The General Toxicity of Novel Platinum Complexes in Rats

  • This study was performed to evaluate the general toxicity of novel Pt(II) complexes, (KHPC-002: [Pt(trans-1-dach) (DPPE)].2NO$_3$, KHPC-005: [Pt(cis-dach)(DPPE)].2NO$_3$ and KHPC-006: [Pt(cis-dach)(DPPP)]. 2NO$_3$). In the acute toxicity study in rats, three dosing groups of Sprague-Dawley male rats in each compounds were given a single intraperitoneal injection of KHPC-002, KHPC-005 and KHPC-006. In order to compare the toxic effects of these novel Pt(II) complexes with those of cisplatin, one group Sprague-Dawley male rats were given 7mg/kg i.p injection of cisplatin. Body weights showed dose-related decrease in all treatment groups when compared wi th the control group.

랫드에 대한 KDRD-010의 급성경구독성시험 (Acute Oral Toxicity of KDRD-010 in Rats)

  • This study was carried out to investigate the acute toxicity of KDRD-010 in Sprague-Dawley rats. KDRD-010 was administratered orally at a dose level of 26, 78, 233, 700, and 2,100 mg/kg. In this study, we daily examined number of deaths, clinical signs, body weights, and pathological examinations for 14 days after administration of KDRD-010. The results indicate that KDRD-010 did not show any toxic effect in rats and oral $LD_50$ value was over 2,100 mg/kg in Sprague-Dawley rats.

GST 추출물의 Sprague-Dawley Rat를 이용한 13주 반복 경구투여 독성시험 및 4주 회복시험 (A Thirteen Week Repeated Oral Dose Toxicity Test and A Four Week Recovery Test of GST in Sprague-Dawley Rats)

  • Objectives: To provide information on the safety of GST (GamiSasangja-tang; CnidiiFructus, Sophora Root, Angelica Gigas Root, Clematidis Radix, Stemonae Radix, Spirodelae Herba), we carried out a 13-week repeated oral dose toxicity and a 4-week recovery test of GST in Sprague-Dawley rats. Methods: Female and male rats were treated with GST at oral doses of 1,250, 2,500, and 5000 mg/kg. The GST was administered for 13 weeks. Mortality, clinical signs, body weight changes, food consumption, ophthalmologic findings, urinalysis, hematological and biochemical parameters, gross findings, organ weights and histological markers were monitored during the study period. The rats were then monitored for 4 extra weeks to determine recovery time after the study period. Results: We found no mortality or abnormalities among clinical signs, body weight, food consumption, ophthalmologic findings, urinalysis, hematological and biochemical parameters, gross findings, organ weights or histological markers in any of the rats tested. Conclusions: The no-observed adverse effects level (NOAEL) is considered as over 5000 mg/kg for male and female rats.

복분자 추출물의 Sprague-Dawley rat를 이용한 단회 경구 투여 독성시험 (Single Oral Dose Toxicity Study of Black Raspberry Extract in Sprague-Dawley Rats)

  • Objective : This study was performed to evaluate the toxicity after a single oral administration of black raspberry extract to male and female Sprague-Dawley (SD) rats and to determine the approximate lethal dose (ALD). Methods : We previously showed that the black raspberry extract repressed the simvastatin-mediated expression of Proprotein convertase subtilisin/kexin type 9 (PCSK9) and improved Low-Density Lipoprotein cholesterol (LDL-C) uptake by hepatocytes through the induction of the Low-Density Lipoprotein Receptor expression in hepatocytes. The groups consisted of black raspberry extract groups, as an oral dose of 2,000 mg/kg and a control group. 5 weeks SD rats were randomly assigned to 4 groups of 5 rats. Each male and female SD rats were administered orally once. For 14 days after the administration, mortality, clinical signs, changes in body weight, and necropsy findings were observed according to the "Standard for Toxicity Study of Pharmaceuticals" of Korea Food and Drug Administration (KFDA) guideline and "Acute Oral Toxicity- Fixed Dose Procedure" of OECD Test Guideline. Results : There were no cases of mortality in the group administered with 2,000 mg/kg of male and female, and no abnormalities in body weight change and clinical signs. Also, no gross abnormalities were observed at the autopsy. Conclusions : As a result of a single oral administration of the black raspberry extract to SD rats, the ALD was determined to exceed 2,000 mg/kg for both male and female SD rats.

