This study was investigated to tile influences of clearcutting of timber harvesting on stream water quality at a natural deciduous forest catchment within the Seoul National University Research Forests in Mt. Baegun, Chunnam province during the periods of 1993 to 2001. Soil chemical ingredients and stream water qualities were monitored at 13 ha clearcutting site, non-treatment site and included in these catchment nearby. After the harvesting first and End years, the levels of total- N, and exchangeable ions ($K^+,\;Na^+,\;Ca^{2+},\;Mg^{2+}$) were decreased the values of before harvesting, and after the harvesting 111th years and eighth years, respectively. But the chemical characteristics of soil were not changed at all. pH of water in the harvesting area was 6.5 in stream water. Among the nutrient, Cd, Pb, Cu, and phosphate were not found, and the level of BOD reached at the level of the domestic use for drinking. Turbidity, odor, taste, $NH_4\;^+-N,\;NO_3\;^--N$, standard plate count, and coliform were also low enough to be used as the domestic use for drinking in the near villagers. Total amount of cation, total amount of anion and total amount of ion in clearcutting site, non- treatment site and included in these catchment nearby were higher than clearcutting site and non- treatment site. But, there was no great difference.
The effects of clearcutting on soil chemical ingredients and stream water quality have been investigated at a natural deciduous forest catchment within the Seoul National University Research Forest in Mt. Paekun, Chunnam province during the periods of 1993 to 1998. Soil chemical ingredients and stream water qualities were monitored at a 13 ha clearcutting site and a non-treatment site nearby. During the first and second years after harvesting, the levels of total-N, and exchangeable ions (K/sup +/, Na/sup +/, Ca/sup 2+/, Mg/sup 2+/) decreased compared to the values of before harvesting. During the fifth years after harvesting, these levels were significantly higher than those during the first and second years after harvesting. But the chemical characteristics of soil were not changed at all. pH of water in the harvesting area was 6.5 in stream water. Among the nutrients, Cd, Pb, Cu, and phosphate were not found, and the level of BOD reached at the level of the domestic use suitable for drinking. Turbidity, odor, taste, NH/sub 4//sup +/ -N, NO/sub 3//sup -/-N, standard plate count, and coliform were also low enough to be used as the domestic use for drinking by the near villagers. During the first and second years after harvesting, BOD increased to about 1 ppm. For that reason, the harvesting planning should be built in the harvesting area in consideration of the control of water quality in the stream.
This study was performed to investigate awareness of hand washing, hand washing behavior, and the levels of indicator microorganisms on hands of food handlers who work in the food court and cafeteria of a university campus. The three methods used were questionnaire survey by interview, direct observation in restrooms, and microbiological examination according to the Food Code of Korea. A positive attitude toward hand washing compliance was reported by the responded food handlers; however, improper hand washing and poor hand hygiene of the food handlers were recognized by the unnoticed direct observation. Significant differences were found between the questionnaire survey and the direct observation (p < 0.05) in hand washing compliance after using the toilet, duration of hand washing, use of hand washing agent, washing different parts of the hands, hand-drying method, temperature of water, and method of turning off the water. Samples taken from their hands before work showed higher level of standard plate count, total and fecal coliforms, and Escherichia coli than those taken after washing with water. After washing hands with antiseptic liquid soap, the bacterial populations including Staphylococcus aureus on hands were dramatically reduced. This study indicates that there is a remarkable difference between the food handlers' awareness of hand washing and their hand washing behavior. Poor hand washing compliance and hand hygiene were indicated by the positive results of total and fecal coliforms, E. coli, and S. aureus on hands of some food handlers. The findings of this study suggest that the hand hygiene of the food handlers need to be improved. More training/education on hand washing and hand hygiene of the food handlers should be necessary.
Methods for the measurement of aquatic bacteria can be divided into two groups. The first group of these methods is based on the 'replicon' concept that live bacterial cells, when diluted and transferred to a suitable medium, produce colonies. These methods distinguish living from dead bacteria, but they massively underestimate bacterial numbers. The second group of enumeration methods uses visual counting technique using specific apparatus such as a microscope. These methods are generally direct and simple, but it is very hard to distinguish between live and dead bacteria and between small particle and bacteria. Recently developed technique in staining methods has provided a reliable method of visual determination of aquatic bacteria. This uses epifluorescence microscopy to measure the total bacterial population. In order to present the fluorescence microscopy as a new methodology for the determination of bacterial numbers in water supplies, data were obtained from chlorine and monochloramine doses added to samples. Total counts by fluorescence microscopy were compared with standard plate count method. The total number of bacteria in water supplies can be determined with fluorescence microscopy. This technique allows better resolution of small bacteria and differentiation of particle from bacteria. Chloramine was found to persist longer in natural waters and prevent bacterial regrowth.
