The Extension of Tofu Shelf-Life with Water-Soluble Degraded Chitosan as Immersion Solution (수용성 키토산분해물질을 침지액으로 이용한 두부의 저장성 증대)

  • For the effect of water-soluble degraded chitosan on the shelf-life of tofu, sterilized distilled water, 0.5% degraded chitosan, 0.5% fumaric acid and 0.5% lactic acid used as an tofu-immersion solutions were investigated by microbial counts, pH, and turbidity during the periods of storage at $4^{\circ}C$. After 2 weeks storage, total aerobic microbial counts in tap water and sterilized distilled water used as an immersion solution were $3.8\;{\times}\;10^8$ and $1.8\;{\times}\;10^8\;CFU/mL$, respectively. In 0.5% fumaric acid and 0.5% lactic acid immersion solutions, the microbial counts were around $10^7\;CFU/mL$ after 2 weeks while the microbial population in 0.5% water-soluble degraded chitosan were, however, $1.6\;{\times}\;10^5\;CFU/mL$ after 2 weeks and $1.7{\times}10^7\;CFU/mL$ after 3 weeks. The lag phase of initial contaminated microbes in 0.5% degraded chitosan solution was longer than those of other treatments. The addition of 0.5% fumaric acid and 0.5% lactic acid decreased the initial pH to pH 5.0, while those of tap water, sterilized distilled water and 0.5% degraded chitosan stabilized the immersion solution at around pH 7.2. All initial pH values were decreased during storage and then slowly increased as storage time was increased. The turbidities in all treatments were increased during storage, but the addition of 0.5% degraded chitosan showed the lowest change, compared to other treatments, showing that the water-soluble degraded chitosan has a good antimicrobial effect and has a potential use to extend the shelf-life of tofu product.

Dietary Supplementation of Resveratrol and Methoxylated Resveratrol Affects on Chicken Thigh Meat Quality (Resveratrol과 Methoxylated Resveratrol이 닭 다리육의 저장 품질에 미치는 영향)

  • This study was carried out to determine the effect of dietary supplementation of resveratrol and methoxylated resveratrol extracted from branch of Morus alba L. on the quality of chicken thigh meat during cold storage. For 35 days, 1-day-old 320 broiler chicks (Ross) were divided into 8 groups and supplemented the diet; basal diet only (BD), BD with antibiotics (AB), vitamin E 20 IU (VE 20), vitamin E 200 IU (VE 200), resveratrol 20 ppm (RV 20), resveratrol 200 ppm (RV 200), methoxylated resveratrol 20 ppm (MR 20), and methoxylated resveratrol 200 ppm (MR 200). After slaughtering the broilers, thighs were collected and analyzed the quality change of the meat during storage at $4^{\circ}C$ for 5 days. The meat quality factors such as pH, color, water holding capacity, and sensory characteristics of thigh meat were determined on the experiment day 1, 3, and 5. AB, VE, and MR increased pH value of chicken thigh compare to BD (p<0.05). Lightness ($L^*$) showed no significant difference during storage day 1 and 5. VE 200 and MR 20 stabilized the redness ($a^*$) of chicken thigh as the value of day 1. Water holding capacity of chicken thigh from VE 20, RV 200, MR 20, and MR 200 on storage day 3 was higher than that of BD (p<0.05). In sensory evaluation, the panelist discriminated the tenderness and gave the higher score on the chicken from AB, VE20, RV 20, and MR 20 compare to BD (p<0.05). These results suggest that the dietary resveratrol and methoxylated resveratrol could be used as chicken meat quality enhancer in broiler industry.

