lower than the standard while those of the high school students were higher. The prevalence rate of scoliosis for the 2nd and 3rd grades of high school is presumed to be higher than that of the 1st grade of high school and the rate for girls will be even higher than the rate for boys of the same age. Thus, scoliosis seems to be an important school health problem. To prevent scoliosis, it is recommeded to reduce the weight of school bag, educate the students to keep a right posture and exercise periodically.

Pediatric Dehydration Assessment at Triage: Prospective Study on Refilling Time

  • Purpose: Dehydration is a paediatric medical emergency but there is no single standard parameter to evaluate it at the emergency department. Our aim was to evaluate the reliability and validity of capillary refilling time as a triage parameter to assess dehydration in children. Methods: This was a prospective pilot cohort study of children who presented to two paediatric emergency departments in Italy, with symptoms of dehydration. Reliability was assessed by comparing the triage nurse's measurements with those obtained by the physician. Validity was demonstrated by using 6 parameters suggestive of dehydration. Comparison between refilling time (RT) and a validated Clinical Dehydration Score (CDS) was also considered. The scale's discriminative ability was evaluated for the outcome of starting intravenous rehydration therapy by using a receiver operating characteristic (ROC) curve. Results: Participants were 242 children. All nurses found easy to elicit the RT after being trained. Interobserver reliability was fair, with a Cohen's kappa of 0.56 (95% confidence interval [CI], 0.41 to 0.70). There was a significant correlation between RT and weight loss percentage (r-squared=-0.27; 95% CI, -0.47 to -0.04). The scale's discriminative ability yielded an area under the ROC curve (AUC) of 0.65 (95% CI, 0.57 to 0.73). We found a similarity between RT AUC and CDS-scale AUC matching the two ROC curves. Conclusion: The study showed that RT represents a fast and handy tool to recognize dehydrated children who need a prompt rehydration and may be introduced in the triage line-up.

Identification of Limiting Amino Acids and Determination of Requirement of Total Sulfur-containing Amino Acids in a Low Protein Diet in Young Chicks. (어린병아리에서 저단백질사료내 제한아미노산의 규명과 함유황아미노산의 요구량 결정)

  • Since a 13% dietary protein level is generally accepted as a standard in evaluating net protein utilization values of protein sources in chicks, limiting amino acids a 13% protein basal diet containing 15% isolated soy-protein as the only source of dietary protein, were identified. Of such amino acids as methionine, lysine, threonine and tryptophan added to the basal diet singly or as a combination, methionine appeared as the only limiting amino acid for optimum growth of the chicks. When the requirement of total sulfur-containing acids (TSAA) was estimated as the point at which the dose-response curve intersected a line representing the plateau for maximum performance, the TSAA requirements for maximum growth and feed intake were 4.73% and 3.73% of dietary protein, respectively. The values, expressed in terms of TSAA intake, required for maximum weight gain, feed intake and gain/feed ratio were 167.1, 136.8 and 159.1 mg/bird/day, respectively.

  • In modern society, various digital devices are being distributed in a wide range of fields due to the fourth industrial revolution and the development of IoT technology. However, noise is generated in the process of acquiring or transmitting an image, and not only damages the information, but also affects the system, causing errors and incorrect operation. AWGN is a representative noise among image noise. As a method for removing noise, prior research has been conducted, and among them, AF, A-TMF, and MF are the representative methods. Existing filters have a disadvantage that smoothing occurs in areas with high frequency components because it is difficult to consider the characteristics of images. Therefore, the proposed algorithm calculates the standard deviation distribution to effectively eliminate noise even in the high frequency domain, and then calculates the final output by applying the probability density function weight of the Laplace distribution using the curve fitting method.
