Soft Rot of Rhizopus oryzae as a Postharvest Pathogen of Banana Fruit in Korea

  • Soft rot on banana fruit caused by Rhizopus oryzae was identified for the first time in Korea. Colonies were white to light brown and formed numerous sporangiospores. Optimum temperature for mycelial growth was $30^{\circ}C$. Sporangia were globose and $30{\sim}200{\mu}m$. Sporangiophores were usually straight, $8{\sim}20{\mu}m$, and rhizoids usually in groups of 3~5. Columella were globose to sub-globose and $90{\sim}110{\mu}m$. Sporangiospores were sub-globose or oval and $4{\sim}10{\mu}m$. Based on its mycological characteristics, molecular analysis, and pathogenicity to host plants, this fungus was identified as Rhizopus oryzae Went & Prisen Geerligs. This is the first report of soft rot on banana caused by Rhizopus oryzae in Korea.

First Report of Gongronella guangdongensis Isolated from Soil in Korea

  • A fungal isolate designated KNU16-033 was isolated from a soil sample in Daegu, Korea. White, short, and felt-like aerial mycelia appeared on the surface of colonies of the isolate. Colonies with a smooth texture developed slowly and reached a diameter of 78 mm after 21 days of incubation on potato dextrose agar. This isolate displayed globose, colorless or light yellow sporangiospores, which differed morphologically from the sporangiospores of Gongronella butleri, which were hyaline, oval to flattened on one side or almost reniform. Based on these morphological characteristics and phylogenetic analysis using internal transcribed spacer regions, the isolated fungus was identified as G. guangdongensis belonging to Cunninghamellaceae. To our knowledge, this is the first record of G. guangdongensis in Korea.

  • A soft rot of fruits caused by Rhizopus nigricans occurred on peach (Prunus persica var. vulgaris) in The Chinju City Agricultural Products Wholesale Marke during in summer season of 2000. The disease infection usually started from wounding after harvest fruits, and then moved to outside. At first, the lesions started with water soaked and rapidly softened and diseased area gradually expanded. In severely infected film house, the rate of infected fruits reached 65.2%. Numerous sporangiospores were produced on the diseased fruits. Most of the sporangiospores were appeared to be readily dispersed in the air. The mycelia grew surface of fruits and produced stolons. Colonies on potato dextrose agar at $25{\sim}30^{\circ}C$ white cottony at first becoming heavily speckled by the presence of sporangia and the browinish black at maturity, spreading rapidly by means of stolons fired at various points to the substrate by rhizoids. Sporangia were $85.3{\sim}243.5{\times}53.4{\sim}219.2\;{\mu}m$ in size and were globose or sub-globose with. somewhat flattened base. The color of sporangia was white at first and then turned black with many spores, and never over-hanging. Sporangiophores were $8.9{\sim}36.6\;{\mu}m$ in width, smooth-walled, non-septate, light brown, simple, long, arising in groups of $3{\sim}5$ from stolons opposite rhizoids. Sporangiospores was $9.7{\sim}24.8{\times}5.9{\sim}15.8\;{\mu}m$, irregular, round, oval, elongate, angular, and browinish-black streaked. Columella was $70.2{\times}149.7{\mu}m$. brownish gray, and umberella-shaped when dehisced. The causal organism was identified as. Rhizopus nigricans Lind on the basis of the morphiogical characteristics of the fungus. Rhizopus soft rot on peach (Prunus persica) caused by the fungi has not been reported in Korea. This is the first report of rhizopus soft rot on peach caused by Rhizopus nigricans in Korea.

Pod Rot of Cowpea (Vigna sinensis) Caused by Choanephora cucurbitarum

  • In August 2001, pod rot of cowpea caused by Choanephora cucurbitarum was found in the experimental fields of the Gyeongsangnam-do Agricultural Research and Extension Services, Korea. Initial symptoms of the disease were the appearance of water-soaked, dark-green lesions and followed by rapid rotting of the infected tissues. As the disease progressed, whitish mycelia and monosporous sporangiophore with monosporous sporangiola were produced on the lesions. The fungus produced white to pale yellowish brown mycelia with scattered monosporous sporangiophore and monosporous sporangia containing sporangiospores on potato dextrose agar (PDA). Monosporous sporangiophore was long, slender and branched at the apex, with each branch bearing a sporangiospore. Sporangium was subglobose in shape and 42.6-112.6 ㎛ in size. Monosporous sporangiola were elliptic, fusiform or ovoid, brown in color, and 9.8-23.4$\times$7.2-12.8 ㎛ in size. Sporangiospores were elliptic, fusiform or ovoid in shape, dark brown or brown in color, 12.9-24.6$\times$8.6-15.4 ㎛ in size, and had three or more appendages. Zygospores were black and 43.6-72.4 ㎛ in size. The fungus grew on PDA at 15-40$\^{C}$, and optimum temperature was 30$\^{C}$. This is the first report on pod rot of cowpea caused by C. cucurbitarum in Korea.

