난자 세포질내 정자 주입술 후 동결보존 배아이식: 고식적 체외수정시술과의 비교 연구 (Clinical Outcome of Transfer of Cryopreserved-Thawed Embryos Obtained after Intracytoplasmic Sperm Injection: Comparison with Conventional In Vitro Fertilization)

  • The objective of this study was to compare retrospectively the survival and pregnancy rates(PR) of cryopresered-thawed embryos obtained from intracytoplasmic sperm injection (ICSI) or conventional in vitro fertilization (IVF). Ninety-six cycles of cryopresered-thawed embryo transfer (ET) were performed in 79 patients from June, 1996 to September, 1997 and grouped as followings: 20 cycles (16 patients) inseminated by ICSI (ICSI Group) and 76 cycles (63 patients) by conventional IVF (IVF Group). Slow-freezing and rapid-thawing protocol was used with 1.5M propanediol (PROH) and 0.1M sucrose as cryoprotectant. All embryos were frozen-thawed at the two pronuclear (2 PN) stage excluding four cycles in which the early cleavage stage embryos were frozen, and allowed to cleave in vitro for one day before ET. The duration from freezing to thawing was comparable in both groups ($mean{\pm}SD$, $112.1{\pm}80.0$ vs. $124.8{\pm}140.1$ days). The age of female ($31.2{\pm}3.4$ vs. $32.6{\pm}3.3$ years) and the endometrial thickness prior to progesterone injection ($9.4{\pm}2.0$ vs. $9.3{\pm}1.8$ mm) were also comparable in both groups. There was no significant difference in the outcomes of cryopreserved-thawed ET between two groups: survival rate ($85.2{\pm}16.1%$ vs. $82.2{\pm}19.7%$), cleavage rate ($96.9{\pm}6.7%$ vs. $94.7{\pm}13.0%$), cumulative embryo score (CES, $54.5{\pm}31.1$ vs. $49.0{\pm}20.0$), preclinical loss rate (5.0% vs. 5.3%), clinical miscarriage rate (0% vs 29.4%), clinical PR per transfer (35.0% vs. 22.4%), implantation rate (9.9% vs. 5.6%), and multifetal PR (42.9% vs. 17.6%). In conclusion, human embryos resulting from ICSI can be cryopreserved-thawed and transferred successfully, and the survival rate and PR are comparable to conventional IVF.

  • Parthenogenesis and culture condition are essential to intracytoplasmic sperm injection and cloning by nuclear transfer. This study investigated apoptosis and in vitro development of parthenogenetic preimplantation porcine embryos. 42∼44 h in vitro matured oocytes derived from a local abattoir were used. Apoptotic cell death was analyzed by using a terminal deoxynucleatidyl transferase mediated deoxyuridine 5-triphoshate nick-end labling (TUNEL) assay. (omitted)

The Production of Transgenic Livestock and Its Applications

  • During the last 20 years, transgenic animal technology has provided revolutionary new opportunities in many aspects of agriculture and biotechnology. Several gene delivery systems including pronuclear injection, retroviral vectors, sperm vectors, and somatic cell cloning have developed for making transgenic animals. In the future major improvements in transgenic animal generation will be mainly covered by somatic cell cloning technology. Many factors affecting integration frequency and expression of the transgenes should be overcome to facilitate the industrial applications of transgenic technology. Transgenic animal technology has settled down in some areas of the biotechnology, especially the mass production of valuable human proteins and xenotransplantation. In the 21st century animal biotechnology will further contribute to welfare of human being.

  • 닭에서 2배수성 배우자(gametes)의 생성을 통해서 4배수성 태아 생산을 위한 본 실험의 결과는 다음과 같다. 1. 2배수성 정자생산 TEM(Tri -ethyleneme-lamine) 0.37mg을 연속 3일 주사한 후 9일째(최초 TEM처리 후 12일째)에 11%로 가장 높았다. 2. 2배수성 배우자를 유도한 후 수정을 통하여 109개의 수정난 중 4배수성 태아를 3개(2.8%) 관찰할 수 있었다. 3. 4배수성 개체의 유전구조는 정상구조에서 변화가 없이 동일한 염색체가 4장으로 구성된 형태였다.

  • Objective: To report the pregnancy which was made by in vitro fertilization using recombinant follicle stimulating hormone and gonadotropin releasing hormone antagonist. Material and Method: Case report. Results: Six oocytes were retrieved and all were fertilized by intracytoplasmic sperm injection. Six embryos were transferred and the pregnancy was confirmed. Conclusion: It is envisaged that the availability of recombinant gonadotropins and gonadotropin releasing hormone antagonists will ultimately lead to shorter, cheaper and safer treatments, using reduced dosages.

Somatic Cell Nuclear Transfer in Rodents, the Little Big Animals

  • Transgenic rats and mice are useful experimental animal models for medical research including human disease model studies. Somatic cell nuclear transfer (SCNT) technology is successfully applied in most mammalian species including cattle, sheep, pig and mouse. SCNT is also considered to increase the efficacy of transgenic/knockout mouse and rat production. However, in the area of reproductive biotechnology, the rodent model is inadequate because of technical obstacles in manipulating the oocytes including intracytoplasmic sperm injection and SCNT. In particular, success of rat SCNT is very limited so far. In this review, the history of rodent cloning is described.

Development of Vitrified Bovine Oocytes following Intracytoplasmic Sperm Injection (ICSI)

  • Oocyte freezing has become a prevalent source for related reproductive technologies. This study was carried out to evaluate viability of post-thawed bovine oocyte injected DTT-treated sperm following by two different activation stimuli (Group 1, 5 M ionomycin, 5 min + CR1aa, 3 h . 1.9 mM dimetylaminopurine (DMP), 3 h; group 2, ionomycin + 10 $\mu\textrm{g}$/$m\ell$ cycloheximide(CHX), 5h). The techniques of ultra-rapid freezing used in this study were essentially similar to those of described by Vajta et al (Theriogenology 1999; 52:939-948), Denuded oocytes at 22 h of culture were exposed to cryoprotectant (3.2 M Ethylene glycol, 2.36 M DMSO, 0.6 M sucrose), and followed by freezing in electron microscopic grid. After thawing the oocytes were transferred back into the drop of maturation medium and cultured for additional 2 h before being subjected to ICSI. All eggs were then cultured in CRlaa medium, and transferred into M199+10% FCS on day 4. The culture was maintained until day 9. In Experiment 1, frozen-ICSI eggs were compared on development into blastocyst to those of unfrozen and IVF control. Those eggs were activated with the method of group 2. A higher proportion of unfrozen-ICSI and IVF eggs developed into cleavage and blastocysts than of frozen-ICSI eggs (65% and 13%; 71% and 23% vs. 39% and 8%; P<0.05). In Experiment 2, development and ploidy of embryos made from group 1 were compared to those from group 2. Between groups there did not differ on the rates of development, however, chromosomal abnormality in group 1 was significantly higher than in group 2 (49% vs. 30%; P<0.05). The present result suggests that frozen bovine oocytes can be used for ICSI.