  • Laver io one of the most necessary and seasonal items in Korean food from oldtimes. Laver is lagely eaten in dried form, and its supply depends entirely upon culture weeds. The history of laver culture in Korea about sixty or seventy years is older than in Japan. Significance of laver culture is divided into two aspects, one is food supply in the nation, and the other is export to other countries. Houses engaged in laver culture are about foully thousands, and laver production in 1972 is estimated as 1, 3 bitten sheets. (1 sheet is a dried laver of 20 cm sq, in the shape of paper) Especcially meaning of layer production is the concentration of labour input, and systematic management of labour. From around 1920, the method of laver culture was introduced by Japanese Imperialism for mono culture in shallow seas, and mass products of laver is provided to Japan market, DOMESTIC MARKET Fundamental consume function calculates at below, $D_{(68_71)}$=16354 $Y^{0.471}$ $P^{-1.0662}$ where D is total layer demand, Y income variable, P price variable. It means income elasticity is 476. in the whole country, and price elasticity is 1, 07. But generally income elasticity is higher in urban area than in rural area, as shown at 1, 3 in Seoul city. But the recent improvement of laver culture techniques, the production of seeds and freezing storage of seeds has been stabilized. Futher new culture farms have been developed by means of break- water fences or by floating culture method. These improvements have been backed up with increased demand of laver products. Import quantity and price of Korean laver products are restrained by three organizations, that is producer, distributor and consumer. This relationship calculated by regression equation shows that import is influenced only producer organization, at the sacrifice of consumer profit. For increase to export of laver products, we urgently require to open foreign trade of laver products for Japanese consumer, .and Japan has political responsibility to solve Korean laver structure. But with long run timeseries, as regards Japanese production and import quantity, importing function shows increasing trend as below, 250 million sheets <3, 947.1674+0.005 $L_{g}$ >) 600 million sheets where $L_{q}$ is relative production quantity of laver in Japan. Operation of High Performance Elutriation-Type Sludge Fermenter and Feasibility for Its Application (고성능 세정식 슬러지 산발효조의 운전 및 적용성 평가)

  • The performance of a novel fermentation process, adopting a sludge blanket type configuration for higher hydrolysis/acidogenesis of the municipal primary sludge, was investigated under batch and semi-continuous conditions with various pH and temperature conditions. This acid elutriation slurry reactor provided higher system performance with a short HRT (5 days) and higher acidogenic effluent quality under pH 9 and thermophilic ($55^{\circ}C$) conditions. The hydrolysis of the sludge was revealed to be significantly dependent on seasonal effects for sludge characteristics but with little impact on acidogenesis. Based on the rainy season at the optimum conditions, VFA production and recovery fraction ($VFA_{COD}/COD$) were $0.18\;g\;VFA_{COD}\;g^{-1}\;VSS_{COD}$ and 63%. As byproducts, nitrogen and phosphorus releasing were $0.006\;g\;N\;g^{-1}\;VSS_{COD}$ and $0.003\;g\;P\;g^{-1}\;VSS_{COD}$, respectively. For the mass balance in a full-scale plant($Q=158,880\;m^3\;day^{-1}$) based on the rainy season, the VFA and non-VFA(as COD) production were $3,110\;kg\;VFA_{COD}\;day^{-1}$ and $1,800\;kg\;COD\;day^{-1}$, resulting in an increase of organics of $31\;mg\;COD\;L^{-1}$ and $20\;mg\;VFA_{COD}\;L^{-1}$ and nutrients of $0.7\;mg\;N\;L^{-1}$ and $0.3\;mg\;P\;L^{-1}$ in the influent sewage. The economical benefit from this process application was estimated to be about $67 per $1,000m^3$ of sewage except for energy requirements and also, better benefits can be expected during the dry season. Also, the results revealed that the process has various additional advantages such as pathogen-free stabilized solids production, excellent solids control and economical benefits.