A Pilot Study on Single-dose Toxicity Testing of Hominis placenta Pharmacopuncture in Sprague-Dawley Rats

  • Objectives: This study was performed to analyze the toxicity and to find the lethal dose of the test substance Hominis placenta pharmacopuncture when used as a single-dose in 6 week old, male and female Sprague-Dawley (SD) rats. Methods: All experiments were conducted at Biotoxtech (Chungwon, Korea), an institution authorized to perform non clinical studies, under the regulations of Good Laboratory Practice (GLP). SD rats were chosen for the pilot study. Doses of Hominis placenta pharmacopuncture extracts, 0.125, 0.25 and 0.5 mL, were administered to the experimental group, and 0.5 mL doses of normal saline solution were administered to the control group. This study was conducted under the approval of the Institutional Animal Ethics Committee. Results: No deaths or abnormalities occurred in any of the groups. Also, no significant changes in body weights were observed among the groups, and no significant differences in hematology/biochemistry, necropsy, and histopathology results were noted. Hematologically, some changes in the male rats in two experimental groups were observed, but those changes had no clinical or toxicological meaning because they were not dose dependent. Histopathological tests on the injected parts showed cell infiltration in the male rats in one of the experimental groups; however, that result was due to spontaneous generation and had no toxicological meaning. Therefore, this study showed that Hominis placenta pharmacopuncture had no effect on the injected parts in terms of clinical signs, body weight, hematology, clinical chemistry, and necropsy. Conclusion: As a result of single-dose tests of the test substance Hominis placenta pharmacopuncture in 4 groups of rats, the lethal dose for both males and females exceeded 0.5 mL/animal. Therefore, the above findings suggest that treatment with Hominis placenta pharmacopuncture is relatively safe. Further studies on this subject are needed.

Sprague-Dawley rats에서 감초 신품종 추출물의 단회투여 독성 평가 (Assessment of the Single Oral dose Toxicity of Glycyrrhiza New Variety Extract in Sprague-Dawley Rats)

  • Glycyrrhiza species (Licorice) are one of the most commonly used medicinal plants in Asian countries such as China, India and Korea. It has been traditionally used to treat many disease including cough, cold, asthma, fatigue, gastritis and respiratory tract infections. Glycyrrhiza new variety, Wongam (WG), have been developed by Korea Rural Development Administration and revealed several pharmacological effects. However, limited data are available on the potential adverse effects of the WG. Here, we evaluated the general toxicity of the WG extract through single oral dose toxicity study in Sprague-Dawley rats. After single oral dose administration, there was no mortality up to 5000 mg/kg during experiment period. In addition, there was no clinical signs including body weight change, gross findings and necropsy findings up to 5000 mg/kg during experiment period. To conclude, the no-observed-adverse-effect level (NOAEL) of WG was higher than 5000 mg/kg and no target organs were identified in male and female Sprague-Dawley rats.

Sprague Dawley 쥐에서 장내 유래 Lactobacillus sp. KP-3의 중금속 축적 저해 효과 (The Inhibitory Effects of Intestine-oriented Lactobacillus sp. KP-3 on the Accumulation of Heavy Metals in Sprague Dawley rats)

