Study on the Characteristics of Dissolved Organic Matters from Diverse Sources by XAD Resin Fractiontion and Microbial Incubation Experiments (XAD 수지분획과 생분해 실험에 의한 기원별 용존유기물질 특성 연구)

  • In this study, characteristics of dissolved organic matter (DOM) from Lake Paldang and seven other DOM sources (lake plankton, plants, soil, composite, treated sewage) were studied using XAD resin fractionation and 28-day microbial incubation experiment. Distribution patterns of DOM-fractions, which include hydrophilic acids (HiA), hydrophilic bases (HiB), hydrophilic neutrals (HiN), hydrophobic acids (HoA), hydrophobic neutrals (HoN) and the extent of DOM biodegradation (i.e., biodegradability) were different depending on the origins of the DOM samples. The DOM distribution pattern and the biodegradability were found to be effective for distinguishing the different DOM sources. The biodegradability (%) had negative correlations with the content (%) of hydrophobic fractions (Ho) and specific UV absorbance of DOM, which indicate that the Ho fractions contain more aromatic carbon structures and relatively stable during biodegradation, irrespective of the sources. To gain additional insight into the microbial transformation of the DOM, we also investigated the changes in the fraction's distribution for plankton, leaf litter and composite samples after the incubation. The results showed that biodegradation of hydrophilic fraction (Hi) causes an increase in the proportion of Ho (HoA, HoN), while biodegradation of HoA increases the HoN production.

  • Characteristics of humic substances on the changes in dissolved organic matter (DOM) characteristics by biodegradation was investigated using three types of the artificial water samples composed of glucose and Suwannee River fulvic acid (SRFA). Some selected DOM characteristics including the specific UV absorbance (SUVA), the synchronous fluorescence spectra and the molecular weight (MW) were compared for the artificial water samples before and after 28-day microbial incubation. The changes of the DOM characteristics were minimal for SRFA during the incubation whereas they were significant for glucose. SUVA, dissolved organic carbon (DOC)-normalized fluorescence intensity, and MW values of glucose increased, suggesting that such labile organic compounds could be exclusively transformed into more humidified materials by biodegradation. For glucose-SRFA mixture, the selected DOM characteristics were greater than those estimated using the assumption that the individual changes of either glucose or SRFA are conservative for the mixture of the two materials. Our results suggest that the presence of humic substances (HS) may lead to the enhancement of the formation of refractory organic materials during biodegradation of labile compounds. Detailed analyses of size exclusion chromatography (SEC) revealed that the enhancement occurred for the DOM mixture with a MW range between 500 Da to 4000 Da.

Monitoring of Changes in Molecular Weight Distribution and Fluorescence Properties of Dissolved Matter (DOM) in Water Treatment Processes (정수처리공정 중 자연유기물질의 분자량 분포 및 형광특성 변화 모니터링)

  • Monitoring of NOM characteristics is important for improving removal efficiency of natural organic matter (NOM) in water treatment processes. In this study, several NOM characteristics, which include specific UV absorbance (SUVA), total carbonate content, molecular weight distribution, and fluorescence properties, were measured using samples collected from a pilot-scale water treatment plant consisting of coagulation/flocculation (C/F), filtration, ozonation and granular activated carbon (GAC) processes. The highest removal of NOM was observed in C/F and filtration processes as demonstrated by the reduction of dissolved organic carbon (DOC) by 25% and 21%, respectively. Despite nearly no change in DOC, however, the lowest SUVA value and the highest total carbohydrate content were observed in the sample from ozonation process. This indicates that non-degradable aromatic compounds become depleted and biodegradable organic compounds are enriched during the process. Comparison of synchronous fluorescence spectra of the samples showed that ozoation process increased protein-like fluorescence while it decreased fulvic-like and terrestrial humic-like fluorescence. Consistently, a slight peak of protein-like fluorescence was observed in the sample from ozonation process. The greatest change in molecular weight distributions of the samples was observed in C/F process. Comparison of size exclusion chromatogram of the samples revealed that NOM fractions with the molecular weight greater than 2000 Da were reduced by over 90% after C/F process. SUVA values and total carbohydrate content of the samples were well correlated with a ratio of protein-like fluorescence and terrestrial humic-like fluorescence intensities with the correlation coefficients of 0.99 and 0.91, respectively. This suggests that synchronous fluorescence properties of NOM could be used as useful tolls for monitoring changes of some NOM characteristics during water treatment processes.

