Solid Dispersion of an HIV Protease Inhibitor

  • LB71350 is an HIV protease inhibitor with poor aqueous solubility and extensive first pass effect. The purpose of the present study was to test the feasibility of solid dosage form of LB71350 with improved bioavailability utilizing solid dispersion. Three different compositions with varying ratio of (LB71350: Gelucire 44/14: Tween 20) were studied. Capsule filling of these solid dispersion compositions was tested using a semi-automatic capsule filling system. Oral bioavailability in dog was tested. Chemical and physical stability at 4, 25 and $40^{\circ}C$ was monitored by HPLC assay, dissolution test, powder XRD and microscopy. The capsule filling system yielded uniform products of drug loading up to 10%. Oral bioavailability in dog was improved compared to the aqueous suspension of crystalline LB71350. Capsules were chemically stable for up to 6 months at $40^{\circ}C$. However, there were temperature and composition dependent physical changes. Decrease in dissolution rates after storage at $40^{\circ}C$ was due to the polymorphic change. In conclusion, manufacturing process, bioavailability, and physico-chemical stability have been considered to propose a solid dispersion capsule formulation for the HIV protease inhibitor with poor physico-chemical properties. A new less soluble crystalline form identified during the physical stability test warrants further study.

Flame Stability and NOx Formation by Micro scale Turbulence (마이크로 스케일 난류에 의한 화염안정성 및 NOx 생성)

  • The effect of micro scale turbulence on flame structure and stability were experimentally investigated by changing the area of micro turbulence generator(MTG) and air velocity in terms of low NOx and high efficiency combustion. NOx and CO concentration were also measured for different MTG areas to investigate whether a vane swirler having MTG has a possibility of using as part for low NOx combustor. From the obtained results, it is shown that flame stability region increases and flame size becomes small as MTG area increases since MTG in itself makes small scale recirculation flow and swirler does large scale recirculation one. It is also shown that low NOx concentration(about 20${\sim}$30ppm@$O_2$ 11%) is achieved for all MTG areas without any increase in CO concentration regardless of air velocity range tested in this study when the equivalence ratio is 0.7. The results obtained in this study can give basic guideline for the design of compact low NOx high efficiency combustor using a vane swirler having MTG.

  • In this study, Standards of Lighting Appliance and Standard of Model Approval and Inspection Technology for EXIT LIGHT are compared so as to analyze stability of a exit light which is fire product. Test items, which are not included in Standard of Model approval and Inspection Technology for EXIT LIGHT, are deduced from aforementioned comparison and analysis. Also the derived test items are experimented with a exit light. For a power factor correction, a power factor correction circuit is designed and power factor, crest factor, asymmetry ratio, luminance variation are analyzed. In order to show the validity of designed circuit, current waveform and voltage waveform are measured.

  • [ $PGE_1$ ] is an endogenous substance of potent vasodialator as well as inhibitor of platelet aggregation. It has been used therapeutically in peripheral arterial occlusive disease and impotence. Intracavernous injection of $PGE_1$ for erectile dysfunction has been established for several years as a treatment option for erectile dysfunction of diverse etiologies, but this mode of administration is limited by penile discomfort, pain at the injection site, inconvenience and noncompliance. As the matter of worse, the $\beta-hydroxy$ moiety of $PGE_1$ is extremely susceptible to dehydration in solution to give inactive $PGA_1$ and $PGB_1$. For the improvement of stability, rapid absorption at action site and the convenience of application, $PGE_1$ was formulated as urethral suppositories of three types of formulations, such as PEG, witepsol, and the mixture of PEG and witepsol. The stability test of $PGE_1$ and the release test in urinary suppositories were performed. Futhermore, the effect of enhancers and vehicle composition on the penetration of $PGE_1$ through excised rat skin was evaluated by permeability coefficient and enhancement ratio.

  • This study aimed to evaluate the stability of steel frames with weak column-strong beam and consider P-effect using the experimental approach. Towards this end, three specimens were tested under monotonic loading condition. Parameters of tests includes the stiffness of the column and the axial load ratio. Result showed that reduction in column stiffness promoted P- effect, which considerably influenced the frame's stability.

