Studies on the RNA nucleotide composition of egg, worm body, pupa and silk-gland(posterior) of Bombyx mori, and spinning gland of spider (가잠(家蠶)의 충체(蟲體), 용체, 잠란(蠶卵) 및 견사선(絹絲腺)(후부(後部))과 지주(蜘蛛) 방적선(紡績腺) RNA의 nucleotide 조성(組成)에 관(關)한 연구(硏究))

  • 가잠(家蠶)(Bombyx mori)의 잠체(蠶體), 용체 및 견사선(絹絲腺)(후부(後部))에서 phenol법(法)으로 RNA를 추출(抽出)하여 RNA의 nucleotide 조성(組成)(mole ratio)을 살피는 한편, 견사선(絹絲腺)(후부(後部))에서 초원심법(超遠沈法)으로 r-RNA, s-RNA를 분리(分離)하여 이에 대(對)한 nucleotide조성(組成)을 조사(調査)하고 또 가잠견사선(家蠶絹絲腺)과 비교(比較)할 목적(目的)으로 거미 방적선(紡績腺)의 t-RNA를 분리(分離)하여 nucleotide성분(成分)을 측정(測定)하여 다음과 같은 결과(結果)를 얻었다. 1) 잠란(蠶卵)에 있어서 이것을 마수(磨粹), 탈지(脫脂) 후(後) lysozyme을 작용(作用)시키고 10% NaCl용액(溶液)으로 가열(加熱) 추출(抽出)하는 새방법(方法)을 고찰(考察)하여 RNA의 추출(抽出)이 극난(極難)한 잠란(蠶卵)에서 RNA를 분리(分離)하는데 성공(成功)하였다. 2) 가잠란(家蠶卵), 잠체(蠶體), 용체 및 견사선(絹絲腺)(후부(後部))의 t-RNA nucleotide 조성(組成)은 다음과 같다. 시료(試料) $\frac{G+C}{A+U}$ $\frac{G+U}{A+C}$ $\frac{Pu}{Py}$ 가잠란(家蠶卵)의 RNA 1.14 1.24 0.99 가잠체(家蠶體)의 RNA 1.40 1.36 0.80 용체의 RNA 1.40 1.33 1.35 후부견사선(後部絹絲腺)의 RNA 1.05 1.32 1.15 이로서 잠체(蠶體). 용체 및 견사선(絹絲腺)의 Pu/Py는 각각(各各) 차이(差異)가 있으나 G+U/A+C는 3자간(者間)에 1.3의 거이 동일(同一)한 수치(數値)를 보여주고 있다. G+C/A+U는 잠체(蠶體)와 용체에 있어서 동일(同一)하나 견사선(絹絲腺)의 그것과는 차이(差異)가 있다. 한편 잠란(蠶卵)에 있어서는 Pu/Py, G+C/A+U, G+U/A+C가 각각(各各) 잠체(蠶體), 용체 및 견사선(絹絲腺)에 있어서와 현저(顯著)한 차이(差異)를 보여주고 있다. G+C/A+U가 1.3이나 되는 RNA의 base ratio를 가진 생물(生物)에 관(關)해서는 아직 보고(報告)된 바 없고 다만 본논문(本論文)의 가잠(家蠶)에 관(關)한 RNA와 속편(續編)인 각종(各種) 패류(貝類) RNA의 nucleotide 조성(組成)에서 모두 1.3에 가까운 수치(數値)를 보여주고 있다. 3) 견사선(絹絲腺)(후부(後部)) t-RNA와 거미 방적선(紡績腺)의 t-RNA의 nucleotide molar ratio 및 견사선(絹絲腺)의 r-RNA, s-RNA nucleotide 조성(組成)은 다음과 같다. 재료(材料) $\frac{G+C}{A+U}$ $\frac{G+U}{A+C}$ $\frac{Pu}{Py}$ 가잠견사선(家蠶絹絲腺)(후부(後部)의 t-RNA 1.05 1.32 1.15의 r-RNA 1.12 1.30 1.20의 s-RNA 1.55 1.33 0.65 지주방적선(蜘蛛紡績腺)의 t-RNA 1.35 1.24 1.16 즉(卽) 가잠견사선(家蠶絹絲腺)(후부(後部))과 거미방적선(紡績腺)의 t-RNA nucleotide 조성(組成)은 Pu/Py가 1.15와 1.16으로서 거이 동일(同一)하지만 G+C/A+U, G+U/A+C에 차이(差異)가 있음을 보았다. 한편 가잠견사선(家蠶絹絲腺)(후부(後部)) r-RNA와 s-RNA의 Pu/Py와 G+C/A+U는 현저(顯著)한 차이(差異)가 있고, G+U/A+C에 있어서는 1.3으로서 거이 동일(同一)한 수치(數値)를 보여주고 있다. 4. 이상(以上)과 같이 잠체(蠶體)에 관(關)한 RNA의 nucleotide 조성(組成)은 소위(所謂) GC-type로서, 현재(現在)까지 문헌(文獻)에 보고(報告)된 각종(各種) 생물(生物)의 RNA의 base ratio에 관(關)하여 비교(比較) 검토(檢討)하였으며, RNA의 nucleotide ratio의 차이(差異)의 의의(意義)에 대(對)하여 고찰(考察)하였다.