  • 유산 균주들을 신생아 및 성인의 분변으로부터 분리하여 그 중에서도 가장 중금속 흡착능이 크고 중금속에 내성이 있다고 사료되는 균주 KP-3를 선별하여 동정한 결과 Lactobacillus sp.으로 밝혀졌다. 동물 실험에서는 생후 4주령의 Sprague dawley male rat에게 선별한 균주 KP-3를 투여하여 혈액 및 장기 내의 중금속 축적 저해효과가 어느 정도인지 조사하였다. 실험군으로는 SD rat 한 마리당 Lactobacillus sp. KP-3와 중금속 10 ppm을 혼합하여 약 100 ml/day씩 7일간 투여하였다. 대조군으로 중금속과 멸균 생리 식염수를 혼합하여 투여한 군과 Lactobacillus sp. KP-3와 멸균 생리 식염수를 혼합하여 투여한 군, 멸균 생리 식염수 만을 투여한 군으로 분리하여 SD rat 한 마리당 약 100 ml/day씩 7일간 투여하였다. 혈액, 간장 및 신장의 시료를 채취하여 카드뮴(Cd), 크롬(Cr), 납(Pb) 등의 중금속 축적량을 측정하였다. 측정 결과 크롬(Cr)은 신장에 주로 축적되었고, 납(Pb)은 간장과 신장에 주로 많은 축적을 보였고, 실험군인 Lactobacillus sp. KP-3와 중금속을 혼합하여 투여한 군에서는 중금속만을 투여한 군보다 중금속 축적량이 낮은 것으로 나타났다. 카드뮴(Cd), 크롬(Cr), 납(Pb)의 평균 중금속 축적 저해율은 각각 41.8%, 33.4% 44.2%로 나타났다. 특히, 혈액에서의 중금속 축적 저해율은 약 70%로 매우 높게 나타났고 혈액에서의 카드뮴 축적 저해율은 100%로 나타났다. 본 실험에서 탐색하여 최적화된 유산균이라고 생각되는 Lactobacillus sp. KP-3균주는 실험 결과에 의하면 중금속 축적 저해에 효과가 있는 것으로 여겨지며, 또한 유산균이 함유된 식품을 지속적으로 섭취함으로써 체내의 중금속의 축적 억제에 도움이 크게 되리라 생각된다.

HRccine(HFRS-virus vaccine)의 급성독성 (Acute Toxicity of HRccine (HFRS virus vaccine) in Rats and Rabbits)

  • HRccine(HFRS-virus vaccine) was administered subcutaneously or intragastrically to Spargue Dawley rats and subcutaneously to Japanese white rabbits to investigate the acute toxicity. $LD_50$ values were above 600 times the expected clinical dose in both animals. There were no differences in body weight changes, clinical signs and autopsy findings between all treated groups and control group in both animals. Therefore, it was concluded that HRccine is a very safe substance.

  • Ag-Os, water treatment agent, was administered orally to ICR mice and Sprague-Dawley rats to investigate the acute oral toxicity. $LD_{50}$ values were above 5 g/kg, 2,000 fold higher than the expected concentration in water, in both species with oral administration. There were also no differences in body weight changes, clinical signs and atopsy findings between all treated groups and control group. Therefore, it was concluded that Ag-Os is a very safe compound.

Sea Tangle Supplementation Alters Intestinal Morphology in Streptozotocin-induced Diabetic Rats and Lowers Glucose Absorption

  • This study examined whether dietary supplementation with sea tangle alters the intestinal morphology of streptozotocin-induced diabetic rats and affects the glucose absorption rate. Forty male Sprague-Dawley rats were divided into 2 groups and fed either a control (AIN76-based) diet or a sea tangle-supplemented diet. After 3 weeks, 10 rats in each group received an intramuscular injection of streptozotocin (45 mg/kg BW), and feeding was continued for 3 additional weeks. Dietary supplementation with sea tangle resulted in a lower fasting plasma glucose level compared with the control diet in diabetic rats. Scanning electron micrographs revealed serious damage to the jejunal villi of diabetic rats fed the control diet, whereas supplementation with sea tangle alleviated the damage. In a separate experiment, 20 male Sprague-Dawley rats were divided into 2 groups and fed either a control diet or a sea tangle-supplemented diet for 5 weeks, and fasted rats were subjected to in situ single-pass perfusion. The glucose absorption rate determined in the absence of digesta was decreased by 34% in the jejunum of rats fed a sea tangle diet compared with those fed a control diet. In conclusion, sea tangle supplementation lowered glucose absorption rate, altered intestinal morphology, and appeared to protect villi from damage caused by diabetes mellitus.
