  • Two cases of spontaneous auricular chondritis were reported in SD rats in a 13-week toxicity study. At necropsy, pinna of each rats had fm, irregular nodules. Based on the anatomical location and histopathological features of the lesion, the disease was diagnosed as auricular chondritis.

비글개와 랫드에서 SDK시제품(가칭)의 4주간 피부도포 반복투여 독성시험에 관한 연구 (Four-Week Topical Toxicity Studies of SDK in Beagle Dogs and Sprague-Dawley Rats)

  • SDK (skin decontamination kit) is new skin decontaminant which is developed by ADD (Agency for defence development). In this study, four-week toxicity of SDK was investigated using beagle dogs and Sprague-Dawley rats. The beagle dogs and Sprague-Dawley rats were dressed topically seven days per week for 28 days, with dosage of 0, 0.25, 0.8 and 1 g/kg/day. respectively. Animals treated with SDK did not cause any death and show any clinical signs. They did not show any significant changes of body weight, feed uptake and water consumption. They were not significantly different from the control group in urinalysis, ocular examination and histopathological examination. In hematological and serum biochemical assay, there were no-dose-defendent changes. Therefore, SDK was not indicated to have any toxic effect in the beagle dogs and Sprague-Dawley rats when it was dressed topically below the dosage 1 g/kg/day for four weeks.

호박분말이 Sprague-Dawley 흰쥐에서 인위적으로 유발한 위암 및 유선암에 미치는 영향 (Effects of Pumpkin Powder on Chemically Induced Stomach and Mammary Cancers in Sprague-Dawley Rats)

  • This study was conducted to investigate the effectiveness of pumpkin powder in the diet of experimental animals on chemically induced stomach and mammary canceers. Three weeks old male SpragueDawley rats were randomly allocated to either 1) basal diet+MNNG, 2) basal diet+MNNG+PMC, 3) 2.5% pumpkin powder supplemented diet+MNNG+PMC, or 4) 5.0% pumpkin powder supplemented diet+MNNG+PMC for stomach cancer experiment. And female Sprague-Dawley(5 weeks old) rats were randomly assigned to either 1) basal diet only, 2) basal diet+DMBA, 3) 2.5% pumpkin powder supplemented diet+DMBA, or 4) 5.0% pumpkin powder supplementd diet+DMBA. In both experiments, supplements of pumpkin powder in basal diet decreased body weight of both male and female experimental animals. Pumpkin powder in rat diet decreased significantly(p<.05) chemically induced stomach cancer. With its suppressing effects on stomach cancer, pumpkin powder in diet of experimental rats had decreasing effects on the initiation and development of DMBA-induced mammary cancer. In conclusiion, current study may provide in vivo data to develop health foods using pumpkin. Further studies, however, are essential to clarify the exact role of pumpkin powder in chemically induced stomach and mammary cancers.

  • The repeated toxicity of Gleditschia-saponin produced and provided by S.S. Bio-Tech Bench Co. was evaluated in Sprague-Dawley rats. Gleditschia-saponin was administered to rats by oral route at dose levels of high (180 mg/kg/day), medium (90 mg/kg/day) and low (45 mg/kg/day) once a day for 14 days. Saline was administered to another group of rats as control. Each group was consisted of 5 male and female rats. There were no dose-related changes in clinical findings, food and water consumption, organ weights, urine analysis, biochemical examination and hematological findings in all groups of animals treated with Gleditschia.- saponin, except body weights. Body weighs in male and female rats were increased significantly (p < 0.05) from day 4 to 14 in low, middle and high dose groups than control group. Body weight in high dose group was increased higher than control or low, middle dose groups on day 14. Gross and histopathological findings revealed no evidence of specific toxicity to Gleditschia.-saponin. Therefore, it was concluded that Gleditschia-saponin had no toxic or side effects in Sprague-Dawley rats in an repeated oral toxicity tests.

Subacute Oral Toxicity Study of a New Type of Cordyceps, Paecilomyces sinclairii, in Sprague-Dawley Rats

  • This study was conducted to investigate the 2 week-oral toxicity of Paecilomyces sinclairii in Sprague-Dawley rats. P. sinclairii was daily administered to male and female rats for 2 weeks with different dose levels (0, 0.008, 0.04, 0.2, 1 and 5 g/kg). There were no clinical signs compared with control group, but a slight increase of white blood cell (WBC) was observed in the males rats receiving all dose levels of P. sinclairii. In biohematological analysis, the levels of glucose and cholesterol in the blood were decreased slightly in the males and females rats at doses of 0.008 or 1 g/ kg. At the all dose groups, there were no significant changes in the body weights, but autopsy findings of all organs showed reduced weights in the thymus of males in the high dose groups of 1 g/ kg and 5 g/kg. These results indicate that P. sinclairii does not induce any significant toxic effect on Sprague-Dawley rats treated for 2 weeks, but the reduced weights of thymus in males may require a further long-term investigation.