  • Rhizopus soft rot caused by Rhizopus stolonifer on June, 2008 occurred on Rubus crataegifolius Bunge in the box at Jinju City Wholesale Market of Agricultural Products at Gyeongnam province in Korea. The infected fruits were rapidly water-soaked, softened, black and eventually rotted. The symptoms occurred after infecting wounds at harvest time. The colony were white to light brown, formed much sporangiospores. The optimum temperature of the fungus on potato dextrose agar was $30^{\circ}C$. Sporangiophores were $12{\sim}25{\mu}m$ in width. Sporangia were globose or hemispheric, white at first and gradually to black, and $82{\sim}195{\mu}m$ in size. Columella were hemispheric, and $70{\sim}92{\mu}m$ in size. Sporangiospores were irregular round or oval, brownishblack and $9{\sim}21{\times}7{\sim}8{\mu}m$ in size. On the basis of symptom, mycological characteristics and pathogenicity of the fungus, the causal fungus was identified as Rhizopus stolonifer. This is the first report of Rhizopus soft rot by R. stolonifer on R. crataegifolius in Korea.

Columella were hemispheric, and $72{\sim}125\;{\mu}m$ in size. Sporangiospores were irregular round or oval, brownish-black and $9{\sim}20{\times}7{\sim}8\;{\mu}m$ in size. On the basis of symptom, mycological characteristics and pathogenicity to banana, the causal fungus was identified as Rhizopus stolonifer. This is the first report of Rhizopus soft rot by R. stolonifer on banana in Korea.

Numerous sporangiospores were formed on the diseased fruits, flower stalk, stem and leaves. Most of the sporangiospores were appeare to be rapidly dispersed in the air. The mycelia grew on the surface of host and formed stolons. Colonies on potato dextrose agar were cottony at first brownish black at maturity. Sporangia were 125.3${\times}$294.2 ${\mu}$m. globose or sub-globose with somewhat flattened base. White at first the black, many spored, and are never overhanging. Sporangiophores were 2.7-6.8${\times}$12.9-33.9 ${\mu}$m, smooth-walled, non-septate, light brown, simple, long, arising in groups of 3-5 from stolons opposite rhizoids. Sporangiophores were 8.6-21.1${\times}$6.41-1.7 ${\mu}$m, irregular, round, oval, elongate, angular and brownish-black streaked. Columella were 63.8${\times}$140.4 ${\mu}$m. brownish gray, umberella-shaped when dehisced. The causal organism was identified as Rhizopus stolonifer Lind on the basis of the morphological characteristics of the fungus. Rhizopus soft rot on squash (Cucurbita moschata) caused by the fungi has not been previously reported in Korea.

Fruit Soft Rot of Sweet Persimmon Caused by Mucor piriformis in Korea

  • A fruit soft rot caused by Mucor piriformis occurred on sweet persimmon storages in Jinju, Changwon and Gimhae, Gyeongnam province, Korea, 2003. The disease infection usually started from wounding after cracking of fruits. At first, the lesions started with water soaked and rapidly softened and diseased lesion gradually expanded. Colonies on potato dextrose agar at $20^{\circ}C$ were whitish to olivaceous-buff Sporangia were globose, black and $96{\sim}153{\mu}m$ in size. Sporangiophores were $26{\sim}42{\mu}m$ in width. Sporangiospores were ellipsoid and $5.8{\sim}10.6{\times}4.3{\sim}7.6{\mu}m$ in size. Columella was obovoid, cylindrical-ellipsoidal, pyriform, subglobose and $80{\sim}125{\mu}m$ in size. Optimal temperature for mycelial growth was $20^{\circ}C$ on PDA. The causal organism was identified as M. piriformis. This is the first report of fruit soft rot on sweet persimmon caused by M. piriformis in Korea.

  • A soft rot of fruits caused by Mucor racemosus occurred on cherry tomato collected in Agricultural Products Wholesale Market in Jinju, Korea. The disease infection usually occurred wounded areas after cracking of fruits. At first, the lesions started with water soaked and rapidly softened and diseased lesion gradually expanded. Colonies were white to brownish to gray in color. Sporangia were $32{\sim}54\;{\mu}m$ in size and globose in shape. Sporangiophores were $8{\sim}14\;{\mu}m$ in width. Sporangiospores were $5{\sim}12\;{\times}\;4{\sim}8\;{\mu}m$ in size, ellipsoidal to subglobose in shape. Columella was $27{\sim}42\;{\mu}m$ in size, obovoid, ellipsoidal, cylindrical-ellipsoidal, slightly pyriform in shape. Chlamydospores were numerous in sporangiophores and barrel-shaped when young, subglobose in old cultures. Optimum growth temperature was about $25^{\circ}C$. The fungus was identified as M. racemosus Fres.. This is the first report of soft rot on cherry tomato caused by M. racemosus in Korea.
