Suppression of Bacterial Wilt with Fuorescent Pseudomonads, TS3-7 strain (Fluorescent siderophore 생산균주, TS3-7에 의한 풋마름병 발병 억제)

  • Among the root colonizing and plant growth promoting bacteria isolated from the bacterial wilt suppressive soil, five strains were detected to produce siderophores by CAS agar assay. The most effective isolate, TS3-7 strain induced significant suppression of bacterial wilt disease in tomato and pepper plants. Seed treatment followed by soil drench application with this strain resulted in over 80% reduction of bacterial wilt disease compared with the control. Significant disease suppression by TS3-7 strain was related to the production of siderophore. Besides iron competition, induction of resistance of the host plant with siderophore was suggested to be another mode of action that suppress bacterial wilt, based on the lack of direct antibiosis against pathogen in vitro. According to Bergey's Manual of Systemic Bacteriology and 16S rDNA sequence data, TS3-7 stain was identified as Pseudomonas sp. TS3-7.

Static measurement of magnetostriction of FeCoGe/phenol composites (FeCoGe/페놀 복합체의 정적 자기변형 측정)

  • The magnetostriction of FeCoGe/phenol composites, which is one of the magnetostrictive materials, measured at the external magnetic field. The measurement was carried out using the electrical-resistance strain gage, the wheaten's Bridge for eliminating the unnecessary voltage, and the lock-in-amp for signal amplification and noise filtering. When the external magnetic field was applied in the longitudinal the samples, the maximum strain of 120ppm was taken with regard to the 10wt.% phenol composite. This results indicate that the FeCoGe/phenol composites can be useful as an actuator because it has larger stain than the other solid state actuators such as piezo electric materials.

  • To make a practical application of a micro-strain sensors with ultrahigh sensitivity, a strain on electrical properties of micro-patterned amorphous (Fe\ulcornerco\ulcorner)\ulcornerSi\ulcornerB\ulcorner films had influenced on the impedance over frequency range from 1 MHz to 1 GHz. Reflecting excellent magnetomachanical couping properties of films, high frequency impedance was subject to change sensitively by a strain : a change in impe-dance of 39% was observed at 200 MHz applied a strain of 224$\times$10\ulcorner. To determine a optimum shape of micro-patterned films, film impedance was analyzed by virtue of its constitutive components of resis-tance and reactance. Result was shown that reduction of the resistance term(hence increase of resultant reactance term) of impedance is more effective for enhancing the strain sensitivity of films at relatively low frequency range.

A Comparative Analysis on The Efficiency of Various Clinical Methods for Diagnosis of Tuberculosis (결핵 진단을 위한 검사 방법간의 효율성에 관한 비교 분석)

  • In recent years continuously increasing number of tuberculosis (TB) cases due to the emergence of strains with multidrug resistance and AIDS is a significant global health problem. Therefore, more rapid and reliable diagnosis of TB may be one of the most urgent needs in efforts to eradicate the disease. The present study was designed to compare and assess the diagnostic values and efficiencies between the conventional methods (X-ray, AFB stain and culture) and PCR for pulmonary TB on 171 cases. Chest X-ray finding and clinical features revealed that 39 (22.8%) of 171 sputum specimens were pulmonary TB cases. The statistical data were taken on the basis of the definitive diagnosis: In X-ray, overall sensitivity, specificity, efficiency and false positive and false negative incidence was respectively 69.2%, 87.1%, 83.0%, 12.9%, and 30.8%; 79.5%, 95.5%, 91.8%,4.6% and 20.5% in AFB-stain; 56.4%, 99.2%,89.5%, 0.8% and 43.6% in culture; 82.1%, 96.2%, 93.0%, 3.8% and 17.9% in PCR. PCR got a highest sensitivity and efficiency as well as a lowest false negative incidence. Culture had a highest specificity with a lowest false positive incidence. These results imply that PCR assay is fast, sensitive and efficient method for diagnosis of pulmonary TB. However, combined use of the conventional methods with thorough quality control may offer more opportunities for detecting Mycobacterium tuberculosis and diagnosting TB although they have some limits.

