Objectives: Excitability o medial vestibular nucleus (MVN) in the brainstem can be affected by changes in the arterial blood pressure. Several animal studies have demonstrated that acute hypotension results in the alteration of multiunit activities and expression of cFos protein in the MVN. In the field of extracellular electrophysiological recording, tetrode technology and spike sorting algorithms can easily identify single unit activity from multiunit activities in the brain. However, detailed properties of electrophysiological changes in single unit of the MVN during acute hypotension have been unknown. Methods: Therefore, we applied tetrode techniques and electrophysiological characterization methods to know the effect of acute hypotension on single unit activities of the MVN of rats. Results: Two or 3 types of unit could be classified according to the morphology of spikes and firing properties of neurons. Acute hypotension elicited 4 types of changes in spontaneous firing of single unit in the MVN. Most of these neurons showed excitatory responses for about within 1 minute after the induction of acute hypotension and then returned to the baseline activity 10 minutes after the injection of sodium nitroprusside. There was also gradual increase in spontaneous firing in some units. In contrast small proportion of units showed rapid reduction of firing rate just after acute hypotension. Conclusions: Therefore, application of tetrode technology and spike sorting algorithms is another method for the monitoring of electrical activity of vestibular nuclear during acute hypotension.
An enzyme-linked immunosorbent assay(ELISA) was developed for the detection of milk proteins in processed foods. The ${\alpha}_{s1}-casein({\alpha}_{s1}-CN)$, a heat stable major milk protein, was immunized into rabbits to produce specific antibodies. When competitive indirect ELISA(ciELISA) using $anti-{\alpha}_{s1}-CN$ antibodies was established, its detection limit was $0.1\;{\mu}g/mL$. The reactivities of the specific antibodies toward ${\alpha}_{s1}-CN$, skim milk, ${\beta}-CN$ and whey protein isolate(WPI) were 100, 37, 0.14 and 0.04%, respectively, as determined by ciELISA. However $anti-{\alpha}_{s1}-CN$ antibodies did not have any reactivity to other milk proteins such as ${\beta}-lactoglobulin,\;{\alpha}-lactalbumin$, bovine serum albumin, and isolated soy protein. When sandwich ELISA was established, its detection limit was $0.01\;{\mu}g/mL$ which was 10 times more sensitive than that of ciELISA. In the spike test which was performed by adding 1-10% of whole CN to market milk, mean assay recovery as determined by sandwich ELISA was 94.8%(CV, 8.2%). Food stuffs and dairy products were assayed by sandwich ELISA to show 29, 0.13, 0.25, and 6.9% of whole CN in skim milk powder, WPI, semi-solid yoghurt, and processed cheese, respectively.
Background: To date, various genotypes of infectious bronchitis virus (IBV) have co-circulated and in Korea, GI-15 and GI-19 lineages were prevailing. The spike protein, particularly S1 subunit, is responsible for receptor binding, contains hypervariable regions and is also responsible for the emerging of novel variants. Objective: This study aims to investigate the putative major amino acid substitutions for the variants in GI-19. Methods: The S1 sequence data of IBV isolated from 1986 to 2021 in Korea (n = 188) were analyzed. Sequence alignments were carried out using Multiple alignment using Fast Fourier Transform of Geneious prime. The phylogenetic tree was generated using MEGA-11 (ver. 11.0.10) and Bayesian analysis was performed by BEAST v1.10.4. Selective pressure was analyzed via online server Datamonkey. Highlights and visualization of putative critical amino acid were conducted by using PyMol software (version 2.3). Results: Most (93.5%) belonged to the GI-19 lineage in Korea, and the GI-19 lineage was further divided into seven subgroups: KM91-like (Clade A and B), K40/09-like, QX-like (I-IV). Positive selection was identified at nine and six residues in S1 for KM91-like and QX-like IBVs, respectively. In addition, several positive selection sites of S1-NTD were indicated to have mutations at common locations even when new clades were generated. They were all located on the lateral surface of the quaternary structure of the S1 subunits in close proximity to the receptor-binding motif (RBM), putative RBM motif and neutralizing antigenic sites in S1. Conclusions: Our results suggest RBM surrounding sites in the S1 subunit of IBV are highly susceptible to mutation by selective pressure during evolution.