Evaluation of Basic Oxygen Furnace Slag as Soil Conditioner in the Rice Paddy Field (논토양 벼재배에서 제강슬래그의 토양개량제로서의 시용 효과)

  • An experiment was conducted to evaluate the possibility of using basic oxygen furnace (BOF) slag as a soil conditioner in rice paddy field. In 1997, rice (Oryza sativa B. cv. Dongjinbyeo) crop was cultivated under different application rates of BOF slag at three different places, Inandong Sunchon city, Youjunglee Bosung province, and Nampyung Najoo city. In each paddy field, five treatments, four application rate of BOF slag (0, 4, 8, $12Mg\;ha^{-1}$) and one application rate of lime ($2Mg\;ha^{-1}$) were tried with three replications. Plant height, number of tillers per hill, leaf area per hill, leaf dry weight, calm dry weight or shoot dry weight per hill were measured five times at the interval of seven days. Chemical contents of rice plants and soil were also measured at the same sampling date Yield were estimated by harvesting $6.6m^2$ per experimental unit and yield components were measured by sampling 10 plants per experimental unit at the harvest date. Application of BOF slag hardly affected contents of soil organic matter, available phosphate and potassium in soil. Soil pH and contents of Ca, Mg, Fe and $SiO_2$ enhanced as BOF slag rate increased. Enhancement of soil pH by ROF slag treatment appeared to be closely related with increase in soil Ca content. Application tate of $2Mg\;ha^{-1}$ of lime showed almost the same effect, in increase of soil Ca content as application rate of $4{\sim}8Mg\;ha^{-1}$ of BOF slag, Fe content in soil decreased sharply as time passed after slag treatment and stabilized more or less at the later sampling date. Contents of inorganic matter in plant such as total nitrogen, phosphate, potassium and Mg were not affected by BOF slag treatment. However, contents of Ca, Fe, and $SiO_2$ in plants increased as slag rate became higher. The growth of rice plants with BOF slag treatment was more or less slower but continued persistently up to the later growth stage, so that growth of plants with BOF slag treatment was almost the same nr even greater than that of control or lime treatment. However, BOF slag rate of $12Mg\;ha^{-1}$ seemed to be too high because all the measurements of plant, growth at this rate showed lower values than those of other treatments at all the sampling dates. Treatments of BOF slag $4Mg\;ha^{-1}$ or $8Mg\;ha^{-1}$ showed higher rough rice yield than other treatments, so that the optimum BOF slag ratein rice paddy field seemed to be in the rage of $4{\sim}8Mg\;ha^{-1}$.

The Effect of Circle Lens and Soft Contact Lens with Identical Material in Clinical Application on the Eyes (써클 및 동일 재질 콘택트렌즈의 임상 적용시 눈에 미치는 영향)

  • Purpose: The present study was conducted to investigate the actual condition of circle lens wear and the differences caused by the pigmentation in blinking rate, non-invasive tear break-up time (NIBUT), and lens surface between circle and soft contact lens wear during clinical application. Methods: Eighty subjects were surveyed to know the actual condition of wearing circle lens. Blink rate, NIBUT of 20 subjects in twenties were measured after separately wearing soft contact lens and circle lens for 5 days. Their lens surfaces were also observed by scanning electron microscope. Results: Fifty percent of circle lens wearers answered that major reason for changing circle lens was discomfort and 67% of answerers wore circle lens more than 6 hours a day. The tendency of increased blink rates in both wearers of circle and soft contact lens at 30 minutes later when tear film stabilized and 3 hour after lens wearing was shown on the 1st, 3rd and 5th day with expanding wearing time. Especially at 3 hours after lens wearing on the 3rd and 5th day, the difference of blink rates was statistically different in circle lens wear and soft contact lens wear. The NIBUTs of soft contact lens wearers and circle lens wearers were 6.0 and 3.7 secs, respectively, at 30 min later on 1st day and were significantly different. NIBUTs of soft contact lens wearers and circle lens wearers were also statistically different 3 hrs after lens wearing on the 1st day and the similar pattern of NIBUTs was shown on the 3rd and 5th day. The roughness of both surfaces in soft contact lens and front surface in circle lens was same. However, pigmented front surface of circle lens was rougher and uneven. Conclusions: As the results, the circle lens wearers might feel discomfort in clinical application since the difference in lens surface of circle lens would change lens wettability during wearing. The difference by tinting contact lens was shown from the result, which could help better wearing circle lens.