  • Flow visualization study has been conducted to simulate the turbulent dispersion behavior of a crossflow jet physically under the conditions of various thermal stratification in a wind tunnel. A smoke jet with the constant ratio of the jet to freestream velocity is injected normally to the cross flow of the thermally stratified wind tunnel(TSWT) for flow visualization. The typical natures of the smoke dispersion under different thermal stratifications such as neutral, weakly stable, strongly stable, weakly unstable, strongly unstable and inversion layer are successfully reproduced in the TSWT. The Instantaneous velocity and temperature fluctuations are measured by using a cold and hot-wire combination probe. The time averaged dispersion behaviors, the centerline trajectories, the spreading angles and the virtual origins of the cross jet are deduced from the edge detected images with respect to the stability parameter. All the general characteristics of the turbulent dispersion behavior reveal that the definitely different dispersion mechanisms are inherent in both stable and unstable conditions. It is conjectured that the turbulent statistics obtained in the various stability conditions quantitatively demonstrate the vertical scalar flux plays a key role in the turbulent dispersion behavior.

Investigation of long-term stability of pentacene thin-film transistors encapsulated with transparent $SnO_2$

  • The long-term stability of pentacene thin-film transistors (TFTs) encapsulated with a transparent $SnO_2$ thin-film prepared by ion beam assisted deposition (IBAD) was investigated. With a buffer layer of thermally evaporated 100 nm $SnO_2$ film deposited prior to IBAD process, our encapsulated OTFTs sustained its initial field-effect mobility up to one month and then gradually degraded showing only 37% reduction compared to 90% reduction of non-encapsulated OTFTs after 100 days in air ambient. The encapsulated OTFTs also exhibited superior on/off current ratio of over $10^5$ to that of the unprotected devices $({\sim}10^4)$ which was reduced from ${\sim}10^6$ before aging. Therefore, the enhanced long-term stability of our encapsulated OTFTs should be attributed to well protection of permeation of $H_2O$ and $O_2$ into the devices by the IBAD $SnO_2$ thin-film which could be used as an effective inorganic gas barrier for transparent organic electronic devices.

  • Combustion stability is one of the most important factors that must be considered in burning of heavy fuel oil, especially low-grade oil. This paper describes the combustion characteristics of petrochemical process by- product in the combustion furnace of heavy fuel oil. Main experimental parameters were combustion load, excess 02, fuel preheating temperature and air/fuel ratio. The capacity of CRF(combustion research facility) used in this study was 1.0 ton/hr and the burner is steam jet type suitable far heavy oil combustion and manufactured by UNIGAS in Italy. The fuel used in this experiment were 0.5 B-C, petrochemical process by-product and 3 kinds of 0.5 B-C/process by-product mixtures. The combustion stability was monitored and exhaust gases such as CO, NOx, SOx and particulates were measured with the excess $O_2$ and combustion load. The main purpose of this study is to clarify whether process by-product can be used as a boiler fuel or not in consideration of flame stability and emission properties.

  • This research presents an analytical model to investigate the stability due to the ball bearing waviness i n a rotating system supported by two ball bearings. The stiffness of a ball bearing changes periodically due to the waviness in the rolling elements as the rotor rotates, and it can be calculated by differentiating the nonlinear contact forces. The linearized equations of motion can be represented as a parametrically excited system in the form of Mathieu's equation, because the stiffness coefficients have time -varying components due to the waviness. Their solution can be assumed as a Fourier series expansion so that the equations of motion can be rewritten as the simultaneous algebraic equations with respect to the Fourier coefficients. Then, stability can be determined by solving the Hill's infinite determinant of these algebraic equations. The validity of this research is proved by comparing the stability chart with the time responses of the vibration model suggested by prior researches. This research shows that the waviness in the rolling elements of a ball bearing generates the time-varying component of the stiffness coefficient, whose frequency is called the frequency of the parametric excitation. It also shows that the instability takes place from the positions in which the ratio of the natural frequency to the frequency of the parametric excitation corresponds to i/2 (i=1,2,3..).

Analysis on productivity and stability of introduced sweet corn hybrids (도입종 단옥수수 교잡종에 대한 생산성 및 안정성 분석)

  • This study was carried out to compare the productivity and to evaluate the stability for the introduced sweet corn hybrids; Sunnychocolat, 7168Y and Gammiok as a check hybrids. The used materials were planted at Anseong and Daejeon by randomized block design with 3 replications on 21 April, 2013. In productivity of the used hybrid corns, plant and ear height showed comparatively stable as a 183.5 cm to 226.5 cm and 45.8 cm to 80.9 cm, respectively. In yield per 10a, number of ears was higher in Sunnychocolat than check, while Gammiok was very poor in kernel setting length and marketability. Beside, stability of the used hybrids also showed highly in the introduced hybrids than check in the ratio of ear to plant height, tolerance to environmental stress and low lodging. As results above, we thought that these introduced hybrids will be well adapted in domestic cultivation and growth habit as like other hybrid corns.