  • The background control data were compiled from rat developmental toxicity studies con-ducted at Toxicology Research Center, KRICT during the 1993-1999 period. These data were assembled in order to provide background in formation for the maternal and fetal data collected in 13 developmental toxicity studies using Sprague-Dawley rats. A total of 325 mated females were used in these studies during the seven-year period and overall pregnancy rate of these females was 93.8%. The present background control data included body weights, food consumption, hematological values, and organ weights of pregnant females, caesarean section data, and fetal examination data. These data can be used not only as a historical database for the meaningful interpretation of data from reproductive and developmental toxicity studies, but also as a contribution to biological characterization oj Sprague-Dawley rats.

Sprague Dawley 랫트에서 자장이 골 창상 치유에 미치는 영향 (Bone Healing Effect of a Magnet in Sprague Dawley Rats)

  • The present study was carried out to examine the bone healing effect of a magnet as the feasibility test for clinical application. The surgical technique of tibia puncture was performed with male Sprague Dawley rats for investigating radiological and histological findings. The treated group was attached the magnet, and control group was attached the placebo magnet for 4, 6, and 8 weeks respectively. The rats were taken a radiograph for examining radiological findings on 2-weekly interval for 8 weeks. Also, the rats were partly euthanized for investigating histological findings on 2-weekly interval from 4 week to 8 week. In the magnet treated rats, the healing process of bone trauma was positively stimulated to compare with control rats.

Dietary Conjugated Linoleic Acid did not Affect on Body Fatness, Fat Cell Sizes and Leptin Levels in Male Sprague Dawley Rats

  • This study was designed to observe the effect of conjugated linoleic acid (CLA) supplementation on body fatness, fat cell sizes and leptin levels in male Sprague Dawley rats. Following weaning, forty rats were divided into 4 groups beef tallow (BT), fish oil (FO). beef tallow with CLA supplementation (BTC), and fish oil with CLA supplementation (FOC) group. For four weeks, all rats were fed experimental diets containing 12% of total dietary fat (w/w) with or without 1% CLA. After 4 weeks, the animals were sacrificed; the total carcass fat, plasma leptin levels, epididymal fat pad weights and fat cell sizes in adipose tissue were measured. CLA supplementation did not significantly affect the rat's body weights, total body fat, epididymal fat pad weights, and fat cell sizes. CLA also did not have a significant effect on plasma leptin levels. These results suggest that CLA supplement was not an effective way to reduce the body weights of male Sprague Dawley rats.

White Matter Damage and Hippocampal Neurodegeneration Induced by Permanent Bilateral Occlusion of Common Carotid Artery in the Rat: Comparison between Wistar and Sprague-Dawley Strain

  • In order to reproduce chronic cerebral hypoperfusion as it occurs in human aging and Alzheimer's disease, we introduced permanent, bilateral occlusion of the common carotid arteries (BCCAO) in rats (Farkas et al, 2007). Here, we induced BCCAO in two different rat strains in order to determine whether there was a strain difference in the pathogenic response to BCCAO. Male Wistar and Sprague-Dawley (SD) rats (250-270 g) were subjected to BCCAO for three weeks. Kluver-Barrera and cresyl violet staining were used to evaluate white matter and gray matter damage, respectively. Wistar rats had a considerably higher mortality rate (four of 14 rats) as compared to SD rats (one of 15 rats) following BCCAO. Complete loss of pupillary light reflex occurred in all Wistar rats that survived, but loss of pupillary light reflex did not occur at all in SD rats. Moreover, BCCAO induced marked vacuolation in the optic tract of Wistar rats as compared to SD rats. In contrast, SD rats showed fewer CA1 hippocampal neurons than Wistar rats following BCCAO. These results suggest that the neuropathological process induced by BCCAO takes place in a region-specific pattern that varies according to the strain of rat involved.

당귀보혈탕(當歸補血湯)의 SD계 흰쥐에 대한 아급성경구독성시험 (A 4-weeks Oral Toxicity of Dangguibohyel-tang in Sprague-Dawley Rats)

  • Objectives : The subacute toxicity was investigated in Sprague-Dawley rats orally treated with Dangguibohyel-tang (DBT). DBT has been used for anemia, in Korean medicine. Methods : Sprague-Dawley rats were administered orally with DBT at the dosages of 1,000 mg/kg for 14 days. We daily examined number of deaths, clinical signs, body weights and gross findings during the 14 days. Results : There were no clinical signs and pathological changes compared with control group. Body weights were not significantly changed between control and treatment groups. In hematological and biochemical serum parameters, all mean values appear to be within the normal range. Conclusion : These results suggest that DBT dose not induce any significant subacute oral toxicities in Sprague-Dawley rats.