In order to investigate the bacteriological contamination of water in Han river, the survey was carried out in eight reservoirs of Seoul water supply during the period from January to December in 1985. 1. The counts by means of total bacteria in eight reservoirs by standard plate count method were as follows: $7.7\times10^2$ per ml in Paldang reservior, $9.6\times10^3$ per ml in Gueiri, $8.4\times10^4$ per ml in Doogdo, $1.6\times10^6$ per ml in Bogwang, $2.5\times10^6$ per ml in Noryangjin, $2.2\times10^6$per ml in Seon yoo, $5.9\times10^6$ per ml in Yungdeungpo and $1.9\times10^7$per ml in Gayang. 2. The average counts of total coliform in eight reservoirs by MPN method were as follows : $2.4\times10$ per 100 ml in Paldang, $5.6\times10^2$ per 100 ml in Gueiri, $2.3\times10^3$ per 100 ml in Doogdo, $5.1\times10^4$ per 100 ml in Noryang-jin, $1.2\times10^5$ per 100 ml in Bogwang, $6.2\times10^4$ per 100 ml in Seonyoo, $1.1\times10^5$ per 100 ml in Yungdeungpo and $2.8\times10^5$ per 100 ml Gayang. 3. The counts by means of fecal coliform in eight reservoirs by MPN method were as follows : non detection per 100 ml in Paldang, 5.2 per 100 ml in Gueiri, $1.2\times10^2$ per 100 ml in Doogdo, $1.6\times10^3$ per 100ml in Bogwang, $2.0\times10^3$per 100ml in Noryangjin, $6.6\times10^2$ per 100ml in Seonyoo, $1.2\times10^3$ per 100 ml in Yungdeungpo and $2.5\times10^3$per 100 ml in Gayang. 4. The counts by means of fecal streptococci in eight reservoirs by MPN method were as follows: non detection per 100 ml in Paldang and Gueiri, $6.9\times10$ per 100 ml in Doogdo, $3.2\times10^2$ 102 per 100 ml in Bogwang, $2.9\times10^2$ per 100 ml in Noryangjin, $3.0\times10^2$ per 100 ml in Seonyoo, $4.0\times10^2$ per 100 ml in Yungdeungpo and $14\times10^3$ per 100 ml in Gayang. 5. The counts means of pseudomonas aeruginosa in eight reservoirs by MPN method were as follows; non detection per 100 ml in Paldang, 2.4 per 100 ml in Gueiri, $1.5\times10$ per 100 ml in Doogdo, $2.0\times10$ per 100ml in Bogwang,$6.2\times10$ per 100mI in Noryangjin, $2.1\times10$ per 100ml in Seonyoo, $6.4\times10$ per 100mI in Yungdeungpo and $7.1\times10$ per 100ml in Gaynag.
This study identified risk factors of cross-contamination of foodborne pathogens and established a good agricultural practice (GAP) system for an agricultural products processing center (APC) for perilla leaves. All samples were collected before and after a standard work shift at the APC, while perilla leaves were also collected after each step in the APC. In addition, the workers and their surroundings were sampled by swabbing. The total plate count (TPC) and coliform count in the water samples increased significantly (p<0.05) to 3.36 and 1.73 log CFU/mL after work, respectively. However, no Escherichia coli and Listeria monocytogenes were detected. The bacterial populations of the workers and their surroundings did not differ significantly (p${\geq}$0.05) before and after work. However, Staphylococcus aureus (<1.66 log CFU) was detected at a high rate (13-50%) in the basket, packing table, gloves and cloth. Although perilla leaves passed through the washing steps, the TPC and coliform bacterial populations on the final products were higher (p${\geq}$0.05) than those of unwashed perilla leaves, which indicates that the washing system was not functioning properly. Accordingly, a GAP system with a better washing system should be employed at this facility.
This experiment was conducted to evaluate the parameters such as nutrient requirements, POY, AnV, Totox, VBN, total plate count, dietary fatty acids and amino acids composition, that are not included in the registered standard composition items required by the Ministry of Agriculture and Forestry, of a moist pellet (MP), three domestic extruded pellets (DEP-1, DEP-2, DEP-3), and two foreign extruded pellets (FEP-1, FEP-2) that are utilized by domestic flounder farms at present. The crude protein was added in excess of the dietary protein requirement in 6 kinds of feeds. When considering the proper PH ratio, it is obvious that protein was added in excess, especially in MP and FEP-2. Crude fat was also added in excess, especially in FEP-1. MP contained a higher dietary phosphorus content than formulated feeds, surpassing the dietary phosphorus requirement and greatly increasing the possibility for causing water pollution. The oxidation of fatty acid and decomposition of protein in MP were higher than in formulated feeds, and may also cause problems on fish farms. Also, it is difficult to store and manage MP, Among the fatty acids, EPA and DHA contents in MP were higher than those in formulated feeds. It is necessary to conduct further studies of EPA and DHA contents in formulated feeds. Lysine content in MP and FEP-2 could meet the dietary lysine requirement of flounder, however, the possibility of insufficient lysine content in the other formulated feeds was high and we considered that extra supplementation was necessary. Therefore, it is necessary to set up quality control standards according to fish species and sizes while considering the specific character of aquatic formulated feeds to restore the confidence of feed companies and aquaculturists to these feeds. This may be an opportunity to make an earlier change from MP to formulated feeds.