Synthesis and Properties of Photo-curable Biomass-based Urethane Acrylate Oligomers (광경화형 바이오매스계 우레탄 아크릴레이트 올리고머의 합성 및 물성 연구)

  • Generally, solvent-type coatings generate a large amount of volatile organic chemicals(VOC), which are carcinogenic substances, in the manufacturing process, and their use is regulated due to environmental problems. There is also the problem of resource depletion due to limited fossil fuels. Therefore, in this study, UV-curable urethane acrylate oligomers were synthesized with different contents of isosorbide, which is a biomass material, and proceeded to evaluate the physical properties of coatings. As the isosorbide contents increased, the viscosity, glass transition temperature, tensile strength, stain resistance, and pencil hardness increased, but elongation and flexibility decreased, and BOI-3 showed the best adhesion. The isosorbide content of the oligomer fixed at 20%, UV-curable urethane acrylate oligomer was synthesized according to the content ratio of polycaprolactone diol(PCL) and Ecoprol H1000(Ecoprol). As the PCL/Ecoprol content ratio increased, the glass transition temperature, elongation, and flexibility increased, but the tensile strength and pencil hardness decreased. It was confirmed that the adhesion and stain resistance increased by improving the surface bonding strength of PCL. All films of oligomers synthesized were transparent without discoloration.

Suppression of Bacterial Wilt with Bacillus subtilis SKU48-2 Strain (Bacillus subtilis SKU48-2에 의한 풋마름병 발병 억제)

  • Bacterial populations from the rhizosphere were obtained and the efficacy of the bacterial wilt suppression, root colonizing ability and resistance to three kinds of chemical pesticides were assayed. According to these results, SKU48-2 was selected as a potential biological agent to control the bacterial wilt caused by Ralstonia solanacearum. SKU48-2 strain at $10^8CFU/ml$ inoculum was able to suppress the bacterial wilt up to 60% in greenhouse trials. Also, the resistance of SKU48-2 to chemical pesticides make possible to use in combination with chemical pesticides for the control of bacterial wilt. Three different powder formulations of SKU48-2 were developed. The shelf-life of powder formulations was effective up to 6 months of storage. Unformulated bacterial suspension could not be stored for 2 weeks, at which time cell viability was completely lost. According to 16S rDNA sequence data, the SKU48-2 stain was identified as Bacillus subtilis.

Anti-proliferative Effect of Paclitaxel in Multicellular Layers of Human Cancer Cells (다층 배양된 암세포에서 파크리탁셀의 항증식효과 분석)

  • Human solid tumors exhibit a multicellular resistance (MCR) resulting from limited drug penetration and decreased sensitivity of tumor cells when interacting with their microenvironments. Multicellular cultures represent solid tumor condition in vivo and provide clinically relevant data. There is little data on antitumor effect of paclitaxel (PTX) in multicellular cultures although its MCR has been demonstrated. In the present study, we evaluated antiproliferative effects of PTX in multicellular layers (MCL) of DLD-1 human colorectal carcinoma cells. BrdU labeling index (LI), thickness of MCL, cell cycle distribution and cellular uptake of calcein were measured before and after exposure to PTX at 0.1 to 50 ${\mu}M$ for 24, 48 and 72 hrs. BrdU LI and thickness of MCL showed a concentration- and time-dependent decrease and the changes in both parameters were similar, i.e., 34.2% and 40.6% decrease in BrdU LI and thickness, respectively, when exposed to $50\;{\mu}M$ for 72 hr. The DLD-1 cells grown in MCL showed increase in $%G_{0}/G_{1}$ and resistance to cell cycle arrest and apoptosis compared to monolayers. Calcein uptake in MCL did not change upon PTX exposure, indicating technical problems in multicellular system. Overall, these data indicate that antitumor activity of PTX may be limited in human solid tumors (a multicellular system) and MCL may be an appropriate model to study further pharmacodynamics of PTX.

