Background: Among the various ginseng strains, Shizhu ginseng is endemic to China, mainly distributed in Kuandian Manchu Autonomous County (Liaoning Province, China); however, not much is known about the compounds (especially saponins) in Shizhu ginseng. Methods: A rapid, sensitive, and reliable ultra-high performance liquid chromatography coupled with MS/MS (UHPLC-MS/MS) method was developed to separate and identify saponins in Shizhu ginseng. Results: The separation was carried out on a Waters ACQUITY UPLC BEH $C_{18}$ column ($100mm{\times}2.1mm$, $1.7{\mu}m$) with acetonitrile and 0.1% formic acid aqueous solution as the mobile phase under a gradient elution at $40^{\circ}C$. The detection was performed on a Micromass Quattro Micro API mass spectrometer equipped with electrospray ionization source in both positive and negative modes. Under the optimized conditions, a total of 31 saponins were identified or tentatively characterized by comparing retention time and MS data with related literatures and reference substances. Conclusion: The developed UHPLC-MS/MS method was suitable for identifying and characterizing the chemical constituents in Shizhu ginseng, which provided a helpful chemical basis for further research on Shizhu ginseng.

An isotope dilution-liquid chromatography/tandem mass spectrometric method was developed as a candidate reference method for the accurate determination of acrylamide in potato chips, starch-rich foodstuff cooked at high temperature. Sample was spiked with 13C3-acrylamide and then extracted with water. The extract was further cleaned up with an Oasis HLB solid-phase extraction (SPE) cartridge and an Oasis mixed-phase cation exchange (MCX) SPE cartridge. The extract was analyzed by using LC/ESI/Tandem MS in positive ion mode. LC with a medium reversed-phase (C4) column was optimized to obtain adequate chromatographic retention and separation of acrylamide. MS was operated to selectively monitor [M+H]+ ions of the analyte and its isotope analogue at m/z 72 and m/z 75, respectively. Sample was also analyzed by the LC/MS with selectively monitoring the collisionally induced dissociation channels of m/z 72 → m/z 55 and m/z 75 → 58. Compared to the LC/MS chromatograms, the LC/MS/MS chromatograms showed substantially reduced background chemical noises coming from solvent clusters formed during ESI spray processes and interferences from sample matrix. Repeatability and reproducibility studies showed that the LC/MS/MS method is a reliable and reproducible method which can provide a typical method precision of 1.0% while the LC/MS results are influenced by chemical interferences.
상수나 하수처리장에서 쓰이는 오존처리는 물질전달에 제약을 받는 공정임은 알려져왔다. 이런 물질전달의 한계를 극복하는 방법으로 매우 효과적인 것은 오존가스를 함유한 기포의 크기를 줄임으로써 물질전달에 필요한 접촉면적을 넓히는 것이다. 오존의 전달을 크게 하기 위한 방법으로서 본 연구에서는 전기장분사(Electrostatic spraying: ES)를 사용하였다. 실험에서는 ES와 기포발생판을 비교하였는데 ES실험에서는 전압을 0 kV에서 4 kV 범위까지 높여주어 물질전달효과를 살펴보았고 기공크기가 다른 두 가지 기포발생판(미세기공: 기공크기 $10{\sim}15{\mu}m$, 중간기공: 기공크기 $40{\sim}60{\mu}m$)을 사용하여 물질전달효율을 비교 분석 하였다. 물질전달효율은 시간의 경과에 따른 용존오존농도의 측정과 페놀의 산화시간으로 간접 계산하였다. 실험 결과 ES의 경우 전압을 0 kV에서 4 kV까지 올렸을 때 기포의 크기가 작아짐으로 인해 오존전달율을 약 40% 정도 향상시킬 수 있었다. 또한 ES와 기포발생판의 물질전달 비교실험에서 미세기공 기포발생판보다 4 kV의 ES가 더 효과적임을 알 수 있었다.
태양열 이용 축열식 직접접촉 열교환기의 실제 적용을 위한 작동조건도출 및 열교환기내 열전달 특성 규명을 위하여 작동유체를 물보다 가벼운 경우(Texaterm)와 무거운 경우 두가지를 실제 태양열 집열회로상에서 실험하였다. 작동유체가 물보다 가벼운 경우와 무거운 경우에 대해 실제 태양열 집열회로상에서 작동하여 최적 열전달을 위한 작동유체드럽형성과 유동을 도출하였고, 작동유체가 무거운 경우가 가벼운 경우보다 열손실계수가 높음을 알 수 있었다. 작동유체가 축열유체보다 가벼운 경우와 무거운 경우에 대해 실제 태양열 직열회로에서 실험한 결과, 작동유체가 가벼운 경우 체적 열전달계수는 $1.848{\times}10^{-3}w/cm^3^{\circ}C$, 효율은 73%이고 무거운 경우는 $0.128{\times}10^{-2}w/cm^3^{\circ}C$, 효율은 82%로 작동유체가 무거운 경우가 보다 좋은 것으로 나타났다.
Objectives : The main aim of this study was to examine the LC-MS/MS used to identify phenolic compounds of CRE(Coptidis Rhizoma 70% EtOH Extract). Also, we investigated antioxidative activities and Anti-inflammatory activities. Methods : LC-MS/MS Analysis HPLC and LC-MS/MS were performed on a 1260 series HPLC system (Agilent Technologies, Inc., California, USA) and 3200 QTrap tandem mass system (Sciex LLC) operated in positive ion mode (spray voltage set at -4.5 kV). The solvent used was DW and Acetonitrile containing 0.1% formic acid, a gradient system was used at a flow rate of 0.5 mL/min for analysis, and a Prontosil C18 column (length, 250 mm; inner diameter, 4.6 mm; particle size, 5 ㎛; Phenomenex Co., Ltd., California, USA, Biochoff Chromatography) was used. The solvent conditions used in the mobile phases were 0-10 min at 10-15% B, 10-20 min at 20% B, 20-30 min at 25%, 30-40 min at 40%, 40-50 min at 70%, 50-60 min at 95%, and 60-70 min at 95%. The analysis was performed at a wavelength of 284 nm and a temperature of 35℃. The cell viability was measured using a 3-(4,5-dimethyethiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. We examined the effects of CRE on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) in a RAW 264.7 cells Results : The chemical analysis CRE by Liquid chromatography-tandem mass spectrometry (LC-MS/MS) confirmed that Rosmarinic acid, Ferrulic acid, 3-O-feruloylquinic acid, and 5-O-feruloylquinic acid as phenolic components. DPPH radical scavenging activity was the inhibitory activity of CRE showed at 200 ㎍/mL a statistically significant level. MTT assay demonstrated that the CRE did not have a cytotoxic effect in RAW 264.7 and LPS-induced RAW264.7 cells. Also, CRE reduced NO production in RAW 264.7 cells stimulated with LPS. Conclusions : Based on these findings, The chemical analysis 4 major components CRE such as Rosmarinic acid, Ferrulic acid, 3-O-feruloylquinic acid, and 5-O-feruloylquinic acid. Moreover, we confirmed that CRE has effects antioxidant and anti-inflammatory. The results demonstrate that CRE can be used as an antioxidant and a powerful chemopreventive ingredient against inflammatory diseases.