A Study on the Stability Test for the Cream Containing Suaeda Asparagoides Extract (나문재 추출물 함유 크림의 안정성 평가에 관한 연구)

  • In the previous study, the anti-oxidant activity of extract/fraction of Sueada asparagoides (SA) was investigated and the results showed that the ethylacetate (EtOAc) fraction and its aglycone fraction had the best performance on the free radical scavenging activity, reactive oxygen species scavenging (ROS) activity and cell protective activity (J. Soc. Cosme. Scientists Korea, 33(3), 145 (2007)). In this study, the stability of cream containing 0.3% SA EtOAc extract (called extract below) was evaluated. pH, viscosity and absorbance (363 nm) were measured under the 4 different temperatures ($0^{\circ}C,\;25{\circ}C,\;37{\circ}C\;and\;45{\circ}C$) and under the sun light at the 4 week intervals during the 12 weeks in total. The control cream without containing the extract did not show pH change under the different temperatures mentioned above. However, the pH of the cream the extract was decreased 0.08 at the temperature ranges of $0^{\circ}C\;to\;37^{\circ}C$. Under the $45^{\circ}C$ and sun light condition, the pH was decreased 0.51 and 0.66, respectively. The cream containing the extract did not show absorbance change at the temperature ranges of 0 to $37^{\circ}C$ for 12 weeks. Instead, the absorbance of the cream treated under $45^{\circ}C$ and sun light condition was decreased 7.6 % and 7.4 %, respectively. This decrease in absorbance is relatively small compared to the 48.3 % decrease of the extract sampled from the cream using ethanol solution. This indicates that the extract is stabilized in the cream. After treating the cream for 12 weeks under the different temperatures, the viscosity was measured for the cream containing the extract and control cream. The values were increased by 1,748 cPs in average compared to the initial value for the former and by 951 cPs in average for the latter. On the other hand, the viscosity of control cream treated under the sun light for 12 weeks was significantly decreased (4,022 cPs) relative to the cream containing the extract, which showed 2,484 cPs increase in viscosity. This indicates that the SA extract contributes to the stability of the emulsion product by protective effect to maintain the viscosity of the cream against sun light. In addition, any change in color or smell was not observed through 12 weeks of the experimental time period. Thus, it is concluded that it is still not clear in the stability of the cream containing the extract when it is stored for the long time. Accordingly, it is suggested that further study is needed to provide more information to the manufactures, who are seeking for the application of the extract to improve the anti-oxidant activity and stability of cosmetic products.

Inhibition of Type II Diabetes in ob/ob Mice and Enhancement of Mitochodrial Biogenesis in C2C12 Myotubes by Korean Mistletoe Extract (한국산 겨우살이 추출물(KME)의 2형 당뇨 억제 및 근육세포 미토콘드리아 생성 증가 효과)

  • In this study, the anti-diabetic activity of a cold water extract of Korean mistletoe (KME) was investigated in C57BL/6J Lep ob (ob/ob) mice. Oral administration of KME (50 or 100 mg/kg/d) significantly inhibited the level of blood glucose of ob/ob mice after 5 days from the beginning of KME treatment. And the anti-diabetic effect of KME was stabilized 10 days after oral administration, showing a substantial reduction of blood glucose levels by more than 20% as compared with control mice. The results of oral glucose tolerance test (OGTT) revealed that oral administration of KME gave rise to a remarkable improvement in overall glucose response. Oral administration of KME in ob/ob diabetic mice also significantly reduced blood total cholesterol (TCHO) and triglyceride (TG) levels compared with the diabetic control mice. Moreover, in an in vitro experiment using C2C12 myotubes, treatment of KME prominently increased glucose uptake. Interestingly, KME significantly increased the expression of peroxisome proliferator-activated receptor gamma coactivator 1-${\alpha}$ ($PGC-1{\alpha}$), a head regulator of mitochondrial biogenesis and oxidative metabolism, and $PGC-1{\alpha}$-associated genes such as glucose transporter type 4 (GLUT4), estrogen-related receptor-${\alpha}$ ($ERR-{\alpha}$), nuclear respiratory factor-1 (NRF-1), and mitochondrial transcription factor A (TmfA) in C2C12 cells. These results suggest that KME has potential as a novel therapeutic agent for diabetes, and its anti-diabetic activity may be related to the regulation of mitochondrial biogenesis.

