Production and Characterization of Lipopeptide Biosurfactant from Bacillus subtilis A8-8

  • A biosurfactant-producing bacterial strain was selected from oil-contaminated soil because of its ability to degrade crude oil and tributyrin $(C_{4:0})$. The strain was identified as Bacillus subtilis A8-8 based on its morphological, biochemical, and physiological characteristics. When B. subtilis A8-8 was grown with crude oil as the sole carbon source, the biosurfactant from the strain emulsified crude oil, vegetable oil, and hydrocarbons. Soybean oil was the optimum substrate for the emulsifying activity and emulsion stability of the biosurfactant, both of which were superior to those of several commercially available surfactants. The biosurfactant was purified by a procedure including HCl precipitation, methanol treatment, and silica-gel chromatography. The partially purified biosurfactant was analyzed by TLC (thin-layer chromatography), SDS-PAGE, and HPLC and it reduced the surface tension of water from 72 mN/m to 26 mN/m at a concentration of 30 mg/l. Therefore, the purified lipopeptide biosurfactant has strong properties as an emulsifying agent and acts as an emulsion-stabilizing agent.

  • Sardine ant mackerel so called dark muscled fish have been underutilized due to the disadvantages in bloody meat color, high content of fat, and postmortem instability of protein. Recent efforts were made to overcome these defects and develope new types of product such as texturized protein concentrates and dark muscle eliminated minced fish. Approach of this study is based on the rapicl dehydration of foamed fish-starch paste by dielectric heating. In process comminuted sardine meat was washed more than three times by soaking and decanting in chilled water and finally centrifuged. The meat was ground in a stone mortar added with adequate amounts of salt, foaming agent, and other ingredients for aid to elasticity and foam stability. The ground meat paste was extruded in finger shape and heated in a microwave oven to give foamed, expanded, and porous solid structure by dehydration. Dielectric constant ($\varepsilon$′) and dielectric loss ($\varepsilon$") vcalues of sardine meat paste were influenced by wavelength and moisture level. Those values at 100KHz and 15MHz were ranged 2.25∼9.86 ; 2.22∼4.18 for $\varepsilon$′ and 0.24∼19.24 ; 0.16∼1.25 for $\varepsilon$", respectively, at the moisture levels of 4.2∼13.8%. For a formula for fish-starch paste preparation, addition of 20∼30% starch (potato starch) to the weight of fish meat, 2∼4% salt, and 5∼10% soybean protein was adequate to yield 4∼5 folds of expansion in volume when heated. Addition of egg yolk was of benefit to micronize foam size and better crispness. In order to provide better foaming and dehydration, addition of 0.2∼0.5% sodium bicarbonate foaming agent, was proper to result in foam size of 0.5∼0.7mm and foam density of 200∼400/$\textrm{cm}^2$ which gave a good crispness.

Improvement of Anthocyanin Encapsulation Efficiency into Yeast Cell by Plasmolysis, Ethanol, and Anthocyanin Concentration Using Response Surface Methodology

  • Anthocyanins are antioxidant compounds susceptible to environmental factors. Anthocyanin encapsulation into yeast cells is a viable solution to overcome this problem. In this study, the optimal factors for anthocyanin encapsulation were investigated, including anthocyanin concentration, plasmolysis contraction agent, and ethanol concentration, and response surface methodology was evaluated, for the first time. Anthocyanin from Hibiscus sabdariffa L. flowers was encapsulated into Saccharomyces cerevisiae using plasmolysis contraction agent (B: 3%-20% w/v), ethanol concentration (C: 3%-20% v/v), and anthocyanin concentration (A: 0.15-0.45 g/ml). The encapsulation yield and anthocyanin loss rate were determined using a spectrometer (520 nm), and color stability evaluation of the capsules was performed at 80℃ for 30 min. The results of the study showed that these factors have a significant impact on the encapsulation of anthocyanin, in which ethanol agents have the highest encapsulation yield compared to other factors in the study. Statistical analysis shows that the independent variables (A, B, C), their squares (A2, B2, C2), and the interaction between B and C have a significant effect on the encapsulation yield. The optimized factors were anthocyanin, 0.25 g/ml; NaCl, 9.5% (w/v); and ethanol, 11% (v/v) with an encapsulation yield of 36.56% ± 0.55% and anthocyanin loss rate of 15.15% ± 0.98%; This is consistent with the expected encapsulation yield of 35.46% and loss rate of 13.2%.