Classification of Cordyceps spp. by Morphological Characteristics and Protein Banding Pattern (동충하초(冬蟲夏草)(Cordyceps) 속균의 형태적인 특징과 단백질 Pattern에 의한 계통 분류)

  • Ten species of Cordyceps species were collected throughout Kangwon province including Chuncheon Dongsanmyun KNU forest experiment from June to September, 1993. Collected Cordyceps species were identified as Cordyceps militaris, C. roseostromata, C. kyushuensis, C. scarabaeicola, Phytocordyceps ninchukiospora, C. nutans, Paecilomyces tenuipes, C. sphecocephala, Hymenostilbe odonatae, Torrubiella sp.. C. militaris, type species of Cordyceps species, was mainly formed on pupae of Lepidoptera and found after the rainy season around July. Fruiting body of C. roseostromata was morphologically similar to those of C. militaris, but relatively small in size and they were also found on lawn or pupa of Lepidoptera. Fruiting body of C. scarabaeicola was found on adult Scarabaeidae specifically and collect fruiting bodies of C. kyushuensis were on larva of moth. C. nutans and C. sphecocephala had host specificity on Hemiptera and Hymenoptera, respectively. Each species formed elliptical fertile part attach to the slim and carneous stalk and they were collected the most in specimen number through whole season of the summer. Ascospore of Phytocordyceps ninchukiospora on seed was characterized by two viable, multiseptate, fusiform units linked end-to-end by a long, filiform connective. Paecilomyces tenuipes, imperfect stage of the genus Cordyceps is multi-infective fungi that attack all stages of all groups of insects. Hymenostilbe odonatae attacks only adult Odonata and Torrubiella sp. formed on spider was difficult to collect because it was found the back side of leaf. As results of cultural test PDA medium showed the best mycelial growth. In the experiment of effect of the acidity inside of the media, C. militaris was good on pH 5, C. nutans and Phytocordyceps ninchukiospora were good on pH 6 and Paecilomyces tenuipes was on pH 7 and C. scarabaeicola was on pH 9. All isolates tested showed the best mycelial growth at $20^{\circ}C$. Morphologically similar isolates were used to analyze protein banding pattern among and within species. As a result, C. militaris, C. roseostromata and C. kyushuensis were clustered into close species and C. scarabaeicola and Phytocordyceps ninchukiospora were relatively distant from those species.

Selection and Quality Evaluation of Sprout Soybean [Glycine max (L.) Merrill] Variety for Environment-Friendly Cultivation in Southern Paddy Field (남부지역 친환경 논 재배를 위한 나물콩 품종 선발 및 품질 평가)

