Properties of the Arterial Pressor Response Induced by Stimulation of the Ventral Root Afferent Fibers in the Cat (고양이 척수 전근내 감각신경 자극으로 유발된 승압반응의 생리학적 특성)

  • In an attempt to characterize the ventral root afferent fibers, arterial blood pressure responses to stimulation of the ventral root (VR) were observed in anesthetized cats. Effects of the morphine administered either intravenously or direct spinally and of the spinal lesions on the pressor responses were compared. Followings are the results obtained. 1) Stimulation of the VR with C-strength, high frequency stimuli evoked a marked pressor response. No depressor response, which had been reported during peripheral nerve stimulation, was observed during VR stimulation with low frequency. 2) Acute cervical spinalization abolished the pressor response, indicating the involvement of supraspinal mechanism. 3) The ascending spinal pathways of the pressor response were located in the dorsolateral funiculus bilaterally. 4) Intravenously administered morphine exaggerated the pressor response to VR stimulation, while direct spinally administered morphine suppressed it. From the above results it was concluded that the ventral root afferent fibers have more similar properties to muscular C-afferent fibers than to cutaneous C-fibers.

Effects of Taurine and ${\beta}-Alanine$ on the Responses of Dorsal Horn Cell to Various Stimuli in Cats (Taurine 및 ${\beta}-alanine$이 고양이 척수후각세포의 Activity에 미치는 효과)

  • In 19 cats anesthetized with ${\alpha}-chloralose$ effects of taurine and ${\beta}-alanine$ on the responses of wide dynamic range (WDR) cells to mechanical, chemical and thermal stimuli were investigated in the lumbar spinal cord of the cat. Also studied was an interaction of strychnine with taurine in affecting the activities of WDR cells. Following intravenous administration of taurine, the responses of WDR cells to all types of mechanical stimuli were markedly enhanced, demonstrating that the response to pressure was most sensitive to taurine action. When the receptive field was exposed to thermal stimuli ($50^{\circ}C$) for 20 sec. taurine increased activity of WDR cell to 169.5% of the control value. The $K^{+}$-induced activation of WDR cells was invariably suppressed after taurine administration. Intravenously administered strychnine remarkably reduced the enhanced response of WDR cell to natural stimuli resulting from intravenous administration of taurine. Also ${\beta}-alanine$ markedly activated the response of spinal dorsal horn cell to natural mechanical stimuli. These findings suggest that neutral amino acid and its derivative such as ${\beta}-alanine$ and taurine can enhance the response of WDR cells to different stimuli in cats.

Electrophysiological Study on Medullospinal Tract Cells Related to Somatosympathetic Reflex in the Cat

  • It is well established that neurons in ventrolateral medulla play a key role in determining the vasomotor tone. The purpose of present study is to identify sympathetic related, medullospinal tract neurons in ventrolateral medulla and to show that these mediate somato-sympathetic reflex. Medullospinal tract cells were identified by antidromic stimulation to intermediolateral nucleus (IML) of the second thoracic ($T_2$) spinal cord in anesthetized cats. Peripheral nerves were stimulated for orthodromic activation of these cells and peripheral receptive fields were determined. Post R wave histogram of unit and spike triggered averaging of sympathetic nerve discharge (SND) were used to define sympathetic related cell. A total of 113 neurons was recorded in ventrolateral medulla that had the axonal projections to $T_2$ spinal cord. Thirty four of these medullospinal cells showed spontaneous discharges and the others not. Between these two groups, rostro-caudal coordinate of the distribution from obex [$4.7{\pm}0.2\;$ (mean S.E.) mm, 4.1 0.1 mm], depth from dorsal surface ($5.5{\pm}0.2mm,\;4.9{\pm}0.1mm$ and conduction velocity ($9.9{\pm}1.7m/sec,\;16.7{\pm}1.9\;m/sec$) were significantly different (p<0.05). In spontaneously discharging group, characteristics of rostral and caudal groups were significantly different and we demonstrated that cells in rostral group mediate somatosympathetic reflex. From these results, we conclude that a certain portion of spontaneously discharging medullospinal tract cells in rostral ventrolateral medulla comprise the efferent outputs of somatosympathetic reflex to sympathetic preganglion neurons.