Annealing Cycle Dependence of MR Properties for Free Layer in $Ni_{25}Mn_{75}-Spin$ Valve Films ($Ni_{25}Mn_{75}-Spin$ Valve 박막 자유층의 열처리 순환수에 따른 자기저항 특성)

  • Annealing cycle number and nonmagnetic layer thickness dependences of interlayer coupling field ( $H_{inf}$ ) and coercivity ( $H_{cf}$ ) of free magnetic layer on NiMn alloy-spin valve films (SVF) were investigated. The SVF is Glass (7059)/N $i_{81}$F $e_{l9}$(70 $\AA$)/Co(10 $\AA$)/Cu(t $\AA$)/Co(15 $\AA$)N $i_{81}$$Fe_{19}$(35 $\AA$)/N $i_{25}$M $n_{75}$(250 $\AA$)Ta(50 $\AA$) films, it were fabricated using the dc sputtering method at different pinning layer thickness and nonmagnetic spacer thickness (Cu thickness; 30 $\AA$, 35 $\AA$, 40 $\AA$) of NiMn alloy with 25 at.%. Ni In case that Cu thickness of SVF is 35 $\AA$ and peak exchange coupling field ( $H_{ex}$) was 620 Oe, while coercivity $H_{c}$ = 280 Oe and MR ratio showed 2.5%. As for $H_{inf}$ and $H_{cf}$ , every SVF increased up to the stabilized values with the increase of annealing cycle number 15, which were $H_{inf}$ of 120 Oe and $H_{cf}$ of 75 Oe. The increase of $H_{cf}$ with the annealing cycle number seems to be caused by the effective reduction of Cu layer thickness due to the increase of interfacial mixing of Cu layer and Co layer. In addition, the $H_{inf}$ and $H_{cf}$ dependences of free NiFe layer by the interfacial mixing effect were appeared the different aspects when Cu layer becomes more thinner and thicker than Cu layer thickness of 35 $\AA$, respectively.ively....

Stabilization of Heavy Metal Contaminated Paddy Soils near Abandoned Mine with Steel Slag and CaO (제강슬래그와 CaO를 이용한 폐광산 주변 중금속 오염 농경지 토양의 안정화 처리 연구)

  • Applicability of CaO and steel slag as stabilizers in the treatment of field and paddy soils near Pungjeong mine contaminated with arsenic and cationic heavy metals was investigated from batch and column experiments. Immobilization of heavy metals was evaluated by TCLP dissolution test. Immobility of heavy metal ions was less than 15% when steel slag alone was used. This result suggests that $Fe_2O_3$ and $SiO_2$, known as the major component of steel slag, have little effect for the immobilization of heavy metal ions due to acidity of TCLP solution. Immobilization of cationic heavy metals was little affected by the ratio of CaO and steel slag while arsenic removal was increased as the ratio of steel slag to CaO increased. In the column test, concentrations of both arsenic and cationic heavy metals in effluents were below the water discharge guideline over the entire reaction period. This result can be explained by the immobilization of cationic heavy metals from the increased pH in soil solution as well as by the formation of insoluble $Ca_3(AsO_4)_2$. From this work, it is possible to suggest that arsenic and cationic heavy metals can be concurrently stabilized by application of both CaO and steel slag.