  • We carried out the experiment to select the suitable sprout soybean varieties for environment-friendly cultivation in paddy field of southern part area, compares of excess moisture injury degree and yield ability among 29 sprout soybean varieties. Plant growth of sprout soybean was generally low in beginning and recovered after flowering due to rainfall. In paddy field cultivation, number of pod per individual and number of seed per individual were less in difference than upland cultivation, and maturing date was delayed 5-14 days than upland cultivation in most species. When environment-friendly cultivation, pest injury was not caused major problem for the growth during the vegetative period of soybean due to ground spider as natural enemy to insect pest. However, damage of stink bugs showed severe during grain filling period, and Dawonkong, Anpyeongkong, Dachaekong and Wonhwangkong showed susceptible to sting bug. SMV infection was weak and showed some necrosis symptoms in Sokangkong, but black root rot was not infected at all. Bacterial pustule began to be infected slowly from pod enlargement stage in most species, displayed severe symptoms in Dawonkong, Pungsannamulkong, Seonamkong and Sobaeknamulkong. The symptoms of pod anthracnose, pod blight and purple spot were greatly appeared after flowering. Disease resistance varieties was Paldokong, Kwangankong, Doremikong, Somyeongkong, Pungsannamulkong, Iksa-namulkong, Seonamkong, Sojinkong, Pureunkong, Bosugkong, Namhaekong and Sorokkong. Lodging index showed 3 in Saebyeolkong, and other species displayed slight lodging in 0-3 degree. 100-seed weight is 9.8-17.2g extent and increased 0.1-3.7g than upland cultivation in most species, but decreased in some species. Government purchase standard, species correspond to small-seed-size namulkong (Sizing screen diameter 4.0-5.6 mm) was Dawonkong, Dachaekong, Bosugkong, Seonamkong, Sokangkong, Hannamkong, Somyeongkong and Wonhwangkong. Species which seed yield was higher than Pungsannamulkong (266kg/10a) were Sorokkong, Hannamkong, Bosugkong and Sowonkong. Considering sprout soybean species, disease endurance, insect resistance, lodging resistance, 100-seed weight, yield ability and excess moisture tolerances synthetically, Seonamkong, Hannamkong, Doremikong, Bosugkong, Pungwonkong, Kwangankong, Sowonkong, Dagikong, Paldokong, Eunhakong and Pungsannamulkong were promising for environment-friendly cultivation in paddy field.

A Short Study on Promoting the Dispersal of Phytoseiulus persimilis using a Bridge on Green Pepper (피망에서 브리지를 이용한 칠레이리응애의 확산 증진 연구)

  • The vertical distribution and dispersion of the two-spotted spider mite, Tetranychus urticae and its predator, Phytoseiulus persimilis, were investigated within and between green pepper plants. We also tested the effect of an artificial bridge with threads on dispersal of P. persimilis between the plants. The extent of spread of T. urticae depended on its infested density and location. When 10 adults of T. urticae were inoculated on upper leaf, they stayed on the upper leaf until 5 days after inoculation. However, when 100 adults were inoculated on an upper leaf, they dispersed to the lower leaf within a day. In condition that 10 adults of T. urticae were inoculated on the lower leaf, they started to move toward upward within a day. In the absence of T. urticae, P. persimilis tended to move to the leaves where T. urticae was present when both T. urticae and P. persimilis were inoculated together within a plant. An artificial bridge with threads between two plants was helpful to disperse P. persimilis to neighboring plants. Therefore, it would be expected that the dispersal of P. persimilis can be promoted by supplying the artificial bridges among plants and by narrowing the distances between plants.

Initial Evaluation using Geochemical Data to infer Tectonic Setting of Mt. Baekdu/Changbaishan Volcano (백두산 화산의 지체구조 추론을 위한 지구화학적 데이터를 이용한 기초 평가)

  • This study aimed to investigate the tectonic setting of the volcanic edifice at Mt. Baekdu by analyzing petrochemical characteristics of Holocene felsic volcanic rocks distributed in the Baekdusan stratovolcano edifice and summit of the Cheonji caldera rim, as well as Pleistocene mafic rocks of the Gaema lava plateau and Changbaishan shield volcano edifice. During the early eruption phases, mafic eruption materials, with composition ranging from alkali basalt to trachybasalt, or from subalkaline (tholeiitic) basalt to basaltic andesite formed the Gaema lava plateau and Changbaishan shield volcanic edifice, whereas the Baekdusan stratovolcano edifice and Holocene tephra deposits near the summit of the Cheonji caldera comprises trachytic and rhyolitic compositions. Analysis results revealed bimodal compositions with a lack of 54-62 SiO2, between the felsic and mafic volcanic rocks. This suggested that magmatic processes occurred at the locations of extensional tectonic settings in the crust. Mafic volcanic rocks were plotted in the field of within-plate volcanic zones or between within-plate alkaline and tholeiite zones on the tectonic discrimination diagram, and it was in good agreement with the results of the TAS diagram. Felsic volcanic rocks were plotted in the field of within-plate granite tectonic settings on discrimination diagrams of granitic rocks. None of the results were plotted in the field of arc islands or continental margin arcs. The primitive mantle-normalized spider diagram did not show negative (-) anomalies of Nb and Ti, which are distinctive characteristics of subduction-related volcanic rocks, but exhibited similar patterns of ocean island basalt. Trace element compositions showed no evidence of, magmatic processes related to subduction zones, indicating that the magmatic processes forming the Baekdusan volcanic field occurred in an intraplate environment. The distribution of shallow earthquakes in this region supports the results. The volcanic rocks of the Baekdusan volcanic field are interpreted as the result of intraplate volcanism originating from the upwelling of mantle material during the Cenozoic era.