Helicobacter pylori Vacuolating Toxin Exhibits Polar Activity of $Cl^-$ Secretion and Secretory Response to Carbachol in T84 Cells

  • To investigate whether VacA (vacuolating toxin) produced by Helicobacter pylori Korean stain 99 induces intestinal secretion, purified VacA was added to T84 cell monolayers mounted in Ussing chambers, and electrical parameters were monitored. Mucosal addition of low pH-pretreated VacA increased short circuit current (Isc). The effect was time- and dose-dependent and saturable. The time-to-peak Isc was concentration-dependent. Chloride channel inhibitors, niflumic acid or 5-nitro-2-(3-phenylpropylamino)-benzoate (NPPB), inhibited VacA-stimulated Isc. Carbachol (CCh)-induced increase of Isc was prolonged by the addition of VacA to the mucosal side only. The effect was unaltered by the addition of niflumic acid. VacA did not show cytopathic effects. These studies indicate that VacA is a nonlethal toxin that acts in a polar manner on T84 monolayers to potentiate $Cl^-$ secretion and the response to CCh secretion without decrease in monolayer resistance. VacA may contribute to diarrhea diseases in human intestinal epithelial cells.

  • Over the past years, a large number of acrylate polymers have been developed and the overcoat thin layer containing acrylate polymers have been used for TFT-LCD color filter. As forming thin layer using acrylate polymers, the existing acrylate polymers have some problems such as low hardness by low Tg temperature, coating uniformity and solubility in organic solvent. To solve these problems, we synthesized new polymer(Scheme.), containing olefin monomer, which has high Tg temperature, good coating uniformity and good solubility in organic solvent. The overcoat thin layer containing new polymer resulted in good coating uniformity, stain, spot, scratch, heat resistance, DOP(Degree Of Planarization) on RGB glass, transparency, hardness, adhesion, anti-chemicals(anti-acid, anti-base, anti-organic solvent), insulation and anti-humidity. Scheme. The structure of new polymer X = Olefin monomer contains ketone, ester, hydroxy, ether, halogen, nitrile, alkoxy, phenyl functional group $R_1$ and $R_2$= H or $CH_3$. Ratio=0<[1/(1+m+n)]<0.7,0.1[$\leq$[n/(1+m+n)]<0.5.

INDENTIFICATION AND ANTIBIOTIC SUSCEPTIBILITY TEST OF MICROORGANISMS ISOLATED FROM INFECTED ROOT CANALS (감염근관에서 분리한 세균의 동정 및 항생제 감수성 검사)

  • Bacteria have been regarded as major etiolgic factors in root canal infections. Infected root canal flora from thirteen patients who had visited to conservative department of Wonkwang dental hospital were cultured on blood agar plates. Cultued microorganisms were isolated and identified with Gram stain and biochemical tests using Vitek Systems(BioMeriux, MO, USA); Antibiotic susceptibillity was performed with disk diffusion and broth microdilution using Vitek Systems. Gram positive cocci(65 %) were predominant, which were composed of 6 Streptococcus viridans group, 5 Staph. spp., and 4 Enterococcus faecium, in the isolatd 23 strains. Gram negative rods (26 %) were the next common bacteria, which were composed of 5 non - fermentative Gram negative rods, and 1 Enterobacter cloacae. Most strains of S. viridans group and E. faecium were susceptible to antibiotics including penicillin. But strains of Staphylococcus spp. and non - fermentative Gram negative rods showed marked resistance to antibiotics except tetrancyclin and cefotaxime. Most results between disk diffusion and microdilution were all agreed, but the results of non - fermentative Gram negative rods were susceptible to cefotaxime in disk diffusion method but resistant in microdilution.
