Effects of Chromium Supplementation and Lipopolysaccharide Injection on the Immune Responses of Weanling Pigs

  • Sixteen specific pathogen free 4-wk-old crossbred weanling pigs were allotted into a $2{\times}2$ factorial experiment to evaluate the effects of chromium (Cr) on the immune responses after lipopolysaccharide (LPS) injection. Two factors included (1) no Cr or 400 ppb Cr supplementation from chromium picolinate (CrPic) and (2) LPS injection ($200{\mu}g/kg$ BW, intraperitoneally) on day 21 (d 21) and 35 (d 35) as compared with saline application. Plasma samples were obtained from all piglets before (0 h) and at 2 h, 4 h, 8 h, and 24 h after LPS injection. The changes in tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and leukocyte populations after LPS injection were not significant on d 21. On d 35, the plasma $TNF-{\alpha}$ level was increased at h 2 postinjection, and supplemental Cr reduced the $TNF-{\alpha}$ level. The leukocyte populations had changed profoundly and lymphocyte subsets of $CD2^+$ and $CD8^+$ were reduced at 8 h postinjection. The blood granulocytes were increased and the percentage of $CD2^+$ was reduced in the Cr-fed group on d 35. Furthermore, Cr supplementation decreased the blastogensis of concanavalin A-stimulated peripheral blood mononuclear cells (PBMC) on d 21. These results suggest that 400 ppb Cr supplementation from CrPic in diets may modulate the immune responses in weanling pigs during LPS injection.

The Influence of Long-term Aloe Supplement on Anti-oxidative Defenses and Cholesterol Content in Brain and Kidney of Aged Rats

  • The present study was investigated the anti-oxidative effects of aloe vera ingestion on brain and kidney in aged rats by monitoring several oxidative-related parameters. Male specific pathogen-free Fischer 344 rats were randomly divided into four groups of five rat each: Group A was fed test chow without aloe supplementation; Group B was fed a diet containing a 1% freeze-dried aloe filet; Group C was fed a diet containing a 1% charcoal-processed, freeze-dried aloe filet; and Group D was fed a diet containing a charcoal-processed, freeze-dried, whole leaf aloe in drinking water. Analyses of tissues were done at 4 months and 16 months of age. Results showed that a long-term intake of aloe, regardless of the preparation used, enhanced antioxidant defenses against lipid peroxidation, as indicated by reduced phosphatidylcholine hydroperoxide levels in both brain and kidney. The additional benefit of aloe intake on the anti-oxidative action was evidenced by enhanced superoxide dismutase and catalase activity in all aloe-ingested groups. Another beneficial effect of aloe shown in this study, although not an anti-oxidative parameter, was its cholesterol-lowering effect as detected in brain and kidney with significant decreases at age16 months of aloe-fed rats. Based on these findings, we conclude that a long-term dietary aloe supplementation modulated the anti-oxidative defense systems and cholesterol level.

Exposure to Cerium Oxide Nanoparticles Is Associated With Activation of Mitogen-activated Protein Kinases Signaling and Apoptosis in Rat Lungs

  • Objectives: With recent advances in nanoparticle manufacturing and applications, potential exposure to nanoparticles in various settings is becoming increasing likely. No investigation has yet been performed to assess whether respiratory tract exposure to cerium oxide ($CeO_2$) nanoparticles is associated with alterations in protein signaling, inflammation, and apoptosis in rat lungs. Methods: Specific-pathogen-free male Sprague-Dawley rats were instilled with either vehicle (saline) or $CeO_2$ nanoparticles at a dosage of 7.0 mg/kg and euthanized 1, 3, 14, 28, 56, or 90 days after exposure. Lung tissues were collected and evaluated for the expression of proteins associated with inflammation and cellular apoptosis. Results: No change in lung weight was detected over the course of the study; however, cerium accumulation in the lungs, gross histological changes, an increased Bax to Bcl-2 ratio, elevated cleaved caspase-3 protein levels, increased phosphorylation of p38 MAPK, and diminished phosphorylation of ERK-1/2-MAPK were detected after $CeO_2$ instillation (p<0.05). Conclusions: Taken together, these data suggest that high-dose respiratory exposure to $CeO_2$ nanoparticles is associated with lung inflammation, the activation of signaling protein kinases, and cellular apoptosis, which may be indicative of a long-term localized inflammatory response.

Two factors included (1) no Cr or 400 ppb Cr supplementation from chromium picolinate and (2) lipopolysaccharide (LPS) injection on day 21 (d 21) and 35 (d 35) compared to saline application. Plasma samples and rectal temperature were obtained from all piglets before (h 0) and at 2 h (h 2), 4 h (h 4), 8 h (h 8), and 24 h (h 24) after LPS injection ($200{\mu}g/kg$ BW, intraperitoneally). The rectal temperature on d 21 was significantly decreased (p<0.05) of about $0.36^{\circ}C$ with Cr supplementation before LPS injection. After LPS injection, the daily gain of piglets was decreased during d 35-38. Supplementation of Cr had no effect in general on growth performance particularly after LPS injection. The plasma glucose, triglycerides and urea nitrogen concentrations were changed in different ways after LPS injection. Plasma cortisol level was significantly elevated at h 2 after LPS injection on d 21 and d 35. The supplementation of Cr in the diet can delayed plasma cortisol release on d 35. The results suggest that 400 ppb Cr supplementation from CrPic may modulate the physiological response during immune stress in weanling pigs.