  • Organic electroluminescence devices(ELD) were fabricated using by molecularly doped method with N,N'-diphenyl-N,N'-bis(3-methylphenyl)-1,1'-biphenyl-4,4'-diamine(TPD) as a hole transport agent, squarylium dye as an emitting agent, and side chain liquid crystalline polymer(MCH) as matrix for TPD. An indium-tin-oxide(ITO) coated glass and an Mg electrode were used as the hole and the electron injecting electrode, respectively. The highest stability of ELD was obtained by spin coating method using dichloroethane as a solvent at a polymer/TPD concentration of 0.005 wt%. For the EL cell with ITO/polymer-TPD/SQ dye/Mg structure, we achieved light red luminescence at a current of 102 mA/$cm^2$ with an applied voltage of 23 V.

  • Scandia stabilized zirconia (ScSZ) is adapted for electrolyte material of solid oxide fuel cell (SOFC) because of its high ionic conductivity and chemical stability. ScMnSZ1 powder having a composition of $((ZrO_2)_{0.89}(Sc_2O_3)_{0.1}(MnO_2)_{0.01})$ is synthesized by ultrasonic spray pyrolysis (USP) method. Porous ScMnSZ1 powder is obtained by using a pore forming agent. Microstructure and morphology, particle size distribution of porous powder synthesized with 3wt% pore forming agent are investigated. Sintered ScMnSZ1 sample with ground fine powder are also investigated their microstructure and electrical conductivity. The electrical conductivity of sintered ScMnSZ1 samples with ground fine powder was 0.082 S/cm, 0.127 S/cm and 0.249 S/cm at $750^{\circ}C$, $800^{\circ}C$ and $900^{\circ}C$, respectively.

The Quality Characteristics of Grape Jelly Made with Various Gelling Agents for Consumption by Elderly Women (여성 고령자를 대상으로 한 포도젤리의 겔화제에 따른 품질 특성 연구)

  • This study was carried out to determine the quality characteristics of grape jellies made with various gelling agents such as agar, $\kappa$-carrageenan, and gellan gum for consumption by elderly women. The concentrations of agar were 0.25, 0.35, and 0.45%, and those of the $\kappa$-carrageenan and gellan gum were 0.15, 0.3, and 0.4%, respectively. The color values, gelling temperatures, melting temperatures, break down rates, textural properties, and consumer acceptance of the grape jellies were measured. The average age of the subjects participating in the acceptance test was 82. The lightness of the grape jelly made with agar was the highest, and its redness was the lowest among the jellies. The gelling and melting temperatures for the grape jelly made with gellan gum were higher than those of the other jellies, and the melting temperatures were 37, 43, and $47^{\circ}C$ based on 0.15, 0.3, and 0.4% gellan gum content, respectively. The break down rate of the grape jelly made with gellan gum was the lowest among the jellies. These results indicate that the stability of the grape jelly made with gellan gum was superior among the jellies. The hardness, adhesiveness, and springiness of the grape jelly made with agar were lowest among the jellies, and the grape jellies made with agar(0.25% and 0.35%) and gellan gum(0.15% and 0.3%) had the highest consumer acceptance among the jellies. However, the gellan gum was deemed an inappropriate gelling agent with regard to the acceptability due to its sour and moldy taste as perceived by the participants.