Responses of Dorsal Horn Neurons to Peripheral Chemical Stimulation in the Spinal Cord of Anesthetized Cats

  • Although nociceptive informations are thought to be processed via different neural mechanisms depending on the types of stimuli, sufficient data have not been accumulated yet. We performed a series of experiments to elucidate the possible neural mechanisms as to chemical stimuli such as formalin, capsaicin and ATP. Single unit activity of wide dynamic range (WDR) neurons and high threshold cells were recorded extracellularly from the lumbosacral enlargement of cat spinal cord before and after chemical stimulation to its receptive field (RF). Each chemical substance - formalin $(20{\mu}l,\;4%),$ capsaicin (33 mM) or Mg-ATP (5 mM)- was injected intradermally into the RFs and then the changes in the spontaneous activity, mechanical threshold and responses to the peripheral mechanical stimuli were observed. In many cases, intradermal injection of formalin (5/11) and capsaicin (8/11) resulted in increase of the spontaneous activity with a biphasic pattern, whereas ATP (8/8) only showed initial responses. Time courses of the biphasic pattern, especially the late response, differed between formalin and capsaicin experiments. One hour after injection of each chemical (formalin, capsaicin, or ATP), the responses of the dorsal horn neurons to mechanical stimuli increased at large and the RFs were expended, suggesting development of hypersensitization (formalin 6/10, capsaicin 8/11, and ATP 15/19, respectively). These results are suggested that formalin stimulates peripheral nociceptor, local inflammation and involvement of central sensitization, capsaicin induces central sensitization as well as affects the peripheral C-polymodal nociceptors and neurogenic inflammation, and ATP directly stimulates peripheral nociceptors.

Effect of Electrical Stimulation of the Spinal Cord on Pressor Response in the Cat (고양이 척수의 전기적 자극에 의한 심맥관 반응)

  • 전기적 자극법은 심맥관계기능의 신경성조절을 이해하기위해 널리 이용되는 방법이며, 일반적으로 중추신경에는 고빈도, 말초신경에서는 저빈도 자극에 의해 최대반응이 유발된다고 알려지고 있으며, 이는 흥분파의 민도가 말초로 내려오며 감소되기 때물이라 해석되고 있다. 또한편 신경계의 어떤 단위위에서건 자극민도가 어느 한계를 넘으면 유발된 반응은 유지되지를 못하고 감쇠소실되는 것으로 이는 주로 시납스간 흥분파전달능 부전에 기인될 것이라고 믿어지고있다. 그러나 고위주추계와 말초를 연결하고 있는 척수에 있어서는 아직 최적전기적 자극조건도 분명히 알려져 있지 않을 뿐 아니라 이 부위에서도 고빈도자극시에 반응이 어느 정도 감쇠하는지에 대한 보고가 없기에 이를 추구코자 저자는 고양이의 상부경수를 면수와 연결부에서 완전 절단하기 전후 여러가지 자극조건으로 자극하여 몇 가지 결론을 얻었기에 보고하는 바이다. 1) 연수와 연결부에서 완전절단을 하기 전에는 경수$(C_1{\sim}C_2)$의 백질, 회백질의 여러부위에서 심맥관반응이 유발되었으나 절단후에는 백질중의 만두부위(좌 $2{\sim}3\;mm$, 배면으로 부터 $0.3{\sim}1.0\;mm$$2.5{\sim}3.5\;mm$)에만 반응이 유발되었으며, 이 두 부위는 심맥관계기능을 지배조절하는 원심성 섬유의 통로라고 사료되었다. 2) 최대반응은 자극조건(빈도 자극파지속시간 강도) 100/sec-1 mesc-3V 및 20/sec-3 msec-3V에서. 최장지속반응은 20/sec-3msec에서 유발되었으므로, 후자가 척수부에 있어 심맥관계반응을 유발키 위한 최적자극조건으로 생각된다. 3) 자극면도 20/sec 이하에서 반응유지가 잘된다는 결과는 생리적 조건하에서 척수를 통과하는 흥분파의 빈도가 20/sec를 넘지 않을 것이란 것을 시사한다. 4) 반응지속정도는 최초의 최대반응의 반으로 감쇠되기까지 소요되는시간 즉 50% 반응감쇠시간으로 표시하였으며, 척수에서는 대뇌피질 및 간뇌에서보다 심맥관계반응의 50%반응감쇠시간이 현저히 긴 것을 알 수 있었다.