Outbreak of Fowl Cholera in native chickens (토종닭에서 가금콜레라(Fowl Cholera) 발생)

  • Fowl cholera(FC), which has been classified as reportable animal disease in Korea, occurred in the backyard farm located in Chungju, Chungbuk province. The farm was not ordinary domestic poultry farm but backyard farm with main cultivation of fruits. The farmer reported unusual mortality of chickens which were grazed in his own fruit farm without any poultry house. There was a lots of opportunities to contact with wild animals including wild birds freely. Clinical signs observed in the chickens were torticolis, mucous excretion from mouth and greenish diarrhea. On the necropsy we found multifocal necrosis on the surface of liver, hemorrhages in the epicardium and ovary and mucopurulent exudates in the joint. The causative agents was isolated from the liver of chickens collected from the farms and identified as Pasteurella multocida using biochemical tests of bacteria. Also, the isolated bacteria were inoculated into specific pathogen free chickens to evaluate the pathogenicity. High mortality and similar pathological lesions compared to those of chickens died in the farm were observed. We report the first case of fowl cholera of the chickens in Korea and further studies are needed to evaluate the serotype, genotype and pathogenicity of isolated Pasteurella multocida.

Repeated Dose 4-Week Oral-Treatment for DRF Toxicity Test of HMC05 in Sprague-Dawley Rats (HMC05의 Sprague-Dawley 흰쥐를 이용한 4주 반복 경구투여 DRF 독성시험)

  • Objectives: HMCO5 is an extract obtained from 8 different herbal mixtures. We undertook a safety evaluation of HMCO5 for a dose range finding (DRF) toxicity test in specific pathogen free (SPF) Sprague-Dawley (SD) male and female rats. Methods: The male and female rats were divided into 4 groups, respectively; G(0), treated with distilled water: G(1), treated with 222 mg/kg HMC05: G(2), treated with 667 mg/kg HMC05, and G(3), treated with 2,000 mg/kg HMC05; HMC05 was administered orally for 4 weeks. The safety evaluation examined clinical signs, mortality, body weight, food consumption, water consumption, ophthalmic findings, urinalysis, hematological values, absolute & relative organ weights, and necropsy findings during the tests. Results: There were no changes in clinical signs, mortality, body weight, food consumption, water consumption, and ophthalmic findings examined during the test periods. In serum biochemical values, triglyceride was increased in male group G(3) and Na$^+$ decreased significantly in male groups G(2), G(3) and G(4). In male group G(4), spleen weight decreased relatively and increases of absolute & relative left ovary weights were found. In addition, an adhesion of liver to diaphragm was found in male group G(2). However, we could not find any dose-interrelationships in these changes. Conclusions: These results indicate that HMC05 extract did not show any toxicity in the DRF toxicity study. Therefore, it suggests that establishment of 1,000, 333 and 111 mg/kg dosages are moderate in a repeated dose 26-week oral toxicity study of HMC05.

Study on natural killer cell activity and its characteristics during hepatocarcinogenesis in rats (랫드의 간암 발생과정에서 분리한 자연살해세포의 활성측정 및 특성연구)

  • The purposes of this study were to set up the method of the natural killer(NK) cell activity assay using the flow cytometer and to examine the characteristics and distribution of the NK cell during rat hepatocarcinogenesis. Forty five male 6 week-old specific pathogen free(SPF) Sprague-Dawley rats were randomly divided into three groups. Group I was the non-treated control and given normal diet and water. Group II was treated with diethylnitrosamine(DEN, 200mg/kg, i.p.) and partial hepatectomy. Group III was treated with DEN, partial hepatectomy and 0.05% phenobarbital sodium in water from 3 to 16 weeks. All animals were examined the morphology of the large granular lymphocyte(LGL), the LGL percent of the total lymphocytes and the LGL conjugation rate with YAC-1 cell in peripheral blood, spleen and liver. Moreover, activity of the LGL isolated from peripheral blood lymphocytes was determined using the flow cytometer. As results, LGL were observed in the peripheral blood, spleen and liver. LGL were observed the relatively faintly staining basophilic cytoplasm with granules, and eccentric, often kidney-shaped nuclei in Giemsa stain. Its size was $11{\sim}13{\mu}m$. LGL percentage of the isolated lymphocytes in peripheral blood, spleen and liver were 1.8~2.3%, 1.3~1.4% and 0.87~0.99%, respectively. LGL conjugation rate with YAC-1 cell was shown to be peripheral blood(9.3~10.3 %) > spleen(7.7~8.7%) > liver(5.6~7.0%). The activity of the LGL isolated from peripheral blood lymphocytes in Group I, II and III was 33.7%, 30.5% and 35.4%, respectively. However, all values were not significantly between groups.