  • Procainamide is the drug of second choice (after lidocaine) in most coronary care units for the treatment of sustained ventricular arrhythmias associated with acute myocardial infarction. The purpose of this study was to develop the efficient assay method of procainamide in human plasma and to assess the pharmacokinetic profile of procainamide in healthy Korean volunteers. The pharmacokinetics of procainamide administered orally was evaluated after a dose of 250 mg. Procainamide in plasma was assayed using a specific HPLC method with UV absorbance at 275 nm. AUC was $4.58{\pm}0.90 {\mu}g/ml-hr$, $C_{max}\;1.34{\pm}0.39{\mu}g/ml$, $T_{max}\;1.06{\pm}0.34 hr$ and half-life $3.07\pm0.34 hr$. $T_{max}$ was slightly shorter than that in Caucasian (1-2 hr), whereas the half-life was similar to that in Caucasian (2.5-4.1 hr).

  • A kinetic study was made to determine the mechanism of the aquation of $cis-[Co(en)_2(NH_3)Cl]^{2+}\;in\;Hg^{2+}$ aqueous solution. The progress of reaction was followed UV/vis-spectrophotometrically by a measurement of the absorbance at a specific wave length (530nm) of $cis-[Co(en)_2(NH_3)Cl]^{2+}$ as a function of time. The experimental results have shown that the reaction rate is dependent upon the concentration of $Hg^{2+}$ that act as a catalyst. And it was found that the overall reaction proceed with second order, first order with respect to Co(III) complex and $Hg^{2+}$. Activation parameters, ${\Delta}H^{\neq}\;and\;{\Delta}S^{\neq}$, were obtained as 12.9 kcal/mol and -19.3 e.u., respectively. We have proposed a plausible reaction mechanism which is consistent with the observed rate equation.

  • Warfarin is a widely used oral anticoagulant agent used to treat thromboembolic disease. The purpose of this study was to develop the efficient assay method of warfarin sodium i n human plasma and to assess the pharmacokinetic profile of the warfarin in healthy Korean volunteers. The pharmacokinetics of warfarin administered orally was evaluated after a dose of 10 mg. Warfarin in plasma was assayed using a specific HPLC method with UV absorbance at 304 nm. AUC was 46.33${\pm}9.95{\mu}g/ml.hr$, $C_{max}$ $1.22{\pm}0.22{\mu}g/ml, $T_{max}$$2.50{\pm}1.41$ hr and half-life $43.49{\pm}4.33$ hr. $T_{max}$ was slightly shorter than that in Caucasian (3~9 hr), whereas the half-life was longer than that in Caucasian (10~45 hr, mean: 36 hr). These results suggest that warfarin may have a longer duration in Korean than in Caucasian.

Fluorescence Properties of Size Fractions of Dissolved Organic Matter Originated From Different Sources (생성 기원에 따른 용존 자연유기물질 분자량별 형광특성 비교)

  • Fluorescence properties and carbohydrate content were investigated using ultrafiltrated size fractions of dissolved organic matters (DOM) originated from different sources. The materials included a treated sewage, an algal organic matter, and a soil leachate, all of which are major constituents of dissolved organic matter in a typical urban river. Four different size fractions were separated from the three sources of each DOM. The size distribution demonstrated that a higher molecular weight fraction was more present in soil leachate compared to two other source DOMs. A higher content of carbohydrates was observed in the following order - algal DOM > treated sewage > soil leachate. A wide range of specific UV absorbance was observed from size fractions of a single source DOM, indicating that aromatic carbon structures are heterogeneously distributed within one source of DOM. The structural heterogeneity was the most pronounced for the soil leachate. The fluorescence index ($F_{450}/F_{500}$) of the treated sewage was similar to that (2.0) typically obtained from autochthonous DOM, suggesting that the treated sewage exhibited autochthonous organic matter-like properties. No protein-like fluorescence intensities were observed for all of the soil leachate size fractions whereas they were observed with two other source DOMs. Based upon the fluorescence peak ratios from fluorescence excitation-emission matrix (EEM), two discrimination indices could be suggested to distinguish three different source DOMs. It is expected that the suggested discrimination indices will be useful to predict the sources of DOM in a typical urban river affected by treated sewage.