Synthesis and In vitro Evaluation of 99mTc-diglucosediethylenetriamine (DGTA) as a Potential Tumor Imaging Agent

  • Using a single step chemical synthesis, we synthesized the potential tumor imaging agent $^{99m}Tc$-diglucose-diethylenetriamine (DGTA) from diethylenetriamine and natural D-glucose. 10 min Incubation of 10 mg of precursor with 50 ${\mu}g$ of $SnCl_2{\cdot}2H_2O$ at room temperature yielded over 95% of $^{99m}Tc$ labeling. The stability for 6 hours in saline or human plasma was over 90%. In vitro tumor cell uptake assays using the SNU-C5 and 9 L cell lines showed that, in 0-400 mg/dL glucose medium, cell uptake of $^{99m}Tc$-DGTA was 1.5-8 times higher than that of [$^{18}F$]FDG. Moreover, [$^{18}F$]FDG uptake was dependent on glucose concentration in the medium, whereas cellular uptake of $^{99m}Tc$-DGTA was not dependent on glucose concentration, suggesting that the two compounds have different uptake mechanisms by tumor cells.

Oxolinic acid Residue in the cultured Eel Tissues and its Change to Heating Process (시판중인 뱀장어중의 Oxolinic acid 잔류량과 가열에 의한 변화)

  • The residual tissue concentraion of the widely used aquatic antibacterial agent, oxolinic acid, was surveyed in eels collected from fish markets of Chonbuk Province, Korea. Their concentrations in the dorsolateral muscle were widely varying. In about 32% of samples examined, oxolinic acid was not detected. In about 16% of those samples in which oxolinic acid was detected, the concentration was above 0.1 ppm. The tissue distrubution of the agent in major organs was in the rank order of kidney>liver>plasma>muscle. When the muscle samples which contained residual oxolinic acid were baked for up to 10 min, there was no change in the drug concentration. Their concentration declined to about 50% by baking for 30 min at which time the tissue turned to the texture of charcoal. The extreme stability of oxolinic acid to heating process was confirmed with muscle samples from eels to which a high dose of oxolinic acid was administered, and also with an aqueous oxolinic acid solution of known concentration. It is suggested that an effective regulatory measure should be initiated to keep eel consumers from residual oxolinic acid impact.

Biomass Waste, Coffee Grounds-derived Carbon for Lithium Storage

  • Biomass waste-derived carbon is an attractive alternative with environmental benignity to obtain carbon material. In this study, we prepare carbon from coffee grounds as a biomass precursor using a simple, inexpensive, and environmentally friendly method through physical activation using only steam. The coffee-derived carbon, having a micropore-rich structure and a low extent of graphitization of disordered carbon, is developed and directly applied to lithium-ion battery anode material. Compared with the introduction of the Ketjenblack (KB) conducting agent (i.e., coffee-derived carbon with KB), the coffee-derived carbon itself achieves a reversible capacity of ~200 mAh/g (0.54 lithium per 6 carbons) at a current density of 100 mA/g after 100 cycles, along with excellent cycle stability. The origin of highly reversible lithium storage is attributed to the consistent diffusion-controlled intercalation/de-intercalation reaction in cycle life, which suggests that the bulk diffusion of lithium is favorable in the coffee-derived carbon itself, in the absence of a conducting agent. This study presents the preparation of carbon material through physical activation without the use of chemical activation agents and demonstrates an application of coffee-derived carbon in energy storage devices.

Agent-Server based Protocol Analyzer for LTE Network (LTE 망을 위한 에이전트-서버 기반의 프로토콜 분석기)

  • Recently, together with the development of wireless communication technologies and the wide use of smart phones, the demand of the next generation mobile communication has been increased. To construct the next generation mobile communication platform efficiently for a short period from protocol development phase to protocol stability phase, an protocol analyzer is required to verify and analyze the protocol. In this paper, we propose the protocol analyzer of LTE network which is the leader of the next generation mobile communication platforms. The protocol analyzer is a software based agent-server architecture and uses XML metadata which defines intercommunication messages to analyze the protocol of the next generation mobile communications. We can analysis the encoding messages applied to LTE network using the loading of the decoding library. We verify the superiority of the proposed analyzer through an integrated test with LTE network.