As various wheat variety for bread, all-purpose, and cake flour have been developed, suitable cultivation method for the end-use of the new variety need to be revised. This study was conducted to suggest an optimal nitrogen(N) fertilizer method for wheat Variety 'Saekeumgang' and 'Hojoong' with good noodle quality. In order to analyze the yield and quality changes of 'Saekeumgang' and 'Hojoong' as nitrogen fertilizer amount and timing, these varieties were sown on paddy soil in jeunju, Republic of Korea. The amount of N fertilizer was divided into 4 levels (7.1, 9.1 11.1,13.1kg/10a). In each levels, N amount in sowing date fixed as 3.6kg/10a, N amount in 10 days after heading(DAH) were treated 0 or 2kg/10a, and the other N amount was treated in regrowing stage. As N amount in regrowing stage increased, culm length of 'Saekkeumgang' was increased, but culm length of 'Hojoong' was not affected. Spike number/m2 was increased when N fertilizer amount in regrowing stage increased as 3.5 to 7.5kg/10a. But, spike number/m2 wasn't increased compared N amount 7.1kg/10a conditions when N fertilizer amount 9.1kg/10a. When the N fertilization amount in regrowing stage was increased by 1kg/10a, grain yield increased by 45.7 kg/10a in 'Saegeumgang' and 21.4kg/10a in 'Hojoong', so the fertilizer effect of 'Saegeumgang' was higher, when N fertilizer amount was increased to 2kg/10a at 10DAH, 1000-grain weight increased, but spike number/m2 and grain yield were not affected by N fertilizer at 10 DAH. Protein content and SDS-sedimentation value were increased as increasing N fertilizer amount in regrowing stage and 10 DAH. Among them, N fertilizer amount in 10 DAH had higher impact on protein content and SDS-sedimentation value. Porcine epidemic diarrhea virus (PEDV) is an infectious and highly contagious virus of swine. In order to develop the transgenic tobacco culture cells producing PEDV antigen protein, four vectors expressing PEDV spike protein (SP) gene under the control of a CaMV 35S promoter were constructed. Four fragments of the SP region of PEDV, SP1 (444 bp, 1487-1930 bp), SP2 (1.7 kb, 2300-3987 bp), SP3 (1.4 kb, 1559-2950 bp), and SP4 (2.6 kb, 9-2643 bp) were amplified by PCR and then C-MYC tag was fused to the end of each SP gene, respectively. These cassettes are inserted into the pCAMBIA2300 (named as 35S::SP1-M, 35S::SP2-M 35S::SP3-M, and 35S::SP4-M, respectively). Tobacco (cv. BY-2) cultured cells were transformed by co-cultivation with Agrobacterium tumefaciens harboring expression vector. We selected kanamycin-resistant calli and checked for the presence of the introduced SP gene using PCR, resulting 70% of them showed the foreign gene. We selected the lines with high-level expression of PEDV antigen protein based on dot blot analysis. Southern blot analysis confirmed that the PEDV SP gene was integrated into the genome of the tobacco cultured cells. Northern blot analysis showed that the introduced gene was highly expressed in transgenic cultured cells. Transgenic tobacco cultured cells-derived antigen induced immunogenicity in mice as determined by a plaque reduction neutralization assay. These results suggest that the vectors expressing PEDV spike protein gene in this study will be useful for the development of transgenic plants and cultured cells producing PEDV antigene protein.
This study was conducted to estimate the effect of the drought on the changes of chlorophyll, protein and proline content of upper three leaves, and the grain yield components of barley plant (CV. baegdong) subjected to water stress at four stages: late vegetative, boot, anthesis early grain filling. 1. In comparison with leaf posititions in water stress, the first leaf below flag leaf maintained the highest relative turgidity, chlorophyll and protein content and showed the least proline accumulation. And, in terms of growth stages in water stress, chlorophyll was shown to be highest at anthesis stage, protein being highest at boot stage and proline being least at boot stage. 2. In boot stages, culm and spike length, and Number of grains per spike were remarkably decreased. And the weight of 1000 grains was at least level in the early grain filling stage, and also the grain size was comparatively decreased at boot and two following stages. 3. The protein content of grain by water stress, apart from early grain filling stage, was not significantly affected by water stress at different growth stages. 4. The rate of sterility was particularly increased at boot and anthesis stages. 5. It was eventually concluded that the boot stage among four growth stages, and the flag leaf in 3 leaf positions were mostly damaged by water stress at reproductive growth stage.