Molecular Characterization and Expression Analysis of Adrenergic Receptor Beta 2 (ADRB2) Gene before and after Exercise in the Horse

  • The adrenergic receptor beta 2 (ADRB2) plays a role in various physiological responses of the muscle to exercise, such as contraction and relaxation. Given its important role in muscle function, we investigated the structure of the horse ADRB2 gene and its expression pattern after exercise to determine if it can serve as a putative biomarker for recovery. Evolutionary analyses using synonymous and non-synonymous mutation ratios, were compared with other species (human, chimpanzee, mouse, rat, cow, pig, chicken, dog, and cat), and revealed the occurrence of positive selection in the horse ADRB2 gene. In addition, expression analyses by quantitative polymerase chain reaction exhibited ubiquitous distribution of horse ADRB2 in various tissues including lung, skeletal muscle, kidney, thyroid, appendix, colon, spinal cord and heart, with the highest expression observed in the lung. The expression of ADRB2 in skeletal muscle was significantly up-regulated about four folds 30 minutes post-exercise compared to pre-exercise. The expression level of ADRB2 in leukocytes, which could be collected with convenience compared with other tissues in horse, increased until 60 min after exercise but decreased afterward until 120 min, suggesting the ADRB2 expression levels in leukocytes could be a useful biomarker to check the early recovery status of horse after exercise. In conclusion, we identified horse ADRB2 gene and analyzed expression profiles in various tissues. Additionally, analysis of ADBR2 gene expression in leukocytes could be a useful biomarker useful for evaluation of early recovery status after exercise in racing horses.

Pressor Action of Physostigmine in the Rabbit (토끼에 있어서의 Physostigmine의 혈압상승작용)

  • The effect of physostigmine (PS), which has been shown to act on the muscarinic receptors in the brains of the rat, dog and cat, on the arterial blood pressure (BP) was investigated in urethane-anesthetized rabbits. Intravenous (iv) PS, $25{\sim}300\;{\mu}g/kg$, caused little change in BP. However, after treatment of rabbits with either of chlorisondamine (CS), hexamethonium, intracerebroventricular (icv) clonidine, icv xylazine and icy reserpine iv PS produced a pressor response. Spinalization of the rabbit also caused iv PS to increase BP. The pressor effect of iv PS in CS-treated rabbits was markedly reduced after prazosin or pirenzepine. Iv PS inhibited the pressor response to McN-A-343 in CS-treated and in spinal rabbits; alternately during the infusion of McN-A-343 iv PS failed to produce the pressor response. The pressor response to DMPP was not affected by iv PS. Icv PS, $12{\sim}200\;{\mu}g/kg$, produced a pressor response which was accentuated after CS-treatment. This pressor effect was inhibited, though not complete, by prazosin or by pirenzepine. A simultaneous treatment of rabbits with both $[Sar^{1},\;Ala^{8}]-angiotensin$ II, an angiotensin II antagonist, and prazosin or pirenzepine almost completely abolished the pressor effect of icv PS, whereas the angiotensin II antagonist did not enhance the inhibitory effect of pirenzepine and prazosin on the pressor response to iv PS . Icv pirenzepine blocked the pressor response to icv PS without affecting that to iv PS. The present results show that the pressor response to iv PS in CS-treated and in spinal rabbits arises from stimulation of the muscarinic receptors in the sympathetic ganglia, whereas the pressor response by icv PS via activation of the muscarinic receptors in the brain which causes an enhancement in the outflow of sympathetic discharge and angiotensin. The results also suggest that iv PS is unable to produce a pressor response in the rabbit unless the sensitivity of the gangionic muscarinic receptors is altered by ganglionic nicotinic blockade, by the decrease of central sympathetic outflow on the sympathetic ganglia or by spinalization.