A pathological study of spontaneous tumors in SPF Ktc : ICR mice (SPF Ktc : ICR 마우스의 자연발생 종양에 대한 병리학적 연구)

  • The study was carried out to obtain the basic data for types, incidence and histopathological features of the spontaneous tumors of the specific pathogen free (SPF) Ktc : ICR mice bred in a barrier system in Technology Research Center, Korea Research Institute of Chemical Technology. One hundred of the mice consisted of 50 males and 50 females were examined for 18 months. 1. The overall incidence rate of spontaneous tumors was 51(51%) of 100 heads tested. The male mice showed slightly higher incidence(28%) than the female(23%), and the incidence rate and the number of affected organs were increased with the increasing age of mice. 2. The incidence rate of primary tumor was 59(59%) of 100 heads tested, consisted of 30 cases (50.8%) of benign tumors and 29 cases(49.2%) of malignant tumors. Among the malignant tumors twenty cases were metastasized to various organs. 3. In tumor incidence rates by systems and organs, the male mice showed the high incidence rate in the liver (18%), hematopoietic system (16% ) and lung (14%), while the female mice, in the hematopoietic system(18%), lung(12%), liver(8%) and uterus(8%). 4. The tumors showing the particularly low incidence rates (<1.0%) were rhabdomyosarcoma in the skeletal muscle, malignant schwannoma in the peripheral nerve, cortical adenoma in the adrenal gland, transitional cell carcinoma in the urinary bladder, tubular cell adenoma in the kidney and adenoma in the pituitary gland and harderian gland.

The comparison of pathogenicity in the SPF chickens challenged with avian reovirus and/or fowl adenovirus (SPF 닭에서 레오바이러스와 아데노바이러스의 단일 혹은 혼합감염에 따른 병원성 비교)

  • Avian reovirus (ARV) and fowl adenovirus (FAdV) were evaluated for pathogenicity in specific pathogen free (SPF) chickens. ARV was isolated from the broilers with history of malabsorption syndrome (MAS). FAdV was isolated from the layer breeders with inclusion body hepatitis and hydropericardium syndrome. Total 6 inoculated groups including 1 un-inoculated group were organized and inoculated with the ARV and/or FAdV by oral route. The minimal pathological lesions and lower viral gene detection rates were present in the ARV inoculated groups compared to those of FAdV or ARV/FAdV inoculated groups. Common gross lesions in the ARV inoculated group were distended intestine with foamy contents and in the FAdV group there were foamy cecal contents and hydropericardium among the evaluation methods such as gross and histological lesion, viral gene detection, body weight and serum chemistry, histopathological lesion score was reliable especially in the liver lesions such as hepatic necrosis and lymphocytic infiltration. However, we did not success to evaluate the synergetic effect of mixed infection of ARV and FAdV in this study. Therefore, we need further study to reproduce malabsorption syndrome of ARV infection using different viral agent such as rotavirus and using different dose of virus.

Pathological evaluation of renal changes induced by multiple nephropathogenic factors in SPF chickens I. Histopathological and electron microscopical observation (신부전 요인에 의해 유발된 닭 신장변화의 병리학적 관찰 I. 병리조직학적 및 전자현미경적 관찰)

  • Renal failure is one of the main causes of economic impacts in the poultry industry and complex syndrome with different severity of clinical signs caused by multiple nephropathogenic factors such as infectious bronchitis viral infection and excess salt and calcium in diet. To evaluate the correlation between severity of renal failure and the causative nephropathogenic factors, one-day-old specific pathogen free chicks were treated with either single causative factor or multiple causative factors described as above. Each group was designed as control for non-treated control, IB for infectious bronchitis virus (IB virus) infection, IBHNa for IB virus infection with high diet salt, IBHCa for IB virus infection with high diet calcium, IBHNC for IB virus infection with high diet salt and calcium, HNa for high diet salt, HCa for high diet calcium and HNC for high diet salt and calcium. Chickens were inoculated with IB virus at 1-day-old and remained on their respective diets until 21 day of age. The high dietary salt feeding groups such as IBHNa, IBHNC, HNa, HNC increased water intake, watery diarrhea, general subcutaneous edema and the high dietary calcium feeding groups such as IBHCa and IBHNC showed severe visceral gout. Two more than treated groups caused high mortality in comparison with the single treated groups. IB virus exposure significantly increased urate deposition and lymphocytic interstitial nephritis. Especially urate deposition dramatically increased when excess diet calcium was combined together. In excess diet salt treated groups enlarged edematous kidneys were observed and hypertrophy of glomeruli were showed. These results suggest that IB virus enhanced the incidence and severity on chicken renal failure clearly related to the quantity of salt and calcium.