Novel Antibacterial, Cytotoxic and Catalytic Activities of Silver Nanoparticles Synthesized from Acidophilic Actinobacterial SL19 with Evidence for Protein as Coating Biomolecule

  • Silver nanoparticles (AgNPs) have potential applications in medicine, photocatalysis, agriculture, and cosmetic fields due to their unique physicochemical properties and strong antimicrobial activity. Here, AgNPs were synthesized using actinobacterial SL19 strain, isolated from acidic forest soil in Poland, and confirmed by UV-vis and FTIR spectroscopy, TEM, and zeta potential analysis. The AgNPs were polydispersed, stable, spherical, and small, with an average size of 23 nm. The FTIR study revealed the presence of bonds characteristic of proteins that cover nanoparticles. These proteins were then studied by using liquid chromatography with tandem mass spectrometry (LC-MS/MS) and identified with the highest similarity to hypothetical protein and porin with molecular masses equal to 41 and 38 kDa, respectively. Our AgNPs exhibited remarkable antibacterial activity against Escherichia coli and Pseudomonas aeruginosa. The combined, synergistic action of these synthesized AgNPs with commercial antibiotics (ampicillin, kanamycin, streptomycin, and tetracycline) enabled dose reductions in both components and increased their antimicrobial efficacy, especially in the case of streptomycin and tetracycline. Furthermore, the in vitro activity of the AgNPs on human cancer cell lines (MCF-7, A375, A549, and HepG2) showed cancer-specific sensitivity, while the genotoxic activity was evaluated by Ames assay, which revealed a lack of mutagenicity on the part of nanoparticles in Salmonella Typhimurium TA98 strain. We also studied the impact of the AgNPs on the catalytic and photocatalytic degradation of methyl orange (MO). The decomposition of MO was observed by a decrease in intensity of absorbance within time. The results of our study proved the easy, fast, and efficient synthesis of AgNPs using acidophilic actinomycete SL19 strain and demonstrated the remarkable potential of these AgNPs as anticancer and antibacterial agents. However, the properties and activity of such particles can vary by biosynthesized batch.

Changes in Spectroscopic and Molecular Weight Characteristics of Dissolved Organic Matter in an Agriculture Reservoir during a Summer Monsoon (장마시기에 따른 농업용 저수지 내 용존 유기물 분광특성과 분자량 변화)

  • In this study, we investigated the variations of dissolved organic matter (DOM) in an agricultural reservoir during the monsoon period (June to October, 2020) with respect to the organic carbon concentration (DOC), molecular weight distribution, and optical properties. The monsoon period was divided into three phases - beginning storm (BS), during storm (DS), and after storm (AS). Our results showed significant differences in the concentrations and characteristics of DOM during the summer monsoon. The DOC concentrations were decreased after the monsoon, probably due to a dilution effect. In contrast, increasing trends were observed in the specific UV absorbance (SUVA), and relative abundances of humic-like fluorescence and larger-sized compounds. These observations implied that the large-sized and humic-like organic components with terrestrial origins strongly affected the reservoir DOM after the summer monsoon. Meanwhile, biopolymer size fraction, which is associated with algal activity, became more abundant after the monsoon. These results suggest that DOM with autochthonous sources became dominant as a result of the inflow of nutrients into the reservoir after the storm. Spatial changes in DOM within the reservoir were not pronounced as much as the temporal variations. All taken, it can be concluded that the summer monsoon simply led to the decrease of DOM concentrations while the sources and the quality of DOM underwent substantial changes, which may enrich refractory organic matter in the reservoir. This study reveals the importance of in-depth DOM quality monitoring before and after summer monsoon for effective water quality management in agricultural reservoirs.

  • An experimental investigation is made to determine the mechanism of the aquation of $[Co(en)_2Cl_2]^+$ in $Hg^{2+}$ aqueous solution. The progress of reaction is followed UV/vis-spectrophotometrically by a measurment of the absorbance at a specific wavelength of Co(III) complex as a function of time. The aquation of cis-$[Co(en)_2Cl_2]^+$ and trans-$[Co(en)_2Cl_2]^+$ has been found to be first order and second order with respect to the concentration of $Hg^{2+}$ catalyst, respectively. It has been found that the reaction rate for aquation of the trans-form is faster than that of cis-form, and that the product of either cis-form or trans-form is always in the mixture ratio of 97 % to 3 %. Plausible reaction mechanism is proposed for the reaction system on the basis of kinetic data and activation parameters. Theoretical rate equation derived from the proposed mechanism is consistent with the observed one.
